2Reconstruction of a hybrid nucleoside antibiotic gene cluster based on scarless modification of large DNA fragments显示文摘Genetic modification of large DNA fragments(gene clusters) is of great importance in synthetic biology and combinatorial biosynthesis as it facilitates rational design and modification of natural products to increase their value and productivity.In this study,we developed a method for scarless and precise modification of large gene clusters by using RecET/RED-mediated polymerase chain reaction(PCR) targeting combined with Gibson assembly.In this strategy,the biosynthetic genes for peptidyl moieties(HPHT) in the nikkomycin biosynthetic gene cluster were replaced with those for carbamoylpolyoxamic acid(CPOAA)from the polyoxin biosynthetic gene cluster to generate a^40 kb hybrid gene cluster in Escherichia coli with a reusable targeting cassette.The reconstructed cluster was introduced into Streptomyces lividans TK23 for heterologous expression and the expected hybrid antibiotic,polynik A,was obtained and verified.This study provides an efficient strategy for gene cluster reconstruction and modification that could be applied in synthetic biology and combinatory biosynthesis to synthesize novel bioactive metabolites or to improve antibiotic production.Jiming Zhuo Binbin Ma Jingjing Xu Weihong Hu Jihui Zhang Huarong Tan Yuqing Tian 2017Science China(Life Sciences)2017,60,9:6
3Polymorphisms of chemokine receptors and its ligand alleles influencing genetic suscepti-bity to HIV-1 infection in eight ethnic groups in Chinese mainland显示文摘Limited genetic information is available concerning the polymorphisms of HIV-1 resistant genes in indigenous Chinese populations. The aim of this study is to identify the allelic frequencies of the chemokine and chemokine receptor genes in the Chinese mainland. Genomic DNA samples extracted from whole blood of 2318 subjects were analyzed by using PCR or PCR/restriction fragment length polymorphism (RFLP) assays, and further confirmed by direct DNA sequencing. Higher frequencies of mutant CCR2-64I (19.15%-28.79%) and SDF1-3’A (19.10% -29.86%) alleles were found in subjects of 8 ethnic groups in the Chinese mainland. In contrast, the △32 mutation in CCRS gene occurs at a very low frequency (0.0016, n=1287) in Han population. A relatively high frequency of CCR5-wt/△32 heterozygotes was observed in Uygurian and Mongolian populations. No A32 mutation allele was detected in Tibetan and other 4 ethnic groups in Yunnan Province. 4Amorphous Co-B/Al nanocomposites prepared by electroless coating technique and their excellent catalytic activity显示文摘To improve the catalytic activity of amorphous Co-B alloys, Co-B coated aluminum (Co-B/Al) nanocomposites were prepared by electroless coating technique and evaluated as additives for the catalytic performance of ammonium perchlorate (AP) and AP-based solid state propellants. X-ray diffractometry (XRD), scanning electron microscopy (SEM), inductive coupled plasma emission spectrometry (ICP), differential scanning calorimetry (DSC) as well as strand burner method were employed to characterize the crystal phase, morphologies, chemical composition, and catalytic activity of the as-synthesized material. The results show that a continuous layer of about 100 nm amorphous Co72.6B27.4 covers the surfaces of Al particles. 5Locomotor activity and anxiety status, but not spatial working memory,are affected in mice after brief exposure to cuprizone显示文摘Chronic long-term exposure to cuprizone causes severe brain demyelination in mice,which leads to changes in locomotion,working memory and anxiety.These findings suggest the importance of intact myelin for these behaviors.This study aimed to investigate the possible behavioral changes in mice with mild oligodendrocyte/myelin damage that parallels the white matter changes seen in the brains of patients with psychiatric disporders.We used the cuprizonetreated mouse model to test both tissue changes and behavioral functions(locomotor activity,anxiety