| 1 | Dihydromyricetin alleviates carbon tetrachloride-induced acute liver injury via JNK-dependent mechanism in mice显示文摘AIM: To assess the effects of dihydromyricetin(DHM) as a hepatoprotective candidate in reducing hepatic injury and accelerating hepatocyte proliferation after carbon tetrachloride(CCl4) treatment.METHODS: C57 BL/6 mice were used in this study. Mice were orally administered with DHM(150 mg/kg) for 4 d after CCl4 treatment. Serum and liver tissue samples were collected on days 1, 2, 3, 5 and 7 after CCl4 treatment. The anti-inflammatory effect of DHM was assessed directly by hepatic histology detection and indirectly by serum levels of aspartate aminotransferase(AST), alanine aminotransferase(ALT), albumin, and superoxide dismutase(SOD). Inflammatory cytokines, such as interleukin(IL)-1β, IL-6 and tumor necrosis factor-α(TNF-α), were detected using ELISA kits. Proliferating cell nuclear antigen(PCNA) staining was used to evaluate the role of DHM in promoting hepatocyte proliferation. Hepatocyte apoptosis wasmeasured by TUNEL assay.Furthermore,apoptosis proteins Caspases-3,6,8,and 9 were detected by Western blot.SP600125 were used to confirm whether DHM regulated liver regeneration through JNK/TNF-αpathways.RESULTS:DHM showed a strong anti-inflammatory effect on CCl4-induced liver injury in mice.DHM could significantly decrease serum ALT,AST,IL-1β,IL-6 and TNF-αand increase serum albumin,SOD and liver SOD compared to the control group after CCl4 treatment(P<0.05).PCNA results indicated that DHM could significantly increase the number of PCNA positive cells compared to the control(348.9±56.0 vs 107.1±31.4,P<0.01).TUNEL assay showed that DHM dramatically reduced the number of apoptotic cells after CCl4 treatment compared to the control(365.4±99.4 vs 90.5±13.8,P<0.01).Caspase activity detection showed that DHM could reduce the activities of Caspases-8,3,6 and 9 compared to the control(P<0.05).The results of Western blot showed that DHM increased the expression of JNK and decreased TNF-αexpression.However,DHM could not affect TNF-αexpression after SP600125 treatment.Furthermore,DHM could significantly improve the survival rate of acute liver failure(ALF)mice(73.3%vs 20.0%,P<0.0001),and SP600125 could inhibit the effect of DHM.CONCLUSION:These findings demonstrate that DHM alleviates CCl4-induced liver injury,suggesting that DHM is a promising candidate for reversing liver injury and ALF. | Jun Xie Jie Liu Tu-Ming Chen Qing Lan Qing-Yu Zhang Bin Liu Dong Dai Wei-Dong Zhang Li-Ping Hu Run-Zhi Zhu | 2015 | World Journal of Gastroenterology2015,21,18: | 17 |

| 3 | Effect of the number of freeze-thaw cycles number on the quality of Pacific white shrimp (Litopenaeus vannamei): An emphasis on moisture migration and microstructure by LF-NMR and SEM显示文摘The objective of this study was to evaluate the effects of the number of freeze-thaw(F-T)cycles on moisture migration,protein degradation,microstructure and quality in Litopenaeus vannamei.The quality of samples with different F-T cycles were determined by low-field nuclear magnetic resonance(LF-NMR),magnetic resonance imaging(MRI)and scanning electron microscopy(SEM),combined with sodium dodecyl sulphate polyacrylamide gel electrophoresis(SDS-PAGE),texture properties analysis(TPA),color difference,polyphenol oxidase(PPO)activity,total volatile basic nitrogen(TVB-N),total viable count(TVC),and sensory evaluation.The results showed that F-T cycles caused a significantly increase in transverse relaxation time in T22 and T23 and a decreased brightness of pseudo-color diagram after 4 F-T cycles,indicating that water mobility increased as immobilized water was shifted to free water.The texture of samples declined as well,especially after 4 F-T cycles.The rapid growth of PPO activity caused a decrease in brightness(L*)and an increase in redness(a∗)after 3 F-T cycles.The increase of TVB-N,TVC,and sensory score explained the changes in shrimp quality which became obvious after 3 F-T cycles and unacceptable after 6 cycles.Meanwhile,fewer than 4 F-T cycles accelerated protein aggregation,while denaturation occurred after 4 cycles.Therefore,repeated F-T cycles could accelerate the changes of protein,microstructure,water distribution,and quality deterioration especially after 3 F-T cycles,with a threshold was 6 F-T cycles. | Weiqing Lan Xiaoyu Hu Xiaohong Sun Xi Zhang Jing Xie | 2020 | Aquaculture and Fisheries2020,5,4: | 15 |

