2Polyamine Accumulation in Transgenic Tomato Enhances the Tolerance to High Temperature Stress显示文摘Polyamines play an important role in plant response to abiotic stress.S-adenosyl-l-methionine decarboxylase(SAMDC) is one of the key regulatory enzymes in the biosynthesis of polyamines.In order to better understand the effect of regulation of polyamine biosynthesis on the tolerance of high-temperature stress in tomato,SAMDC cDNA isolated from Saccharomyces cerevisiae was introduced into tomato genome by means of Agrobacterium tumefaciens through leaf disc transformation.Transgene and expression was confirmed by Southern and Northern blot analyses,respectively.Transgenic plants expressing yeast SAMDC produced 1.7-to 2.4-fold higher levels of spermidine and spermine than wild-type plants under high temperature stress,and enhanced antioxidant enzyme activity and the protection of membrane lipid peroxidation was also observed.This subsequently improved the efficiency of CO2 assimilation and protected the plants from high temperature stress,which indicated that the transgenic tomato presented an enhanced tolerance to high temperature stress(38℃) compared with wild-type plants.Our results demonstrated clearly that increasing polyamine biosynthesis in plants may be a means of creating high temperature-tolerant germplasm.Lin Cheng Yijing Zou Shuli Ding Jiajing Zhang Xiaolin Yu Jiashu Cao Gang Lu 2009Journal of Integrative Plant Biology2009,51,5:16
3Thermodynamic conditions of framework grain dissolution of clastic rocks and its application in Kela 2 gas field显示文摘Feldspar and clastic debris are the most important constituent framework grains of sedimentary clastic rocks and their chemical dissolution plays an essential role in the formation and evolution of the secondary pore in the reservoir rocks. On the basis of thermodynamic phase equilibrium, this study investigates the chemical equilibrium relationships between fluid and various plagioclase and K-feldspar in diagenesis of the sediments, particularly, the impact of temperature and fluid compositions (pH, activity of K+, Na+, Ca2+ and so on) on precipitation and dissolution equilibria of feldspars. Feldspar is extremely easily dissolved in the acid pore water with a low salinity when temperature decreases. The dissolution of anorthite end-member of plagioclase is related to the Ca content of the mineral and the fluid, higher Ca either in the mineral or in the fluid, easier dissolution of the feldspar. Moreover, the dissolution of albite end-member of plagioclase is related to Na of both the mineral and fluid, lower Na out of both the mineral and fluid, easier dissolution of the mineral. Similarly, lower-K fluid is more powerful to dissolve K-feldspar than the higher. The anorthite component of plagioclase is most easily dissolved in ground water-rock system, the albite is the secondary, and K-feldspar is the most stable. Selective dissolution of plagioclase occurs in diagenesis because of the plagioclase solid solution that is mainly composed of albite and anorthite end-members, Ca-rich part of which is preferentially dissolved by the pore water, in contrast to the Na-rich parts. Based on investigation of reservoir quality, secondary pore, dissolution structures of feldspar, clay minerals and ground water chemistry of the Kela 2 gas field of Kuqa Depression in the Tarim basin of Western China, we discovered that the secondary pore is very well developed in the highest quality section of the reservoir, and the plagioclase of which was obviously selectively dissolved, in contrast to the overgrowth of K-feldspar. Chemistry of the ground water out of the highest quality section of the reservoir is consistent with the complete dissolution field of anorthite, partial dissolution field of albite component of plagioclase, and the precipitation of K-feldspar on the temperature-logK phase equilibrium diagram drawn from this study. The thermodynamic modeling calculation gave us an insight into formation of the secondary pore within the reservoir, and the dissolution of plagioclase an important cause of the secondary pore. 5Stratigraphy of the Triassic-Jurassic Boundary Successions of the Southern Margin of the Junggar Basin,Northwestern China显示文摘The Triassic-Jurassic(Tr-J) boundary marks a major extinction event,which(~200 Ma) resulted in global extinctions of fauna and flora both in the marine and terrestrial realms.There prevail great challenges in determining the exact