1Segmentation of Hematoxylin-Eosin stained breast cancer histopathological images based on pixel-wise SVM classifier显示文摘Hematoxylin-Eosin(HE) staining is the routine diagnostic method for breast cancer(BC), and large amounts of HE stained histopathological images are available for analysis. It is emergent to develop computational methods to efficiently and objectively analyze these images, with the aim of providing potentially better diagnostic and prognostic information for BC. This work focuses on analyzing our in-house HE stained histopathological images of breast cancer tissues. Since tumor nests(TNs) and stroma morphological characteristics can reflect the biological behaviors of breast invasive ductal carcinoma(IDC), accurate segmentation of TNs and the stroma is the first step towards the subsequent quantitative analysis. We first propose a method based on the pixel-wise support vector machine(SVM) classifier for segmenting TNs and the stroma, then extract four morphological characters related to the TNs from the images and investigate their relationships with the patients' 8-year disease free survival(8-DFS). The evaluation result shows that the classification based segmentation method is able to distinguish between TNs and stroma with 87.1% accuracy and 80.2% precision,suggesting that the proposed method is promising in segmenting HE stained IDC histopathological images. 3Stress-induced CsPbBr3 nanocrystallization on glass surface:Unexpected mechanoluminescence and applications显示文摘In this work,we discovered an unexpected mechanoluminescence (ML) phenomena occurring when transforming amorphous into crystalline,due to the stress-induced precipitation of CsPbBr3 perovskite nanocrystals on glass surface.It is revealed that,unlike the conventional thermal-induced phase transformation mechanism,the breakage of bonding of glass network provides the energy for nucleation and growth,and the shear stress avoids the long-range migration of structural units for crystallization.Such unique ML phenomenon enables the visualization of dynamical force that is inaccessible by common strategy,and so,opens up some novel applications,such as the pressure-sensitive 'glassy pencil' to learn people's writing habits,and the pb^2+-detection with good sensitivity and selectivity.These findings not only demonstrate an effective route for the preparation of perovskite materials in a green,time-saving,low cost,and scalable way,enrich the knowledge of glass crystallization mechanism,but also exploit a useful avenue to quantitatively visualize the dynamical force.Xiaoqiang Xiang Hang Lin Renfu Li Yao Cheng Qingming Huang Ju Xu Congyong Wang Xueyuan Chen Yuansheng Wang 2019Nano Research2019,12,5:8
4Construction and visualization of high-resolution three-dimensional anatomical structure datasets for Chinese digital human显示文摘The objective of the China Digital Human Project (CDH) is to digitize and visualize the anatomical structures of human body. In the project, a database with information of morphology, physical charac-teristics and physiological function will be constructed. The raw data of CDH which was completed in the Southern Medical University is employed. In Huazhong University of Science and Technology (HUST), the frozen section images are preprocessed, segmented, labeled in accordance with the major organs and tissues of human beings, and reconstructed into three-dimensional (3D) models in parallel on high performance computing clusters (HPC). Some visualization software for 2D atlas and 3D mod-els are developed based on the new dataset with high resolution (0.1mm×0.1mm×0.2mm). In order to share, release and popularize the above work, a website (www.vch.org.cn) is online. 5Q-switching of waveguide lasers based on graphene/WS_2 van der Waals heterostructure显示文摘We report on the operation of passively Q-switched waveguide lasers at 1 μm wavelength based on a graphene∕WS_2 heterostructure as a saturable absorber(SA). The gain medium is a crystalline Nd:YVO_4 cladding waveguide produced by femtosecond laser writing. The nanosecond waveguide laser operation at 1064 nm has been realized with the maximum average output power of 275 m W and slope efficiency of 37%. 