| 1 | Somatic Embryogenesis Receptor Kinases Control Root Development Mainly via Brassinosteroid-Independent Actions in Arabidopsis thaliana显示文摘Brassinosteroids(BRs),a group of plant steroidal hormones,play critical roles in many aspects of plant growth and development.Previous studies showed that BRI1-mediated BR signaling regulates cell division and differentiation during Arabidopsis root development via interplaying with auxin and other phytohormones.Arabidopsis somatic embryogenesis receptor-like kinases(SERKs),as co-receptors of BRI1,were found to play a fundamental role in an early activation step of BR signaling pathway.Here we report a novel function of SERKs in regulating Arabidopsis root development.Genetic analyses indicated that SERKs control root growth mainly via a BR-independent pathway.Although BR signaling pathway is completely disrupted in the serk1 bak1 bkk1 triple mutant,the root growth of the triple mutant is much severely damaged than the BR deficiency or signaling null mutants.More detailed analyses indicated that the triple mutant exhibited drastically reduced expression of a number of genes critical to polar auxin transport,cell cycle,endodermis development and root meristem differentiation,which were not observed in null BR biosynthesis mutant cpd and null BR signaling mutant bri1-701. | Junbo Du Hongju Yin Shasha Zhang Zhuoyun Wei Baolin Zhao Jinghua Zhang Xiaoping Gou Honghui Lin Jia Li | 2012 | Journal of Integrative Plant Biology2012,54,6: | 22 |
| 2 | T cell–associated immunoregulation and antiviral effect of oxymatrine in hydrodynamic injection HBV mouse model显示文摘Although oxymatrine(OMT) has been shown to directly inhibit the replication of hepatitis B virus(HBV) in vitro, limited research has been done with this drug in vivo. In the present study, the antiviral effect of OMT was investigated in an immunocompetent mouse model of chronic HBV infection.The infection was achieved by tail vein injection of a large volume of DNA solution. OMT(2.2, 6.7 and20 mg/kg) was administered by daily intraperitoneal injection for 6 weeks. The efficacy of OMT was evaluated by the levels of HBV DNA, hepatitis B surface antigen(HBs Ag), hepatitis B e antigen(HBe Ag)and hepatitis B core antigen(HBc Ag). The immunoregulatory activity of OMT was evaluated by serum ELISA and flow cytometry. Results shows that OMT at 20 mg/kg inhibited HBV replication, and it was more efficient than entecavir(ETV) in the elimination of serum HBs Ag and intrahepatic HBc Ag. Inaddition, OMT accelerated the production of interferon-γ(IFN-γ) in a dose-dependent manner in CD4^+T cells. Our findings demonstrate the beneficial effects of OMT on the enhancement of immunological function and in the control of HBV antigens. | 5 | Cable-truss supporting system for gob-side entry driving in deep mine and its application显示文摘In order to solve the large deformation controlling problem for surrounding rock of gob-side entry driving under common cable anchor support in deep mine, site survey, physical modeling experiment, numerical simulation and field measurement were synthetically used to analyze the deformation and failure characteristics of surrounding rock. Besides, applicability analysis, prestress field distribution characteristics of surrounding rock and the control effect on large deformation of surrounding rock were also further studied for the gob-side entry driving in deep mine using the cable-truss supporting system. The results show that, first, compared with no support and traditional bolt anchor support, roof cable-truss system can effectively restrain the initiation and propagation of tensile cracks in the roof surrounding rock and arc shear cracks in the two sides, moreover, the broken development of surrounding rock, roof separation and extrusion deformation between the two sides of the roadway are all controlled; second, a prestressed belt of trapezoidal shape is generated in the surrounding rock by the cable-truss supporting system, and the prestress field range is wide. | 6 | Biosensors based on acetylcholinesterase immobilized on mesoporous silica thin films显示文摘Using tetraethoxysilane and 3-aminopropyltriethoxysilane as the silica sources, amino-functionalized mesoporous silica thin films with 3-dimensional cubic structure have been deposited on conducting ITO substrate in the presence of surfactant F127 templates under acid conditions. The acetylcholinesterase (AchE) and cytochrome c (Cyt c) were incorporated into the pores of mesoporous thin films, and an amperometric biosensor was obtained. After adsorption of AchE and Cyt c, the ordered cubic structure of mesoporous silica and the bioactivities and electrochemical activities of the immobilized protein and enzyme molecules were retained. The sensor properties of the biosensor were investigated by using acetylthiocholine iodide as the substrate and Cyt c as the electron transfer mediator. | 11 | Improved observation of colonized roots reveals the regulation of arbuscule development and senescence by drought stress in the arbuscular mycorrhizae of citrus显示文摘Citrus is the typical mycorrhizal fruit tree species establishing symbiosis with arbuscular mycorrhizal (AM) fungi. However, arbuscule development and senescence in colonized citrus roots, especially in response to drought stress, remain unclear, which is mainly due to the difficulty in clearing and staining lignified roots with the conventional method. Here, we improved the observation of colonized roots of citrus plants with the sectioning method, which enabled the clear observation of AM fungal structures. Furthermore, we investigated the effects of one week of drought stress on arbuscule development and senescence with the sectioning method. Microscopy observations indicated that drought stress significantly decreased mycorrhizal colonization (F%and M%) although it did not affect plant growth performance. Fluorescence