10Neuropharmacological evaluation of methanolic extract of Costus speciosus Linn. rhizome in Swiss albino mice显示文摘Objective: To evaluate the neuropharmacological properties of Costus speciosus(C. speciosus) rhizome using different experimental mouse models. Methods: The anxiolytic effect was investigated by hole-board test, elevated plus maze and light/dark test, while central nervous system(CNS) depressant effect was evaluated by thiopental sodium-induced sleep test. Finally, antidepressant effect was evaluated by forced swimming test and tail suspension test. Results: In both elevated plus maze and hole board test, 400 mg/kg C. speciosus showed more significant CNS depressant effect than 1 mg/kg diazepam. Both 200 mg/kg and 400 mg/kg C. speciosus extract produced a significant dose-dependent decrease in onset of sleep. In forced swimming test, C. speciosus rhizome showed a decrease in duration of immobility in a dose-dependent manner. Imipramine(10 mg/kg) and C. speciosus extract at 400 mg/kg dose exhibited a significant reduction in duration of immobility in tail suspension test which provided additional evidence of antidepressant effect of C. speciosus rhizome. Conclusions: Our study indicates that C. speciosus rhizome possesses CNS depressant, anxiolytic and antidepressant-like activities. 11New drug delivery system for liver sinusoidal endothelial cells for ischemia-reperfusion injury显示文摘AIM: To investigate the cytoprotective effects in hepatic ischemia-reperfusion injury, we developed a new formulation of hyaluronic acid(HA) and sphingosine 1-phophate.METHODS: We divided Sprague-Dawley rats into 4 groups: control, HA, sphingosine 1-phosphate(S1P), and HA-S1 P. After the administration of each agent, we subjected the rat livers to total ischemia followed by reperfusion. After reperfusion, we performed the following investigations: alanine aminotransferase(ALT), histological findings, Td T-mediated d UTP-biotin nick end labeling(TUNEL) staining, and transmission electron microscopy(TEM). We also investigated the expressionof proteins associated with apoptosis, hepatoprotection, and S1 P accumulation. RESULTS: S1 P accumulated in the HA-S1 P group livers more than S1 P group livers. Serum ALT levels, TUNEL-positive hepatocytes, and expression of cleaved caspase-3 expression, were significantly decreased in the HA-S1 P group. TEM revealed that the liver sinusoidal endothelial cell(LSEC) lining was preserved in the HA-S1 P group. Moreover, the HA-S1 P group showed a greater increase in the HO-1 protein levels compared to the S1 P group.CONCLUSION: Our results suggest that HA-S1 P exhibits cytoprotective effects in the liver through the inhibition of LSEC apoptosis. 20PIN FORMED 2 Modulates the Transport of Arsenite in Arabidopsis thaliana显示文摘Arsenic contamination is a major environmental issue,as it may lead to serious health hazard.The reduced trivalent formof inorganic arsenic,arsenite,is in generalmore toxic to plants comparedwith the fully oxidized pentavalent arsenate.Theuptakeof arsenite inplants hasbeenshown tobemediatedthrough a large subfamily of plant aquaglyceroporins,nodulin 26-like intrinsic proteins(NIPs).However,the efflux mechanisms,as well as themechanismof arsenite-induced root growth inhibition,remain poorly understood.Usingmolecular physiology,synchrotron imaging,and root transport assay approaches,we show that the cellular transport of trivalent arsenicals inArabidopsis thalianais stronglymodulatedbyPINFORMED2(PIN2)auxin efflux transporter.Root transport assay using radioactive arsenite,X-ray fluorescence imaging(XFI)coupled with X-ray absorption spectroscopy(XAS),and inductively coupled plasma mass spectrometry analysis revealed that pin2 plants accumulate higher concentrations of arsenite in roots comparedwith the wild-type.At the cellular level,arsenite specifically targets intracellular sorting of PIN2 and thereby alters the cellular auxin homeostasis.Consistently,loss of PIN2 function results in arsenite hypersensitivity in roots.XFI coupled with XAS further revealed that loss of PIN2 function results in specific accumulation of arsenical species,but not the other metals such as iron,zinc,or calcium in the root tip.Collectively,these results suggest that PIN2 likely functions as an arsenite efflux transporter for the distribution of arsenical species in planta.Mohammad Arif Ashraf Kana Umetsu Olena Ponomarenko Michiko Saito Mohammad Aslam Olga Antipova Natalia Dolgova Cheyenne D.Kiani Susan Nehzati Keitaro Tanoi Katsuyuki Minegishi Kotaro Nagatsu Takehiro Kamiya Toru Fujiwara Christian Luschnig Karen Tanino Ingrid Pickering Graham N.George Abidur Rahman 2020Plant Communications2020,1,3:0