6Fabrication technique of micro/nano-scale speckle patterns with focused ion beam显示文摘The fabrication technique of micro/nano-scale speckle patterns with focused ion beam (FIB) system is studied for digital image correlation (DIC) measurement under a scanning electron microscope (SEM).The speckle patterns are fabricated by directly etching the counterpart of the specimen to the black part of a template.Mean intensity gradient is used to evaluate the quality of these SEM images of speckle patterns fabricated based on different templates to select an optimum template.The pattern size depending on the displacement measurement sensitivity is adjusted by altering the magnification of FIB according to the relation curve of the etching size versus magnification.The influencing factors including etching time and ion beam current are discussed.Rigid body translation tests and rotation tests are carried out under SEM to verify the reliability of the fabricated speckle patterns.The calculated values are in good agreement with the imposed ones.LI YanJie XIE HuiMin LUO Qiang GU ChangZhi HU ZhenXing CHEN PengWan ZHANG QingMing 2012Science China(Physics,Mechanics & Astronomy)2012,55,6:7
7Firing patterns of long-term cultured neuronal network on multi-electrode array显示文摘Spontaneous neuronal activity plays an important role in the development and plasticity of brain. To explore the developmental changes in the firing pattern of the neuronal networks in vitro, the hippocampal neurons were cultured on the multi-microelectrode array dish for over 14 weeks and the spontaneous activity was recorded. The results showed that random firing was observed in the 1st week and transformed into synchronized activity after two weeks, then tightly synchronized activity appeared in week 2 to 7 and finally the activities transformed into the random firing pattern. These results suggested three stages in the long-term development of neuronal network in vitro: the stage for connection, the stage of synchronized activity and the mature stage. 8Oxygenation,inflammatory response and lung injury during one lung ventilation in rabbits using inspired oxygen fraction of 0.6 vs.1.0显示文摘Maintaining adequate oxygenation during one-lung ventilation(OLV) requires high inspired oxygen fraction(FiO_2).However,high FiO_2 also causes inflammatory response and lung injury.Therefore,it remains a great interest to clinicians and scientists to optimize the care of patients undergoing OLV.The aim of this study was to determine and compare oxygenation,inflammatory response and lung injury during OLV in rabbits using FiO_2 of 0.6 vs.1.0.After 30 minutes of two-lung ventilation(TLV) as baseline,30 rabbits were randomly assigned to three groups receiving mechanical ventilation for 3 hours:the sham group,receiving TLV with 0.6 FiO_2;the 1.0 FiO_2 group,receiving OLV with 1.0 FiO_2;the 0.6 FiO_2 group,receiving OLV with 0.6 FiO_2.Pulse oximetry was continuously monitored and arterial blood gas analysis was intermittently conducted.Histopathologic study of lung tissues was performed and inflammatory cytokines and the mRNA and protein of nuclear factor kappa B(NF-κB) p65 were determined.Three of the 10 rabbits in the 0.6 FiO_2 group suffered hypoxemia,defined by pulse oximetric saturation(SpO_2) less than 90%.Partial pressure of oxygen(PaO_2),acute lung injury(ALI) score,myeloperoxidase(MPO),tumor necrosis factor-a(TNF-α),interleukin-6(IL-6),mRNA and protein of NF-kB p65 were lower in the 0.6 FiO_2group than in the 1.0 FiO_2 group.In conclusion,during OLV,if FiO_2 of 0.6 can be tolerated,lung injury associated with high FiO_2 can be minimized.Further study is needed to validate this finding in human subjects.Zeping Xu Lianbing Gu Qingming Bian Pengyi Li Lijun Wang Jingyuan Zhang Yanning Qian 2017The Journal of Biomedical Research2017,31,1:7
9Phase I study of CAR-T cells with PD-1 and TCR disruption in mesothelin-positive solid tumors显示文摘Programmed cell death protein-1(PD-1)-mediated immunosuppression has been proposed to contribute to the limited clinical efficacy of chimeric antigen receptor T(CAR-T)cells in solid tumors.We generated PD-1 and T cell receptor(TCR)deficient mesothelin-specific CAR-T(MPTK-CAR-T)cells using CRISPR-Cas9 technology and evaluated them in a dose-escalation study.A total of 15 patients received one or more infusions of MPTK-CAR-T cells without prior lymphodepletion.No dose-limiting toxicity or unexpected adverse events were observed in any of the 15 patients.The best overall response was stable disease(2/15 patients).Circulating MPTK-CAR-T cells peaked at days 7–14 and became undetectable beyond 1 month.TCR-positive CAR-T cells rather than TCR-negative CAR-T cells were predominantly detected in effusion or peripheral blood from three patients after infusion.We further confirmed the reduced persistence of TCR-deficient CAR-T cells in animal models.Our results establish the preliminary feasibility and safety of CRISPR-engineered CAR-T cells with PD-1 disruption and suggest that the natural TCR plays an important role in the persistence of CAR-T cells when treating solid tumors.Zhenguang Wang Na Li Kaichao Feng Meixia Chen Yan Zhang Yang Liu Qingming Yang Jing Nie Na Tang Xingying Zhang Chen Cheng Lianjun Shen Jiaping He Xun Ye Wei Cao Haoyi Wang Weidong Han 2021Cellular & Molecular Immunology2021,18,9:7
