1BMAL1 knockout macaque monkeys display reduced sleep and psychiatric disorders显示文摘Circadian disruption is a risk factor for metabolic, psychiatric and age-related disorders, and non-human primate models could help to develop therapeutic treatments. Here, we report the generation of BMAL1 knockout cynomolgus monkeys for circadian-related disorders by CRISPR/Cas9 editing of monkey embryos. These monkeys showed higher nocturnal locomotion and reduced sleep, which was further exacerbated by a constant light regimen. Physiological circadian disruption was reflected by the markedly dampened and arrhythmic blood hormonal levels. Furthermore, BMAL1-deficient monkeys exhibited anxiety and depression, consistent with their stably elevated blood cortisol, and defective sensory processing in auditory oddball tests found in schizophrenia patients. Ablation of BMAL1 up-regulated transcriptional programs toward inflammatory and stress responses, with transcription networks associated with human sleep deprivation, major depressive disorders, and aging. 2Commissioning progress of the FAST显示文摘The Five-hundred-meter Aperture Spherical radio Telescope(FAST) was completed with its main structure installed on September 25, 2016, after which it entered the commissioning phase. This paper aims to introduce the commissioning progress of the FAST over the past two years. To improve its operational reliability and ensure effective observation time, FAST has been equipped with a real-time information system for the active reflector system and hierarchical commissioning scheme for the feed support system, which ultimately achieves safe operation of the two systems. For meeting the high-performance indices, a highprecision measurement system was set up based on the effective control methods that were implemented for the active reflector system and feed support system. Since the commissioning of the FAST, a low-frequency ultra-wideband receiver and 19-beam1.05-1.45 GHz receiver have been mainly used. Telescope efficiency, pointing accuracy, and system noise temperature were completely tested and ultimately achieved the acceptance indices of the telescope. The FAST has been in the process of national acceptance preparations and has begun to search for pulsars. 3Postoperative intussusception in infants and children:a report of seven cases显示文摘Postoperative intussusception is an uncommon but serious condition in infants and children.Here,we report seven cases of postoperative intussusception in infants and children who were seen at our institution over the last 13 y.The patients showed increased nasogastric drainage,vomiting,lack of stool,and/or growing abdominal distension 2 to 9 d following abdominal surgery.Manual reduction was successful in five cases.In two cases,necro-sis was found and intestinal resection and anastomosis were carried out.No recurrence was observed at six months of follow-up.Postoperative intussusception should be suspected in pediatric surgical patients who showed signs of intestinal obstruction in the early postoperative period.Weiwei Jiang Weibing Tang Qiming Geng Xiaoqun Xu 2012The Journal of Biomedical Research2012,26,1:3

5Chronic hypoperfusion due to intracranial large artery stenosis is not associated with cerebralβ-amyloid deposition and brain atrophy显示文摘Background:Insufficient cerebral perfusion is suggested to play a role in the development of Alzheimer disease(AD).However,there is a lack of direct evidence indicating whether hypoperfusion causes or aggravates AD pathology.We investigated the effect of chronic cerebral hypoperfusion on AD-related pathology in humans.Methods:We enrolled a group of cognitively normal patients(median age:64 years)with unilateral chronic cerebral hypoperfusion.Regions of interest with the most pronounced hypoperfusion changes were chosen in the hypoperfused region and were then mirrored in the contralateral hemisphere to create a control region with normal perfusion.11C-Pittsburgh compound-positron emission tomography standard uptake ratios and brain atrophy indices were calculated from the computed tomography images of each patient.Results:The median age of the 10 participants,consisting of 4 males and 6 females,was 64 years(47-76 years).We found that there were no differences in standard uptake ratios of the cortex(volume of interest[VOI]:P=0.721,region of interest[ROI]:P=0.241)and grey/white ratio(VOI:P=0.333,ROI:P=0.445)and brain atrophy indices(Bicaudate,Bifrontal,Evans,Cella,Cella media,and Ventricular index,P>0.05)between the hypoperfused regions and contralateral normally perfused regions in patients with unilateral chronic cerebral hypoperfusion.Conclusion:Our findings suggest that chronic hypoperfusion due to large vessel stenosis may not directly induce cerebralβ-amyloid deposition and neurodegeneration in humans.Dongyu Fan Huiyun Li Dongwan Chen Yang Chen Xu Yi Heng Yang Qianqian Shi Fangyang Jiao Yi Tang Qiming Li Fangyang Wang Shunan Wang Rongbing Jin Fan Zeng Yanjiang Wang 2022Chinese Medical Journal2022,,5:1