| 5 | Trabecular-like Ti-6Al-4V scaffolds for orthopedic: fabrication by selective laser melting and in vitro biocompatibility显示文摘Porous metal scaffolds play an important role in the orthopedic field, due to their wide applications in prostheses implantation. Some previous studies showed that the scaffolds with trabecular bone structure reconstructed via computed tomography had satisfactory biocompatibility. However, the reverse modeling scaffolds were inflexible for customized design. Therefore, a top-down designing biomimetic bone scaffold with favorable mechanical performances and cytocompatibility is urgently demanded for orthopedic implants. An emerging additive manufacturing technique, selective laser melting, was employed to fabricate the trabecular-like porous Ti-6Al-4 V scaffolds with varying irregularities(0.05-0.5) and porosities(48.83%–74.28%) designed through a novel Voronoi-Tessellation based method. Micro-computed tomography and scanning electron microscopy were used to characterize the scaffolds’ morphology.Quasi-static compression tests were performed to evaluate the scaffolds’ mechanical properties. The MG63 cells culture in vitro experiments, including adhesion, proliferation, and differentiation, were conducted to study the cytocompatibility of scaffolds. Compressive tests of scaffolds revealed an apparent elastic modulus range of 1.93–5.24 GPa and an ultimate strength ranging within 44.9–237.5 MPa, which were influenced by irregularity and porosity, and improved by heat treatment. Furthermore, the in vitro assay suggested that the original surface of the SLM-fabricated scaffolds was favorable for osteoblasts adhesion and migration because of micro scale pores and ravines. The trabecular-like porous scaffolds with full irregularity and higher porosity exhibited enhanced cells proliferation and osteoblast differentiation at earlier time, due to their preferable combination of small and large pores with various shapes. | 6 | Reproductive management through integration of PGD and MPS-based noninvasive prenatal screening/diagnosis for a family with GJB2-associated hearing impairment显示文摘A couple with a proband child of GJB2(encoding the gap junction protein connexin 26)-associated hearing impairment and a previous pregnancy miscarriage sought for a reproductive solution to bear a healthy child. Our study aimed to develop a customized preconception-to-neonate care trajectory to fulfill this clinical demand by integrating preimplantation genetic diagnosis(PGD), noninvasive prenatal testing(NIPT), and noninvasive prenatal diagnosis(NIPD) into the strategy. Auditory and genetic diagnosis of the proband child was carried out to identify the disease causative mutations. The couple then received in-vitro-fertilization treatment, and eight embryos were obtained for day 5 biopsy. PGD was performed by short-tandem-repeat linkage analysis and Sanger sequencing of GJB2 gene. Transfer of a GJB2 c.235del C heterozygous embryo resulted in a singleton pregnancy. At the 13 th week of gestation, genomic DNA(g DNA) from the trio family and cell-free DNA(cf DNA) from maternal plasma were obtained for assessment of fetal chromosomal aneuploidy and GJB2 mutations. NIPT and NIPD showed the absence of chromosomal aneuploidy and GJB2-associated disease in the fetus, which was later confirmed by invasive procedures and postnatal genetic/auditory diagnosis. | 7 | Antibacterial mechanism of Ginkgo biloba leaf extract when applied to Shewanella putrefaciens and Saprophytic staphylococcus显示文摘The antimicrobial mechanism of Ginkgo biloba leaf extracts(GBLE)when applied to predominant spoilage bacteria(Shewanella putrefaciens and Saprophytic staphylococcus)on refrigerated pomfret and minimal inhibitory concentrations(MICs)were measured by the plate counting method.GBLE at MIC and 2MIC were prepared in tryptic soy broth(TSB)medium and equivalent amounts of sterile distilled water were used in place of GBLE as a control group.The impact of GBLE on the growth of bacteria,the permeability of cell membrane,and cell wall were also investigated by growth curve of bacteria,alkaline phosphates activity(AKP),and electrical conductivity.A scanning electron microscope(SEM)was used to study the effects of GBLE on the cellular structure of S.putrefaciens and S.staphylococcus.The results showed that the MICs of GBLE when applied to S.putrefaciens and S.staphylococcus were 100 mg/mL,the inhibitory rates of MIC and 2MIC concentrations of GBLE when applied to S.putrefaciens were 36.11%and 100%,while 27.78%and 62.22%for S.staphylococcus.Meanwhile,GBLE inhibited the growth of S.putrefaciens and S.staphylococcus until the number of cells at 2MIC values decreased to 0 and 4.29 log CFU/mL,respectively,after 24 h.The electrical conductivity of bacteria increased with GBLE treatment,which was followed by an increased leakage of AKP.The SEM revealed that the structure of bacterial cells was destroyed and the bacteria began to be adhere to each other.The inhibition effect of GBLE when applied to S.putrefaciens and S.staphylococcus was related to the damage of cell membrane and cell wall.It was also revealed that GBLE damages the morphology of bacteria and had stronger effects on the cell membrane of S.putrefaciens than that of S.staphylococcus. | Nannan Zhang Weiqing Lan Qian Wang Xiaohong Sun Jing Xie | 2018 | Aquaculture and Fisheries2018,3,4: | 11 |