status,and spatial working memory).The results showed that mice given cuprizone in their diet for 7 days had no significant myelin breakdown as evaluated by immunohistochemical staining for myelin basic protein,while the number of mature oligodendrocytes was reduced.The number and length of Caspr protein clusters,a structural marker of the node of Ranvier,did not change.The locomotor activity of the cuprizonetreated mice increased whereas their anxiety levels were lower than in normal controls;spatial working memory,however,did not change.These results,for the first time,link emotion-related behavior with mild white matter damage in cuprizone-treated mice.Handi Zhang Yanbo Zhang Haiyun Xu Lingyan Wang Jinsong Zhao Junhui Wang Zhijun Zhang Qingrong Tan Jiming Kong Qingjun Huang Xin-Min Li 2013Neuroscience Bulletin2013,29,5:4
6Impact of energy structure adjustment on air quality:a case study in Beijing,China显示文摘Energy consumption is a major cause of air pollution in Beijing,and the adjustment of the energy structure is of strategic importance to the reduction of carbon intensity and the improvement of air quality.In this paper,we explored the future trend of energy structure adjustment in Beijing till 2020,designed five energy scenarios focusing on the fuel substitution in power plants and heating sectors,established emission inventories,and utilized the Mesoscale Modeling System Generation 5(MM5)and the Models-3/Community Multiscale Air Quality Model(CMAQ)to evaluate the impact of these measures on air quality.By implementing this systematic energy structure adjustment,the emissions of PM_(10),PM_(2.5),SO_(2),NO_(x),and non-methane volatile organic compounds(NMVOCs)will decrease distinctly by 34.0%,53.2%,78.3%,47.0%,and 30.6%respectively in the most coalintensive scenario of 2020 compared with 2005.Correspondingly,MM5-Models-3/CMAQ simulations indicate significant reduction in the concentrations of major pollutants,implying that energy structure adjustment can play an important role in improving Beijing’s air quality.By fuel substitution for power plants and heating boilers,PM_(10),PM_(2.5),SO_(2),NO_(x),and NMVOCs will be reduced further,but slightly by 1.7%,4.5%,11.4%,13.5%,and 8.8%respectively in the least coal-intensive scenario.The air quality impacts of different scenarios in 2020 resemble each other,indicating that the potential of air quality improvement due to structure adjustment in power plants and heating sectors is limited.However,the CO_(2) emission is 10.0%lower in the least coal-intensive scenario than in the most coal-intensive one,contributing to Beijing’s ambition to build a low carbon city.Except for energy structure adjustment,it is necessary to take further measures to ensure the attainment of air quality standards.Bin ZHAO Jiayu XU Jiming HAO 2011Frontiers of Environmental Science & Engineering2011,5,3:4

11A New Lizard(Lepidosauria:Squamata) from the Upper Cretaceous of Henan,China显示文摘Funiusaurus luanchuanensis gen.et sp.nov.was described on the basis of an incomplete skull from the Upper Cretaceous Qiupa Formation of the Tantou Basin in Luanchuan County,Henan Province.It is the second representative of lizards known from Luanchuan and adds a new member to the Luanchuan Fauna.F.luanchuanensis is a small-sized lizard and systematically assigned to the Polyglyphanodontidae of the Teiioidea because of the presence of a caniniform tooth and an elongate posterior process of the postorbital.It is distinctive in that the heterodont dentition bears 19 teeth in both the upper and lower jaws,the 3rd maxillary tooth is large and caniniform,the post-caniniform teeth in maxilla and those posterior to the 3rd dentary tooth are chisel-like in lateral view,the prefrontal possesses a fossa on its lateral surface,the postorbital with an extremely elongate posterior process and the well-developed retroarticular process has a deep fossa on its dorsal surface.In phylogeny,our analysis suggests a close relationship of Funiusaurus to the large-sized Tianyusaurus from the same basin within the Tuberocephalosaurinae.The discovery of Funiusaurus is significant in confirming the status of the Tuberocephalosaurinae,which includes a group of the Asian members of the Polyglyphanodontidae only.XU Li WU Xiaochun Lü Junchang JIA Songhai ZHANG Jiming PU Hanyong ZHANG Xingliao 2014Acta Geologica Sinica(English Edition)2014,88,4:2
