1Metabonomic analysis of plasma biochemical changes in pyrexia rats after treatment with Gegenqinlian decoction,aspirin and itraconazole by UHPLC-FT-ICR-MS显示文摘A metabonomic approach involving an ultrahigh-performance liquid chromatography combined with Fourier transform ion cyclotron resonance mass spectrometry(UHPLC-FT-ICR-MS)was used to investigate the changes in the endogenous metabolites in the plasma of rats with yeast-induced pyrexia treated with Gegenqinlian decoction(GQLD),aspirin and itraconazole.The differences in the small molecule profiles of treatment using traditional Chinese medicine,etiological treatment and symptomatic treatment were elucidated.Thirty-six plasma metabolites were identified or putatively identified,and the effects of the three medicines on the thirty-six metabolites were studied.Their metabolic pathways indicated that GQLD,aspirin and itraconazole ameliorated the rats with yeast-induced pyrexia predominantly by regulating the metabolisms of phospholipid,sphingolipid,fatty acid oxidation,fatty acid amides,amino acid and glycerolipid in vivo.The pharmacodynamics and metabonomic results showed that the three medicines exhibited the therapeutic effects on pyrexia by regulating the perturbations of multiple metabolisms.The study provided a scientific basis for an in-depth understanding of the therapeutic effects of GQLD,aspirin and itraconazole on rats with yeast-induced pyrexia.Ting Liu Ruiyun Li Yue Cui Zhiguo Yu Yunli Zhao 2020Journal of Pharmaceutical Analysis2020,10,6:6
2Efficient lung cancer-targeted drug delivery via a nanoparticle/MSC system显示文摘Low targeting efficiency limits the applications of nanoparticles in cancer therapy. The fact that mesenchymal stem cells(MSC) trapped in the lung after systemic infusion is a disadvantage for cell therapy purposes. Here, we utilized MSC as lung cancer-targeted drug delivery vehicles by loading nanoparticles(NP)with anti-cancer drug. MSC showed a higher drug intake capacity than fibroblasts. In addition, MSC showed predominant lung trapping in both rabbit and monkey. IR-780 dye, a fluorescent probe used to represent docetaxel(DTX) in NP, delivered via MSC accumulated in the lung. Both in vitro MSC/A549 cell experiments and in vivo MSC/lung cancer experiments validated the intercellular transportation of NP between MSC and cancer cells. In vivo assays showed that the MSC/NP/DTX drug delivery system exerted primary tumor inhibition efficiency similar to that of a NP/DTX drug system. 3Composition analysis and antioxidant activities of the Rhus typhina L.stem显示文摘The present investigation reports the chemical composition of the Rhus typhina L. stem identified via mass spectrometry and NMR as gallic acid, 1-O-galloyl-β-D-glucose, tryptophan, scopolin, methyl gallate, fustin, quercetin, rutin, and 1,2,3,4,6-penta-O-galloyl-β-D-glucose. The antioxidant properties and the chemical composition contents of the R. typhina L. stem grown in different regions in China were determined. To determine the antioxidant activity, a total phenolic content analysis, 2, 2-diphenyl-1-picrylhydrazyl radical scavenging activity assay, ferric reducing antioxidant power assay, and β-carotene linoleic acid model system were conducted. The results showed that the Rhus typhina L. stem possessed high antioxidant capacities due to its high phenolic content. The contents of the nine isolated compounds were determined by UPLC-ESI-MS/MS. The calibration curves of the nine isolated compounds were linear within the concentration range and the average recoveries were high. The result showed that 1-Ogalloyl-β-D-glucose, gallic acid, methyl gallate, and 1,2,3,4,6-penta-O-galloyl-β-D-glucose could be the compounds mainly responsible for the antioxidant capacity of the R. typhina L. stem. 8Analysis of Imaging Characteristics and Dynamic Changes of 3 Cases of Severe Novel Coronavirus Pneumonia in Qinghai Province显示文摘Objective:To analyze the characteristics,dynamic changes,and outcomes of the first imaging manifestations of 3 patients with severe COVID-19 in our hospital.Methods:Computed tomography(CT)findings of 3 patients with severe COVID-19 who tested positive by the nucleic acid test in our hospital were selected,mainly focusing on the morphology,distribution characteristics,and dynamic changes of the first CT findings.Results:3 patients with severe pneumonia were older,with one aged 80.The first chest CT examination for all 3 patients differed.Imaging showed a leafy distribution of consolidation,primarily affecting the lower lobes of both lungs and extending subpleurally.A grid-like pattern was observed,along with changes in the consolidation and air bronchogram.These changes had slower absorption,especially in patients with underlying diseases.Conclusion:CT manifestations of severe COVID-19 have specific characteristics and the analysis of their characteristics and dynamic changes provide valuable insights for clinical treatment.Yingfang Yu Ruiyun Zhao Changde Li Fuqiang Ma Lingyun Guo Yang Li 2024Journal of Clinical and Nursing Research2024,8,3:0
