1Drift and deposition of ultra-low altitude and low volume application in paddy field显示文摘Field trials were performed to evaluate various techniques for measuring spray deposition and aerial drift during spray application to paddy field.The application of a spraying agent containing the fluorescent dye Rhodamine-B was applied by an unmanned aerial vehicle(UAV)which flew at a height of 5 m,a speed of 3 m/s,and the wind speed of 3 m/s.The results showed that because the downdraft produced by a helicopter rotor increased the penetrability of crops,there is a higher deposition on the upper layer and the under layer than the traditional spraying.The average deposition on the upper layer accounts for 28% of the total spraying,the deposition on the under layer accounts for 26% of the total spraying.The deposition on the under layer takes up 92.8% of the deposition on the upper layer.Droplets drift data showed that the drift of non-target area took up 12.9% of the total liquid spray.The 90% drifting droplets were located within a range of 8 m of the target area;the drift quantity was almost zero at a distance of 50 m away from the treated area.Xue Xinyu Tu Kang Qin Weicai Yubin Lan Huihui Zhang 2014International Journal of Agricultural and Biological Engineering2014,7,4:40
2Ionic liquid based dispersive liquid-liquid microextraction of aromatic amines in water samples显示文摘In this work,a new microextraction method termed ionic liquid based dispersive liquid-liquid microextraction(IL-DLLME) was demonstrated for the extraction of 2-methylaniline,4-chloroaniline,1-naphthylamine and 4-aminobiphenyl in aqueous matrices.After extraction the ionic liquid(IL)phase was injected directly into the high performance liquid chromatography(HPLC) system for determination.Some parameters that might affect the extraction efficiency were optimized.Under the optimum conditions,good linear relationship,sensitivity and reproducibility were obtained.The limits of detection(LOD,S/N=3)for the four analytes were in the range of 0.45-2.6μg L^(-1).The relative standard deviations(R.S.D.,n=6)were in the range of 6.2-9.8%. 3Electroacupuncture pretreatment exhibits antidepressive effects by regulating hippocampal proteomics in rats with chronic restraint stress显示文摘The clinical effect of electroacupuncture on depression is widely recognized. However, the signal transduction pathways and target proteins involved remain unclear. In the present study, rat models of chronic restraint stress were used to explore the mechanism by which electroacupuncture alleviates depression. Rats were randomly divided into control, model, and electroacupuncture groups. Chronic restraint stress was induced in the model and electroacupuncture groups by restraining rats for 28 days. In the electroacupuncture group, electroacupuncture pretreatment at Baihui(GV20) and Yintang(GV29) acupoints was performed daily(1 m A, 2 Hz, discontinuous wave, 20 minutes) prior to restraint for 28 days. Open field tests and body weight measurements were carried out to evaluate the depressive symptoms at specific time points. On day 28, the crossing number, rearing number, and body weights of the model group were significantly lower than those in the control group. Behavior test results indicated that rat models of depressive-like symptoms were successfully established by chronic restraint stress combined with solitary raising. On day 28, an isobaric tag for a relative and absolute quantitation-based quantitative proteomic approach was performed to identify differentially expressed proteins in hippocampal samples obtained from the model and electroacupuncture groups. The potential function of these differential proteins was predicted through the use of the Cluster of Orthologous Groups of proteins(COG) database. Twenty-seven differential proteins(uncharacteristic proteins expected) were selected from the model and electroacupuncture groups. In addition to unknown protein functions, COG are mainly concentrated in general prediction function, mechanism of signal transduction, amino acid transport and metabolism groups. 