1A Resource for Inactivation of MicroRNAs Using Short Tandem Target Mimic Technology in Model and Crop Plants显示文摘microRNAs (miRNAs)are endogenous small non-coding RNAs that bind to mRNAs and target them for cleavage and/or translational repression,leading to gene silencing.We previously developed short tandem target mimic (STTM)technology to deactivate endogenous miRNAs in Arabidopsis.Here,we created hundreds of STTMs that target both conserved and species-specific miRNAs in Arabidopsis,tomato,rice,and maize,providing a resource for the functional interrogation of miRNAs.We not only revealed the functions of several miRNAs in plant development,but also demonstrated that tissue-specific inactivation of a few miRNAs in rice leads to an increase in grain size without adversely affecting overall plant growth and development.RNA-seq and small RNAseq analyses of STTM156/157 and STTM165/166 transgenic plants revealed the roles of these miRNAs in plant hormone biosynthesis and activation,secondary metabolism,and ion-channel activity-associated electrophysiology,demonstrating that STTM technology is an effective approach for studying miRNA functions.To facilitate the study and application of STTM transgenic plants and to provide a useful platform for storing and sharing of information about miRNA-regulated gene networks,we have established an online Genome Browser (http://gffzzc750bc0108fa4044suxqfqb6ou0wp6wbw.ffgz.tsg.suse.edu.cn/designindex2.php) to display the transcriptomic and miRNAomic changes in STTMinduced miRNA knockdown plants.Ting Peng Mengmeng Qiao Haiping Liu Sachin Teotia Zhanhui Zhang Yafan Zhao Bobo Wang Dongjie Zhao Lina Shi Cui Zhang Brandon Le Kestrel Rogers Chathura Gunasekara Haitang Duan Yiyou Gu Lei Tian Jinfu Nie Jian Qi Fanrong Meng Lan Huang Qinghui Chen Zhenlin Wang Jinshan Tang Xiaoqing Tang Ting Lan Xuemei Chen Hairong Wei Quanzhi Zhao Guiliang Tang 2018Molecular Plant2018,11,11:7
2Regulative effect of Taohong Siwu decoction(桃红四物汤) on extracellular matrix of endometrium in drug-induced abortion显示文摘OBJECTIVE: To explore the effects of Taohong Siwu decoction(桃红四物汤, THSWD) on the extracellular matrix of endometrium in rats following drug-induced abortion.METHODS: Thirty-six pregnant female rats were administered mifepristone and misoprostol to induce abortion, and amounts of uterine bleeding were recorded. Pathological damage and collagen accumulation were detected by hematoxylin-eosin staining and Masson’s trichrome staining in uterus, respectively. Myeloperoxidase was evaluated by immunohistochemistry.The expression levels of fibronectin, laminin, matrix metalloproteinase 9(MMP-9), and tissue inhibitor of metalloproteinase 1(TIMP-1) were quantified using western blotting.RESULTS: THSWD could promote endometrial protection in rats following drug-induced abortion.The contents of cellulose and myeloperoxidase were significantly decreased in uterine tissue of THSWD-treated groups. Moreover, THSWD significantly decreased the expression levels of fibronectin, laminin, and TIMP-1. THSWD also significantly increased MMP-9 expression and the MMP-9/TIMP-1 ratio.CONCLUSION: THSWD plays a critical role in endometrial protection by reducing extracellular matrix deposition and uterine fibrosis. 3Curcumin alleviates TGF-β1-induced fibrosis in NRK-49F cells via suppression of miR-21 expression,and regulation of the TGF-β1/smad3 signaling pathway显示文摘Objective:Overexpression of microRNA-21(miR-21)has been well-recognized during fibrosis in a wide range of tissue types through regulation of fibrogenesis via diverse pathways.Curcumin exhibits antifibrotic effects in various organs.We and others have previously demonstrated that curcumin has positive effects on the protection against fibrosis.However,the effect of curcumin on miR-21 expression has not been reported.Methods:In this study,transforming growth factor-b1(TGF-b1)-stimulated rat renal fibroblast cells(NRK-49F)were transfected with miR-21 inhibitors,then