9A novel microcurrent dressing for wound healing in a rat skin defect model显示文摘Background:The exogenous application of low-intensity electric stimulation(ES)may mimic a natural endogenous bioelectric current and accelerate the repair process of skin wounds.This study designed a novel microcurrent dressing(MCD)and evaluated its potential effects on wound healing in a rat skin defect model.Methods:First,wireless ES was integrated into a medical cotton cushion to fabricate the MCD,and its electrical property was examined by using a universal power meter.Then,animal experiments were conducted to evaluate the MCD’s effect.Forty-five rats were randomized into control(Con)group,Vaseline gauze(VG)group and MCD group.A full-thickness round skin incision 1.5 cm in diameter was made on the back of each animal.Apart from routine disinfection,the Con rats were untreated,whereas the other two groups were treated with VG or MCD.On days 3,7 and 14 post injury,the wound areas were observed and measured using image analysis software following photography,and the skin samples were harvested from wound tissue.Then,histopathological morphology was observed routinely by hematoxylin and eosin(HE)staining;tumor necrosis factorα(TNF-α)and interleukin(IL)-1βexpression were detected by Western blotting.Vascular endothelial growth factor(VEGF)and epidermal growth factor(EGF)expression were detected with immunohistochemistry.Results:The MCD generated a sf electric potential greater than 0.95 V.Animal experiments showed that the woundhealing rate in the MCD group was significantly increased compared with the Con and VG groups(P<0.05 or P<0.01).Histopathological observation revealed an alleviated inflammatory response,induced vascular proliferation and accelerated epithelization in the MCD group.Moreover,samples from the MCD group expressed reduced TNF-αand IL-1βlevels and increased VEGF and EGF levels compared with those of the other two groups(P<0.05 or P<0.01).However,no significant difference was noted between the Con and VG groups at each time point.Conclusions:The MCD generates a stable and lasting ES and significantly promotes wound healing by reducing inflammation duration and increasing growth factors expression.Thus,MCD may act as a promising biomaterial device for skin wound healing.Chao Yu Zhi-Xiu Xu Yan-Hui Hao Ya-Bing Gao Bin-Wei Yao Jing Zhang Bing Wang Zong-Qian Hu RuiYun Peng 2020Military Medical Research2020,7,2:0
10Electric field-driven folding of single molecules显示文摘Folding of molecules is an essential process in nature,and various molecular machines achieve their chemical and mechanical function via controlled folding of molecular conformations.The electric field offers a unique strategy to drive the folding of molecular conformation and to control charge transport through single molecules but remains unexplored.The single-molecule break junction technique provides access to detect the conformational changes via the monitoring of single-molecule conductance,and the electric field between two metal electrodes with nanoscale spacing can provide an extremely strong to achieve in-situ control and detection of molecular folding at the single-molecule level.Here,we use the electric field to control the single-molecule folding using the scanning tunneling microscope break junction(STM-BJ)technique.The electric fields induced folding could lead to a∼1400%conductance change of the single-molecule junctions,and the folding/unfolding process can be in-situ switched at the scale of milliseconds.DFT calculations suggest the conformational control originates from the electric fieldinduced charge injection,and the formation of homoconjugated conformation with the overlapped orbitals.This work provides the first demonstration of electric field-driven molecular folding,which is essential for the understanding of molecular machines in nature and for the design of artificial molecular machines.Saisai Yuan Yu Zhou Tengyang Gao Lichuan Chen Wei Xu Ping Duan Juejun Wang Zhichao Pan Chun Tang Yang Yang Ruiyun Huang Zongyuan Xiao Wenjing Hong 2024Chinese Chemical Letters2024,35,1:0
