1Comparative study of the net exchange of CO_(2) in 3 types of vegetation ecosystems on the Qinghai-Tibetan Plateau显示文摘Using the eddy covariance method,from 1 July2003 to 30 June 2004, we conducted the observation andanalysis of ecosystem CO2 flux in 3 types of alpine meadowvegetation (Kobresia humilis, Potentilla fruticosa shrub andKobresia tibetica swamp meadows) on the Qinghai-TibetanPlateau. The results show that the Kobresia humilis meadow,the shrub meadow and the swamp meadow’s highest CO2uptake rates are 16.78, 10.42 and 16.57 μmol·m?2·s?1respectively, while their highest CO2 release rates are 8.22,7.73 and 18.67 μmol·m?2·s?1 respectively. The Kobresiahumilis meadow and shrub meadow’s annual atmosphericuptakes are 282 g CO2/m2 and 53 g CO2/m2, respectively,while swamp meadow’s annual atmospheric release is 478 gCO2/m2. This proves that the Kobresia humilis meadow andthe shrub meadow on the Qinghai-Tibetan Plateau haverelatively low potential for CO2 uptake and release comparedto C4 grasslands, a number of lowland grasslands, andforests. Moreover, swamp meadow has relatively high releasepotential. This, in turn, reveals clear differences in carbonsource/sink between different types of vegetation in theQinghai-Tibetan Plateau alpine meadow ecosystem. 2Characterizing CO_2 fluxes for growing and non-growing seasons in a shrub ecosystem on the Qinghai-Tibet Plateau显示文摘To assess carbon budget for shrub ecosystems on the Qinghai-Tibet Plateau, CO2flux was measured with an open-path eddy covariance system for an alpine shrub ecosystem during growing and non-growing seasons. CO2 flux dynamics was distinct between the two seasons. During the growing season from May to September, the ecosystem exhibited net CO2uptake from 08:00 to 19:00 (Beijing Standard Time), but net CO2 emission from 19:00 to 08:00.Maximum CO2 uptake appeared around 12:00 with values of 0.71, 1.19, 1.46 and 0.67 g CO2m-2 h-1 for June, July, August and September, respectively. Diurnal fluctuation of CO2 flux showed higher correlation with photosynthetic photon flux density than temperature. The maximum net CO2 influx occurred in August with a value of 247 g CO2 m-2. The total CO2 uptake by the ecosystem was up to 583 g CO2 m-2 for the growing season. During the non-growing season from January to April and from October to December, CO2 flux showed small fluctuation with the largest net CO2 efflux of 0.30 g CO2 m-2 h-1 in April. The diurnal CO2 flux was close to zero during most time of the day, but showed a small net CO2 efflux from 11:00 to 18:00. Diurnal CO2 flux, is significantly correlated to diurnal temperature in the non-growing season. The maximum monthly net CO2 efflux appeared in April, with a value of 105 g CO2 m-2. 3Spatial heterogeneity of soil chemical properties between Haloxylon persicum and Haloxylon ammodendron populations显示文摘Spatial heterogeneity is a ubiquitous feature in natural ecosystems, especially in arid regions. Different species and their discontinuous distribution, accompanied by varied topographic characteristics, result in soil resources distributed differently in different locations, and present significant spatial heterogeneity in desert ecosystems. In this study, conventional and geostatistical methods were used to identify the heterogeneity of soil chemical properties in two desert populations, Haloxylon persicum Bunge ex Boss., which dominates on the slopes and tops of sand dunes and Haloxylon ammodendron (C. A. Mey.) Bunge, which inhabits interdunes in the Gurbantunggut Desert of Xinjiang, China. The results showed that soil pH, electrical conductivity (EC), soil organic carbon (SOC), available nitrogen (AN) and available phosphorus (AP) were significantly higher in H. ammodendron populations than that in H. persicum. The coefficient of variation (CV) indicated that (1) most parameters presented a moderate degree of variability (10% < CV < 100%) except pH in both plots, (2) the variability of soil pH, EC and AP in H. ammodendron populations was higher than that in H. persicum populations, and (3) SOC and AN in H. ammodendron populations were lower than that in H. persicum populations. Geostatistical analysis revealed a strong spatial dependence (C0/(C0+C) < 25%) within the distance of ranges for all tested parameters in both plots. The Kriging-interpolated figures showed that the soil spatial distribution was correlated with the vegetation distribution, individual size of plants, and the topographic features, especially with the plants nearest to sampling points and the topographic features. In each plot, soil EC, SOC, AN and AP presented similar distributions, and fertile islands and salt islands occurred in both plots but did not affect every individual plant, since the sampling distance was larger than the size of such fertile islands. The results of topographic effects on soil heterogeneity suggested significant differences between the interdunes and dune-tops. Different topographic characteristics (physical factors) between plots result in the differences in SOC, AN and AP, while the heterogeneity of soil pH and EC arise from plant species and their distribution (biotic factor). Such biotic and physical factors did not occur in isolation, but worked together on soil heterogeneity, and played important parts in improving the soil properties. 