4The signal pathways and treatment of cytokine storm in COVID-19显示文摘The Coronavirus Disease 2019(COVID-19)pandemic has become a global crisis and is more devastating than any other previous infectious disease.It has affected a significant proportion of the global population both physically and mentally,and destroyed businesses and societies.Current evidence suggested that immunopathology may be responsible for COVID-19 pathogenesis,including lymphopenia,neutrophilia,dysregulation of monocytes and macrophages,reduced or delayed type I interferon(IFN-I)response,antibody-dependent enhancement,and especially,cytokine storm(CS).The CS is characterized by hyperproduction of an array of pro-inflammatory cytokines and is closely associated with poor prognosis.These excessively secreted pro-inflammatory cytokines initiate different inflammatory signaling pathways via their receptors on immune and tissue cells,resulting in complicated medical symptoms including fever,capillary leak syndrome,disseminated intravascular coagulation,acute respiratory distress syndrome,and multiorgan failure,ultimately leading to death in the most severe cases.Therefore,it is clinically important to understand the initiation and signaling pathways of CS to develop more effective treatment strategies for COVID-19.Herein,we discuss the latest developments in the immunopathological characteristics of COVID-19 and focus on CS including the current research status of the different cytokines involved.We also discuss the induction,function,downstream signaling,and existing and potential interventions for targeting these cytokines or related signal pathways.We believe that a comprehensive understanding of CS in COVID-19 will help to develop better strategies to effectively control immunopathology in this disease and other infectious and inflammatory diseases.Lan Yang Xueru Xie Zikun Tu Jinrong Fu Damo Xu Yufeng Zhou 2021Signal Transduction and Targeted Therapy2021,6,8:7

6ITS2 barcoding DNA region combined with high resolution melting(HRM) analysis of Hyoscyami Semen, the mature seed of Hyoscyamus niger显示文摘Hyoscyami Semen, the mature dried seed of Hyoscyamus niger L., has long been used as a traditional Chinese medicine to treat human diseases. Hyoscyami Semen is found in local markets in China. In markets, sellers and buyers commonly inadvertently mix the seeds of H. niger with the seeds of related species such as Hygrophila salicifolia(Vahl) Nees, Astragalus complanatus R. Br., Cuscuta australis R. Br., Cuscuta chinensis Lam., and Impatiens balsamina L. because of their similar morphologies or similar names. Thus, developing a reliable method for discriminating H. niger seeds from its adulterants is necessary to reduce confusion and ensure the safe use of Hyoscyami Semen. The present study was designed to evaluate the efficiency of high-resolution melting analysis combined with DNA barcoding(Bar-HRM) with internal transcribed spacer 2 to discriminate H. niger. Our results show that Bar-HRM successfully identified the adulterants and detected the proportion of H. niger DNA extract within an admixture. In particular, HRM detected H. niger DNA extract in A. complanatus DNA extract at concentrations as low as 1%. In conclusion, the Bar-HRM method developed in the present study for authenticating H. niger is rapid and cost-effective. 7Exogenous nerve growth factor protects the hypoglossal nerve against crush injury显示文摘Studies have shown that sensory nerve damage can activate the p38 mitogen-activated protein kinase(MAPK)pathway,but whether the same type of nerve injury after exercise activates the p38MAPK pathway remains unclear.Several studies have demonstrated that nerve growth factor may play a role in the repair process after peripheral nerve injury,but there has been little research focusing on the hypoglossal nerve injury and repair.In this study,we designed and established rat models of hypoglossal nerve crush injury and gave intraperitoneal injections of exogenous nerve growth factor to rats for 14 days.p38MAPK activity in the damaged neurons was increased following hypoglossal nerve crush injury;exogenous nerve growth factor inhibited this increase in acitivity and increased the survival rate of motor neurons within the hypoglossal nucleus.Under transmission electron microscopy,we found that the injection of nerve growth factor contributed to the restoration of the morphology of hypoglossal nerve after crush injury.Our experimental findings indicate that exogenous nerve growth factor can protect damaged neurons and promote hypoglossal nerve regeneration following hypoglossal nerve crush injury.Li-yuan Fan Zhong-chao Wang Pin Wang Yu-yan Lan Ling Tu 2015Neural Regeneration Research2015,10,12:4
