| 1 | Experimental Study of the Pressure Fluctuations in a Pump Turbine at Large Partial Flow Conditions显示文摘Frequent shifts of output and operating mode require a pump turbine with excellent stability. Current researches show that large partial flow conditions in pump mode experience positive-slope phenomena with a large head drop. The pressure fluctuation at the positive slope is crucial to the pump turbine unit safety. The operating instabilities at large partial flow conditions for a pump turbine are analyzed. The hydraulic performance of a model pump turbine is tested with the pressure fluctuations measured at unstable operating points near a positive slope in the performance curve. The hydraulic performance tests show that there are two separated positive-slope regions for the pump turbine, with the flow discharge for the first positive slope from 0.85 to 0.91 times that at the maximum efficiency point. The amplitudes of the pressure fluctuations at these unstable large partial flow conditions near the first positive slope are much larger than those at stable operating condtions. A dominant frequency is measured at 0.2 times the impeller rotational frequency in the flow passage near the impeller exit, which is believed to be induced by the rotating stall in the flow passage of the wicket gates. The test results also show hysteresis with pressure fluctuations when the pump turbine is operated near the first positive slope. The hysteresis creates different pressure fluctuations for those operation points even though their flow rates and heads are similar respectively. | 2 | Programmed death-1 expression is associated with the disease status in hepatitis B virus infection显示文摘AIM: To define the potential role of programmed death-1/programmed death-ligand (PD-1/PD-L) pathway in different hepatitis B virus (HBV) infection disease status; we examined the expression of PD-1 on antigen specific CD8+ T cells in peripheral blood of patients with chronic hepatitis B (CHB) and acute exacerbation of hepatitis B (AEHB) infection. METHODS: The PD-1 level on CD8+ T lymphocytes and the number of HBV specifi c CD8+ T lymphocytes in patients and healthy controls (HCs) were analyzed by staining with pentameric peptide-human leukocyte antigen2 (HLA2) complexes combined with flow cytometry. Real-time quantitative polymerase chain reaction (PCR) was used to measure the serum HBV-DNA levels. RESULTS: The level of PD-1 expression on total CD8+ T cells in CHB patients (13.86% ± 3.38%) was significantly higher than that in AEHB patients (6.80%± 2.19%, P < 0.01) and healthy individuals (4.63% ± 1.23%, P < 0.01). Compared to AEHB patients (0.81% ± 0.73%), lower frequency of HBV-specific CD8+ T cells was detected in chronic hepatitis B patients (0.37% ± 0.43%, P < 0.05). There was an inverse correlation between the strength of HBV-specific CD8+ T-cell response and the level of PD-1 expression. Besides, there was a significant positive correlation between HBV viral load and the percentage of PD-1 expression on CD8+ T cells in CHB and AEHB subjects (R = 0.541, P < 0.01). However, PD-1 expression was not associated with disease flare-ups as indicated by alanine aminotransferase (ALT) levels (R = 0.066, P > 0.05). CONCLUSION: Our results confirm previous reports that HBV specific CD8+ T-cell response in the peripheral blood is more intense in patients with AEHB than in chronic hepatitis B with persistent viral infection. | 3 | Highly efficient generation of arbitrary vector beams with tunable polarization, phase, and amplitude显示文摘We propose an efficient and robust method to generate tunable vector beams by employing a single phasetype spatial light modulator(SLM). With this method, a linearly polarized Gaussian beam can be converted into a vector beam with arbitrarily controllable polarization state, phase, and amplitude. The energy loss during the conversion is greatly reduced and depends mainly on the reflectivity of the SLM. We experimentally demonstrate that conversion efficiency of about 47% is achieved by using an SLM with reflectivity of 62%.Several typical vector beams, including cylindrical vector beams, vector beams on higher order Poincaré spheres,and arbitrary vector beams attached with phases and