1Dynamic Investigation on Chromosome Aberration of a Human Retinoblastoma Cell Line SO-Rb_(50)显示文摘G-banding and karyotype analyses of cells in seventeen passages of SO-Rb_(50) during a long period of culture for about four years were performed. Three chromosome markers 13q14^-, 1p36^+ and 12p13^+ were found. Cells possessed 13q14^- reduced to zero after the 200th passage while 1p^+ and 12p^+ cells increased to 100% after 30 and 200 passages respectively. 2Experimental Study of Effect of Perfluorohexyloctane to Retina显示文摘Objective: To investigate the effect of perfluorohexvloctane to the retina of rabbit eyes.Methods: Perfluoroh-exyloctane (experimental group) or BSS(control group) intovitreous cavities of fifteen vitrectomized New Zealand white rabbits. A slit-lampbiomicroscope and an indirect ophthalmoscope were used to examine all the eyes pre- andpostoperation. Histopathological examination was performed after the rabbits weresacrificed. Results:Perfluorohexyloctane was injected into the vitreous cavity forming a single largeclear globe. No retinal detachment and cataract were found. The edema of outer plexiformlayer was significant, and then get thin, inner nuclear layer and ganglion cell bodies hada darker cytop: lasm and nucleoplasm. Conclusions: Perfluorohexyloctane in vitreous cavity had significant side effects on retinaAs a silicone solvent, it should be removed out completely after injection. 4Alloy-Electrode-Assisted High-Performance Enhancement-Type Neodymium-Doped Indium-Zinc-Oxide Thin-Film Transistors on Polyimide Flexible Substrate显示文摘Flexible thin-film transistors with high current-driven capability are of great significance for the next-generation new display technology.The effect of a Cu-Cr-Zr(CCZ)copper alloy source/drain(S/D)electrode on flexible amorphous neodymiumdoped indium-zinc-oxide thin-film transistors(NdIZO-TFTs)was investigated.Compared with pure copper(Cu)and aluminum(Al)S/D electrodes,the CCZ S/D electrode changes the TFT working mode from depletion mode to enhancement mode,which is ascribed to the alloy-assisted interface layer besides work function matching.X-ray photoelectron spectroscopy(XPS)depth profile analysis was conducted to examine the chemical states of the contact interface,and the result suggested that chromium(Cr)oxide and zirconium(Zr)oxide aggregate at the interface between the S/D electrode and the active layer,acting as a potential barrier against residual free electron carriers.The optimal NdIZO-TFT exhibited a desired performance with a saturation mobility(μsat)of 40.3 cm^(2)·V-1·s^(-1),an Ion/Ioff ratio of 1:24×10^(8),a subthreshold swing(SS)value of 0.12 V·decade^(-1),and a threshold voltage(Vth)of 0.83 V.This work is anticipated to provide a novel approach to the realization of highperformance flexible NdIZO-TFTs working in enhancement mode.Kuankuan Lu Rihui Yao Wei Xu Honglong Ning Xu Zhang Guanguang Zhang Yilin Li Jinyao Zhong Yuexin Yang Junbiao Peng 2021Research2021,,1:1

6Pathological,Ultrastructural and Immunohistochemical Observations of Adenoma of Retinal Pigment Epithelium显示文摘Purpose: To Study the clinical, pathological, ultrastructural and immunohistchemicalcharacters of adenoma of the retinal pigment epithelium in order to offer evidence todiagnose this tumor.Methods: Routine paraffin slices HE stain, histochemistry PAS and VG stain,transmission electron microscopy, and immunohistochemistry for S-100 and vimentinwith LSAB method were used.Results: The tumor cells were oval and cuboidal in shape. Part of the tumor had atubular arrangement. Around the sheets of tumors cells there was a large amount ofuniform red stick-like substances. The above matter represented positive in PAS stain.Most of the above matter was yellow, while less of the matter showed red in VG stain.Transmission electron microscopy showed that there were tight junctions between tumorcells. 11Cloning and Sequence Analysis of Light Variable Region Gene of Anti-human Retinoblastoma Monoclonal Antibody显示文摘Purpose: To clone the variable region gene of light chain of monoclonal antibody against human retinoblastoma and to analyze the characterization of its nucleotide sequence as well as amino acid sequence.Methods: Total RNA was extracted from 3C6 hybridoma cells secreting specific monoclonal antibody(McAb)against human retinoblastoma(RB), then transcripted reversely into cDNA with olig-dT primers.The variable region of the light chain (VL) gene fragments was amplified using polymeerase chain reaction(PCR) and further cloned into pGEM(R) -T Easy vector. Then, 3C6 VL cDNA was sequenced by Sanger's method.Homologous analysis was done by NCBI BLAST.Results: The complete nucleotide sequence of 3C6 VL cDNA consisted of 321 bp encoding 107 amino acid residues, containing four workframe regions(FRs)and three complementarity-determining regions (CDRs) as well as the typical structure of two cys residues. The sequence is most homological to a member of the Vk9 gene family, and its chain utilizes the Jkl gene segment.Conclusion: The light chain variable region gene of the McAb against human RB was amplified successfully , which belongs to the Vk9 gene family and utilizes Vk-Jk1 gene rearrangement. 