| 1 | Cationic conjugated polymers for homogeneous and sensitive fluorescence detection of hyaluronidase显示文摘The cationic charged water-soluble polyfluorenes containing 2,1,3-benzothiadiazole (BT) units (P1-3) have been synthesized and characterized. These polymers demonstrate intramolecular energy transfer from the fluorene units to the BT sites when oppositely charged hyaluronan is added due to the formation of electrostatic complexes, followed by a shift in emission color from blue to green or brown. Upon adding hyaluronidase, the hyaluronan is cleaved into fragments. The relatively weak electrostatic interactions of hyaluronan fragments with polyfluorenes keep their main chains separated and energy transfer from the fluorene units to the BT sites is inefficient, and the polyfluorenes recover their blue emissions. The complexes of conjugated polymers/hyaluronan can be utilized as probes for sensitive and facile fluorescence assays for hyaluronidase. Consequently, it is urgent to find new antibiotics with efficient broad-spectrum antimicrobial activity. By virtue of versatility in molecular design, a new peptide-like cell membrane-broken molecule, oligo-(7,7′-bifluoren-benzo[c][1,2,5]thiadiazole)(OFBT) possessing a conjugated backbone and eight pendant guanidyl moieties was designed and synthesized. OFBT exhibits favorable broad-spectrum of antimicrobial activity to pathogens including Gram-negative and Gram-positive bacteria, and fungi with the minimum inhibitory concentration(MIC)below 3.0 μM. | 8 | Multiplex detection of KRAS and BRAF mutations using cationic conjugated polymers显示文摘The mutation detections of KRAS and BRAF genes are of significant importance to predict the responses to anti-cancer therapy and develop new drugs. In this paper, we developed a multi-step fluorescence resonance energy transfer (FRET) assay for multiplex detection of KRAS and BRAF mutations using cationic conjugated polymers (CCP). The newly established detection system could detect as low as 2% mutant DNAs in DNA admixtures. By triggering the emission intensity change of CCP and the dyes labeled in the DNA, four possible statuses (three mutations and one wildtype) can be differentiated in one extension reaction. The detection efficiency of this new method in clinical molecular diagnosis was validated by determining KRAS and BRAF mutations of 51 formalin-fixed paraffin-embedded (FFPE) ovary tissue samples. Furthermore, the result of the CCP-based multi-step FRET assay can be directly visualized under UV light so that no expensive instruments and technical expertise are needed. | 17 | Synthesis of amphiphilic poly(fluorene) derivatives for selective imaging of Staphylococcus aureus显示文摘Three amphiphilic poly(fluorene-co-phenylene) derivatives with different side chains(PFP-1, PFP-2, PFP-3) were designed and synthesized for exploring their detection and imaging of pathogens. Upon incubation with six kinds of different pathogens, it was found the three polymers could selectively interact with Staphylococcus aureus(S. aureus). Their selective imaging towards S. aureus were thus realized. The selective imaging towards S. aureus was also confirmed even under the blend of microbes. PFP-3 shows stronger fluorescence imaging signal than PFP-1 and PFP-2. Zeta potential and isothermal titration microcalorimetry(ITC) tests demonstrated that both electrostatic interactions and hydrophobic interactions played important roles in the binding between PFPs and pathogens. This material was designed to facilitate the nucleic acid binding, encapsulation and efficient cellular uptake. PFP can effectively protect siRNA against nuclease degradation, which is necessary for gene carriers. PFP can be used for multidrug resistance gene-targeted siRNA delivery in doxorubicin (Dox)-resistant human breast cancer cells (MCF7) cells. As a siRNA transfection agent, PFP can efficiently achieve the reversal of drug resistance and enhance the drug sensitivity. | 19 | Selective biocompatibility and responsive imaging property of cationic conjugated polyelectrolyte to cancer cells显示文摘Optical imaging, as an important molecular imaging modality, has emerged many attractions in studying the biological or molecular events both in cell level and living subject because of its high resolution and sensitivity, noninvasive manner and low cost. Herein, we bring novel insights into a water-soluble conjugated polyelectlyte by deeply studying its properties in cells. Poly(9, 9-bis(6'-N,N,N-trimethylammonium hexyl)fluorene phenylene)(PFP), a good biosensing material, was studied in this paper. The biocompatibility of PFP was investigated in different cells, and cell cycle analysis was carried out to explore the reasons of different biocompatibility of PFP to cells. After irradiation, fluorescence enhancement of blue emission and turn-on of long-wavelength emission of PFP in HepG2 cells was observed, which was first reported as far as we know.