location of the terrestrial Tr-J boundary,because of endemism of taxa and the scarcity of fossils in terrestrial settings leading to difficulties in linking marine and terrestrial sedimentary successions.Investigation based on palynology and bivalves has been carried out over a 1113 m thick section,which is subdivided into 132 beds,along the Haojiagou valley on the southern margin of the Junggar Basin of the northern Xinjiang,northwestern China.The terrestrial Lower Jurassic is conformably resting on the Upper Triassic strata.The Upper Triassic covers the Huangshanjie Formation overlaid by the Haojiagou Formation,while the Lower Jurassic comprises the Badaowan Formation followed by the Sangonghe Formation.Fifty six pollen and spore taxa and one algal taxon were identified from the sediments.Based on the key-species and abundance of spores and pollen,three zones were erected:the Late Triassic(Rhaetian) Aratrisporites-Alisporites Assemblage,the Early Jurassic(Hettangian) Perinopollenites-Pinuspollenites Assemblage,and the Sinemurian Perinopollenites-Cycadopites Assemblage.The Tr-J boundary is placed between bed 44 and 45 coincident with the boundary between the Haojiagou and Badaowan formations.Beds with Ferganoconcha(?),Unio-Ferganoconcha and Waagenoperna-Yananoconcha bivalve assemblages are recognized.The Ferganoconcha(?) bed is limited to the upper Haojiagou Formation, Unio-Ferganoconcha and Waagenoperna-Yananoconcha assemblages are present in the middle and upper members of the Badaowan Formation.The sedimentary succession is interpreted as terrestrial with two mainly lake deposit intervals within Haojiagou and Badaowan formations,yielding fresh water algae and bivalves.However,the presence of brackish water algae Tasmanites and the marine-littoral facies bivalve Waagenoperna from the Badaowan Formation indicate that the Junggar Basin was influenced by sea water caused by transgressions from the northern Tethys,during the Sinemurian.SHA Jingeng Vivi VAJDA PAN Yanhong Linda LARSSON YAO Xiaogang ZHANG Xiaolin WANG Yaqiong CHENG Xiansheng JIANG Baoyu DENG Shenghui CHEN Siwei PENG Bo 2011Acta Geologica Sinica(English Edition)2011,85,2:7
6Dose-effect analysis of treatment by modified Zhibaidihuang decoction on polycystic ovary syndrome hyperandrogenism显示文摘OBJECTIVE: To observe the efficacy and safety of different dosages of modified Zhibaidihuang decoction(MZBDD) on polycystic ovary syndrome(PCOS) hyperandrogenism(HA) patients.METHODS: Ninety PCOS HA patients and 30 infertile patients due to tubal factor were selected. Ninety PCOS HA patients were randomly divided into three groups: low dosage group(LDG), medium dosage group(MDG) and high dosage group(HDG) and infertile patients were selected as normal control group(NCG). PCOS HA patients were treated with different dosage of MZBDD for 4 weeks. For HA patients, serum total testosterone(T), estrodial(E2), follicle stimulating hormone(FSH), luteinizing hormone(LH), prolatin(PRL), alanine aminotransferase(AST), aspartate amino transferase(AST) were determined before and after treatment, while acne scores(Rosenfield) were signed. Basal body temperatures(BBT) were asked to be measured every day. And for the else, T, E2,FSH, LH, PRL, AST, and AST were determined before treatment.RESULTS: Totally 111 patients completed the clinical research. There were no differences among the four groups on serum T before treatment(P =0.221). Serum T concentration of both MDG and HDG after treatment significantly were lower than that of before treatment(P = 0.039, P = 0.000),while there was no obvious difference in LDG(P =0.829). Serum T concentration of both MDG and HDG were significantly lower than that of LDG after treatment(P = 0.048, P = 0.006). To compared with before treatment, there were no differences in Serum FSH, LH, E2, P and PRL among the three groups(LDG, MDG, HDG)(as for FSH, P = 0.136, P = 0.503,P = 0.062; as for LH, P = 0.473, P = 0.513, P = 0.096;as for E2, P = 0.206, P = 0.927, P = 0.076; as for PRL,P = 0.120, P = 0.903, P = 0.407, as for P, P = 0.308, P =0.866, P = 0480). Acne scores of all the three groups were obviously lower than that of before treatment(P = 0.031; P = 0.033; P = 0.002). 39.5% of the patients had biphasic BBT, but there were no differences among the three groups(P = 0.510). 