1A multiplex PCR for differential detection of Echinococcus granulosus sensu stricto,Echinococcus multilocularis and Echinococcus canadensis in China显示文摘Background:Echinococcosis caused byEchinococcus is one of the most major infectious diseases in north-west highland of China.E.granulosus sensu strict,E.multilocularis,andE.canadensis are known to be the only three species related to human health transmitting in the areas.To achieve targeted treatment and control of echinococcosis,the accurate identification and discrimination of the species are important.However,currently the available diagnostic approaches do not present ideal results either in accuracy or efficiency.Methods:In the study,a set of primers were designed to aim at the three human-pathogenicEchinococcus species in China.The one-step multiplex PCR assay was developed and evaluated for the specificity and sensitivity.A total of 73parasitic lesions and 41 fecal materials obtained from human and various animals collected in the clinic and the field were tested to assess the applicability of this method.Results:The multiplex PCR effectively detected the individual DNA from the targeted species and their random mixtures generating with distinguishable expected size of products.The detection limit of the assay for each of the three species was 5 pg/μl when they were tested separately.When DNA mixtures of the targeted species containing the same concentration were used as templates,the lowest amount of DNA which can be detected was 50 pg/μl,10 pg/μl and 5 pg/μl forE.granulosus s.s.,E.multilocularis,andE.canadensis respectively.No cross-reactivity was observed when DNA from eight genetically close species was used as control templates.The multiplex PCR identifications of all samples were in line with the original sequencing results except for those infected withE.shiquicus,which showed negative signals in the developed assay.Of all the tested stool materials,16 were previously found positive forEchinococcus by visual and microscopic examination.Among these 16 samples,13 were confirmed by the multiplex PCR,and the other three tested negative.Additionally,the multiplex PCR identified another 14 positive feces from the remained 25 stool samples which absence of worms.Conclusions:The developed multiplex PCR shows advantages in fast diagnosis and large-scale epidemiological investigation,which proven to be a promising tool utilized in clinic and surveillance system.Jing-Ye Shang Guang-Jia Zhang Sha Liao Yan Huang Wen-Jie Yu Wei He Guang-You Yang Tiao-Ying Li Xing-Wang Chen Bo Zhong Qian Wang Qi Wang Rui-Rui Li Hao Wang 2019Infectious Diseases of Poverty2019,8,4:11

3FluxExplorer: A general platform for modeling and analyses of metabolic net-works based on stoichiometry显示文摘Stoichiometry-based analyses of meta- bolic networks have aroused significant interest of systems biology researchers in recent years. It is necessary to develop a more convenient modeling platform on which users can reconstruct their network models using completely graphical operations, and explore them with powerful analyzing modules to get a better understanding of the properties of metabolic systems. Herein, an in silico platform, FluxExplorer, for metabolic modeling and analyses based on stoichiometry has been developed as a publicly available tool for systems biology research. This platform integrates various analytic approaches, in- cluding flux balance analysis, minimization of meta- bolic adjustment, extreme pathways analysis, shadow prices analysis, and singular value decom- position, providing a thorough characterization of the metabolic system. Using a graphic modeling process, metabolic networks can be reconstructed and modi- fied intuitively and conveniently. The inconsistencies of a model with respect to the FBA principles can be proved automatically. In addition, this platform sup- ports systems biology markup language (SBML). FluxExplorer has been applied to rebuild a metabolic network in mammalian mitochondria, producing meaningful results. 4Dmrt1 regulates the immune response by repressing the TLR4 signaling pathway in goat male germline stem cells显示文摘Double sex and mab-3-related transcription factor 1(Dmrt1),which is expressed in goat male germline stem cells(mGSCs)and Sertoli cells,is one of the most conserved transcription factors involved in sex determination.In this study,we highlighted the role of Dmrt1 in balancing the innate immune response in goat mGSCs.Dmrt1 recruited promyelocytic leukemia zinc finger(Plzf),also known as zinc finger and BTB domain-containing protein 16(Zbtb16),to repress the Toll-like receptor 4(TLR4)-dependent inflammatory signaling pathway and nuclear factor(NF)-κB.Knockdown of Dmrt1 in seminiferous tubules resulted in widespread degeneration of germ and somatic cells,while the expression of proinflammatory factors were significantly enhanced.We also demonstrated that Dmrt1 stimulated proliferation of mGSCs,but repressed apoptosis caused by the immune response.Thus,Dmrt1 is sufficient to reduce inflammation in the testes,thereby establishing the stability of spermatogenesis and the testicular microenvironment.Yu-Dong Wei Xiao-Min Du Dong-Hui Yang Fang-Lin Ma Xiu-Wei Yu Meng-Fei Zhang Na Li Sha Peng Ming-Zhi Liao Guang-Peng Li Chun-Ling Bai Wei-Shuai Liu Jin-Lian Hua 2021Zoological Research2021,42,1:5
5LIN28A inhibits DUSP family phosphatases and activates MAPK signaling pathway to maintain pluripotency in porcine induced pluripotent stem cells显示文摘LIN28A,an RNA-binding protein,plays an important role in porcine induced pluripotent stem cells(piPSCs).However,the molecular mechanism underlying the function of LIN28A in the maintenance of pluripotency in piPSCs remains unclear.Here,we explored the function of LIN28A in piPSCs based on its overexpression and knockdown.We performed total RNA sequencing(RNA-seq)of piPSCs and detected the expression levels of relevant genes by quantitative real-time polymerase chain reaction(qRT-PCR),western blot analysis,and immunofluorescence staining.Results indicated that piPSC proliferation ability decreased following LIN28A knockdown.Furthermore,when LIN28A expression in the shLIN28A2 group was lower(by 20%)than that in the negative control knockdown group(shNC),the pluripotency of piPSCs disappeared and they differentiated into neuroectoderm cells.Results also showed that LIN28A overexpression inhibited the expression of DUSP(dual-specificity phosphatases)family phosphatases and activated the mitogen-activated protein kinase(MAPK)signaling pathway.Thus,LIN28A appears to activate the MAPK signaling pathway to maintain the pluripotency and proliferation ability of piPSCs.Our study provides a new resource for exploring the functions of LIN28A in piPSCs.Xiao-Long Wu Zhen-Shuo Zhu Xia Xiao Zhe Zhou Shuai Yu Qiao-Yan Shen Ju-Qing Zhang Wei Yue Rui Zhang Xin He Sha Peng Shi-Qiang Zhang Na Li Ming-Zhi Liao Jin-Lian Hua 2021Zoological Research2021,42,3:5

