1Toxicity of graphene oxide and multi-walled carbon nanotubes against human cells and zebrafish显示文摘Graphene possesses unique physical and chemical properties, which have inspired a wide range of potential biomedical applications. However, little is known about the adverse effects of graphene on the human body and ecological environment. The purpose of our work is to make assessment on the toxicity of graphene oxide (GO) against human cell line (human bone marrow neuroblastoma cell line and human epithelial carcinoma cell line) and zebrafish (Danio rerio) by comparing the toxic effects of GO with its sister, multi-walled carbon nanotubes (MWNTs). The results show that GO has a moderate toxicity to organisms since it can induce minor (about 20%) cell growth inhibition and slight hatching delay of zebrafish embryos at a dosage of 50 mg/L, but did not result in significant increase of apoptosis in embryo, while MWNTs exhibit acute toxicity leading to a strong inhibition of cell proliferation and serious morphological defects in developing embryos even at relatively low concentration of 25 mg/L. The distinctive toxicity of GO and MWNTs should be ascribed to the different models of interaction between nanomaterials and organisms, which arises from the different geometric structures of nanomaterials. 2Jasmonic acid carboxyl methyltransferase regulates development and herbivory-induced defense response in rice显示文摘Jasmonic acid(JA) and related metabolites play a key role in plant defense and growth. JA carboxyl methyltransferase(JMT) may be involved in plant defense and development by methylating JA to methyl jasmonate(Me JA) and thus influencing the concentrations of JA and related metabolites. However, no JMT gene has been well characterized in monocotyledon defense and development at the molecular level. After we cloned a rice JMT gene,Os JMT1, whose encoding protein was localized in the cytosol, we found that the recombinant Os JMT1 protein catalyzed JA to Me JA. Os JMT1 is up-regulated in response to infestation with the brown planthopper(BPH; Nilaparvata lugens). Plants in which Os JMT1 had been overexpressed(oeJMT plants) showed reduced height and yield. These oe-JMT plants also exhibited increased Me JA levels but reduced levels of herbivore-induced JA and jasmonoyl-isoleucine(JAIle). The oe-JMT plants were more attractive to BPH female adults but showed increased resistance to BPH nymphs,probably owing to the different responses of BPH female adults and nymphs to the changes in levels of H_2O_2 and Me JA in oe-JMT plants. 4The R2R3-MYB transcription factor GhMYB1a regulates flavonol and anthocyanin accumulation in Gerbera hybrida显示文摘Anthocyanins and flavonols have vital roles in flower coloration,plant development,and defense.Because anthocyanins and flavonols share the same subcellular localization and common biosynthetic substrates,these pathways may compete for substrates.However,the mechanism regulating this potential competition remains unclear.Here,we identified GhMYB1a,an R2R3-MYB transcription factor involved in the regulation of anthocyanin and flavonol accumulation in gerbera(Gerbera hybrida).GhMYB1a shares high sequence similarity with that of other characterized regulators of flavonol biosynthesis.In addition,GhMYB1a is also phylogenetically grouped with these proteins.The overexpression of GhMYB1a in gerbera and tobacco(Nicotiana tabacum)resulted in decreased anthocyanin accumulation and increased accumulation of flavonols by upregulating the structural genes involved in flavonol biosynthesis.We further found that GhMYB1a functions as a homodimer instead of interacting with basic helix-loop-helix cofactors.These results suggest that GhMYB1a is involved in regulating the anthocyanin and flavonol metabolic pathways through precise regulation of gene expression.The functional characterization of GhMYB1a provides insight into the biosynthesis and regulation of flavonols and anthocyanins.Chunmei Zhong Yi Tang Bin Pang Xukun Li Yuping Yang Jing Deng Chengyong Feng Lingfei Li Guiping Ren Yaqin Wang Jianzong Peng Shulan Sun Shan Liang Xiaojing Wang 2020Horticulture Research2020,7,1:7