7Electrical stimulation modulates injury potentials in rats after spinal cord injury显示文摘An injury potential is the direct current potential difference between the site of spinal cord injury and the healthy nerves.Its initial amplitude is a significant indicator of the severity of spinal cord injury,and many cations,such as sodium and calcium,account for the major portion of injury potentials.This injury potential,as well as injury current,can be modulated by direct current field stimulation;however,the appropriate parameters of the electrical field are hard to define.In this paper,injury potential is used as a parameter to adjust the intensity of electrical stimulation.Injury potential could be modulated to slightly above 0 mV(as the anode-centered group) by placing the anodes at the site of the injured spinal cord and the cathodes at the rostral and caudal sections,or around -70 mV,which is resting membrane potential(as the cathode-centered group) by reversing the polarity of electrodes in the anode-centered group.In addition,rats receiving no electrical stimulation were used as the control group.Results showed that the absolute value of the injury potentials acquired after 30 minutes of electrical stimulation was higher than the control group rats and much lower than the initial absolute value,whether the anodes or the cathodes were placed at the site of injury.This phenomenon illustrates that by changing the polarity of the electrical field,electrical stimulation can effectively modulate the injury potentials in rats after spinal cord injury.This is also beneficial for the spontaneous repair of the cell membrane and the reduction of cation influx.Guanghao Zhang Xiaolin Huo Aihua Wang Changzhe Wu Cheng Zhang Jinzhu Bai 2013Neural Regeneration Research2013,8,27:4

9Antibody Cocktail Exhibits Broad Neutralization Activity Against SARS-CoV-2 and SARS-CoV-2 Variants显示文摘Severe acute respiratory syndrome coronavirus 2(SARS-Co V-2)has precipitated multiple variants resistant to therapeutic antibodies.In this study,12 high-affinity antibodies were generated from convalescent donors in early outbreaks using immune antibody phage display libraries.Of them,two RBD-binding antibodies(F61 and H121)showed high-affinity neutralization against SARS-Co V-2,whereas three S2-target antibodies failed to neutralize SARS-Co V-2.Following structure analysis,F61 identified a linear epitope located in residues G446–S494,which overlapped with angiotensinconverting enzyme 2(ACE2)binding sites,while H121 recognized a conformational epitope located on the side face of RBD,outside from ACE2 binding domain.Hence the cocktail of the two antibodies achieved better performance of neutralization to SARS-Co V-2.Importantly,these two antibodies also showed efficient neutralizing activities to the variants including B.1.1.7 and B.1.351,and reacted with mutations of N501 Y,E484 K,and L452 R,indicated that it may also neutralize the recent India endemic strain B.1.617.The unchanged binding activity of F61 and H121 to RBD with multiple mutations revealed a broad neutralizing activity against variants,which mitigated the risk of viral escape.Our findings revealed the therapeutic basis of cocktail antibodies against constantly emerging SARS-Co V-2 variants and provided promising candidate antibodies to clinical treatment of COVID-19 patients infected with broad SARS-Co V-2 variants.Yuanyuan Qu Xueyan Zhang Meiyu Wang Lina Sun Yongzhong Jiang Cheng Li Wei Wu Zhen Chen Qiangling Yin Xiaolin Jiang Yang Liu Chuan Li Jiandong Li Tianlei Ying Dexin Li Faxian Zhan Youchun Wang Wuxiang Guan Shiwen Wang Mifang Liang 2021Virologica Sinica2021,36,5:3

15Design and test of 2ZLX-2 transplanting machine for oil peony显示文摘In order to improve the quality of oil peony transplanting,based on the characteristics of peony seedlings and the transplanting effect,the transplanting process was analyzed.A peony seedling transplanting machine was designed,which was mainly composed of a chain-clamp type planting mechanism(clamp,transmission chain,sprocket),slideway,profiling wheels,trencher,the power transmission system and rack.Through the movement analysis of the planting mechanism and its operation process,its structural parameters were optimized.The effective length of the clamp was determined as L_(1)=235 mm,the adjustment range was 235-285 mm,and the height of the planting trajectory was H=340 mm.Based on the physical characteristics and mechanical characteristics of the peony seedlings,the parameters of the slideway and the putter were determined.The effective length of the slideway was determined as L_(2)=420 cm,the width T_(2)=105 mm,the distance between the slideways S=45 mm.The initial angle difference of the putter wasθ=13°,and the putter rotation angleβ=37°,putter height ratioε=2.Based on the above parameters and design requirements,the radius of the planting sprocket R_(1)=42 mm,R_(2)=80 mm.