2Nature inspired hierarchical structures in nano-cellular epoxy/graphene-Fe3O4 nanocomposites with ultra-efficient EMI and robust mechanical strength显示文摘Hierarchical layered structures,whether in a compact form like nacre or a porous manner like bone,are well known for their combined features of high stiffness,strength,and lightweight,inspiring many man-made materials and structures for high performance applications.The use of nacre/bone like hierarchical structures in polymer nanocomposites can achieve excellent mechanical and functional properties with high filler volume fractions after carefully aligning functional nanofillers,although the fabrication and processing remain a great challenge.In this work,a bio-inspired lightweight nano-cellular epoxy/graphene-Fe_(3)O_(4) nanocomposite with high nanofiller loading of 75 wt.%was successfully fabricated by combining features from both nacre and bone structures,via a simple compression molding process together with an eco-friendly supercritical CO_(2) foaming process to achieve robust mechanical strength and excellent electromagnetic interference(EMI)shielding effectiveness(SE)simultaneously.Highly aligned graphene-Fe_(3)O_(4) nanoplatelets with well controlled nanoscale porous structures(52.6 nm)enabled both low density(1.26 g/cm^(3))and high specific EMI SE>5200 dB/cm^(2)/g,as well as preserved tensile strength of 67 MPa.This study provides a sustainable route to fabricate nature mimicked structures with high performance and high flexibility for a wide range of applications,from portable electronics to healthcare devices.Xun Fan Fengchao Wang Qiang Gao Yu Zhang Fei Huang Ronglin Xiao Jianbin Qin Han Zhang Xuetao Shi Guangcheng Zhang 2022Journal of Materials Science & Technology2022,,8:2

4Differentiation of embryonic stem cells in adult bone marrow显示文摘Embryonic stem cells (ESCs) are a potential source of generating transplantable hematopoietic stem and progenitor cells,which in turn can serve as 'seed' cells for hematopoietic regeneration.In this study,we aimed to gauge the ability of mouse ESCs directly differentiating into hematopoietic cells in adult bone marrow (BM).To this end,we first derived a new mouse ESC line that constitutively expressed the green fluorescent protein (GFP) and then injected the ESCs into syngeneic BM via intra-tibia.The progeny of the transplanted ESCs were then analyzed at different time points after transplantation.Notably,however,most injected ESCs differentiated into non-hematopoietic cells in the BM whereas only a minority of the cells acquired hematopoietic cell surface markers.This study provides a strategy for evaluating the differentiation potential of ESCs in the BM micro-environment,thereby having important implications for the physiological maintenance and potential therapeutic applications of ESCs.Yueying Li Jing He Fengchao Wang Zhenyu Ju Sheng Liu Yu Zhang Zhaohui Kou Yanfeng Liu Tao Cheng Shaorong Gao 2010Journal of Genetics and Genomics2010,37,7:1

