1Antidiabetic Effects of Gegen Qinlian Decoction via the Gut Microbiota Are Attributable to Its Key Ingredient Berberine显示文摘Gegen Qinlian Decoction(GQD),a traditional Chinese medicine(TCM)formula,has long been used for the treatment of common metabolic diseases,including type 2 diabetes mellitus.However,the main limitation of its wider application is ingredient complexity of this formula.Thus,it is critically important to identify the major active ingredients of GQD and to illustrate mechanisms underlying its action.Here,we compared the effects of GQD and berberine,a hypothetical key active pharmaceutical ingredient of GQD,on a diabetic rat model by comprehensive analyses of gut microbiota,short-chain fatty acids,proinflammatory cytokines,and ileum transcriptomics.Our results show that berberine and GQD had similar effects on lowering blood glucose levels,modulating gut microbiota,inducing ileal gene expression,as well as relieving systemic and local inflammation.As expected,both berberine and GQD treatment significantly altered the overall gut microbiota structure and enriched many butyrate-producing bacteria,including Faecalibacterium and Roseburia,thereby attenuating intestinal inflammation and lowering glucose.Levels of short-chain fatty acids in rat feces were also significantly elevated after treatment with berberine or GQD.Moreover,concentration of serum proinflammatory cytokines and expression of immune-related genes,including Nfkb1,Stat1,and Ifnrg1,in pancreatic islets were significantly reduced after treatment.Our study demonstrates that the main effects of GQD can be attributed to berberine via modulating gut microbiota.The strategy employed would facilitate further standardization and widespread application of TCM in many diseases.Xizhan Xu Zezheng Gao Fuquan Yang Yingying Yang Liang Chen Lin Han Na Zhao Jiayue Xu Xinmiao Wang Yue Ma Lian Shu Xiaoxi Hu Na Lyu Yuanlong Pan Baoli Zhu Linhua Zhao Xiaolin Tong Jun Wang 2020Genomics, Proteomics & Bioinformatics2020,18,6:29
2Association of serum levels of perfluoroalkyl substances with gestational diabetes mellitus and postpartum blood glucose显示文摘This study was conducted to examine the association of perfluoroalkyl substance(PFAS)exposure with gestational diabetes mellitus(GDM) risk and postpartum fasting blood glucose.We used a 1:2 matched case–control study with 84 GDM subjects and 168 healthy pregnant women from Beijing, China. The maternal blood was collected at 1–2 days before delivery, and eight linear isomers and fourteen branched isomers were determined in maternal serum.Logistic regression analyses were performed to evaluate the associations after adjusting for potential confounders. The median of the sum of levels of total PFASs was 4.24 ng/m L with a interquartile range(IQR) of 2.82–6.54 ng/m L. Although maternal PFAS exposure was not associated with risk of GDM, significant positive associations were observed between evaluated exposure to specific PFAS congeners and increasing blood glucose. The odds ratio(ORs) of the highest category of postpartum fasting blood glucose for perfluoro-1-metylheptylsulfonat(1 m-PFOS), perfluoro-3/4-metylheptylsulfonat(3 m+4 m-PFOS), perfluoro-5-metylheptylsulfonat(5 m-PFOS), and perfluorohexane sulfonate(PFHx S) were 2.03(95% CI: 1.09–3.77), 1.93(95% CI:1.04–3.58), 2.48(95% CI: 1.33–4.65), and 2.26(95% CI: 1.21–4.21), respectively, suggesting negative effects of maternal exposure to specific PFAS compounds on glucose metabolism.Yuxin Wang Lei Zhang Yue Teng Jiayu Zhang Lin Yang Jingguang Li Jianqiang Lai Yunfeng Zhao Yongning Wu 2018Journal of Environmental Sciences2018,30,7:8

4Benthic Habitat Quality Assessment Based on Biological Indices in Xiaoqing River Estuary and Its Adjacent Sea of Laizhou Bay,China显示文摘Based on the macrofauna data(2008-2011) in Xiaoqing River estuary and its adjacent sea, Laizhou Bay of Bohai Sea, China, the AZTI Marine Biotic Index(AMBI) and Multivariate AMBI(M-AMBI) were used for benthic habitat quality(BHQ) assessment. Results showed that BHQ presented an obvious trend of improvement along the direction of stream channel and river mouth, and in the coastal areas. AMBI and M-AMBI were significantly related to environmental pressure gradient data. Therefore, the two indices can well indicate BHQ in the studied area. However, there were significant differences between results of the two indices. In the cases of low taxa number and high abundance of single species, AMBI might overestimate BHQ. We thus adjusted its thresholds to solve this problem. And M-AMBI might overestimate BHQ when benthic assemblage was dominated by the opportunistic species. 