12Numerical analysis of the effects of downhole dynamic conditions on formation testing while drilling显示文摘Formation testing while drilling is an innovative technique that is replacing conventional pressure testing in which the fluid sampling is conducted in a relatively short time following the drilling. At this time, mud invasion has just started, mudcake has not formed entirely and the formation pressure is not stable. Therefore, it is important to study the influence of the downhole dynamic environment on pressure testing and fluid sampling. This paper applies an oil-water two phase finite element model to study the influence of mudcake quality and mud filtrate invasion on supercharge pressure, pretest and sampling in the reservoirs of different permeability. However, the study is only for the cases with water based mud in the wellbore. The results illustrate that the mudcake quality has a significant influence on the supercharge pressure and fluid sampling, while the level of mud filtrate invasion has a strong impact on pressure testing and sampling. In addition, in-situ formation pressure testing is more difficult in low permeability reservoirs as the mud filtrate invasion is deeper and therefore degrades the quality of fluid sampling. 13A facile ionic liquid approach to prepare cellulose fiber with good mechanical properties directly from corn stalks显示文摘It is very difficult to directly spin the lignocellulose without pretreatment.Ionic liquids(ILs)are promising solvent to dissolve lignocellulose to prepare cellulose fiber.However,the degree of cellulose polymerization(DP)is reduced when lignocellulose is dissolved in ILs,and the lignin removal rate is low.The elongation at break and tensile strength of the fibers obtained by spinning the lignocellulose dissolved in ILs are poor.In this paper,preparing cellulose fiber directly from lignocellulose based on dissolving corn stalk via[C4mim]Cl-L-arginine binary system is achieved.It shows that the removal rate of lignin can reach 92.35%and the purity of cellulose can reach 85.32%after corn stalk was dissolved at 150℃C for 11.5 h when the mass fraction of arginine is 2.5%.The elongation at break of fiber reached 10.12%and the tensile strength reached 420 MPa.It is mainly due to the fact that L-arginine not only inhibits the degradation of cellulose but also promotes the delignination.Without any pulping or pretreatment,preparing cellulose fibers via direct dissolution and extrusion may provide a simple and effective way to prepare many novel cellulose materials.Jiming Yang Xingmei Lu Yanqiang Zhang Junli Xu Yongqing Yang Qing Zhou 2020Green Energy & Environment2020,5,2:2

15Zircon U-Pb ages and Hf isotopic characteristics of the Dehe biotite monzonitic gneiss pluton in the North Qinling orogen and their geological significance显示文摘The Dehe granitic pluton intruded the Xiahe Group which is in the core complex of the North Qinling Orogenic Belt(NQOB).It shows gneissic bedding and possesses typical S-type granite minerals such as muscovite and garnet.LA-ICP-MS U-Pb isotopic dating of the Dehe granite yielded a weighted average age of 925±23 Ma which represents the emplacement age of the pluton.Most of the εHf(t) values are negative,and the two-stage model ages are consistent with the age of the Qinling Group.The isotope data show that the Dehe granite was formed in the following geological setting:in the syn-collision setting of the NQOB in the Neoproterozoic,crustal thickening induced partial melting of materials derived from the Qinling complex,and then the maga upwelled and intruded into the Xiahe Group.The formation of the Dehe S-type granite implied the occurrence of a convergent event in the QOB during the Neoproterozoic.Wang, Fei Zhu, Laimin Li, Jiming Lee, Ben Gong, Hujun Yang, Tao Wang, Wei Xu, Ao 2011Chinese Journal Of Geochemistry2011,30,2:1