probes (WGA 488 and/or Nile red) revealed that drought stress inhibited arbuscule development by increasing the percentage of arbuscules at the early stage and decreasing the percentages of arbuscules at the midterm and mature stages. Meanwhile, drought stress accelerated arbuscule senescence, which was characterized by the increased accumulation of neutral lipids. Overall, the sectioning method developed in this study enables the in-depth investigation of arbuscule status, and drought stress can inhibit arbuscule development but accelerate arbuscule senescence in the colonized roots of citrus plants. | 12 | Rapid fatty acids detection of vegetable oils by Raman spectroscopy based on competitive adaptive reweighted sampling coupled with support vector regression显示文摘Objectives:The composition and content of fatty acids are critical indicators of vegetable oil quality.To overcome the drawbacks of traditional detection methods,Raman spectroscopy was investigated for the fast determination of the fatty acids composition of oil.Materials and Methods:Rapeseed and soybean oil at different depths of the oil tank at different storage times were collected and an eighth-degree polynomial function was used to fit the Raman spectrum.Then,the multivariate scattering correction,standard normal variable transformation(SNV),and Savitzky–Golay convolution smoothing methods were compared.Results:Polynomial fitting combined with SNV was found to be the optimal pretreatment method.Characteristic wavelengths were selected by competitive adaptive reweighted sampling.For monounsaturated fatty acids(MUFAs),polyunsaturated fatty acids(PUFAs),and saturated fatty acids(SFAs),44,75,and 92 characteristic wavelengths of rapeseed oil,and 60,114,and 60 characteristic wavelengths of soybean oil were extracted.Support vector regression was used to establish the prediction model.The R^(2)values of the prediction results of MUFAs,PUFAs,and SFAs for rapeseed oil were 0.9670,0.9568,and 0.9553,and the root mean square error(RMSE)values were 0.0273,0.0326,and 0.0340,respectively.The R^(2)values of the prediction results of fatty acids for soybean oil were respectively 0.9414,0.9562,and 0.9422,and RMSE values were 0.0460,0.0378,and 0.0548,respectively.A good correlation coefficient and small RMSE value were obtained,indicating the results to be highly accurate and reliable.Conclusions:Raman spectroscopy,based on competitive adaptive reweighted sampling coupled with support vector regression,can rapidly and accurately analyze the fatty acid composition of vegetable oil. | Linjiang Pang Hui Chen Liqing Yin Jiyu Cheng Jiande Jin Honghui Zhao Zhihao Liu Longlong Dong Huichun Yu Xinghua Lu | 2022 | Food Quality and Safety2022,6,4: | 0 |

| 16 | Degradation prediction of proton exchange membrane fuel cell stack using semi-empirical and data-driven methods显示文摘Degradation prediction of proton exchange membrane fuel cell(PEMFC)stack is of great significance for improving the rest useful life.In this study,a PEMFC system including a stack of 300 cells and subsystems has been tested under semi-steady operations for about 931 h.Then,two different models are respectively established based on semi-empirical method and data-driven method to investigate the degradation of stack performance.It is found that the root mean square error(RMSE)of the semi-empirical model in predicting the stack voltage is around 1.0 V,while the predicted voltage has no local dynamic characteristics,which can only reflect the overall degradation trend of stack performance.The RMSE of short-term voltage degradation predicted by the DDM can be less than 1.0 V,and the predicted voltage has accurate local variation characteristics.However,for the long-term prediction,the error will accumulate with the iterations and the deviation of the predicted voltage begins to fluctuate gradually,and the RMSE for the long-term predictions can increase to 1.63 V.Based on the above characteristics of the two models,a hybrid prediction model is further developed.The prediction results of the semi-empirical model are used to modify the input of the data-driven model,which can effectively improve the oscillation of prediction results of the data-driven model during the long-term degradation.It is found that the hybrid model has good error distribution(RSEM=0.8144 V,R2=0.8258)and local performance dynamic characteristics which can be used to predict the process of long-term stack performance degradation. | Yupeng Wang Kangcheng Wu Honghui Zhao Jincheng Li Xia Sheng Yan Yin Qing Du Bingfeng Zu Linghai Han Kui Jiao | 2023 | Energy and AI2023,11,1: | 0 |
| 17 | Enhanced genome editing to ameliorate a genetic metabolic liver disease through co-delivery of adeno-associated virus receptor显示文摘Genome editing through adeno-associated viral(AAV) vectors is a promising gene therapy strategy for various diseases,especially genetic disorders. However, homologous recombination(HR) efficiency is extremely low in adult animal models. We assumed that increasing AAV transduction efficiency could increase genome editing activity, especially HR efficiency, for in vivo gene therapy. Firstly, a mouse phenylketonuria(PKU) model carrying a pathogenic R408W mutation in phenylalanine hydroxylase(Pah) was generated. Through co-delivery of the general AAV receptor(AAVR), we found that AAVR could dramatically increase AAV transduction efficiency in vitro and in vivo. Furthermore, co-delivery of SaCas9/sgRNA/donor templates with AAVR via AAV8 vectors increased indel rate over 2-fold and HR rate over 15-fold for the correction of the single mutation in Pah;mice. Moreover, AAVR co-injection successfully increased the site-specific insertion rate of a 1.4 kb Pah cDNA by 11-fold, bringing the HR rate up to 7.3% without detectable global off-target effects. Insertion of Pah cDNA significantly decreased the Phe level and ameliorated PKU symptoms.