5PbGA2ox8 induces vascular-related anthocyanin accumulation and contributes to red stripe formation on pear fruit显示文摘Fruit with stripes,which are generally longitudinal,can occur naturally,but the bioprocesses underlying this phenomenon are unclear.Previously,we observed an atypical anthocyanin distribution that caused red-striped fruit on the spontaneous pear bud sport“Red Zaosu”(Pyrus bretschneideri Rehd.).In this study,comparative transcriptome analysis of the sport and wild-type“Zaosu”revealed that this atypical anthocyanin accumulation was tightly correlated with abnormal overexpression of the gene-encoding gibberellin(GA)2-beta-dioxygenase 8,PbGA2ox8.Consistently,decreased methylation was also observed in the promoter region of PbGA2ox8 from“Red Zaosu”compared with“Zaosu”.Moreover,the GA levels in“Red Zaosu”seedlings were lower than those in“Zaosu”seedlings,and the application of exogenous GA4 reduced abnormal anthocyanin accumulation in“Red Zaosu”.Transient overexpression of PbGA2ox8 reduced the GA4 level and caused anthocyanin accumulation in pear fruit skin.Moreover,the presence of red stripes indicated anthocyanin accumulation in the hypanthial epidermal layer near vascular branches(VBs)in“Red Zaosu”.Transient overexpression of PbGA2ox8 resulting from vacuum infiltration induced anthocyanin accumulation preferentially in calcium-enriched areas near the vascular bundles in pear leaves.We propose a fruit-striping mechanism,in which the abnormal overexpression of PbGA2ox8 in“Red Zaosu”induces the formation of a longitudinal array of anthocyanin stripes near vascular bundles in fruit.Rui Zhai Zhigang Wang Chengquan Yang Kui Lin-Wang Richard Espley Jianlong Liu Xieyu Li Zhongying Wu Pengmin Li Qingmei Guan Fengwang Ma Lingfei Xu 2019Horticulture Research2019,6,1:5
6The interplay of aging,genetics and environmental factors in the pathogenesis of Parkinson’s disease显示文摘Background:Parkinson’s disease(PD)is characterized by dopaminergic neuronal loss in the substantia nigra pars compacta and intracellular inclusions called Lewy bodies(LB).During the course of disease,misfoldedα-synuclein,the major constituent of LB,spreads to different regions of the brain in a prion-like fashion,giving rise to successive non-motor and motor symptoms.Etiology is likely multifactorial,and involves interplay among aging,genetic susceptibility and environmental factors.Main body:The prevalence of PD rises exponentially with age,and aging is associated with impairment of cellular pathways which increases susceptibility of dopaminergic neurons to cell death.However,the majority of those over the age of 80 do not have PD,thus other factors in addition to aging are needed to cause disease.Discovery of neurotoxins which can result in parkinsonism led to efforts in identifying environmental factors which may influence PD risk.Nevertheless,the causality of most environmental factors is not conclusively established,and alternative explanations such as reverse causality and recall bias cannot be excluded.The lack of geographic clusters and conjugal cases also go against environmental toxins as a major cause of PD.Rare mutations as well as common variants in genes such as SNCA,LRRK2 and GBA are associated with risk of PD,but Mendelian causes collectively only account for 5%of PD and common polymorphisms are associated with small increase in PD risk.Heritability of PD has been estimated to be around 30%.Thus,aging,genetics and environmental factors each alone is rarely sufficient to cause PD for most patients.Conclusion:PD is a multifactorial disorder involving interplay of aging,genetics and environmental factors.This has implications on the development of appropriate animal models of PD which take all these factors into account.Common converging pathways likely include mitochondrial dysfunction,impaired autophagy,oxidative stress and neuroinflammation,which are associated with the accumulation and spread of misfoldedα-synuclein and neurodegeneration.Understanding the mechanisms involved in the initiation and progression of PD may lead to potential therapeutic targets to prevent PD or modify its course.Shirley Yin-Yu Pang Philip Wing-Lok Ho Hui-Fang Liu Chi-Ting Leung Lingfei Li Eunice Eun Seo Chang David Boyer Ramsden Shu-Leong Ho 2019Translational Neurodegeneration2019,8,1:5

