| 1 | Lx2-32c,a novel semi-synthetic taxane,exerts antitumor activity against prostate cancer cells in vitro and in vivo显示文摘Tubulin has been shown to be an effective target for the development of cytotoxic agents against prostate cancer. Previously, we reported that Lx2-32 c is an anti-tubulin agent with high binding affinity to tubulin. In this study, we investigated the potential of Lx2-32 c to act as an effective cytotoxic agent in the treatment of prostate cancer. MTT assays showed that Lx2-32 c was cytotoxic to all tested prostate cancer cell lines. The Lx2-32c-treated cells typically exhibited a rounded morphology associated with the onset of apoptosis, as evidenced by immunocytochemical staining. Human prostate cancer cell lines treated with Lx2-32 c arrest in the G2/M phase of the cell cycle and the treatment is associated with an increased ratio of cells in the sub-G0/G1 phase as determined by flow cytometry. Furthermore, expression of the cleaved form of poly(ADP-ribose) polymerase in prostate cancer cell lines treated with Lx2-32 c was shown by Western blotting assay. Xenograft implants of LNCa P and PC3-derived tumors in nude mice showed that Lx2-32 c treatment significant inhibited tumor growth with effects equivalent to those of docetaxel. | 2 | Integration of high-fidelity model of forward variable area bypass injector into zero-dimensional variable cycle engine model显示文摘Forward Variable Area Bypass Injector(FVABI)is one of key components which contributes to modulate the cycle parameters of Variable Cycle Engine(VCE)under various operation conditions.The modeling method of zero-dimensional FVABI was reviewed and its deficiency was analyzed based on FVABI flow characteristic.In order to improve the accuracy of VCE performance simulation,the high-fidelity modeling method of FVABI was developed based on its working characteristics.Then it was coupled with the zero-dimensional VCE model and the multi-level VCE model was built.The results indicate that the geometric and aerodynamic parameters can affect the interaction between the two airflows and the zero-dimensional FVABI model is too simple to predict the component performance accurately,especially when the FVABI inner bypass is chocked.Based on the performance curves for single bypass mode and the regression model of multi-scale support vector regression for double bypass mode,the high-fidelity model can predict FVABI performance accurately and rapidly.The integration of high-fidelity FVABI model into zerodimensional VCE model can be done by adjusting iterative variables and balance equations.The multi-level model has good convergence and it can predict VCE performance when the FVABI inner bypass is chocked. | Fu SONG Li ZHOU Zhanxue WANG Zhifu LIN Jingwei SHI | 2021 | Chinese Journal of Aeronautics2021,34,8: | 3 |
| 3 | Surveillance of enteric pathogens in imported seafood and environmental surfaces in five seafood markets before the outbreak of COVID-19显示文摘To monitor the presence of enteric pathogens in imported seafood,a total of 140 seafood samples imported from eight overseas countries were collected from Beijing,Dalian,Shanghai,Guangzhou,and Wuhan seafood markets from June to November 2019.Additionally,116 viral,environmental swab samples were also collected from the Wuhan and Guangzhou seafood markets.Five typical enteric bacterial pathogens(Aeromonas spp.,Shigella spp.,Salmonella spp.,Vibrio spp.,and Listeria monocytogenes)and four viruses(Rotavirus,Norovirus,Astrovirus,and Sapovirus)were detected positive.Results showed that eight Vibrio parahaemolyticus isolates appeared in seafood imported to Dalian,Wuhan,Shanghai,Guangzhou,and Beijing.In contrast,Vibrio fluvialis and Aeromonas were isolated in another two samples.Norovirus was detected in one oyster sample imported from France and environmental surface in Guangzhou.The remaining pathogens were negative in all the samples being tested.With 120 V.parahaemolyticus isolates from the above countries,the genomic analysis revealed that sequence type ST1152 isolates imported from Canada were clustered with two V.parahaemolyticus isolates from Canada.This study presented the first microbiological analysis of the Wuhan seafood market before the outbreak of COVID-19,which demonstrated that supervision should be strengthened to prevent enteric pathogens via imported seafood. | Songzhe Fu Wei Wang Qingyao Wang Fenglan He Jingwei Hao Bo Pang | 2021 | Biosafety and Health2021,3,4: | 1 |

| 5 | Isolation and identification of a novel coronavirus from wild bird显示文摘A novel coronavirus strain was isolated from laryngotracheal swab of wild partridge and designated as partridge/GD/S14/2003 (S14). Its whole genomic sequence was obtained (GenBank Accession number: AY646283) through RT-PCR amplification, cloning, nucleotide sequencing, and analysis by the DNASTAR program. To investigate the origin of the virus, we further analyzed the nucleotide sequences of the main structural proteins, and compared those with other available virus isolates. Our results showed that the highest nucleotide homologies between the S1 gene of S14 strain and those of nephrogenic-type strains JX1-99 and TJ2-96 were 94.6% and 93.4%, respectively. In addition, a relatively high genetic identity, 85% and 84.3%, respectively, was detected between S1 gene of S14 and those of strains QXIBV and LX4. The results suggested that the S14 strain may be originated from or related to nephrogenic-type and proventriculus-type infectious bronchitis virus (IBV). The highest nucleotide homology between the S2 gene of S14 strain and those of QXIBV and LX4 was 85% and 84.3%, respectively and all of them belonged to group II coronaviruses. The highest nucleotide homology between the M gene of S14 strain and those of strains SAIB20 and GD6-98 was 90.6% and 90.2%, respectively by which S14 belonged to group III. Although they displayed high level of genetic identity in S1 and S2 gene, there was lower homology of M coding sequences between S14 and BJ, and between S14 and QXIBV strains. Phylogenetic analysis of N gene indicated that group I strains might evolve from RNA recombination between strain H52 and Gray; while group II strains from strain H120 and D1466. S14 strain had the highest N gene homology with strain QXIBV which was 95.7%, thus classified as a group III member. Strains SAIB20 and GD6-98 which were closely related to the M gene of S14 strain belonged to group I and IV, respectively.