8Vegetation Restoration Technique System and Its Ecological Functions of the Degraded Ecosystem in the Arid-Hot Valleys——Taking the Example of Typical Mode in Small Watershed of Yuanmou显示文摘This paper concluded the vegetation restoration technique system in the arid-hot valleys and studied the anti-erosion function, environmental function and biological diversity effects of vegetation restoration on the ecosystem in the arid-hot valleys. The results showed that the soil erodibility decreased significantly after the vegetation restoration. The climate environment of the small watershed had a great improvement after the vegetation restoration, of which the temperature decreased, the humidity increased, the harsh environment of dry and hot in this region changed. The studies of the ecosystem biodiversity were mainly on the analysis of the relations between biodiversity and ecological function of the artificial ecological forest pattern and the natural enclosed treatment mode on the severely of degraded land. It could conclude that the natural en-closed treatment mode is helpful to the biodiversity of the ecosystem and the improvement and stability of the ecosystem, and Leucaena artificial forest restoration pattern reduced the species diversity but optimized the ecological function. 9Landscape Ecological Evaluation on the Artificial Forest of Leucaena leucocephala——Taking the Example of Xiaokua Hill of Yuanmou显示文摘Based on the theory of ecology, this article conducted a landscape ecological evaluation on Leucaena leucocephala artificial forest in the arid-hot valley, Yuanmou Xiaokua hill,using a combination method of qualitative analysis and landscape comprehensive evaluation. The comprehensive evaluation indexes of landscape include forest coverage, biodiversity, stability, soil erosion, environmental quality, availability, scientific research characteristic, economic benefits. Using AHP (analytical hierarchy process), determine the grade of ecological evaluation factors and the weight of evaluation index and calculate the CEI (comprehensive evaluation index) on ecological evaluation. Evaluation results showed that the area CEI was 0.714 225, a better quality of the eco-logical level, but biodiversity indicators was the lowest. To sum up, it was difficult to maintain ecological restoration effectiveness and process but also guarantee a good biodiversity in the arid-hot valleys. 12Rainfall-Interception Effect of Vegetation Restored Region of Gully in Arid-Hot Valley of Jinshajiang River显示文摘This paper divides the process of rainfall redistribution into four levels in vegetation restored region of gully in arid-hot valley of Jinshajiang River. These levels include arbor interception, litter interception, soil interception and surface runoff. The results showed that the maximum of interception was 8.10 mm, the mean interception was 4.53 mm, the maximum of interception rate was 56.10 % and the mean interception rate was 32.63 %.When the rainfall is over 20 mm, the interception rate descends rapidly along with the rainfall increasing. The maximum of water-holding content of litter was 8.14-20.33 t/hm2, the mean of which is 12.52 t/hm2 and the maximum of water-holding content was over two times of it own weight.The rainfall of litter interception was 150.24 mm, accounted for the rainfall income 24.48%.The maximum water-holding capacity of soil achieves 2 198.54 t/hm2, but that of the barren land is 1 825.60 t/hm2. The rainfall of soil interception was 219.87 mm, accounted for the rainfall income 35.82%. The surface runoff of vegetation restored region was 43.41 mm, accounted for 7.07% of the rainfall income, but the surface runoff of non-forest land was 413.24 mm, accounts for 70.26% of the rainfall income.The surface runoff of the planted forest community was by 63.19% lower than that of the non-forest land. 13Estimation method for random sonic fatigue life of thin-walled structure of a combustor liner based on stress probability distribution显示文摘As to the sonic fatigue problem of an aero-engine combustor liner structure under the random acoustic loadings,an effective method for predicting the fatigue life of a structure under random loadings was studied.Firstly,the probability distribution of Von Mises stress of thin-walled structure under random loadings was studied,analysis suggested that probability density function of Von Mises stress process accord approximately with two-parameter Weibull distribution.The formula for calculating Weibull parameters were given.Based on the Miner linear theory,the method for predicting the random sonic fatigue life based on the stress probability density was developed,and the model for fatigue life prediction was constructed.As an example,an aero-engine combustor liner structure was considered.The power spectrum density(PSD) of the vibrational stress response was calculated by using the coupled FEM/BEM(finite element method/boundary element method) model,the fatigue life was estimated by using the constructed model.And considering the influence of the wide frequency band,the calculated results were modified.Comparetive analysis shows that the estimated results of sonic fatigue of the combustor liner structure by using Weibull distribution of Von Mises stress are more conservative than using Dirlik distribution to some extend.The results show that the methods presented in this paper are practical for the random fatigue life analysis of the aeronautical thin-walled structures.SHA Yun-dong GUO Xiao-peng LIAO Lian-fang XIE Li-juan 2011航空动力学报2011,26,4:1