19Tenofovir disoproxil fumarate therapy in patients with chronic hepatitis B and advanced fibrosis or compensated cirrhosis显示文摘Background and aims:Tenofovir disoproxil fumarate(TDF)is the first-line therapy for chronic hepatitis B.This interim analysis presents the efficacy and safety data for TDF at Week 144 in patients with chronic hepatitis B and advanced fibrosis or compensated cirrhosis from China.Methods:Patients were assessed for incidence of newly diagnosed hepatocellular carcinoma(HCC)and disease progression,liver stiffness measurement(LSM),virological suppression(serum hepatitis B virus DNA<20 IU/mL),alanine aminotransferase normalization,hepatitis B e antigen(HBeAg)loss and seroconversion,histological liver fibrosis score,and safety at Week 144.Results:Overall,197 patients were enrolled.At Week 144,the incidence of newly diagnosed HCC was observed in 2.1%patients,and the incidence of disease progression was observed in 3.6%patients.The mean(standard deviation)change in LSM from baseline was5.1(5.85)kPa.Serum hepatitis B virus DNA<20 IU/mL was observed in 94.1%patients,alanine aminotransferase normalization in 33.5%patients,HBeAg loss in 35.6%patients,and HBeAg seroconversion in 14.4%patients.Among patients with stage F3 or F4 fibrosis at baseline by LSM,38.3%patients regressed to stage F0/1,and 22.0%of patients regressed to stage F2 at Week 144.Overall,67.7%patients experienced≥1 adverse events,13.8%patients experienced TDF-related adverse events,and 16.4%patients experienced serious(none were TDF-related).Conclusions:At Week 144 of TDF treatment,low incidence of HCC and disease progression were reported.Virological suppression was observed in 94.1%patients,which was associated with fibrosis regression.No new safety events were identified.Huiying Rao Jia Shang Qing Xie Jianqi Lian Pujun Gao Junping Shi Xinyue Chen Jiefei Wang Min Xu Liaoyun Zhang Yingren Zhao QingMao Maorong Wang Wei Zhao Zong Zhang Jidong Jia Hong Tang Jiming Zhang Xin Zheng Chang Liu Lai Wei 2022iLIVER2022,1,3:1
20The miR157-SPL-CNR module acts upstream of bHLH101 to negatively regulate iron deficiency responses in tomato显示文摘Iron(Fe)homeostasis is critical for plant growth,development,and stress responses.Fe levels are tightly controlled by intricate regulatory networks in which transcription factors(TFs)play a central role.A series of basic helix-loop-helix(b HLH)TFs have been shown to contribute to Fe homeostasis,but the regulatory layers beyond b HLH TFs remain largely unclear.Here,we demonstrate that the SQUAMOSA PROMOTER-BINDING PROTEIN-LIKE(SPL)TF Sl SPL-CNR negatively regulates Fe-deficiency responses in tomato(Solanum lycopersicum)roots.Fe deficiency rapidly repressed the expression of Sl SPL-CNR,and Fe deficiency responses were intensified in two clustered regularly interspaced palindromic repeats(CRISPR)/CRISPRassociated protein 9-generated Sl SPL-CNR knockout lines compared to the wild-type.Comparative transcriptome analysis identified 47 Fe deficiencyresponsive genes the expression of which is negatively regulated by Sl SPL-CNR,one of which,Slb HLH101,helps regulate Fe uptake genes.Sl SPLCNR localizes the nucleus and interacts with the GTAC and BOX 4(ATTAAT)motifs in the Slb HLH101 promoter to repress its expression.Inhibition of Sl SPL-CNR expression in response to Fe deficiency was well correlated with the expression of the micro RNA Slymi R157.Slymi R157-overexpressing tomato lines displayed enhanced Fe deficiency responses,as did Sl SPL-CNR loss-of-function mutants.We propose that the Slymi R157-Sl SPL-CNR module represents a novel pathway that acts upstream of Slb HLH101 to regulate Fe homeostasis in tomato roots.Huihui Zhu Jiayi Wang Dan Jiang Yiguo Hong Jiming Xu Shaojian Zheng Jianli Yang Weiwei Chen 2022Journal of Integrative Plant Biology2022,64,5:1