10Metastatic status of sentinel lymph nodes in breast cancer determined with photoacoustic microscopy via dual-targeting nanoparticles显示文摘Detection of sentinel lymph nodes(SLNs)is critical to guide the treatment of breast cancer.However,distinguishing metastatic SLNs from normal and inflamed lymph nodes(LNs)during surgical resection remains a challenge.Here,we report a CD44 and scavenger receptor class B1 dual-targeting hyaluronic acid nanoparticle(5K-HA-HPPS)loaded with the near-infra-red fluorescent dye DiR-BOA for SLN imaging in breast cancer.The small sized(~40 nm)self-assembled 5K-HA-HPPSs accumulated rapidly in the SLNs after intradermal injection.Compared with normal popliteal LNs(N-LN),there were ~3.2-fold and ~2.4-fold increases in fluorescence intensity in tumour metastatic SLNs(T-MLN)and inflamed LNs(Inf-LN),respectively,6 h after nanoparticle inoculation.More importantly,photoacoustic microscopy(PAM)of 5KHA-HPPS showed a significantly distinct distribution in T-MLN compared with N-LN and Inf-LN.Signals were mainly distributed at the centre of T-MLN but at the periphery of N-LN and Inf-LN.The ratio of PA intensity(R)at the centre of the LNs compared with that at the periphery was 5.93±0.75 for T-MLNs of the 5K-HA-HPPS group,which was much higher than that for the Inf-LNs(R=0.2±0.07)and N-LNs(R=0.45±0.09).These results suggest that 5K-HA-HPPS injection combined with PAM provides a powerful tool for distinguishing metastatic SLNs from pLNs and inflamed LNs,thus guiding the removal of SLNs during breast cancer surgery.Yanfeng Dai Xiang Yu Jianshuang Wei Fanxin Zeng Yiran Li Xiaoquan Yang Qingming Luo Zhihong Zhang 2020Light(Science & Applications)2020,9,1:6
11FluxExplorer: A general platform for modeling and analyses of metabolic net-works based on stoichiometry显示文摘Stoichiometry-based analyses of meta- bolic networks have aroused significant interest of systems biology researchers in recent years. It is necessary to develop a more convenient modeling platform on which users can reconstruct their network models using completely graphical operations, and explore them with powerful analyzing modules to get a better understanding of the properties of metabolic systems. Herein, an in silico platform, FluxExplorer, for metabolic modeling and analyses based on stoichiometry has been developed as a publicly available tool for systems biology research. This platform integrates various analytic approaches, in- cluding flux balance analysis, minimization of meta- bolic adjustment, extreme pathways analysis, shadow prices analysis, and singular value decom- position, providing a thorough characterization of the metabolic system. Using a graphic modeling process, metabolic networks can be reconstructed and modi- fied intuitively and conveniently. The inconsistencies of a model with respect to the FBA principles can be proved automatically. In addition, this platform sup- ports systems biology markup language (SBML). FluxExplorer has been applied to rebuild a metabolic network in mammalian mitochondria, producing meaningful results. 12An analytical biomarker for treatment of patients with recurrent B-ALL after remission induced by infusion of anti-CD19 chimeric antigen receptor T(CAR-T) cells显示文摘Anti-CD19 chimeric antigen receptor-modified T(CAR-T-19) cells have emerged as a powerful targeted immunotherapy for B-cell lineage acute lymphoblastic leukemia with a remarkable clinical response in recent trials. Nonetheless, few data are available on the subsequent clinical monitoring and treatment of the patients, especially those with disease recurrence after CAR-T-19 cell infusion. Here, we analyzed three patients who survived after our phase I clinical trial and who were studied by means of biomarkers reflecting persistence of CAR-T-19 cells in vivo and predictive factors directing further treatment. One patient achieved 9-week sustained complete