“Feng Dan”peony seedlings were used as transplantation test objects.The results showed that:when the planting frequency was 60-75 plants/min,the upright rate was 89.2%-92.8%,the leakage rate was 0-0.28%,the injury rate of seedlings was 0,and the qualified rate of planting depth was 91.7%-95.3%,the variation coefficient of plant spacing was 0.25%-0.54%.This machine can effectively improve the quality and efficiency of transplanting,meeting the requirements of mechanical design and production needs.Jiangtao Ji Qun Cheng Xin Jin Zhihong Zhang Xiaolin Xie Mingyong Li 2020International Journal of Agricultural and Biological Engineering2020,13,4:1
16An adaptive cell-based domain integration method for treatment of domain integrals in 3D boundary element method for potential and elasticity problems显示文摘An adaptive cell-based domain integration method(CDIM) is proposed for the treatment of domain integrals in 3D boundary element method(BEM). The domain integrals are computed in background cells rather than volume elements. The cells are created from the boundary elements based on an adaptive oct-tree structure and no other discretization is needed. Cells containing the boundary elements are subdivided into smaller sub-cells adaptively according to the sizes and levels of the boundary elements; and the sub-cells outside the domain are deleted to obtain the desired accuracy. 17Acoustic Goos-H?nchen effect显示文摘We report two models of the lateral displacement of acoustic-wave scattering on a fluid-solid interface that reveal an acoustic analog of the Goos-H?nchen effect in optics. This acoustic analog is called the acoustic Goos-H?nchen effect. Using newly proposed models, we made numerical calculations for the system of a water-Perspex interface. Specifically, in the post-critical-angle region,we observed a lateral displacement(and transition time) of the reflected P-wave with respect to the incident P-wave. The first arrival of the acoustic signal from the interface is found to be a reflected P-wave rather than the sliding-refraction P-wave usually described in traditional acoustic-logging sliding P-wave theory. 20Sustainability of SARS-CoV-2 Induced Humoral Immune Responses in COVID-19 Patients from Hospitalization to Convalescence Over Six Months显示文摘Understanding the persistence of antibody in convalescent COVID-19 patients may help to answer the current major concerns such as the risk of reinfection,the protection period of vaccination and the possibility of building an active herd immunity.This retrospective cohort study included 172 COVID-19 patients who were hospitalized in Wuhan.A total of404 serum samples were obtained over six months from hospitalization to convalescence.Antibodies in the specimens were quantitatively analyzed by the capture chemiluminescence immunoassays(CLIA).All patients were positive for the anti-SARS-Co V-2 Ig M/Ig G at the onset of COVID-19 symptoms,and the Ig G antibody persisted in all the patients during the convalescence.However,only approximately 25%of patients can detect the Ig M antibodies,Ig M against N protein(NIg M)and receptor binding domain of S protein(RBD-Ig M)at the 27 th week.The titers of Ig M,N-Ig M and RBD-Ig M reduced to 16.7%,17.6%and 15.2%of their peak values respectively.In contrast,the titers of Ig G,N-Ig G and RBD-Ig G peaked at 4–5 th week and reduced to 85.9%,62.6%and 87.2%of their peak values respectively at the end of observation.Dynamic behavior of antibodies and their correlation in age,gender and severity groups were investigated.In general,the COVID-19 antibody was sustained at high levels for over six months in most of the convalescent patients.Only a few patients with antibody reducing to an undetectable level which needs further attention.The humoral immune response against SARS-Co V-2 infection in COVID-19 patients exhibits a typical dynamic of acquired immunity.Yang Zheng Qing Zhang Ashaq Ali Ke Li Nan Shao Xiaoli Zhou Zhiqin Ye Xiaomin Chen Shanshan Cao Jing Cui Juan Zhou Dianbing Wang Baidong Hou Min Li Mengmeng Cui Lihua Deng Xinyi Sun Qian Zhang Qinfang Yang Yong li Hui Wang Yake Lei Bo Yu Yegang Cheng Xiaolin Tong Dong Men Xian-En Zhang 2021Virologica Sinica2021,36,5:1
