1Blockade of cholecystokinin-2 receptor and cyclooxygenase-2 synergistically induces cell apoptosis, and inhibits the proliferation of human gastric cancer cells in vitro显示文摘Gastrin and cyclooxygenase-2(COX-2) playimportant roles in the carcinogenesis and progression ofgastric cancer.However,it remains unknown whether the combination of cholecystokinin-2(CCK-2) receptor antagonist plus COX-2 inhibitor exerts synergistic anti-tumor effects on human gastric cancer.Here,we demonstrated that the combination of AG-041R(a CCK-2 receptor antagonist) plus NS-398(a selective COX-2 inhibitor) treatment had synergistic effects on proliferation inhibition,apoptosis induction,down-regulation of Bcl-2 and up-regulation of Bax expression in MKN-45 cells.These results indicate that simultaneous targeting of CCK-2 receptor and COX-2 may inhibit gastric cancer development more effectively than targeting either molecule alone.(C)2008 Elsevier Ireland Ltd.All rights reserved.Sun, W. 4Polymorphisms of MTHFD,plasma homocysteine levels, and risk of gastric cancer in ahigh-risk Chinese population显示文摘Purpose:Accumulative evidence suggests that folate has a protective effect on gastric cancer. The methylenetetrahyd-rofolate dlehydrogenase(MTHFD) plays an important role in folate and homocysteine metabolisms, and polymorphisms of MTHFD may result in disturbance of the folate-mediated homocysteine pathway. The aim of this study is to test the hypothesis that genetic variants of MTHFD and plasma homocysteine levels are associated with risk of gastric cancer and modulated by genotypes of methylenetetrahydrofolate reductase(MTHFR). Experimental Design: We genotyped G1958A and T401C in MTHFD and C677T in MTHFR and detected total plasma homocysteine(tHcy) levels in a case-control study of 589 gastric cancer cases and 635 cancer-free controls in a high-risk Chinese population. Results:The variant genotypes of MTHFD 1958AA and 401CC were associated with a significantly increased risk of gastric canceradjusted odds ratio(OR), 2.05; 95% confidence interval(95% CI),1.34-3.13 for 1958AA; adjusted OR,1.43; 95% CI,1.14-1.80 for 401CC compared with 1958GG/GA and 401TT/TC genotypes, respectively. Both of the effects were more evident in the subjects carrying MTHFR 677CT/TT genotypes. The average tHcy level was significantly higher in gastric cancer cases than in controls(P < 0.01), and the upper quartile of tHcy (> 13.6 mu mol/L) was associated with an 82% significantly increased risk of gastric cancer, compared with the lowest quartile of tHcy(<= 8.0 pmol/L;adjusted OR,1.82; 95% CI,1.20-2.75). Conclusions:The strong associations between MTHFD variants and the plasma tHcy levels and gastric cancer risk suggest, for the first time, a possible gene-environment interaction between genetic variants of folate-metabolizing genes and high tHcy levels in gastric carcinogenesis.Wang, L. 7K-ATP channel openers facilitate glutamate uptake by GluTs in rat primary cultured Astrocytes显示文摘Increasing evidence, including from our laboratory, has revealed that opening of ATP sensitive potassium channels(K-ATP channels) plays the neuronal protective roles both in vivo and in vitro. Thus K-ATP channel openers(KCOs) have been proposed as potential neuroprotectants. Our previous studies demonstrated that K-ATP channels could regulate glutamate uptake activity in PC12 cells as well as in synaptosomes of rats. Since glutamate transporters(GluTs) of astrocytes play crucial roles in glutamate uptake and KATP channels are also expressed in astrocytes, the present study showed whether and how KATP channels regulated the function of GluTs in primary cultured astrocytes. The results showed that nonselective KCO pinacidil, selective mitochondrial KCO diazoxide, novel, and blood-brain barrier permeable KCO iptakalim could enhance glutamate uptake, except for the sarcolemmal KCO P1075. Moreover pinacidil, diazoxide, and iptakalim reversed the inhibition of glutamate uptake induced by 1-methyl-4-phenylpyridinium(MPP+). These potentiated effects were completely abolished by mitochondrial K-ATP blocker 