remission and subsequently received an allogeneic hematopoietic stem cell transplant. Another patient who showed relapse after 20 weeks without detectable leukemia in the cerebrospinal fluid after CAR-T-19 cell treatment was able to achieve a morphological remission under the influence of stand-alone low-dose chemotherapeutic agents. The third patient gradually developed extensive extramedullary involvement in tissues with scarce immune-cell infiltration during a long period of hematopoietic remission after CAR-T-19 cell therapy. Long-term and discontinuous increases in serum cytokines(mainly interleukin 6 and C-reactive protein) were identified in two patients(Nos. 1 and 6) even though only a low copy number of CAR molecules could be detected in their peripheral blood. This finding was suggestive of persistent functional activity of CAR-T-19 cells. 13Monitor and control of neuronal activities with femtosecond pulse laser显示文摘Combined with the fluorescence labeling technique, two-photon microscopy excited with femtosecond pulse laser has become an important tool for neuroscience research. In this research, the calcium signals from neurons in rat cortex slice were monitored by a custom-built two-photon microscopy, and the spontaneous calcium signals and the pharmacological responses as well as the responses to femtosecond pulse laser stimulation were recorded. The results showed that the amplitude of the cal-cium signals increased in direct proportion to the corresponding electrical activities. Glutamate induced a calcium transient, but continuous application resulted in smaller response. Simultaneous monitoring of neuronal populations distinguished the neurons of different microcircuits. The femtosecond pulse laser induced local or global calcium signals in the pyramidal neurons. 15Attribution analysis based on Budyko hypothesis for land evapotranspiration change in the Loess Plateau, China显示文摘Land evapotranspiration(ET) is an important process connecting soil, vegetation and the atmosphere, especially in regions that experience shortage in precipitation.Since 1999, the implementation of a large-scale vegetation restoration project has significantly improved the ecological environment of the Loess Plateau in China.However, the quantitative assessment of the contribution of vegetation restoration projects to long-term ET is still in its infancy.In this study, we investigated changes in land ET and associated driving factors from 1982 to 2014 in the Loess Plateau using Budyko-based partial differential methods.Overall, annual ET slightly increased by 0.28 mm/a and there were no large fluctuations after project implementation.An attribution analysis showed that precipitation was the driving factor of inter-annual variability of land ET throughout the study period;the average impacts of precipitation, potential evapotranspiration, and vegetation restoration on ET change were 61.5%, 11.5% and 26.9%, respectively.These results provide an improved understanding of the relationship between vegetation condition change and climate variation on terrestrial ET in the study area and can support future decision-making regarding water resource availability.HE Guohua ZHAO Yong WANG Jianhua GAO Xuerui HE Fan LI Haihong ZHAI Jiaqi WANG Qingming ZHU Yongnan 2019Journal of Arid Land2019,11,6:5

17Deflagration-to-detonation transition process for spherical aluminum dust/epoxypropane mist/air mixtures in a large-scale experimental tube显示文摘The deflagration-to-detonation transitions (DDTs) for clouds of spherical aluminum dust (SAD) mixed with air or epoxypropane mist (EPM) and air were investigated in a 29.6-m-long experimental tube of 199 mm in diameter. The clouds formed through the injection of SAD and SAD/liquid epoxypropane samples into the experimental tube. Explosions of the SAD/air mixture were initiated using a 7-m-long EPM/air cloud explosion ignited by a 40-J electric spark. Explosions in SAD/EPM/air clouds were initiated using a 1.2-m EPM/air cloud explosion ignited by a 40-J electric spark initiated using a 40-J electric spark. Self-sustained detonation waves formed in SAD/EPM/air mixtures instead of in SAD/air mixtures. The stages and characteristics of the DDT process in SAD/air and SAD/EPM/air mixtures were studied and analyzed. Self-sustained detonation was evident from the existence of a transverse wave and a cellular structure. 