5Polymorphisms of MTHFR and TYMS predict capecitabine-induced hand-foot syndrome in patients with metastatic breast cancer显示文摘Background:Breast cancer is a global problem,and a large number of new cases are diagnosed every year.Capecit-abine is effective in patients with metastatic breast cancer(MBC).Hand-foot syndrome(HFS)is a common adverse effect of capecitabine.In this study,we investigated the association between single nucleotide polymorphisms(SNPs)in genes involved in capecitabine metabolism pathways and capecitabine-induced HFS in Chinese patients with MBC to identify some predictive genetic biomarkers.Methods:We selected 3 genes involved in capecitabine metabolism and screened genetic variants in these target genes.We genotyped a total of 22 SNPs in the thymidylate synthase gene(TYMS),the methylene tetrahydrofolate reductase gene(MTHFR),and the ribonucleotide reductase M1 gene(RRM1)in 342 MBC patients treated with capecit-abine-based chemotherapy.The genotype distributions of each SNP in patients with and without HFS were assessed using Pearson’sχ^(2)test,and the relationship between HFS and genotypes of SNPs was determined using logistic regression analysis.The association between SNPs and their corresponding gene expression was analyzed using the Blood expression quantitative trait loci(eQTL)browser online tools.Results:We found 4 positive sites for HFS in the TYMS and MTHFR genes:TYMS rs2606241(P=0.022),TYMS rs2853741(P=0.019),MTHFR rs3737964(P=0.029),and MTHFR rs4846048(P=0.030).Logistic regression analyses showed that the genotype AG of MTHFR rs3737964[odds ratio(OR)=0.54,95%confidence interval(CI)0.31-0.97,P=0.038]and MTHFR rs4846048(OR=0.54,95%CI 0.30-0.98,P=0.042)were protective factors of HFS,whereas the genotype CT of TYMS rs2853741(OR=2.25,95%CI 1.31-3.87,P=0.012)increased the risk of HFS.The association between the genotype GT of TYMS rs2606241(OR=1.27,95%CI 0.73-2.23,P=0.012)and HFS was uncertain.Further eQTL analyses confirmed that the alleles of rs3737964 and rs4846048 affected the gene expression levels of MTHFR in cis.Conclusions:We have identified four potentially useful pharmacogenetic markers,TYMS rs2606241,TYMS rs2853741,MTHFR rs3737964,and MTHFR rs4846048 to predict capecitabine-induced HFS in MBC patients.Shaoyan Lin Jian Yue Xiuwen Guan Peng Yuan Jiayu Wang Yang Luo Ying Fan Ruigang Cai Qiao Li Shanshan Chen Pin Zhang Qing Li Fei Ma Binghe Xu 2019Cancer Communications2019,39,1:3
6Enhancing photovoltaic performance via aggregation dynamics control in fused-ring electron acceptor显示文摘A new fused-ring electron acceptor FNIC3 with dynamics controlled aggregation behavior was synthesized.FNIC3 shows strong absorption in 600–900 nm,HOMO/LUMO energy levels of−5.59/−4.04 eV,and electron mobility of 1.2×10^(−3) cm^(2) V^(−1) s^(−1).The aggregation of FNIC3 shows strong dependency on film formation time.Prolongation of film formation time promotes the crystallization of FNIC3,leading to improved crystallinity and enlarged aggregate sizes.Aggregation of FNIC3 significantly influences the photovoltaic device parameters.Appropriate aggregation red-shifts the absorption and improves the mobilities of the blend,which contributes to high photocurrent and fill factor thus high power conversion efficiency(PCE).Overaggregation leads to increased nonradiative energy loss and insufficient charge generation,resulting in decreased open-circuit voltage and short-circuit current density.The blends based on PM6:FNIC3 fabricated under proper film formation time exhibit a PCE of 12.3%,higher than those fabricated under short and long film formation time(10.0–10.5%).Jiayu Wang Runyu Zhu Shijie Wang Yawen Li Boyu Jia Jiadong Zhou Peiyao Xue Susanne Seibt Yuze Lin Zengqi Xie Wei Ma Xiaowei Zhan 2021Aggregate2021,2,3:1

9Improved in situ sequencing for high-resolution targeted spatial transcriptomic analysis in tissue sections显示文摘Spatial transcriptomics enables the study of localization-indexed gene expression activity in tissues,providing the transcriptional landscape that in turn indicates the potential regulatory networks of gene expression.In situ sequencing(ISS)is a targeted spatial transcriptomic technique,based on padlock probe and rolling circle amplification combined with next-generation sequencing chemistry,for highly multiplexed in situ gene expression profiling.Here,we present improved in situ sequencing(IISS)that exploits a new probing and barcoding approach,combined with advanced image analysis pipelines for high-resolution targeted spatial gene expression profiling.We develop an improved combinatorial probe