5-hydroxydecanoate. Furthermore, either diazoxide or iptakalim could inhibit MPP+-induced elevation of reactive oxygen species (ROS) and phosphorylation of protein kinases C(PKC). These findings are the first to demonstrate that activation of K-ATP channel, especially mitochondrial K-ATP channel, improves the function of GluTs in astrocytes due to reducing ROS production and downregulating PKC phosphorylation. Therefore, the present study not only reveals a novel pharmacological profile of KCOs as regulators of GluTs, but also provides a new strategy for neuroprotection.Sun, X. L. Zeng, X. N. Zhou, F. 10Interfacial bonding mechanism in Al/coated steel dissimilar refill friction stir spot welds显示文摘Defect-free dissimilar Al/zinc coated steel and Al/AlSi coated steel welds were successfully fabricated by refill friction stir spot welding. However, Al alloy and uncoated steel could not be welded under the same welding condition. Al-Zn eutectic layer formed at the Al/zinc coated steel interface showed nonuniformity in thickness and nanoscale intermetallic(IMC) produced was discontinuous. The bonding formation between the Al-Zn layer and the surrounding materials was attributed to a liquid/solid reaction mechanism. Bonding formation at Al alloy and AlSi coated steel interface was attributed to a solid/solid reaction mechanism, as the joining process did not involve with melting of base metals or AlSi coating materials. Kissing bond formed at the weld boundary acted as a crack initiation and propagation site,and the present study showed that weld strength of Al 5754/AlSi coated steel was greatly influenced by properties of original IMC layer.Z. 11Mechanisms of High Coercivity in Ni/NiO Composite Films by Post Annealing显示文摘A coercivity as large as 2.4 kOe has been achieved in the Ni/NiO composite film after an annealing under a magnetic field of 10 kOe and an O_2 partial pressure of 0.001 torr.The coercivity was attributed to the strong exchange coupling of Ni and NiO.Small grain size of Ni and NiO was observed after the post-annealing.The enhanced coercivity is probably associated with the domain wall pinning by local energy minima,the distribution of Ni and NiO,and the domain structure in the interface of Ni/NiO generated under the presence of the magnetic field during the post-annealing.J.B. 14Constraints on sedimentary ages of the Chuanlinggou Formation in the Ming Tombs, Beijing, North China Craton: LA-ICP-MS and SHRIMP U–Pb dating of detrital zircons显示文摘Detrital zircons in five sedimentary samples,MC1 to MC5, from the bottom of the Chuanlinggou Formation in the Ming Tombs District, Beijing, were dated with the LA-ICP-MS and SHRIMP U–Pb methods. Age spectra of the five samples show a major peak at 2500 Ma and a secondary peak at 2000 Ma, suggesting their provenances were mainly from the crystalline basement of the North China Craton and the Trans-North China Orogen.The youngest zircon has an age of 1673 ± 44 Ma, indicating that the Chuanlinggou Formation was deposited after this age. From sample MC4 to MC5, lithology changed from a clastic rock(fine-grained sandstone) to a carbonate rock(fine-grained dolomite), suggesting that the depositional basin became progressively deeper. The age spectrum of sample MC5 shows a major peak at 2500 Ma and a secondary peak at 2000 Ma. Sample MC4, which is stratigraphically lower than sample MC5, only had one peak at 2500 Ma. We conclude that there was a transgressive event when sediments represented by MC5 was deposited, and seawater carried ca. 2000 Ma clastic materials to the basin where the Chuanlinggou Formation was deposited, leading to the addition of ca. 2000 Ma detritus.Our research indicates that the source area for the sediments became more extensive with time. We conclude thatthe Chuanlinggou Formation in the Ming Tombs District was deposited in a low-energy mud flat sedimentary environment in the inter-supra tidal zone because it is mainly composed of silty mudstone and fine-grained sandstone with relatively simple sedimentary structures.Jing Ding Yuruo Shi Alfred Kroner J.