18High-resolution mapping of brain vasculature and its impairment in the hippocampus of Alzheimer’s disease mice显示文摘Accumulating evidence indicates the critical importance of cerebrovascular dysfunction in the pathogenesis of Alzheimer’s disease(AD).However,systematic comparative studies on the precise brain vasculature of wild-type and AD model mice are still rare.Using an image-optimization method for analysing Micro-Optical Sectioning Tomography(MOST)data,we generated cross-scale whole-brain 3 D atlases that cover the entire vascular system from large vessels down to smallest capillaries at submicron resolution,for both wild-type mice and a transgenic(APP/PS1)mouse model of AD.In addition to distinct vascular patterns in different brain regions,we found that the main vessels of the molecular layer of the hippocampal dentate gyrus(DG-ml)undergo abrupt changes in both diameter and branch angle,spreading a unique comb-like pattern of capillaries.By using a quantitative analysis workflow,we identified in the hippocampus of AD mice an overall reduction of the mean vascular diameter,volume fraction and branch angle,with most significant impairment in the DG-ml.In addition,virtual endoscopy revealed irregular morphological features in the vessel lumen of the AD mice,potentially contributing to the impairment of blood flow.Our results demonstrate the capability of high-resolution cross-scale evaluation of brain vasculature and underscore the importance of studying hippocampal microcirculation for understanding AD pathogenesis.Xiaochuan Zhang Xianzhen Yin Jingjing Zhang Anan Li Hui Gong Qingming Luo Haiyan Zhang Zhaobing Gao Hualiang Jiang 2019National Science Review2019,6,6:4
19A digitally generated ultrafine optical frequency comb for spectral measurements with 0.01-pm resolution and 0.7-μs response time显示文摘Optical spectral measurements are crucial for optical sensors and many other applications,but the prevailing methods,such as optical spectrum analysis and tunable laser spectroscopy,often have to make compromises among resolution,speed,and accuracy.Optical frequency combs are widely used for metrology of discrete atomic and molecular spectral lines.However,they are usually generated by optical methods and have large comb spacing,which limits the resolution for direct sampling of continuous spectra.To overcome these problems,this paper presents an original method to digitally generate an ultrafine optical frequency comb(UFOFC)as the frequency ruler for spectral measurements.Each comb line provides one sampling point,and the full spectrum can be captured at the same time using coherent detection.For an experimental demonstration,we adopted the inverse fast Fourier transform to generate a UFOFC with a comb spacing of 1.46 MHz over a 10-GHz range and demonstrated its functions using a Mach–Zehnder refractive index sensor.The UFOFC obtains a spectral resolution of 0.01 pm and response time of 0.7 μs;both represent 100-fold improvements over the state of the art and could be further enhanced by several orders of magnitude.The UFOFC presented here could facilitate new label-free sensor applications that require both high resolution and fast speed,such as measuring binding kinetics and single-molecule dynamics.Yuan Bao Xingwen Yi Zhaohui Li Qingming Chen Jianping Li Xudong Fan Xuming Zhang 2015Light(Science & Applications)2015,4,1:4
20PINPOINT SOURCE LOCALIZATION FOR OCULAR NONSELECTIVE ATTENTION WITH COMBINATION OF ERP AND fNIRI MEASUREMENTS显示文摘Compared with event-related potential(ERP)which is widely used in psychology research,functional near-infrared imaging(fNIRI)is a new technique providing hemodynamic information related to brain activity,except for electrophysiological signals.Here,we use both these techniques to study ocular attention.We conducted a series of experiments with a classic paradigm of ocular nonselective attention,and monitored responses with fNIRI and ERP respectively.The results showed that fNIRI measured brain activations in the left prefrontal lobe,while ERPs showed activation in frontal lobe.More importantly,only with the combination measurements of fNIRI and ERP,we were then able to find the pinpoint source of ocular nonselective attention,which is in the left and upper corner in Brodmann area 10.These results demonstrated that fNIRI is a reliable technique in psychology,and the combination of fNIRI and ERP can be promising to reveal more information in the research of brain mechanism.TING LI LI LI PENG DU QINGMING LUO HUI GONG 2008Journal of Innovative Optical Health Sciences2008,1,2:3