the expression of Smad3,a-smooth muscle actin(a-SMA),type I collagen(COL1A1),and type III collagen(COL3A1)was determined using quantitative real-time-PCR(qRT-PCR)and western blot analysis.The effect of curcumin on production of the extracellular matrix was evaluated using immunofluorescence,qRT-PCR,and western blot analysis.Results:TGF-b1 stimulation upregulated the expression of miR-21 and induced fibrogenesis in NRK-49F cells.Transfection with miR-21 inhibitors selectively decreased smad3 activity,and noticeably reduced the expression of a-SMA,COL1A1,and COL3A1.Curcumin treatment significantly inhibited the expressions of smad3,a-SMA,COL1A1,COL3A1,as well as miR-21 expression in NRK-49F cells in a dosedependent manner.Conclusion:Curcumin exerted its anti-fibrotic effects by targeting the TGF-b1/smad3 signaling pathway and suppressing miR-21 expression,thereby provides novel insight in the protective effects of curcumin against fibrosis in various organs.Lan Zhang Yi Liu Xiting Wang Cheng Li Mengmeng Shen Liming Di Shuangshuang He Yu Li 2020Journal of Traditional Chinese Medical Sciences2020,7,1:2

5GHM-FKNN:a generalized Heronian mean based fuzzy k-nearest neighbor classifier for the stock trend prediction显示文摘Stock trend prediction is a challenging problem because it involves many variables.Aiming at the problem that some existing machine learning techniques, such as random forest(RF), probabilistic random forest(PRF), k-nearest neighbor(KNN), and fuzzy KNN(FKNN), have difficulty in accurately predicting the stock trend(uptrend or downtrend) for a given date, a generalized Heronian mean(GHM) based FKNN predictor named GHM-FKNN was proposed.GHM-FKNN combines GHM aggregation function with the ideas of the classical FKNN approach.After evaluation, the comparison results elucidated that GHM-FKNN outperformed the other best existing methods RF, PRF, KNN and FKNN on independent test datasets corresponding to three stocks, namely AAPL, AMZN and NFLX.Compared with RF, PRF, KNN and FKNN, GHM-FKNN achieved the best performance with accuracy of 62.37% for AAPL, 58.25% for AMZN, and 64.10% for NFLX.吴振峰 WANG Mengmeng LAN Tian ZHANG Anyuan 2023High Technology Letters2023,29,2:0
6Ultrafast Laser Fabrication of Novel Fiber Mach-Zehnder Interferometer Sensors and Its Cost-Effective Alternative Manufacturing Methods (Invited Paper)显示文摘The recent progresses of fiber sensor fabrication in our group are reviewed.Novel inline fiber Mach-Zehnder interferometer(MZI) sensors with various structures are proposed and manufactured by femtosecond laser fabrication and fusion splicing for high-quality sensing of refractivity-sensitive parameters such as temperature,concentration,humidity,pressure,stress and strain: a) for an MZI sensor with a trench on a single-mode fiber,the refractive index(RI) sensitivity of acetone vapor is about 104 nm/RIU(refractive index unit) and the temperature sensitivity is 51.5 pm/℃ from 200 to 875 ℃;b) For an MZI consisting of two micro-air-cavities,the sensitivity is 501.5 nm/RIU and the detection limit is 1.994×10-6 RIU at the refractive index of 1.4;c) to reduce the fabrication cost,a new fusion-splicing based method is proposed to fabricate MZI sensors;the sensitivity is 664.57 nm/RIU with a detection limit of 1.5×10-6 RIU and its cost is tens of times cheaper than those of commercialized long period fiber Gratings;Also,5×10-5 acetone vapors are successfully detected by the MZI sensors coated with zeolite thin films.Wang Sumei Jiang Lan Li Benye Zhao Longjiang Yang Jinpeng Wang Mengmeng Xiao Hai Lu Yongfeng Hai-Lung Tsai 2011中国激光2011,38,6:0
