2Expression profile of cholecystokinin type-A receptor in gallbladder cancer and gallstone disease显示文摘BACKGROUND:Regulatory peptide receptors have attracted the interest of oncologists as a new promising approach for cancer pathology,imaging and therapy.Although cholecystokinin (CCK) is a potent modulator of gallbladder contractility and plays a potential role in pancreatic carcinogenesis through CCK type-A receptor (CCKAR),its role in gallbladder cancer (GBC) is still unknown and immunohistochemical detection of CCKAR in the gallbladder has not yet been reported.This novel case-control study aimed to investigate the expression profile of CCKAR in GBC and gallstone disease (GSD).METHODS:This study included 162 samples of gallbladder:94 from GBC and 68 from GSD.Expression of CCKAR was analyzed by immunohistochemistry and immunoblotting.The results were statistically correlated with disease history including age,sex,presence of gallstone,stage and differentiation.RESULTS:CCKAR was positive in 30/68 (44.1%) of GSD and 72/94 (76.6%) of GBC samples.Fifty-one of the 72 (70.8%) CCKAR-positive GBC samples showed over-expression.Interestingly,consistent results also appeared in the immunoblotting study.CONCLUSIONS:CCKAR expression was significantly increased in GBC compared to GSD.Moreover,CCKAR expression was associated with the degree of tumor differentiation,i.e.,less expression in poorly-differentiated tumors.Thus,it has future prognostic and therapeutic implications in the management of GBC.Rajani Rai Mallika Tewari Mohan Kumar Tej Bali Singh Hari S Shukla 2011Hepatobiliary & Pancreatic Diseases International2011,10,4:7
3Autologous bone marrow derived stem cell therapy in patients with type 2 diabetes mellitus-defining adequate administration methods显示文摘AIM To carry out randomized trial for evaluating effects of autologous bone marrow derived stem cell therapy(ABMSCT) through different routes.METHODS Bone marrow aspirate was taken from the iliac crest of patients. Bone marrow mononuclear cells were separatedand purified using centrifugation. These cells were then infused in a total of 21 patients comprising three groups of 7 patients each. Cells were infused into the superior pancreaticoduodenal artery(Group Ⅰ), splenic artery(Group Ⅱ) and through the peripheral intravenous route(Group Ⅲ). Another group of 7 patients acted as controls and a sham procedure was carried out on them(Group Ⅳ). The cells were labelled with the PET tracer F18-FDG to see their homing and in vivo distribution. Data for clinical outcome was expressed as mean ± SE. All other data was expressed as mean ± SD. Baseline and post treatment data was compared at the end of six months, using paired t-test. Cases and controls data were analyzed using independent t-test. A probability(P) value of < 0.05 was regarded as statistically significant. Measures of clinical outcome were taken as the change or improvement in the following parameters:(1) C-peptide assay;(2) HOMA-IR and HOMA-B;(3) reduction in Insulin dose; subjects who showed reduction of insulin requirement of more than 50% from baseline requirement were regarded as responders; and(4) reduction in HbA 1c. RESULTS All the patients, after being advised for healthy lifestyle changes, were evaluated at periodical intervals and at the end of 6 mo. The changes in body weight, body mass index, waist circumference and percentage of body fat in all groups were not significantly different at the end of this period. The results of intra-group comparison before and after ABMSCT at the end of six months duration was as follows:(1) the area under C-peptide response curve was increased at the end of 6 mo however the difference remained statistically non-significant(P values for fasting C-peptide were 0.973, 0.103, 0.263 and 0.287 respectively and the P values for stimulated C-peptide were 0.989, 0.395, 0.325 and 0.408 respectively for groups Ⅰ?to Ⅳ);(2) the Insulin sensitivity indices of HOMA IR and HOMA B also did not show any significant differences(P values for HOMA IR were 0.368, 0.223, 0.918 and 0.895 respectively and P values for HOMA B were 0.183, 0.664, 0.206 and 0.618 respectively for groups Ⅰto Ⅳ);(3) Group Ⅰshowed a significant reduction in Insulin dose requirement(P < 0.01). Group Ⅱ patients also achieved a significant reduction in Insulin dosages(P = 0.01). The Group Ⅰand Group Ⅱ patients together constituted the targeted group wherein the feeding arteries to pancreas were used for infusing stem cells. Group Ⅲ, which was the intravenous group, showed a non-significant reduction in Insulin dose requirement(P = 0.137). Group Ⅳ patients which comprised the control arm also showed a significant reduction in Insulin dosages at the end of six months(P < 0.05); and(4) there was a non-significant change in the Hb A1 c levels at the end of 6 mo across all groups(P = 0.355, P = 0.351, P = 0.999 and P = 0.408 respectively for groups Ⅰto Ⅳ). 