8Using zebrafish as the model organism to understand organ regeneration显示文摘The limited regenerative capacity of several organs, such as central nervous system(CNS), heart and limb in mammals makes related major diseases quite difficult to recover. Therefore, dissection of the cellular and molecular mechanisms underlying organ regeneration is of great scientific and clinical interests. Tremendous progression has already been made after extensive investigations using several model organisms for decades. Unfortunately, distance to the final achievement of the goal still remains. Recently, zebrafish became a popular model organism for the deep understanding of regeneration based on its powerful regenerative capacity, in particular the organs that are limitedly regenerated in mammals. Additionally, zebrafish are endowed with other advantages good for the study of organ regeneration. This review summarizes the recent progress in the study of zebrafish organ regeneration, in particular regeneration of fin, heart, CNS, and liver as the representatives. 9Synthesis of novel β-amino ketones containing a p-aminobenzoic acid moiety and evaluation of their antidiabetic activities显示文摘The synthesis of two series of β-amino ketones containing a p-aminobenzoic acid moiety (TM-1 and TM-2) using a modified protocol of the Mannich reaction is reported. The molecular structures of a total of tweenty three new target compounds were characterized by 1H NMR, 13C NMR, ESI-MS and HR-MS. Subsequently, their antidiabetic activities were screened in vitro. The α-glucodase inhibition (α-GI) activity of compound 1e reached a remarkable level of 66.50%. The peroxisome proliferatoractivated receptor (PPAR) relative activation activities of six compounds are above 80%, and in particular 2i displays an unprecedentedly high PPAR of 130.91%. 10Apple SERRATE negatively mediates drought resistance by regulating MdMYB88 and MdMYB124 and microRNA biogenesis显示文摘The function of serrate(SE)in miRNA biogenesis in Arabidopsis is well elucidated,whereas its role in plant drought resistance is largely unknown.In this study,we report that MdSE acts as a negative regulator of apple(Malus×domestica)drought resistance by regulating the expression levels of MdMYB88 and MdMYB124 and miRNAs,including mdm-miR156,mdm-miR166,mdm-miR172,mdm-miR319,and mdm-miR399.MdSE interacts with MdMYB88 and MdMYB124,two positive regulators of apple drought resistance.MdSE decreases the transcript and protein levels of MdMYB88 and MdMYB124,which directly regulate the expression of MdNCED3,a key enzyme in abscisic acid(ABA)biosynthesis.Furthermore,MdSE is enriched in the same region of the MdNECD3 promoter where MdMYB88/MdMYB124 binds.Consistently,MdSE RNAi transgenic plants are more sensitive to ABA-induced stomatal closure,whereas MdSE OE plants are less sensitive.In addition,under drought stress,MdSE is responsible for the biogenesis of mdm-miR399,a negative regulator of drought resistance,and negatively regulates miRNAs,including mdm-miR156,mdm-miR166,mdm-miR172,and mdm-miR319,which are positive regulators of drought resistance.Taken together,by revealing the negative role of MdSE,our results broaden our understanding of the apple drought response and provide a candidate gene for apple drought improvement through molecular breeding.Xuewei Li Pengxiang Chen Yinpeng Xie Yan Yan Liping Wang Huan Dang Jing Zhang Lingfei Xu Fengwang Ma Qingmei Guan 2020Horticulture Research2020,7,1:4
11Linkage map and QTL mapping of red flesh locus in apple using a R1R1×R6R6 population显示文摘The red flesh in apple fruit is a desired trait by consumers and it is associated to the anthocyanin content,which is mainly controlled by MdMYB10 with a R6 promoter.In this study,a high-density linkage group was constructed using the‘Fuji’x‘Red3’population which contained homozygous alleles R1R1 and R6R6,respectively.The linkage group consists of 7630 SNPs along 17 linkage groups,spanning 2270.21 cM,with an average density of 0.30 cM permarker.The cyanidin-3-galactoside concentration was used as the phenotypic data in QTL analysis.Moreover,one QTL peak which was flaked by two markers,marker2187260 to marker2173766,with LOD scores of 4.49 was detected.This QTL ranged from 0 to 40.79 cM on the top of linkage group(LG16).In addition one candidate molecular marker(marker2175442)in this QTL was identified,which was significant correlated with the flesh cyanidin-3-galactoside concentration.These genetic findings enrich the breeding basis of fruit flesh coloration in apple.Chengquan Yang Guangya Sha Tao Wei Baiquan Ma Cuiying Li Pengmin Li Yangjun Zou Lingfei Xu Fengwang Ma 2021Horticultural Plant Journal2021,7,5:4