anchor ligation chemistry using a 2-base encoding strategy for barcode interrogation.The new encoding strategy results in higher signal intensity as well as improved specificity for in situ sequencing,while maintaining a streamlined analysis pipeline for targeted spatial transcriptomics.We show that IISS can be applied to both fresh frozen tissue and formalin-fixed paraffin-embedded tissue sections for single-cell level spatial gene expression analysis,based on which the developmental trajectory and cell-cell communication networks can also be constructed.Xinbin Tang Jiayu Chen Xinya Zhang Xuzhu Liu Zhaoxiang Xie Kaipeng Wei Jianlong Qiu Weiyan Ma Chen Lin Rongqin Ke 2023Journal of Genetics and Genomics2023,50,9:1

11Unprecedented Heatwave in Western North America during Late June of 2021: Roles of Atmospheric Circulation and Global Warming显示文摘An extraordinary and unprecedented heatwave swept across western North America(i.e.,the Pacific Northwest)in late June of 2021,resulting in hundreds of deaths,a massive die-off of sea creatures off the coast,and horrific wildfires.Here,we use observational data to find the atmospheric circulation variabilities of the North Pacific and Arctic-Pacific-Canada patterns that co-occurred with the development and mature phases of the heatwave,as well as the North America pattern,which coincided with the decaying and eastward movement of the heatwave.Climate models from the Coupled Model Intercomparison Project(Phase 6)are not designed to simulate a particular heatwave event like this one.Still,models show that greenhouse gases are the main reason for the long-term increase of average daily maximum temperature in western North America in the past and future.Chunzai WANG Jiayu ZHENG Wei LIN Yuqing WANG 2023Advances in Atmospheric Sciences2023,40,1:1
12Modeling Impact of Natural Hazard-Induced Disasters on Income Distribution in the United States显示文摘Economic damage due to hurricane activities has been shown to impact income inequality in the coastal states of the United States. We consider 17 other natural hazards, in addition to hurricanes, that affected the entire United States for the period 1970–2013. Two fixed effects models were developed to quantify the relationship between income inequality and economic and demographic variables, including crop and property losses from natural hazard-induced disasters. These models include state-byyear and region-by-year fixed effects models. 14Enhancing Photovoltaic Performance of Asymmetric Fused-Ring Electron Acceptor by Expanding Pyrrole to Pyrrolo[3,2-b]pyrrole显示文摘We propose a strategy to improve performance of unidirectionally extended fused-ring electron acceptors by using pyrrolo[3,2-b]pyrrole to replace pyrrole ring, and design two asymmetric nonfullerene acceptors 1PIC and 2PIC. Replacing pyrrole in 1PIC with pyrrolo[3,2-b]pyrrole remarkably red-shifts absorption peak by 109 nm, elevates the HOMO and LUMO energy levels, and improves electron mobility. 18氯霉素及其产物降解菌群在长期驯化过程中的微生物群落和关键基因演替模式显示文摘As a widespread emerging contaminant,chloramphenicol(CAP)adversely impacts ecological communities in the water environment.Biological treatment is widely used for aquatic pollutant removal,and the performance of functional microbes determines its outcome.Herein,a consortium with a powerful CAPdegrading capacity was domesticated from activated sludge.As the common degradation products of CAP,4-nitrobenzoic acid(PNB)and 2,2-dichloroacetic acid(DCA)were also used as the sole substrates for long-term domestication.The successional pattern of the microbial community and critical functional genes through the 2.5-year domestication was revealed by metagenomic analysis.Sphingomonas,Caballeronia,and Cupriavidus became the most dominant populations in the CAP-,PNB-,and DCAdegrading consortia,respectively,and they were crucial degraders of PNB and DCA.Their collaboration contributed to the high mineralization rate of CAP.PNB was transformed into protocatechuic acid(PCA)and then mineralized through meta-cleavage and ortho-cleavage pathways.Crucial functional genes involved in CAP,PNB,and DCA metabolism,including CAP acetyltransferase,CAP oxidoreductase,haloacid dehalogenases,and protocatechuate dioxygenases,were significantly enriched in consortia.p H and carbon source had significant impacts on CAP biodegradation efficiency.The domesticated consortia and isolated strains are necessary microbial resources to enhance the bioremediation of CAP-,PNB-,or DCA-polluted environments.Jiayu Zhang Kaiyan Zhou Fangliang Guo Huaxin Lei Renxin Zhao Lin Lin Xiaoyan Li Bing Li 2023Engineering2023,31,12:0