4Effect of homogenization treatment on microstructure evolution and the distributions of RE and Zr elements in various MgeLieREeZr alloys显示文摘Mge3Lie0.4Zr alloys containing RE elements(Gd,La,Nd)(Mge3LieREe0.4Zr alloys)are investigated to reveal the influence of homogenization treatment on microstructures and distributions of RE,Zr elements.It is found that 300C24 h homogenization treatment shows better improvement on the microstructure including the refinement of grain size,the dispersion of cellular dendrite and low melting point particles.Before treatment,La and Nd segregate effectively at grain boundary and Zr segregates in the form of precipitates.Homogenization treatment induces the reduction of RE segregation.However,the segregation of Zr in precipitates cannot be abated due to the relatively low diffusion rate compared with RE elements.Lei Bao Qichi Le Zhiqiang Zhang Jianzhong Cui Qinxue Li 2013Journal of Magnesium and Alloys2013,1,2:10
5The involvement of wheat U-box E3 ubiquitin ligase TaPUB1 in salt stress tolerance显示文摘U-box E3 ubiquitin ligases play important roles in the ubiquitin/26 S proteasome machinery and in abiotic stress responses. Ta PUB1-overexpressing wheat(Triticum aestivum L.) were generated to evaluate its function in salt tolerance. These plants had more salt stress tolerance during seedling and flowering stages, whereas the Ta PUB1-RNA interference(RNAi)-mediated knock-down transgenic wheat showed more salt stress sensitivity than the wild type(WT). Ta PUB1 overexpression upregulated the expression of genes related to ion channels and increased the net root Na+efflux, but decreased the net K+efflux and H+influx, thereby maintaining a low cytosolic Na+/K+ratio, compared with the WT. However,RNAi-mediated knock-down plants showed the opposite response to salt stress. Ta PUB1 could induce the expression of some genes that improved the antioxidant capacity of plants under salt stress. Ta PUB1 also interacted with Ta MP(Triticum aestivum α-mannosidase protein), a regulator playing an important role in salt response in yeast and in plants. Thus, low cytosolic Na+/K+ratios and better antioxidant enzyme activities could be maintained in wheat with overexpression of Ta PUB1 under salt stress. 6Automatic Method for Synchronizing Workpiece Frames in Twin-robot Nondestructive Testing System显示文摘The workpiece frames relative to each robot base frame should be known in advance for the proper operation of twin-robot nondestructive testing system. However, when two robots are separated from the workpieces, the twin robots cannot reach the same point to complete the process of workpiece frame positioning. Thus, a new method is proposed to solve the problem of coincidence between workpiece frames. Transformation between two robot base frames is initiated by measuring the coordinate values of three non-collinear calibration points. The relationship between the workpiece frame and that of the slave robot base frame is then determined according to the known transformation of two robot base frames, as well as the relationship between the workpiece frame and that of the master robot base frame. Only one robot is required to actually measure the coordinate values of the calibration points on the workpiece. This requirement is beneficial when one of the robots cannot reach and measure the calibration points. The coordinate values of the calibration points are derived by driving the robot hand to the points and recording the values of top center point(TCP) coordinates. The translation and rotation matrices relate either the two robot base frames or the workpiece and master robot. The coordinated are solved using the measured values of the calibration points according to the Cartesian transformation principle. An optimal method is developed based on exponential mapping of Lie algebra to ensure that the rotation matrix is orthogonal. Experimental results show that this method involves fewer steps, offers significant advantages in terms of operation and time-saving. 9HIV-1 viral cores enter the nucleus collectively through the nuclear endocytosis-like pathway显示文摘It is recognized that HIV-1 capsid cores are disassembled in the cytoplasm, releasing their genomes into the nucleus through nuclear pores, but there is also evidence showing the capsid(CA) exists in the nucleus. Whether HIV-1 enters the nucleus and how it enters the nucleus through the undersized nuclear pore remains mysterious. Based on multicolor labeling and real-time imaging of the viral and cellular components, our observations via light and electron microscopy suggest that HIV-1 selectively gathered at the microtubule organization center(MTOC), leading the nearby nuclear envelope(NE) to undergo deformation,invagination and restoration to form a nuclear vesicle in which the viral particles were wrapped;then, the inner membrane of the nuclear vesicle ruptured to release HIV-1 into the nucleus. 