First, a comprehensive dynamical model of the permanent magnet synchronous motor(PMSM) in d-q frame and its space-state equation are established. The slide model control method is used to estimate the electromotive force of PMSM under static frame, while the position of rotor and its actual speed are estimated by using phase loop lock(PLL) method. Next,using Lyapunov stability theorem, the asymptotical stability condition of the slide model observer is presented. Furthermore, based on the backstepping control theory, the PMSM rotor speed and current tracking backstepping controllers are designed, because such controllers display excellent speed tracking and anti-disturbance performance. | 12 | Changes of plasma C-reactive protein in patients with craniocerebral injury before and after hyperbaric oxygenation: A randomly controlled study显示文摘BACKGROUND: Plasma inflammatory factor, such as C-reactive protein, whose content is regarded as a sensitively pathological marked protein and quantitative indexes of central nervous system injury, has been paid more and more attention in clinic. OBJECTIVE: To observe the effects and clinical significance of C-reactive protein in patients with craniocerebral injury after hyperbaric oxygenation. DESIGN: Randomized controlled study. SETTING: Departments of Neurosurgery, Laboratory and Hyperbaric Oxygen, the Second Affiliated Hospital, Medical College of Shantou University. PARTICIPANTS: A total of 60 patients with craniocerebral injury were selected from Department of Neurosurgery, the Second Affiliated Hospital, Medical College of Shantou University from October 2006 to April 2007. There were 37 males and 23 females and the mean age was 26 years. All subjects were certainly diagnosed as history of craniocerebral injury. Patients hospitalized at 24 hours after injury, Glasgow Coma Score ranged from 3 to 12 points, and all patients were certainly diagnosed with CT or MR scanning. Patients and their relatives provided confirmed consent. All the subjects were randomly divided into hyperbaric oxygenation group and control group with 30 in each group. METHODS: Patients in the control group were treated with routinely neurosurgical therapy after hospitalization; however, based the same basic treatment in the control group, patients in the hyperbaric oxygenation group received hyperbaric oxygenation by using iced-wheel four-door 2-cabin air-compression chamber (made in Yantai) from 24 hours to 10 days after operation or injury. After entering the cabin, patients who had a clear consciousness breathed the oxygen by using face mask; contrarily, patients directly breathed the oxygen. Therapeutic project: Expression was increased for about 15–20 minutes, maintained for about 70–80 minutes, and decreased for 20 minutes. Otherwise, pressure was maintained from 0.2 to 0.25 MPa. Hyperbaric oxygenation took an hour for once a day and 10 times were regarded as a course. Venous blood was collected before treatment and on the next day of the first course end. Content of C-reactive protein in plasma was measured with immune turbidimetry in hyperbaric oxygenation group; in addition, content of C-reactive protein in plasma was directly measured with the same method at the corresponding time in the control group. If the content was less or equal to 8 mg/L, it was regarded as normal value. Effects of the two groups were evaluated based on Glasgow Coma Score before and after treatment. MAIN OUTCOME MEASURES: Content of plasma C-reactive protein and Glasgow Coma Score in the two groups before and after treatment. RESULTS: All 60 patients were involved in the final analysis. ① Content of plasma C-reactive protein: The two contents were obviously higher than normal value after craniocerebral injury. There was no significant difference in the two groups before treatment (P > 0.05), but both contents were decreased after treatment, and there was significant difference between HBOT group and control group after treatment (t =4.756, P < 0.01). In addition, there was significant difference in hyperbaric oxygen therapy group before and after treatment (t =5.236, P < 0.01). ② Glasgow Coma Score: There was no significant difference in the two groups before treatment (P > 0.05), but scores were increased in both groups after treatment (t =9.92, 2.51, P < 0.01, 0.05); on the other hand, therefore, there was significant difference between the two groupsafter