8Prognostic value of pre-and post-transplantation 18F-fluorodeoxyglucose positron emission tomography results in non-Hodgkin lymphoma patients receiving autologous stem cell transplantation显示文摘Objective: High-dose chemotherapy(HDC) followed by autologous stem cell transplantation(ASCT) is the standard of care in the upfront or relapsed/refractory setting in some patients with non-Hodgkin lymphoma(NHL).However,a proportion of patients do not respond to ASCT.^(18)F-fluorodeoxyglucose(FDG) positron emission tomography(PET)/computed tomography(CT) has been widely used for staging,response evaluation,and prognosis prediction.Here,we investigated the prognostic role of PET/CT in NHL patients before and after ASCT.Methods: A retrospective study was conducted at Peking University Cancer Hospital.All NHL patients who underwent ASCT between March 2010 and July 2016 were identified.Patients who had PET/CT scan before and after ASCT were included.Deauville criteria(5-point scale) were used to interpret PET scans.Univariate and multivariate survival analyses were performed using Cox regression.The predictive value of PET scanning was estimated by comparing the area under the receiver operating characteristic(ROC) curve.Results: In total,79 patients were enrolled in this study.In univariate analysis,pre-and post-ASCT PET result was identified as prognostic factors for 3-year progression-free survival(PFS) and overall survival(OS).Patients with negative pre-ASCT PET result demonstrated significantly better PFS(84.2% vs.54.2%) and OS(89.2% vs.63.6%) than patients with positive pre-ASCT PET result.PFS(91.6% vs.25.3%) and OS(96.5% vs.36.8%) were also significantly different between patients with negative and positive post-ASCT PET result.Multivariate analysis also showed a significant association between survival and post-ASCT PET result.ROC analysis revealed that the predictive value of post-ASCT PET result was superior to that of pre-ASCT PET result alone.Combined pre-and post-ASCT PET result is better for predicting outcomes in patients with NHL receiving transplantation.Deauville criteria score >3 was identified as the best cutoff value for post-ASCT PET.Conclusions: Post-ASCT PET result was more important than pre-ASCT PET result in predicting outcomes for NHL patients who underwent ASCT.The prognostic significance can be improved when combining preASCT PET result with post-ASCT PET result.Deauville criteria can be used for interpreting PET scans in this scenario.Zhitao Ying Lan Mi Xuejuan Wang Yuewei Zhang Zhi Yang Yuqin Song Xiaopei Wang WenZheng Ningjing Lin Meifeng Tu Yan Xie Lingyan Ping Chen Zhang Weiping Liu LijuanDeng Jun Zhu 2017Chinese Journal of Cancer Research2017,29,6:3
9Resveratrol promotes the survival and neuronal differentiation of hypoxia-conditioned neuronal progenitor cells in rats with cerebral ischemia显示文摘Hypoxia conditioning could increase the survival of transplanted neuronal progenitor cells(NPCs)in rats with cerebral ischemia but could also hinder neuronal differentiation partly by suppressing mitochondrial metabolism.In this work,the mitochondrial metabolism of hypoxia-conditioned NPCs(hcNPCs)was upregulated via the additional administration of resveratrol,an herbal compound,to resolve the limitation of hypoxia conditioning on neuronal differentiation.Resveratrol was first applied during the in vitro neuronal differentiation of hcNPCs and concurrently promoted the differentiation,synaptogenesis,and functional development of neurons derived from hcNPCs and restored the mitochondrial metabolism.Furthermore,this herbal compound was used as an adjuvant during hcNPC transplantation in a photothrombotic stroke rat model.Resveratrol promoted neuronal differentiation and increased the long-term survival of