7Capturing the hierarchically assorted modules of protein–protein interactions in the organized nucleome显示文摘Nuclear proteins are major constituents and key regulators of nucleome topological organization and manipulators of nuclear events.To decipher the global connectivity of nuclear proteins and the hierarchically organized modules of their interactions,we conducted two rounds of cross-linking mass spectrometry(XL-MS)analysis,one of which followed a quantitative double chemical cross-linking mass spectrometry(in vivo qXL-MS)workflow,and identified 24,140 unique crosslinks in total from the nuclei of soybean seedlings.This in vivo quantitative interactomics enabled the identification of 5340 crosslinks that can be converted into 1297 nuclear protein–protein interactions(PPIs),1220(94%)of which were non-confirmative(or novel)nuclear PPIs compared with those in repositories.There were 250 and 26 novel interactors of histones and the nucleolar box C/D small nucleolar ribonucleoprotein complex,respectively.Modulomic analysis of orthologous Arabidopsis PPIs produced 27 and 24 master nuclear PPI modules(NPIMs)that contain the condensate-forming protein(s)and the intrinsically disordered region–containing proteins,respectively.These NPIMs successfully captured previously reported nuclear protein complexes and nuclear bodies in the nucleus.Surprisingly,these NPIMs were hierarchically assorted into four higher-order communities in a nucleomic graph,including genome and nucleolus communities.This combinatorial pipeline of 4C quantitative interactomics and PPI network modularization revealed 17 ethylene-specific module variants that participate in a broad range of nuclear events.The pipeline was able to capture both nuclear protein complexes and nuclear bodies,construct the topological architectures of PPI modules and module variants in the nucleome,and probably map the protein compositions of biomolecular condensates.Shuaijian Dai Shichang Liu Chen Zhou Fengchao Yu Guang Zhu Wenhao Zhang Haiteng Deng Al Burlingame Weichuan Yu Tingliang Wang Ning Li 2023Molecular Plant2023,16,5:0
8The hydrophobic cluster on the surface of protein is the key structural basis for the SDS-resistance of chondroitinase VhChlABC显示文摘The application of chondroitinase requires consideration of the complex microenvironment of the target.Our previous research reported a marine-derived sodium dodecyl sulfate(SDS)-resistant chondroitinase VhChlABC.This study further investigated the mechanism of VhChlABC resistance to SDS.Focusing on the hydrophobic cluster on its strong hydrophilic surface,it was found that the reduction of hydrophobicity of surface residues Ala181,Met182,Met183,Ala184,Val185,and Ile305 significantly reduced the SDS resistance and stability.Molecular dynamics(MD)simulation and molecular docking analysis showed that I305G had more conformational flexibility around residue 305 than wild type(WT),which was more conducive to SDS insertion and binding.The affinity of A181G,M182A,M183A,V185A and I305G to SDS was significantly higher than that of WT.In conclusion,the surface hydrophobic microenvironment composed of six residues was the structural basis for SDS resistance.This feature could prevent the binding of SDS and the destruction of hydrophobic packaging by increasing the rigid conformation of protein and reducing the binding force of SDS-protein.The study provides a new idea for the rational design of SDS-resistant proteins and may further promote chondroitinase research in the targeted therapy of lung diseases under the pressure of pulmonary surfactant.Juanjuan Su Hao Wu Chengying Yin Fengchao Zhang Feng Han Wengong Yu 2024Marine Life Science & Technology2024,6,1:0
9Aged hematopoietic stem cells entrap regulatory T cells to create a prosurvival microenvironment显示文摘Although DNA mutation drives stem cell aging,how mutation-accumulated stem cells obtain clonal advantage during aging remains poorly understood.Here,using a mouse model of irradiation-induced premature aging and middle-aged mice,we show that DNA mutation accumulation in hematopoietic stem cells(HSCs)during aging upregulates their surface expression of major histocompatibility complex class II(MHCII).MHCII upregulation increases the chance for recognition by bone marrow(BM)-resident regulatory T cells(Tregs),resulting in their clonal expansion and accumulation in the HSC niche.On the basis of the establishment of connexin 43(Cx43)-mediated gap junctions,BM Tregs transfer cyclic adenosine monophosphate(cAMP)to aged HSCs to diminish apoptotic priming and promote their survival via activation of protein kinase A(PKA)signaling.Importantly,targeting the HSC–Treg interaction or depleting Tregs effectively prevents the premature/physiological aging of HSCs.These findings show that aged HSCs use an active self-protective mechanism by entrapping local Tregs to construct a prosurvival niche and obtain a clonal advantage.Weinian Liao Chaonan Liu Ke Yang Jun Chen Yiding Wu Shuzhen Zhang Kuan Yu Lisha Wang Li Ran Mo Chen Fang Chen Yang Xu Song Wang Fengchao Wang Qian Zhang Jinghong Zhao Lilin Ye Changhong Du Junping Wang 2023Cellular & Molecular Immunology2023,20,10:0

11Long-term exposure to fine particulate matter modifies the association between physical activity and hypertension incidence显示文摘Background:The trade-off between the benefits of regular physical activity(PA)and the potentially detrimental effects of augmented exposure to air pollution in highly polluted regions remains unclear.This study aimed to examine whether ambient fine particulate matter(PM_(2.5))exposure modified the impacts of PA volume and intensity on hypertension risk.Methods:We included 54,797 participants without hypertension at baseline in a nationwide cohort of the Prediction for Atherosclerotic Cardiovascular Disease Risk in China(China-PAR)project.PA volume and intensitywere assessed by questionnaire,and high-resolution(1 km×1 km)PM_(2.5)estimates were generated using a satellite-based model.Results:During 413,516 person-years of follow-up,12,100 incident hypertension cases were identified.PM_(2.5)significantly modified the relationship between PA and hypertension incidence(p_(interaction)<0.001).Increased PA volume was negatively associated with incident hypertension in the low PM_(2.5)stratum(<59.8μg/m^(3),ptrend<0.001),with a hazard ratio of 0.81(95%confidence interval(95%CI):0.74-0.88)when comparing the fourth with the first quartile of PA volume.However,the health benefits were not observed in the high PM_(2.5)stratum(≥59.8μg/m^(3),p_(trend)=0.370).Moreover,compared with light PA intensity,vigorous intensity was related to a 20%(95%CI:9%-29%)decreased risk of hypertension for participants exposed to low PM_(2.5),but a 17%(95%CI:4%-33%)increased risk for those with high PM_(2.5)levels.Conclusion:PA was associated with a reduced risk of hypertension only among participants with low PM_(2.5)exposure.Our findings recommended regular PA to prevent hypertension in less polluted regions and reinforced the importance of air quality improvement.Qiong Liu Keyong Huang Fengchao Liang Xueli Yang Jianxin Li Jichun Chen Xiaoqing Liu Jie Cao Chong Shen Ling Yu Yingxin Zhao Ying Deng Ying Li Dongsheng Hu Xiangfeng Lu Yang Liu Dongfeng Gu Fangchao Liu Jianfeng Huang 2022Journal of Sport and Health Science2022,11,6:0
