| 3 | Serum uric acid and risk of incident diabetes in middle-aged and elderly Chinese adults: prospective cohort study显示文摘The association between serum uric acid and the risk of incident diabetes in Chinese adults remains unknown.This study aimed to investigate this association in a community-dwelling population aged≥40 years in Shanghai,China.Oral glucose tole3rance test was conducted during baseline and follow-up visits.Relative risk regression was utilized to examine the associations between baseline gender-specific serum uric acid levels and incident diabetes risk.A total of 613(10.3%)incident diabetes cases were identified during the follow-up visit after 4.5 years.Fasting plasma glucose,postload glucose,and glycated hemoglobin A1c during the follow-up visit progressively increased across the sex-specific quartiles of serum uric acid(all Ps<0.05).The incidence rate of diabetes increased across the quartiles of serum uric acid(7.43%,8.77%,11.47%,and 13.43%).Multivariate adjusted regression analysis revealed that individuals in the highest quartile had 1.36-fold increased risk of diabetes compared with those in the lowest quartile of serum uric acid(odds ratio(95%confidence interval)=1.36(1.06−1.73)).Stratified analysis indicated that the association was only observed in women.Accordingly,serum uric acid was associated with the increased risk of incident diabetes among middle-aged and elderly Chinese women. | Di Cheng Chunyan Hu Rui Du Hongyan Qi Lin Lin Xueyan Wu Lina Ma Kui Peng Mian Li Min Xu Yu Xu Yufang Bi Weiqing Wang Yuhong Chen Jieli Lu | 2020 | Frontiers of Medicine2020,14,6: | 3 |

| 10 | NAT10 promotes cell proliferation by acetylating CEP170 mRNA to enhance translation efficiency in multiple myeloma显示文摘Multiple myeloma(MM) is still an incurable hematologic malignancy, which is eagerly to the discovery of novel therapeutic targets and methods. N-acetyltransferase 10(NAT10) is the first reported regulator of mRNA acetylation that is activated in many cancers. However, the function of NAT10 in MM remains unclear. We found significant upregulation of NAT10 in MM patients compared to normal plasma cells, which was also highly correlated with MM poor outcome. Further enforced NAT10 expression promoted MM growth in vitro and in vivo, while knockdown of NAT10 reversed those effects. The correlation analysis of acetylated RNA immunoprecipitation sequencing(ac RIP-seq) and ribosome profiling sequencing(Ribo-seq) combined with RIP-PCR tests identified centrosomal protein 170(CEP170) as an important downstream target of NAT10. Interfering CEP170 expression in NAT10-OE cells attenuated the acceleration of cellular growth caused by elevated NAT10. Moreover,CEP170 overexpression promoted cellular proliferation and chromosomal instability(CIN) in MM.Intriguingly, remodelin, a selective NAT10 inhibitor, suppressed MM cellular growth, induced cellular apoptosis in vitro and prolonged the survival of 5TMM3VT mice in vivo. We screened 74 infants and randomizedand analyzed results in 20 infants [9 receivingLGG (LGG+) and 11 not receiving LGG (LGG-)]. LGG wasincorporated in the formula (Nutramigen?) (minimum of3 × 10^7 CFU/d) in the LGG+ group. Fecal microbiota andinflammatory biomarkers, including fecal calprotectin(FC), plasma cytokines, circulating regulatory T cells(Tregs), and crying + fussing time were analyzed todetermine optimal time points and effect sizes for alarger trial.RESULTS: Recruitment in this population was slow, withabout 66% of eligible infants willing to enroll; subjectretention was better (75%). These rates were influencedby parents' reluctance to volunteer their infant for aclinical trial and by their tendency to change formulas.The maximal difference of crying + fussing time wasobserved at day 14, comparing the 2 groups, with amean difference of -91 (95%CI: -76, 259) min (P = NS).FC showed no significant difference, but the optimaltime to determine a potential effect was at day 90 [witha mean difference of 121 (95%CI: -48, 291) μg/g stool],observing a lower level of FC in the LGG+ group. Thefecal microbial communities were chaotic, as determinedby Shannon's diversity index and not apparently influencedby the probiotic. Pure 1,2-aceanthryenedione, which is used as intermediate of functional aromatic polymer material, was obtained by recrystalling the reaction mixture with aether and was determined by GC/MS, 1HNMR and FTIR analysis. The influences of various parameters, such as the contents of AlCl3 in [Emim]C1-A1C13, the amount of acylation agent, amount of [Emim]C1-A1C13, reaction temperature and reaction time were investigated. The optimum conditions were as follows: the molar fraction of A1C13 in ionic liquid [x(AlCl3)] being 0.67 , molar ratio of ionic liquid to anthracene being 2:1 , molar ratio of oxalyl chloride to anthracene being 2:1, reaction temperature being 40℃ and reaction time being 6h. Under above conditions, the yield and selectivity of 1,2-aceanthrylenedione can reach 91.5% and 98.3% respectively. Pure 3,3’-biacenaphthene was obtained by recrystalling and column chromatography from the reaction mixture and was determined by GC/MS, 1HNMR and FTIR analysis. The influence of various reaction conditions on the yield of 3,3’-biacenaphthene were studied by GC analysis. The result shows that the optimum synthesis conditions of the coupling reaction are as following: the molar ratio of FeCl3 to [Bmim]Cl being 3, the mole ratio of FeCl3 in [Bmim]Cl/FeCl3 to acenaphthene being 4, the reaction temperature being 20 ℃, the reaction time being 4h and the solvent of the reaction system being PhNO2. Under those conditions, the yield of the 3,3’-biacenaphthene will be 48.71 % and selectivity of that will be 78.56 %. The structure of 1,2-aceanthrylenedione was identified by measure of melting point, GC/MS, FTIR and 1HNMR analyses. The influences of various parameters, such as solvent, dosage of catalyst, molar ratio of anthracene to oxalyl chloride, reaction temperature, and reaction time were studied. The anhydrous AlCl3 was found to catalyze the reaction with high yeild and good selectivity.