7Parallel evolution of twoAIM24 protein subfamilies and their conserved functions in ER stress tolerance in land plants显示文摘Despite decades of efforts in genome sequencing and functional characterization,some important protein families remain poorly understood.In this study,we report the classification,evolution,and functions of the largely uncharacterized AIM24 protein family in plants,including the identification of a novel subfamily.We show that two AIM24 subfamilies(AIM24-A and AIM24-B)are commonly distributed in major plant groups.These two subfamilies not only have modest sequence similarities and different gene structures but also are of independent bacterial ancestry.We performed comparative functional investigations on the two AIM24 subfamilies using three model plants:the moss Physcomitrium patens,the liverwort Marchantia polymorpha,and the flowering plant Arabidopsis thaliana.Intriguingly,despite their significant differences in sequence and gene structure,both AIM24 subfamilies are involved in ER stress tolerance and the unfolded protein response(UPR).In addition,transformation of the AIM24-A gene from P.patens into the AIM24-B null mutant of A.thaliana could at least partially rescue ER stress tolerance and the UPR.We also discuss the role of AIM24 genes in plant development and other cellular activities.This study provides a unique example of parallel evolution in molecular functions and can serve as a foundation for further investigation of the AIM24 family in plants.Yanlong Guan Guanxiao Chang Jinjie Zhao Qia Wang Jiali Qin Mengmeng Tang Shuanghua Wang Lan Ma Jianchao Ma Guiling Sun Yun Zhou Jinling Huang 2023Plant Communications2023,4,3:0
8Unprecedent green macroalgae bloom:mechanism and implication to disaster prediction and prevention显示文摘Green macroalgae bloom(GMB),with the dominant species of Ulva prolifera,has regularly occurred since 2007 along the China coast.Although disaster prevention and control achieved favorable results in 2020,the satellite-observed GMB annual maximum coverage(AMC)rebounded sharply in 2021 to an unprecedented level.The reasons for this rebound and the significant interannual variability over past 15 years are still open questions.Here,by using long-term time-series(2007-2022)optical and Synthetic Aperture Radar satellite observations(1000+scenes),meteorological data and water quality statistics,the mechanism analysis was performed by exploring effects from natural factors and human activities.Two key determinants for AMC are successfully identified from numerous potential factors which are the macroalgae distribution in a key area(the Subei Shoal)during a critical period(from April to May 20)and the nutrient availability.Furthermore,by using these two parameters,a novel model for AMC prediction(R^(2)=0.87,p<0.01)is proposed and independently validated,which can reasonably explain the significant interannual variability(2014-2021)and agree well with the latest observation in 2022(percentage difference 12%).Finally,suggestions are proposed for future disaster prevention and alleviation.This work may aid future bloom prediction and management measure optimization.Mengmeng Cao Xuyan Li Tingwei Cui Xinliang Pan Yan Li Yanlong Chen Ning Wang Yanfang Xiao Xingai Song Yuzhu Xu Runa A Bing Mu Song Qing Rongjie Liu Wenjing Zhao Yuhai Bao Jie Zhang Lan Wei 2023International Journal of Digital Earth2023,16,1:0
9Stem cell microencapsulation maintains stemness in inflammatory microenvironment显示文摘Maintaining the stemness of the transplanted stem cell spheroids in an inflammatory microenvironment is challenging but important in regenerative medicine. Direct delivery of stem cells to repair periodontal defects may yield suboptimal effects due to the complexity of the periodontal inflammatory environment. Herein, stem cell spheroid is encapsulated by interfacial assembly of metal-phenolic network(MPN) nanofilm to form a stem cell microsphere capsule. Specifically, periodontal ligament stem cells(PDLSCs) spheroid was coated with FeⅢ/tannic acid coordination network to obtain spheroid@[FeⅢ-TA] microcapsules. The formed biodegradable MPN biointerface acted as a cytoprotective barrier and exhibited antioxidative, antibacterial and anti-inflammatory activities, effectively remodeling the inflammatory microenvironment and maintaining the stemness of PDLSCs.