with tunable amplitude, are generated and verified experimentally. | 4 | Development of synthetic volatile attractant for male Ectropis obliqua moths显示文摘The tea geometrid Ectropis obliqua is one of the most serious leaf-feeding insect pests in tea(Camellia sinensis) in East Asia. Although several volatile chemicals emitted from tea plants have been reported to be attractive to E. obliqua moths, no synthetic attractants for E. obliqua moths have been developed. By measuring the behavioral responses of the moth to a series of chemicals in the lab, we found that a blend containing a ternary mixture containing(Z)-3-hexenal,(Z)-3-hexenyl hexanoate and benzyl alcohol clearly attracted to E. obliqua moths of both sex and that(Z)-3-hexenyl acetate could enhance the attractiveness of the ternary blend. Moreover, we found that the volatiles emitted from the plant-E. obliqua larva complex have the same attractiveness as: 1) the blend of volatiles containing the ternary mixture and 2) the blend containing(Z)-3-hexenyl acetate plus the ternary mixture to both male and female moths. In a field bioassay, more male moths were observed on traps that were baited with the blend containing(Z)-3-hexenyl acetate plus the ternary mixture than on control traps. | 7 | Effects of H pylori infection on gap-junctional intercellular communication and proliferation of gastric epithelial cells in vitro显示文摘AIM: To explore the effects of H pylori infection on gap-junctional intercellular communication (GJIC) and proliferation of gastric epithelial cells in vitro. METHODS: A human gastric epithelial cell line (SGC-7901) cultured on coverslips was exposed overnight to intact H pylori (CagA+ or CagA-strains) and sonicated extracts,respectively. GJIC between the cells was detected by fluorescence redistribution after photobleaching (FRAP) technique. Proliferation of SGC cells was determined by methylthiazolyl tetrazolium (MTT) assay. RESULTS: When compared with control in which cells were cultured with simple medium alone,both CagA+ and CagA-H pylori isolates could inhibit GJIC (CagA+: F = 57.98,P < 0.01; CagA-: F = 29.59,P < 0.01) and proliferation (CagA+: F = 42.65,P < 0.01; CagA-: F = 58.14,P < 0.01) of SGC-7901 cells. Compared with CagA-strains,CagA+ H pylori more significantly down-regulated GJIC of gastric cells (intact H pylori: t = 13.86,P < 0.01; sonicated extracts: t = 11.87,P < 0.01) and inhibited proliferation gastric cells to a lesser extent in vitro (intact H pylori: t = 3.06,P < 0.05; sonicated extracts: t = 3.94,P < 0.01). CONCLUSION: Compared with CagA-H pylori strains,CagA+ strains down-regulate GJIC of gastric epithelia cells more significantly and inhibit proliferation of gastric cells to a lesser extent in vitro . | 9 | Effect of moxa smoke produced during combustion of Aiye(Folium Artemisiae Argyi) on behavioral changes in mice inhaling the smoke显示文摘OBJECTIVE: To evaluate the effects that inhalation of the combustion products of the traditional Chinese herb Aiye(Folium Artemisiae Argyi) has on the central nervous system.METHODS: Forty Kunming mice(half male) were randomly assigned(n = 10/group) to a control group(C) or one of three moxa smoke concentration groups(% opacity): low(L1; 0.4%), medium(M1; 2%), and high(H1; 15%). Mice in the latter three groups were exposed to moxa smoke in a dynamic gas exposure cabinet for 20 min per day for7 days. Mice in control group were placed in the same cabinet without any intervention. For the sleep experiments, another 50 mice were divided into five groups of 10 mice each: a saline-injected control group, L1 + pentobarbital sodium(PS)-injected group, M1+PS group, H1(15%)+PS group,and a positive control group(10 mg/kg, chlorpromazine, p.o.). The weight, general activities, locomotor activities, rotarod performance, sleep duration,and sleeping rate induced by a subthreshold dose of pentobarbital sodium were recorded in the mice,and the composition of moxa smoke was analyzed using headspace gas chromatography(GC-HS).RESULTS: A low concentration of smoke significantly decreased the frequency of locomotor activities and the time for which the mice remaining on the rotarod; however, a high smoke concentration significantly prolonged the pentobarbital-induced sleeping