4Analysis of soil moisture variation by forest cover structure in lower western Himalayas, India显示文摘Soil moisture affects various hydrological processes, including evapotranspiration, infiltration, and runoff. Forested areas in the lower western Himalaya in India constitute the headwater catchments for many hill streams and have experienced degradation in forest cover due to grazing, deforestation and other human activities. This change in forest cover is likely to alter the soil moisture regime and, consequently, flow regimes in streams. The effect of change in forest cover on soil moisture regimes of this dry region has not been studied through long term field observations. We monitored soil matric potentials in two small watersheds in the lower western Himalaya of India. The watersheds consisted of homogeneous land covers of moderately dense oak forest and moderately degraded mixed oak forest. Observations were recorded at three sites at three depths in each watershed at fortnightly intervals for a period of three years. The soil moisture contents derived from soil potential measurements were analyzed to understand the spatial, temporal and profile variations under the two structures of forest cover. The analysis revealed large variations in soil moisture storage at different sites and depths and also during different seasons in each watershed. Mean soil moisture storage during monsoon, winter and summer seasons was higher under dense forest than under degraded forest. Highest soil moisture content occurred at shallow soil profiles, decreasing with depth in both watersheds. A high positive correlation was found between tree density and soil moisture content. Mean soil moisture content over the entire study period was higher under dense forest than under degraded forest. This indicated a potential for soil water storage under well managed oak forest. Because soil water storage is vital for sustenance of low flows, attention is needed on the management of oak forests in the Himalayan region.J. V. Tyagi Nuzhat Qazi S. P. Rai M. P. 6FORECASTING AUTOMOBILE WARRANTY PERFORMANCE IN PRESENCE OF ‘MATURING DATA’ PHENOMENA USING MULTILAYER PERCEPTRON NEURAL NETWORK显示文摘Automobile companies that spend billions of dollars annually towards warranty cost, give high priority to warranty reduction programs. Forecasting of automobile warranty performance plays an important role towards these efforts. The forecasting process involves prediction of not only the specific months-in-service (MIS) warranty performance at certain future time, but also at future MIS values. However, ’maturing data’ (also called warranty growth) phenomena that causes warranty performance at specific MIS values to change with time, makes such a forecasting task challenging. Although warranty forecasting methods such as log-log plots and dynamic linear models appear in literature, there is a need for applications addressing the well recognized issue of ‘maturing data’. In this paper we use an artificial neural network for the forecasting of warranty performance in presence of ‘maturing data’ phenomena. The network parameters are optimized by minimizing the training and testing errors using response surface methodology. 8Seasonal Inhomogeneity of Soot Particles over the Central Indo-Gangetic Plains,India:Influence of Meteorology显示文摘Black carbon(BC) particles play a unique and important role in earth's climate system.BC was measured(in-situ) in the central part of the Indo-Gangetic Plains(IGP) at Varanasi,which is a highly populated and polluted region due to its topography and extensive emission sources.The annual mean BC mass concentration was 8.92 ± 7.0 μg m^(-3),with 34%of samples exceeding the average value.Seasonally,BC was highest during the post-monsoon and winter periods(approximately 18 μg m^(-3)) and lower in the premonsoon/monsoon seasons(approximately 6 μg m^(-3)).The highest frequency(approximately 46%) observed for BC mass was in the interval from 5 to 10 μg m^(-3).However,during the post-monsoon season,the most common values(approximately 23%) were between 20 and 25 μg m^(-3).The nighttime concentrations of BC were approximately twice as much as the daytime values because of lower boundary layer heights at nighttime.The Angstrom exponent was significantly positively correlated(0.55) with ground-level BC concentrations,indicating the impact of BC on the columnar aerosol properties.The estimated mean absorption Angstrom exponent was 1.02 ± 0.08 μg m^(-3),indicating that the major source of BC was from fossil fuel combustion.Significant negative correlations between BC mass and meteorological parameters indicate a pronounced effect of atmospheric dynamics on the BC mass in this region.The highest mean BC mass concentration(18.1 ± 6.9 μg m^(-3)) as a function of wind speed was under calm wind conditions(38%of the time).SINGH B.P.