12The role of China's terrestrial carbon sequestration 2010-2060 in offsetting energy-related CO_(2) emissions显示文摘Energy consumption dominates annual CO_(2) emissions in China.It is essential to significantly reduce CO_(2) emissions from energy consumption to reach national carbon neutrality by 2060,while the role of terrestrial carbon sequestration in offsetting energy-related CO_(2) emissions cannot be underestimated.Natural climate solutions(NCS),including improvements in terrestrial carbon sequestration,represent readily deployable options to offset anthropogenic greenhouse gas emissions.However,the extent to which China’s terrestrial carbon sequestration in the future,especially when target-oriented managements(TOMs)are implemented,can help to mitigate energy-related CO_(2) emissions is far from certain.By synthesizing available findings and using several parameter-sparse empirical models that have been calibrated and/or fitted against contemporary measurements,we assessed China’s terrestrial carbon sequestration over 2010-2060 and its contribution to offsetting national energy-related CO_(2) emissions.We show that terrestrial C sequestration in China will increase from 0.375±0.056(mean±standard deviation)Pg C yr^(-1) in the 2010s to 0.458±0.100 Pg C yr^(-1)under RCP2.6 and 0.493±0.108 Pg C yr^(-1) under the RCP4.5 scenario in the 2050s,when TOMs are implemented.The majority of carbon sequestration comes from forest,accounting for 67.8-71.4%of the total amount.China’s terrestrial ecosystems can offset 12.2-15.0%and 13.4-17.8%of energy-related peak CO_(2) emissions in 2030 and 2060,respectively.The implementation of TOMs contributes 11.9%of the overall terrestrial carbon sequestration in the 2020s and 23.7%in the 2050s.The most likely strategy to maximize future NCS effectiveness is a full implementation of all applicable cost-effective NCS pathways in China.Our findings highlight the role of terrestrial carbon sequestration in offsetting energy-related CO_(2) emissions and put forward future needs in the context of carbon neutrality.Yao Huang Wenjuan Sun Zhangcai Qin Wen Zhang Yongqiang Yu Tingting Li Qing Zhang Guocheng Wang Lingfei Yu Yijie Wang Fan Ding Ping Zhang 2022National Science Review2022,9,8:4

14Assessment of the risk of infectious aerosols leaking to the environment from BSL-3 laboratory HEPA air filtration systems using model bacterial aerosols显示文摘To assess the risk of infectious bacterial aerosols leaking to the environment, the filtration efficiency of a biosafety level 3(BSL-3) laboratory high-efficiency particulate(HEPA) filter was investigated using the aerosolized bacteria Serratia marcescens. The aerosol size was measured using an Andersen sampler. Eight first stage HEPA filters(numbered 1-8) were distributed in contaminated labs and exhausts from each0f the first stage HEPA filters were aggregated and filtered through one second stage HEPA filter before being released to the environment. In total, 8 first-stage and 1 second-stage HEPA filters from the BSL-3 air purification system were analyzed. No S. marcescens was detected in first stage filters 1,2,4,5,7 and 8 and the second stage HEPA filter. The filtration efficiencies against aerosolized S. marcescens were >99.9999%.First stage filter numbers 3 and 6 had filtration efficiencies of 99.9825% and 99.9906%, respectively. When filter number 3 was replaced by a new filter and the bracket for filter number 6 was sealed, no aerosolized S. marcescens was detected in the filtered air. 15Zebrafish hhex-null mutant develops an intrahepatic intestinal tube due to de-repression of cdx1b and pdx1显示文摘The hepatopancreatic duct (HPD) system links the liver and pancreas to the intestinal tube and is composed of the extrahepatic biliary duct, gallbladder, and pancreatic duct. Haematopoietically expressed-homeobox (Hhex) protein plays an essential role in the establishment of HPD;however, the molecular mechanism remains elusive. Here, we show that zebrafish hhex-null mutants fail to develop the HPD system characterized by lacking the biliary marker Annexin A4 and the HPD marker sox9b. The hepatobiliary duct part of the mutant HPD system is replaced by an intrahepatic intestinal tube characterized by expressing the intestinal marker fatty acid-binding protein 2a (fabp2a). Cell lineage analysis showed that this intrahepatic intestinal tube is not originated from hepatocytes or cholangiocytes. Further analysis revealed that cdx1b and pdx1 are expressed ectopically in the intrahepatic intestinal tube and knockdown of cdx1b and pdx1 could restore the expression of sox9b in the mutant. Chromatin-immunoprecipitation analysis showed that Hhex binds to the promoters of pdx1 and cdx1b genes to repress their expression. 