time and sleeping rate.CONCLUSION: The concentration-dependent relaxing effects of moxa smoke on the Central Nervous System(CNS) were confirmed. Moreover, GC-HS analysis showed that the component present in the highest concentration in moxa smoke was eucalyp-tol, an essential oil well recognized for its soothing effects on the CNS. | 10 | A comparison of atmospheric CO_2 concentration GOSAT-based observations and model simulations显示文摘Satellite observations of atmospheric CO2 are able to truly capture the variation of global and regional CO2 concentration.The model simulations based on atmospheric transport models can also assess variations of atmospheric CO2 concentrations in a continuous space and time,which is one of approaches for qualitatively and quantitatively studying the atmospheric transport mechanism and spatio-temporal variation of atmospheric CO2 in a global scale.Satellite observations and model simulations of CO2 offer us two different approaches to understand the atmospheric CO2.However,the difference between them has not been comprehensively compared and assessed for revealing the global and regional features of atmospheric CO2.In this study,we compared and assessed the spatio-temporal variation of atmospheric CO2 using two datasets of the column-averaged dry air mole fractions of atmospheric CO2(XCO2)in a year from June 2009 to May 2010,respectively from GOSAT retrievals(V02.xx)and from Goddard Earth Observing System-Chemistry(GEOS-Chem),which is a global 3-D chemistry transport model.In addition to the global comparison,we further compared and analyzed the difference of CO2 between the China land region and the United States(US)land region from two datasets,and demonstrated the reasonability and uncertainty of satellite observations and model simulations.The results show that the XCO2 retrieved from GOSAT is globally lower than GEOS-Chem model simulation by 2 ppm on average,which is close to the validation conclusion for GOSAT by ground measures.This difference of XCO2 between the two datasets,however,changes with the different regions.In China land region,the difference is large,from 0.6 to 5.6 ppm,whereas it is 1.6 to 3.7 ppm in the global land region and 1.4 to 2.7 ppm in the US land region.The goodness of fit test between the two datasets is 0.81 in the US land region,which is higher than that in the global land region(0.67)and China land region(0.68).The analysis results further indicate that the inconsistency of CO2concentration between satellite observations and model simulations in China is larger than that in the US and the globe.This inconsistency is related to the GOSAT retrieval error of CO2 caused by the interference among input parameters of satellite retrieval algorithm,and the uncertainty of driving parameters in GEOS-Chem model. | LEI LiPing GUAN XianHua ZENG ZhaoCheng ZHANG Bing RU Fei BU Ran | 2014 | Science China Earth Sciences2014,57,6: | 6 |

| 13 | Determination of thermal expansion coefficients for unidirectional fiber-reinforced composites显示文摘In the present work, the coefficients of thermal expansion(CTEs) of unidirectional(UD)fiber-reinforced composites are studied. First, an attempt is made to propose a model to predict both longitudinal and transverse CTEs of UD composites by means of thermo-elastic mechanics analysis. The proposed model is supposed to be a concentric cylinder with a transversely isotropic fiber embedded in an isotropic matrix, and it is subjected to a uniform temperature change. Then a concise and explicit formula is offered for each CTE. Finally, some finite element(FE) models are created by a finite element program MSC. Patran according to different material systems and fiber volume fractions. In addition, the available experimental data and results of other analytical solutions of CTEs are presented. Comparisons are made among the results of the cylinder model,the finite element method(FEM), experiments, and other solutions, which show that the predicted CTEs by the new model are in good agreement with the experimental data. RESULTS:Compared with HepG2 cells,HepG2.2.15 cells showed organelle degeneration and filopodia disappearance under electron microscope.HepG2.2.15 cells proliferated and migrated slowly in