transplanted hcNPCs.18-fluorine fluorodeoxyglucose positron emission tomography and rotarod test showed that resveratrol and hcNPC transplantation synergistically improved the neurological and metabolic recovery of stroke rats.In conclusion,resveratrol promoted the neuronal differentiation and therapeutic efficiency of hcNPCs in stroke rats via restoring mitochondrial metabolism.This work suggested a novel approach to promote the clinical translation of NPC transplantation therapy.Yao Yao Rui Zhou Rui Bai Jing Wang Mengjiao Tu Jingjing Shi Xiao He Jinyun Zhou Liu Feng Yuanxue Gao Fahuan Song Feng Lan Xingguo Liu Mei Tian Hong Zhang 2021Frontiers of Medicine2021,15,3:3
10Construction of sperm-specific lactate dehydrogenase DNA vaccine and experimental study of its immuno-contraceptive effect on mice显示文摘Lactate dehydrogenase C4 (LDHC4) is a key enzyme for sperm metabolism. It is distributed specifically in testis and is highly immunogenic. In this study, two DNA vaccines pVAX1-hLDHC and pVAX1-mLDHC were constructed by inserting coding sequences of human and mice LDHC4 into the eukaryotic ex-pression vector pVAX1. The production of LDHC4 specific antibodies was induced in the sera of vac-cinated mice and the reproductive tract secretions of vaccinated female mice through immunization by mucosal surface instillation. Furthermore, the antibody titer increased with the times of immunization. In the mating experiment, the number of newborns of the vaccinated mice reduced significantly and some immunized female mice even lost the ability to bear any offsprings, suggesting that the difference between the immunized and control mice was statistically significant. Sperm agglutination analysis indicated that both the antisera from immunized mice and the reproductive tract secretions of vacci-nated female mice could agglutinate normal sperms. Results of immunohistochemistry showed that the antibodies present in the sera of immunized mice and the reproductive tract secretions of vaccinated female mice could specifically react with LDHC4 antigen, which mainly locates in the cytoplasm, acrosome membrane externa and acrosome capsule of the sperm. 11Distributions of Sr and Fe and their influence on modification of hypoeutectic Al-Si alloy显示文摘The influence of cooling rate and Fe-containing phases on Sr-modification of Si phases in hypoeutectic Al-Si alloys, a problem with great industrial importance, was investigated. The microstructures of samples were examined using scanning electron microscopy(SEM) with energy-dispersive X-ray spectroscopy(EDX). A new method of electron probe microanalysis(EPMA) map scanning was used to analyze the Sr distribution, which gave quantitative results covering more Si particles. EPMA map scanning, together with SEM with EDX, was also used in analyzing the distribution of Fe phases. Results show that Fe-containing phase was related to the unmodified Si particles in samples with partial modification failure and the plate-like Si phases in samples without modification failure. 13Significance of serum fibroblast growth factor-23 and miR-208b in pathogenesis of atrial fibrillation and their relationship with prognosis显示文摘BACKGROUND The incidence and prevalence of atrial fibrillation are increasing each year,and this condition is one of the most common clinical arrhythmias.AIM To investigate the levels and significance of serum fibroblast growth factor 23(FGF-23)and miR-208 b in patients with atrial fibrillation and their relationship with prognosis.METHODS From May 2018 to October 2019,240 patients with atrial fibrillation were selected as an observation group,including 134 with paroxysmal atrial fibrillation and 106 with persistent atrial fibrillation;150 patients with healthy sinus rhythm were selected as a control group.The serum levels of FGF-23 and miR-208 b in the two groups were measured.In the observation group,cardiac parameters were determined by echocardiography.RESULTS The serum levels of FGF-23 and miR-208 b in the observation group were 210.20±89.60 ng/mL and 5.30±1.22 ng/mL,which were significantly higher than the corresponding values in the control group(P<0.05).In the observation group,the serum levels of FGF-23 and