9Evolutionary Topology Optimization for Heat Conduction Fields显示文摘An effective evolutionary method for solving the structural topology design problems of heat conductive fields is presented in this paper.The topology optimization model based on minimizing the heat transport potential capacity dissipation of heat conductive field is then established and the corresponding sensitivity of objective function is derived to determine which elements would be removed of the heat conductive field for having the increment of the objective heat transport potential capacity dissipation minimized.A Filtering technique is employed in sensitivity field to eliminate numerical instabilities in the evolutionary procedure. 11USP9x promotes CD8^(+)T-cell dysfunction in association with autophagy inhibition in septic liver injury显示文摘Sepsis is a life-threatening condition manifested by concurrent inflammation and immunosuppression.Ubiquitin-specific peptidase 9,X-linked(USP9x),is a USP domain-containing deubiquitinase which is required in T-cell development.In the present study,we investigate whether USP9x plays a role in hepatic CD8^(+)T-cell dysfunction in septic mice.We find that CD8^(+)T cells are decreased in the blood of septic patients with liver injury compared with those without liver injury,the CD4/CD8 ratio is increased,and the levels of cytolytic factors,granzyme B and perforin are downregulated.The number of hepatic CD8^(+)T cells and USP9x expression are both increased 24 h after cecal ligation and puncture-induced sepsis in a mouse model,a pattern similar to liver injury.The mechanism involves promotion of CD8^(+)T-cell dysfunction by USP9x associated with suppression of cell cytolytic activity via autophagy inhibition,which is reversed by the USP9x inhibitor WP1130.In the in vivo studies,autophagy is significantly increased in hepatic CD8^(+)T cells of septic mice with conditional knockout of mammalian target of rapamycin.This study shows that USP9x has the potential to be used as a therapeutic target in septic liver injury.Lulu Sheng Juntao Chen Yiqing Tong Yi Zhang Qiming Feng Zhenghao Tang 2022Acta Biochimica et Biophysica Sinica2022,54,12:0
12Similarities and Differences of CT Features between COVID-19 Pneumonia and Heart Failure显示文摘Aims:During the COVID-19 epidemic,chest computed tomography(CT)has been highly recommended for screen-ing of patients with suspected COVID-19 because of an unclear contact history,overlapping clinical features,and an overwhelmed health system.However,there has not been a full comparison of CT for diagnosis of heart failure or COVID-19 pneumonia.Methods:Patients with heart failure(n=23)or COVID-19 pneumonia(n=23)and one patient with both diseases were retrospectively enrolled.Clinical information and chest CT images were obtained and analyzed.Results:There was no difference in ground-glass opacity,consolidation,crazy paving pattern,the lobes affected,and septal thickening between heart failure and COVID-19 pneumonia.However,a less rounded morphology(4%vs.70%,P=0.00092),more peribronchovascular thickening(70%vs.35%,P=0.018)and fi ssural thickening(43%vs.4%,P=0.002),and less peripheral distribution(30%vs.87%,P=0.00085)were found in the heart failure group than in the COVID-19 group.Importantly,there were also more patients with upper pulmonary vein enlargement(61%vs.4%,P=0.00087),subpleural effusion(50%vs.0%,P=0.00058),and cardiac enlargement(61%vs.4%,P=0.00075)in the heart failure group than in the COVID-19 group.Besides,more fi brous lesions were found in the COVID-19 group,although there was no statistical difference(22%vs.4%,P=0.080).Conclusions:Although there is some overlap of CT features between heart failure and COVID-19,CT is still a useful tool for differentiating COVID-19 pneumonia.Zhaowei Zhu Jianjun Tang Xiangping Chai Zhenfei Fang Qiming Liu Xinqun Hu Danyan Xu Jia He Liang Tang Shi Tai Yuzhi Wu Shenghua Zhou 2021Cardiovascular Innovations and Applications2021,,3:0