10DC-SIGN promotes Japanese encephalitis virus transmission from dendritic cells to T cells via virological synapses显示文摘Skin-resident dendritic cells(DCs) likely encounter incoming viruses in the first place, and their migration to lymph nodes following virus capture may promote viral replication. However, the molecular mechanisms underlying these processes remain unclear. In the present study, we found that compared to cell-free viruses, DC-bound viruses showed enhanced capture of JEV by T cells.Additionally, JEV infection was increased by co-culturing DCs and T cells. Blocking the C-type lectin receptor DC-specific intercellular adhesion molecule-3-grabbing non-integrin(DC-SIGN) with neutralizing antibodies or antagonists blocked JEV transmission to T cells. Live-cell imaging revealed that DCs captured and transferred JEV viral particles to T cells via virological synapses formed at DC-T cell junctions. 11Binding of HIV-1 virions to α_4β_7 expressing cells and impact of antagonizing α_4β_7 on HIV-1 infection of primary CD4^+ T cells显示文摘HIV-1 envelope glycoprotein is reported to interact with α4β7, an integrin mediating the homing of lymphocytes to gut-associated lymphoid tissue, but the significance of α4β7 in HIV-1 infection remains controversial. Here, using HIV-1 strain Ba L, the gp120 of which was previously shown to be capable of interacting with α4β7, we demonstrated that α4β7 can mediate the binding of whole HIV-1 virions to α4β7-expressing transfectants. We further constructed a cell line stably expressing α4β7 and confirmed the α4β7-mediated HIV-1 binding. In primary lymphocytes with activated α4β7 expression, we also observed significant virus binding which can be inhibited by an anti-α4β7 antibody. Moreover, we investigated the impact of antagonizing α4β7 on HIV-1 infection of primary CD4+ T cells. In α4β7-activated CD4+ T cells, both anti-α4β7 antibodies and introduction of shorthairpin RNAs specifically targeting α4β7 resulted in a decreased HIV-1 infection. Our findings indicate that α4β7 may serve as an attachment factor at least for some HIV-1 strains. 15Research on the changes and significance of serum RBP4 level in patients with senior coronary heart disease accompanying diabetes mellitus显示文摘Objective:To explore the significance of serum Retinol binding protein 4(RBP4)and its relationship with coronary artery lesion in patients with senior coronary heart disease accompanying T2DM by determining the level of serum RBP4 and Gensini score.Methods:30 cases of patients who were 60 years old above with coronary heart disease accompanying T2DM were selected and included in the experimental group,and 30 cases of patients of 60 years old above with coronary heart disease alone were included in the control group.Both groups of patients were given CAG examinations.In addition,Gensini score was calculated according to different degrees and parts of coronary artery lesion.It was required to record each patient’s age,gender,fasting blood glucose(FPG),triglycerides(TG),total cholesterol(TC),high density lipoprotein cholesterol(HDL-C),low density lipoprotein cholesterol(LDL-C),glycated hemoglobin(HbA1c)and other laboratory examination indexes.ELISA was used to detect the level of serum RBP4 in each group,and statistical analysis was performed to the data in each group.Results:(1)RBP4 level,GS score,FPG and LDL-C in the experimental group were all higher than those in the control group,and the difference was of statistical significance(p<.05).There was no statistically significant difference in age,gender,TC,TG and HDL-C between two groups.(2)RBP4 was positively correlated to FPG and HbA1c.(3)In patients with senior coronary heart disease accompanying diabetes mellitus,HbA1c was positively correlated to GS score,RBP4 and FPG.(4)RBP4 was a risk factor for coronary artery stenosis in patients with senior coronary heart disease accompanying diabetes mellitus.Conclusions:The level of serum RBP4 in patients with senior coronary heart disease accompanying diabetes mellitus is higher than that in patients with coronary heart disease alone,with a deeper degree of coronary artery lesion.The level of serum RBP4 is increased with the degree of lesion deepened in patients with senior coronary heart disease,suggesting that the level of serum RBP4 is expected to be an early predictor of coronary artery lesion for patients with senior coronary heart disease.Qinxue Li Biao Ge Ruijing Yan Yinlong Bai Yan Liu 2019Discussion of Clinical Cases2019,6,1:0