miR-208 b in patients with persistent atrial fibrillation were 234.22±70.05 ng/mL and 5.83±1.00 ng/mL,which were significantly higher than the corresponding values in patients with paroxysmal atrial fibrillation(P<0.05).The left atrial dimension(LAD)of patients with persistent atrial fibrillation was 38.81±5.11 mm,which was significantly higher than that of patients with paroxysmal atrial fibrillation(P>0.05).The serum levels of FGF-23and miR-208 b were positively correlated with the LAD(r=0.411 and 0.382,P<0.05).In the observation group,the serum levels of FGF-23 and miR-208 b in patients with a major cardiovascular event(MACE)were 243.30±72.29 ng/mL and 6.12±1.12 ng/mL,which were significantly higher than the corresponding values in patients without a MACE(P<0.05).CONCLUSION The serum levels of FGF-23 and miR-208 b are increased in patients with atrial fibrillation and are related to the type of disease,cardiac parameters,and prognosis.Jie-Min Chen Yao-Tang Zhong Chang Tu Jun Lan 2020World Journal of Clinical Cases2020,8,16:2
14Prognostic value of ^(18)F-fluorodeoxyglucose positron emission tomography using Deauville criteria in diffuse large B cell lymphoma treated with autologous hematopoietic stem cell transplantation显示文摘Objective: High-dose chemotherapy(HDC) followed by autologous hematopoietic stem cell transplantation(auto-HSCT) plays an important role in improving outcomes of diffuse large B cell lymphoma(DLBCL) patients.18 F-fluorodeoxyglucose(18 F-FDG) positron emission tomography(PET)/computed tomography(CT) has been widely accepted in response assessment and prediction of prognosis in DLBCL. Here, we report the value of 18 FFDG PET/CT pre-and post-HSCT in predicting outcomes of patients with DLBCL.Methods: DLBCL patients who had PET/CT scan before and after HSCT were included. PET results were interpreted based upon Deauville criteria. The prognostic value of 18 F-FDG PET/CT in auto-HSCT was evaluated.Results: Eighty-four patients were enrolled. In univariate analysis, pre-and post-HSCT PET findings were correlated with 3-year progression-free survival(PFS) [hazard ratio(HR)=4.391, P=0.001; HR=7.607, P<0.001] and overall survival(OS)(HR=4.792, P=0.008; HR=26.138, P<0.001). Patients receiving upfront auto-HSCT after firstline treatment had better outcomes than relapsed/refractory DLBCL patients(3-year PFS, P<0.001; 3-year OS,P<0.001). In the relapsed/refractory patients, pre-and post-HSCT PET findings were also associated with 3-year PFS(P=0.003 vs. P<0.001) and OS(P=0.027 vs. P<0.001). A significant correlation was observed between clinical response to chemotherapy before auto-HSCT and outcomes of patients in the entire cohort(3-year PFS, P<0.001;3-year OS, P<0.001) and in the subgroup of 21 patients with positive pre-HSCT PET(3-year PFS, P=0.084; 3-year OS, P=0.240). A significant association between survival and post-HSCT PET findings was observed in multivariate analysis(HR=5.168, P<0.001).Conclusions: PET results before and after HSCT are useful prognostic factors for DLBCL patients receiving HSCT. 20Integrative Analyses of Lung Squamous Cell Carcinoma in Ten Chinese Patients with Transcriptome Sequencing显示文摘Few effective therapies have been developed for the treatment of lung squamous cell carcinoma(SQCC), in part due to a lack of understanding regarding the mechanisms underlying the initiation and development of this disease. Whole transcriptome sequencing not only provides insight into the expression of all transcribed genes, but offers an efficient approach for identifying genetic variations,including gene fusions, mutations and alternative splicing. In this study, we performed whole transcriptome sequencing of 10 patients with stage IIIA lung SQCC, and discovered a large number of single nucleotide variants(SNVs; mean of 12.2 SNVs/Mb), with C>T/G>A and A>G/T>C transitions being the most frequently observed. Additionally, a total of 132 gene fusions were identified based upon Top Hat alignments, 70.5%(93/132) of which occurred as a result of intra-chromosomal rearrangements. Based on the number of supporting reads for each fusion, we further validated 20 of the 26 top gene fusions by RT-PCR and Sanger sequencing.