16Prevalence of short stature among children in China:A systematic review显示文摘Importance The prevalence and characteristics of short stature(SS)among children in China should be assessed to provide guidance for planning and implementation of nationwide public health policies.Thus far,there have been no accurate estimates of the prevalence of SS in China.Objective To analyze the prevalence of SS among children in China and to explore the influences of sex,area,age,study year,and study site on prevalence rates.Methods Relevant literature was identified by searching the following databases:PubMed,Embase,The Cochrane Library,Chinese Biomedical Literature,China Knowledge Resource Integrated,WeiPu,and WanFang databases.Meta-analysis was carried out using STATA 11.2.Results This meta-analysis included 39 studies with 348326 Chinese participants;the studies covered 20 provinces,municipalities,and autonomous regions.The pooled prevalence of SS was 3.2%(95%confidence interval[CI],2.6%–3.7%;I2=99.8%).The prevalence of SS in boys and girls were 3.1%(95%CI,2.5%–3.7%)and 3.2%(95%CI,2.6%–3.9%),respectively.The sex difference was not statistically significant(P>0.05).The prevalence of SS was higher in rural areas than in urban areas(4.7%[95%CI,3.6%–5.8%]vs.2.8%[95%CI,2.2%–3.4%];P<0.001).The prevalence of SS was higher in West China(5.2%;95%CI,4.4%–6.0%)than in Northeast China(0.6%;95%CI,0.3%–0.8%),East China(2.3%;95%CI,1.9%–2.8%),or Central China(2.9%;95%CI,1.9%–3.9%).Interpretation The prevalence of SS among children was higher in western and rural areas of China.Close attention to children’s growth and development is needed to prevent the occurrence of SS.Fulun Li Ke Liu Qianlong Zhao Junyi Chen Lingfei Liu Qingmu Xie Jing Yang 2021Pediatric Investigation2021,5,2:2

19Cardiac proteomics reveals the potential mechanism of microtubule associated protein 4 phosphorylation-induced mitochondrial dysfunction显示文摘Background:Our previous work suggested that microtubule associated protein 4(MAP4)phosphorylation led to mitochondrial dysfunction in MAP4 phosphorylation mutant mice with cardiomyopathy,but the detailed mechanism was still unknown.Thus,the aim of this study was to investigate the potential mechanism involved in mitochondrial dysfunction responsible for cardiomyopathy.Methods:The present study was conducted to explore the potential mechanism underlying the mitochondrial dysfunction driven by MAP4 phosphorylation.Strain of mouse that mimicked constant MAP4 phosphorylation(S737 and S760)was generated.The isobaric tag for relative and absolute quantitation(iTRAQ)analysis was applied to the heart tissue.Gene Ontology(GO),Kyoto Encyclopedia of Genes and Genomes(KEGG),and protein-protein interaction(PPI)were all analyzed on the basis of differential expressed proteins(DEPs).Results:Among the 72 cardiac DEPs detected between the two genotypes of mice,12 were upregulated and 60 were downregulated.GO analysis showed the biological process,molecular function,and cellular component of DEPs,and KEGG enrichment analysis linked DEPs to 96 different biochemical pathways.In addition,the PPI network was also extended on the basis of DEPs as the seed proteins.Three proteins,including mitochondrial ubiquitin ligase activator of NF-κB 1,reduced form of nicotinamide adenine dinucleotide(NADH)-ubiquinone oxidoreductase 75 kDa subunit,mitochondrial and growth arrest,and DNA-damage-inducible proteins-interacting protein 1,which play an important role in the regulation of mitochondrial function,may correlate with MAP4 phosphorylationinduced mitochondrial dysfunction.Western blot was used to validate the expression of the three proteins,which was consistent with iTRAQ experiments.Conclusions:These findings revealed that the DEPs caused by MAP4 phosphorylation in heart tissue using iTRAQ technique and may provide clues to uncover the potential mechanism of MAP4 phosphorylation-induced mitochondrial dysfunction.Lingfei Li Junhui Zhang Qiong Zhang Yuesheng Huang Jiongyu Hu 2019Burns & Trauma2019,7,1:1