12Detection of Tumor Suppressor Gene and Oncogene in SO-Rb_(50) Human Retinoblastoma Cell Line显示文摘Retinoblastoma (Rb) is the most common malignant'cancer of eye. So-Rb_(50) is the first Rb cell line established in China in 1988. It has passed to the 387th passage now. We collected cells of the 327th passage of SO-Rb_(50), purified its genomic DNA and detected it with Rb and c-myc cDNA probes respectively (normal human white blood cells DNA was the control). 13Experiment Study of Effect of Perfluorohexyloctane on Corneal Endothelial Cells显示文摘Purpose: To investigate the effect of Perfluorohexyloctane (F6H8) on corneal endothelial cells(CEC) of rabbit eyes. Methods: Fifteen New Zealand white rabbits were derided into two groups: experimental group(F6H8) and control group(BSS) . All rabbits underwent anterior chamber injection of 0. 15ml F6H8 or BSS. Slit-lamp biomicroscopy and corneal endothelium photography were performed pre-operatively and postoperatively. Histopa-thological examination and Transmission electron microscopy(TEM) were done afterthe rabbits were sacrificed. Results : All the corneas were clear. Since 4 weeks after operation , the endothelial cells were markedly irregular in size and shape and the number of endothelial cells was markedly decreased. Multilayered retrocorneal membranes(RCM)grew gradually 2 weeks after surgely. Vacuolar degeneration was seen in some endothelial cells. Nuclear degeneration and edema of plasma were seen in TEM. Conclusion: Corneal endothelial cell degenerated after contacting with F6H8 for 2 -4 weeks. As a silicone solvent, it should be removed completely after injection. We don't recommend it to be used as a new intraocular temponade. 14Pluripotent Embryonic Stem Cells Developed into Medulloepithelioma in Nude Mice Eyes显示文摘Purpose: The pluripotent embryonic stem cells can differentiate into various kinds offormal tissues. There is no previous report on the differentiation of embryonic stem cellin the intraocular environment. In this paper, the authors tried to investigate theintraocular growth character of mice embryonic stem cells in nude mice.Methods: Murine embryonic stem cells were cultured and maintained in anundifferentiated state in vitro. They were transplanted into the right eyes of 20 nude miceby microinjection under operating microscope. Animal eye observation, light microscopeand immunohistochemical examinations were implemented.Results: Two to three days after transplantation, small pieces of gray-white materialcould be viewed in the vitreous cavity. Between the 15th and 20th day, the gray-whitemass grew into the anterior chamber in 4 nude mice eyes. Then, the mass at the anteriorchamber extended extraocularly. On the 30th day, a remarkable proptosis was observedin two of the four nude mice. In 6 to 45 days, the mice were executed for morphologicalexamination which showed the following typical structures: (1) Undifferentiated cellswith prominent nucleolius. (2) Flexner-Wintersteiner-like rosettes. (3) Medulloepithe-lioma-like structure: the cells were arranged in sheets, cords, tubes, and cysts. (4) Large,spindle-or astrocyte-like cells. (5) Cartilage-like structure. 15Observation of Surface Structure of Retinoblastoma Cell Line显示文摘Objective: We study the adhesion moleclues on the surface of SO-Rb50 retinoblastoma cell line.Motheds : The distribution of proteoglycans in the retinoblastoma SO-Rb50 cell line was analyzed by histochem-electron microscopy, using Colloidal Iron in combination with a series of enzyme digestions. In addition, immunohistochemistry was performed using a panel of specific antibodies including neuron specific enolase(NSE) ,glial fibrillary acidic protein(GFAP) , S-100 protein, fibronectin, laminin, and collagen IV.Results: Immunohistochemical stains showed the most marked cytoplasmic reactivity of SO-Rb50 cells with anti-NSE and anti-S100. The cells member and surface was postive with anti-NSE. No reactivity was noted with antibodies against laminin, GFAP, and collagen IV.