vitro,and hardly formed tumor and lung metastasis in nude mice.Flow cytometry showed that the majority of HepG2.2.15 cells were arrested in G1 phase,and apoptosis was minor in both cell lines.Furthermore,the levels of cytoskeletal proteins F-actin and Ezrin were decreased in HepG2.2.15 cells. | 17 | Functional assembly of root-associated microbial consortia improves nutrient efficiency and yield in soybean显示文摘Root-associated microbes are critical for plant growth and nutrient acquisition. However, scant information exists on optimizing communities of beneficial root-associated microbes or the mechanisms underlying their interactions with host plants. In this report, we demonstrate that rootassociated microbes are critical influencers of host plant growth and nutrient acquisition. Three synthetic communities(SynComs) were constructed based on functional screening of 1,893 microbial strains isolated from root-associated compartments of soybean plants. Functional assemblage of SynComs promoted significant plant growth and nutrient acquisition under both N/P nutrient deficiency and sufficiency conditions.Field trials further revealed that application of SynComs stably and significantly promoted plant growth, facilitated N and P acquisition, and subsequently increased soybean yield. Among the tested communities, SynCom1 exhibited the greatest promotion effect, with yield increases of up to 36.1% observed in two field sites. Further RNA-seq implied that SynCom application systemically regulates N and P signaling networks at the transcriptional level, which leads to increased representation of important growth pathways, especially those related to auxin responses. | 18 | Serine metabolism orchestrates macrophage polarization by regulating the IGF1–p38 axis显示文摘Serine metabolism is reportedly involved in immune cell functions, but whether and how serine metabolism regulates macrophage polarization remain largely unknown. Here, we show that suppressing serine metabolism, either by inhibiting the activity of the key enzyme phosphoglycerate dehydrogenase in the serine biosynthesis pathway or by exogenous serine and glycine restriction, robustly enhances the polarization of interferon-γ-activated macrophages (M(IFN-γ)) but suppresses that of interleukin-4-activated macrophages (M(IL-4)) both in vitro and in vivo. Mechanistically, serine metabolism deficiency increases the expression of IGF1 by reducing the promoter abundance of S-adenosyl methionine-dependent histone H3 lysine 27 trimethylation. IGF1 then activates the p38-dependent JAK–STAT1 axis to promote M(IFN-γ) polarization and suppress STAT6-mediated M(IL-4) activation. | 20 | Milk production and composition and metabolic alterations in the mammary gland of heat-stressed lactating dairy cows显示文摘This experiment was conducted to investigate the effects of heat stress(HS) on the feed intake, milk production and composition and metabolic alterations in the mammary gland of dairy cows. Twenty Holstein cows were randomly assigned to one of two treatments according to a completely randomized design. Half of the cows were allocated to the HS group in August(summer season), and the other half were assigned to the HS-free group in November(autumn season). HS reduced(P<0.01) dry matter intake(DMI), milk yield, milk protein and milk urea nitrogen(MUN) of cows compared with HSfree control, but increased(P<0.01) milk somatic cell counts(SCC). We determined the HS-induced metabolic alterations and the relevant mechanisms in dairy cows using liquid chromatography mass spectrometry combined with multivariate analyses. Thirty-four metabolites were identified as potential biomarkers for the diagnosis of HS in dairy cows. Ten of these metabolites, glucose, lactate, pyruvate, lactose, β-hydroxybutyrate, citric acid, α-ketoglutarate, urea, creatine, and orotic acid, had high sensitivity and specificity for HS diagnoses, and seven metabolites were also identified as potential biomarkers of HS in plasma, milk, and liver. These substances are involved in glycolysis, lactose, ketone, tricarboxylic acid(TCA), amino acid and nucleotide metabolism, indicating that HS mainly affects lactose, energy and nucleotide metabolism in the mammary gland of lactating dairy cows.