treatment (t =9.21, P < 0.01). Creation and functional annotation of point mutations has been limited by the low efficiency of conventional methods based on random mutation. An efficient tool for generating targeted single-base mutations is desirable for both functional genomics and precise genetic improvement. The objective of this study was to test the efficiency of targeted C-to-T base editing of two non-allelic acetolactate synthase(ALS) in generating sulfonylurea herbicide-resistant mutants. A CRISPR/Cas9 nickase-cytidine deaminase fused with uracil DNA glycosylase inhibitor(UGI) was employed to achieve targeted conversion of cytosine to thymine in ZmALS1 and ZmALS2. Both protoplasts and recovered mutant plants showed the activity of the cytosine base editor, with an in vivo efficiency of up to 13.8%. Transgene-free edited plants harboring a homozygous ZmALS1 mutation or a ZmALS1 and ZmALS2 double mutation were tested for their resistance at a dose of up to 15-fold the recommended limit of chlorsulfuron, a sulfonylurea herbicide widely used in agriculture. | 19 | Association of Overlapped and Un-overlapped Comorbidities with COVID-19 Severity and Treatment Outcomes: A Retrospective Cohort Study from Nine Provinces in China显示文摘Objective Several COVID-19 patients have overlapping comorbidities. The independent role of each component contributing to the risk of COVID-19 is unknown, and how some non-cardiometabolic comorbidities affect the risk of COVID-19 remains unclear.Methods A retrospective follow-up design was adopted. A total of 1,160 laboratory-confirmed patients were enrolled from nine provinces in China. Data on comorbidities were obtained from the patients’ medical records. Multivariable logistic regression models were used to estimate the odds ratio(OR) and 95% confidence interval(95% CI) of the associations between comorbidities(cardiometabolic or non-cardiometabolic diseases), clinical severity, and treatment outcomes of COVID-19.Results Overall, 158(13.6%) patients were diagnosed with severe illness and 32(2.7%) had unfavorable outcomes. Hypertension(2.87, 1.30–6.32), type 2 diabetes(T2 DM)(3.57, 2.32–5.49),cardiovascular disease(CVD)(3.78, 1.81–7.89), fatty liver disease(7.53, 1.96–28.96), hyperlipidemia(2.15, 1.26–3.67), other lung diseases(6.00, 3.01–11.96), and electrolyte imbalance(10.40, 3.00–26.10)were independently linked to increased odds of being severely ill. T2 DM(6.07, 2.89–12.75), CVD(8.47,6.03–11.89), and electrolyte imbalance(19.44, 11.47–32.96) were also strong predictors of unfavorable outcomes. Women with comorbidities were more likely to have severe disease on admission(5.46,3.25–9.19), while men with comorbidities were more likely to have unfavorable treatment outcomes(6.58, 1.46–29.64) within two weeks.Conclusion Besides hypertension, diabetes, and CVD, fatty liver disease, hyperlipidemia, other lung diseases, and electrolyte imbalance were independent risk factors for COVID-19 severity and poor treatment outcome. | 20 | Efficacy and safety of transcutaneous auricular vagus nerve stimulation combined with conventional rehabilitation training in acute stroke patients: a randomized controlled trial conducted for 1 year involving 60 patients显示文摘Transcutaneous auricular vagus nerve stimulation(ta-VNS)is a novel noninvasive treat-ment for stroke that directly stimulates the peripheral auricular branch of the vagus nerve.There have been recent reports that ta-VNS combined with conventional rehabilitation training promotes the recovery of neurological function of patients with acute stroke.However,these were small-sample-sized studies on the recovery of neurological function in patients after percutaneous vagus nerve stimulation in the subacute and chronic phases after stroke.This double-blinded randomized controlled trial involved 60 acute ischemic or hemorrhagic stroke patients aged 18-80 years who received treatment in the Second Affiliated Hospital of Chongqing Medical University.The subjects were randomly assigned to receive ta-VNS or sham ta-VNS combined with conventional rehabilitation training.The follow-up results over 1 year revealed that ta-VNS combined with conventional rehabilitation training greatly improved the recovery of motor and sensory functions and emotional responses compared with sham ta-VNS combined with conventional rehabilitation training.There were no obvious side effects.These findings suggest that ta-VNS combined with conventional rehabilitation training for the treatment of acute ischemic or hemorrhagic stroke patients is safe and effective. | Jia-Ni Li Chen-Chen Xie Chang-Qing Li Gui-Fang Zhang Hao Tang Chuan-Na Jin Jing-Xi Ma Lan Wen Ke-Ming Zhang Ling-Chuan Niu | 2022 | Neural Regeneration Research2022,17,8: | 7 |