14Relationship of GSTM1 and GSTT1 genetic variant and markers of oxidative stress and inflammation in smokers with coronary artery disease显示文摘Objective:To investigate the role of glutathione S-transferase (GST) genetic variants and markers of oxidative stress and inflammation in smoking-related coronary artery disease (CAD) patients. Methods:Five hundred and thirty-five Chinese CAD patients were successfully genotyped. Plasma total antioxidant status (TAOS), glutathione, C-reactive protein (CRP), fibrinogen(FIB) and white blood cell count (WBC) were determined to evaluate the oxidative stress and inflammatory response. Results: GSTM1-0/GSTT1-0 subjects had a higher CRP, FIB, WBC and GSH and a lower TAOS compared to patients with wild-type GSTM1/GSTT1 genes, but there was significant difference only with regards to TAOS. Smokers with the null genotype of GSTT1 had the highest CRP and the lowest TAOS and GSH when compared to the GSTT1-1 genotype with smoking status, or the GSTT1-0 genotype with non-smoking status, or the GSTT1-1 genotype with non-smoking status. However, we found no significant difference between these groups. Also, no significant interaction was observed between genotypes and smoking status in determining CRP levels. 16Evolution of gene regulatory network of C_(4) photosynthesis in the genus Flaveria reveals the evolutionary status of C_(3)-C_(4) intermediate species显示文摘C_(4) photosynthesis evolved from ancestral C_(3) photosynthesis by recruiting pre-existing genes to fulfill new functions.The enzymes and transporters required for the C_(4) metabolic pathway have been intensively studied and well documented;however,the transcription factors(TFs)that regulate these C_(4) metabolic genes are not yet well understood.In particular,how the TF regulatory network of C_(4) metabolic genes was rewired during the evolutionary process is unclear.Here,we constructed gene regulatory networks(GRNs)for four closely evolutionarily related species from the genus Flaveria,which represent four different evolutionary stages of C_(4) photosynthesis:C_(3)(F.robusta),type Ⅰ C_(3)-C_(4)(F.sonorensis),type Ⅱ C_(3)-C_(4)(F.ramosissima),and C_(4)(F.trinervia).Our results show that more than half of the co-regulatory relationships between TFs and core C_(4) metabolic genes are species specific.The counterparts of the C_(4) genes in C_(3) species were already co-regulated with photosynthesis-related genes,whereas the required TFs for C_(4) photosynthesis were recruited later.The TFs involved in C_(4) photosynthesis were widely recruited in the type Ⅰ C_(3)-C_(4) species;nevertheless,type Ⅱ C_(3)-C_(4) species showed a divergent GRN from C_(4) species.In line with these findings,a ^(13)CO_(2) pulse-labeling experiment showed that the CO_(2) initially fixed into C_(4) acid was not directly released to the Calvin–Benson–Bassham cycle in the type Ⅱ C_(3)-C_(4) species.Therefore,our study uncovered dynamic changes in C_(4) genes and TF co-regulation during the evolutionary process;furthermore,we showed that the metabolic pathway of the type Ⅱ C_(3)-C_(4) species F.ramosissima represents an alternative evolutionary solution to the ammonia imbalance in C_(3)-C_(4) intermediate species.Ming-Ju Amy Lyu Qiming Tang Yanjie Wang Jemaa Essemine Faming Chen Xiaoxiang Ni Genyun Chen Xin-Guang Zhu 2023Plant Communications2023,4,1:0

18A flexible solid polymer electrolyte enabled with lithiated zeolite for high performance lithium battery显示文摘Solid-state lithium batteries using composite polymer electrolytes(CPEs)have attracted much attention owing to their higher safety compared to liquid electrolytes and flexibility compared to ceramic electrolytes.However,their unsatisfactory lithium-ion conductivity still limits their development.Herein,a high ion conductive CPE with multiple continuous lithium pathways is designed.This new electrolyte consists of poly(vinylidene fluorideco-hexafluoropropylene)(PVDF-HFP)and lithiated X type zeolite(Li-X),which possesses a high ionic conductivity(1.98×10^(-4)S/cm),high lithium transference number(t_(Li^(+))=0.55),wide electrochemical window(4.7 V),and excellent stability against the lithium anode.Density functional theory(DFT)calculation confirms that the Lewis acid sites in zeolite can graft with N,N-dimethylformamide(DMF)and PVDF-HFP chains,resulting in decreased crystallinity of polymer and providing rapid Li+transmission channels.When used in a full cell,the solid Li|Li-X-3%|LiFePO_(4) cell displays excellent cycling stability and rate performance at room temperature and 60℃.Furthermore,pouch cells with the Li-X-3%electrolyte exhibit brilliant safety under extreme conditions,such as folding and cutting.Thus,this proposed zeolite-PVDF-HFP CPE represents a promising potential in the application of making a safer,higher performing,and flexible solid-state lithium battery.Zhiyu Ding Qiming Tang Qi Zhang Penghui Yao Xingjun Liu Junwei Wu 2023Nano Research2023,16,7:0
