1Jianpi Qingchang decoction alleviates ulcerative colitis by inhibiting nuclear factor-κB activation显示文摘AIM To inve s t igat e t he t he r ape ut ic e f f e c t of Jianpi Qingchang decoction(JPQCD) on dextran sulfate sodium(DSS)-induced ulcerative colitis(UC) in mice.METHODS C57BL/c mice were injected intragastrically with 5% DSS instead of drinking water for 7 d, and their body weight, diarrhea severity and fecal bleeding were monitored, while the mice in the control group were treated with standard drinking water, without DSS. After 7 d, the DSS drinking water was changed to normal water and the DSS group continued with DSS water. The control and DSS groups were given normal saline by intragastric injection. The 5-aminosalicylic acid(5-ASA) group was treated orally with 5-ASA at a dose of 100 mg/kg daily. The JPQCD group was treated orally with JPQCD at a dose of 17.1 g/kg daily. On day 14, the colon length was measured, the colorectalhistopathological damage score was assessed, and protein levels of interleukin(IL)-1β, IL-8 and tumor necrosis factor-alpha(TNF-α) in colon supernatants were measured by enzyme-linked immunosorbent assay. m RNA expression of IL-1β, IL-8, TNF-α and nuclear factor-kappa B(NF-κB) was detected by realtime quantitative polymerase chain reaction. Western blotting was used to detect the protein expression of NF-κB and inhibitor of kappa B. RESULTS Acute inflammation occurred in the mice administered DSS, including the symptoms of losing body weight, loose feces/watery diarrhea and presence of fecal blood; all these symptoms worsened at 7 d. The colons of mice treated with DSS were assessed by histological examination, and the results confirmed that acute inflammation had occurred, as evidenced by loss of colonic mucosa and chronic inflammatory cell infiltration, and these features extended into the deeper layer of the colon walls. The expression levels of IL-1β, IL-8 and TNF-α in the DSS group were higher than those in the control group(P < 0.05), and the expression levels of IL-1β, IL-8 and TNF-α in the JPQCD and 5-ASA groups were lower than those in the DSS group after treating with JPQCD and 5-ASA. Comparing with the DSS group, the mR NA level of IL-1β, IL-8, TNF-α and NF-κB was significantly reduced by 5-ASA and JPQCD. The difference between JPQCD and 5-ASA groups was not statistically significant(P > 0.05). Comparing with the DSS group, due to using JPQCD and 5-ASA, significant suppression of activation in DSSinduced NF-κB and increased phosphorylation of IκB in mice with experimental colitis occurred(P < 0.05). The difference between the JPQCD group and the 5-ASA group was not statistically significant(P > 0.05). 2Jianpi Qingchang decoction regulates intestinal motility of dextran sulfate sodium-induced colitis through reducing autophagy of interstitial cells of Cajal显示文摘AIM To investigate the underlying effect of Jianpi Qingchang decoction(JQD) regulating intestinal motility of dextran sulfate sodium(DSS)-induced colitis in mice. METHODS C57BL/6 mice were randomly divided into four groups: the control group, the DSS group, the JQD group, and the 5-aminosalicylic acid group. Except for the control group, colitis was induced in other groups by giving distilled water containing 5% DSS. Seven days after modeling, the mice were administered corresponding drugs intragastrically. The mice were sacrificed on the 15^(th) day. The disease activity index, macroscopic and histopathologic lesions, and ultrastructure of colon interstitial cells of Cajal(ICC) were observed. The levels of tumor necrosis factor-alpha(TNF-α), interleukin(IL)-1β, IL-10 and interferon gamma(IFN-γ), the expression of nuclear factor-kappa B(NF-κB) p65, c-kit, microtubule-associated protein 1 light chain 3(LC3-Ⅱ) and Beclin-l m RNA, and the colonic smooth muscle tension were assessed. RESULTS Acute inflammation occurred in the mice administered DSS. Compared with the control group, the levels of IL-1β, TNF-α, IL-10 and IFN-γ, the expression of LC3-Ⅱ, Beclin-1 and NF-κB p65 m RNA, and the contractile frequency increased(P < 0.05), the expression of c-kit m RNA and the colonic smooth muscle contractile amplitude decreased in the DSS group(P < 0.05). Compared with the DSS group, the levels of IL-10 and IFN-γ, the expression of c-kit m RNA, and the colonic smooth muscle contractile amplitude increased(P < 0.05), the levels of TNF-α and IL-1β, the expression of LC3-Ⅱ, Beclin-1 and NF-κB p65 m RNA, and the contractile frequency decreased in the JQD group(P < 0.05).CONCLUSION JQD can regulate the intestinal motility of DSS-induced colitis in mice through suppressing intestinal inflammatory cascade reaction, reducing autophagy of ICC, and regulating the network path of ICC/smooth muscle cells.Yan-Cheng Dai Lie Zheng Ya-Li Zhang Xuan Chen De-Liang Chen Li-Juan Wang Zhi-Peng Tang 2017World Journal of Gastroenterology2017,23,26:26
3Salt-responsive genes in rice revealed by cDNA microarray analysis显示文摘We used cDNA microarrays containing ~9,000 unigenes to identify 486 salt responsive expressed sequence tags (ESTs) (representing ~450 unigenes) in shoots of the highly salt-tolerant rice variety, Nona Bokra (Oryza sativa L. Ssp.Indica pv. Nona). Some of the genes identified in this study had previously been associated with salt stress. Howeverthe majority were novel, indicating that there is a great number of genes that are induced by salt exposure. Analysis of the salt stress expression profile data of Nona provided clues regarding some putative cellular and molecular processes that are undertaken by this tolerant rice variety in response to salt stress. Namely, we found that multiple transcription factors were induced during the initial salt response of shoots. Many genes whose encoded proteins are implicated in detoxification, protectant and transport were rapidly induced. Genes supporting photosynthesis were repressed and those supporting carbohydrate metabolism were altered. Commonality among the genes induced by salt exposure with those induced during senescence and biotic stress responses suggests that there are shared signaling pathways among these processes. We further compared the transcriptome changes of the salt-sensitive cultivar, IR28, with that of Nona rice. Many genes that are salt responsive in Nona were found to be differentially regulated in IR28. 4Effects of radix curcumae-derived diterpenoid C on Helicobacter pylori-induced inflammation and nuclear factor kappa B signal pathways显示文摘AIM:To study effect of diterpenoid C extracted from radix curcumae on Helicobacter pylori(H.pylori)-infected inflammation,intestinal metaplasia,and nuclear factor kappa B(NF-κB)signaling pathway in vitro.METHODS:We used I-type H.pylori to infect human gastric epithelial gastric epithelium cell line(GES-1)cell lines,and then H.pylori-infected GES-1 cells were treated with radix curcumae(RC)-derived diterpenoid C of different concentrations(5,10,20μg/mL)and amoxicillin.The expression of p65,IκB kinase(IKK)αand IKKγproteins was detected with Western blotting,and the expression of interleukin(IL)-8,IL-6 and IL-4 was determined with enzyme-linked immunosorbent assay method.Data were analyzed using SPSS software ver18.0.For comparisons between groups of more than two unpaired values,one-way analysis of variance(ANOVA)was used.If an ANOVA F value was significant,post hoc comparisons were performed between groups.If results were not normally distributed,the Mann-Whitney U test was used to compare two groups of unpaired values,whereas for comparisons between groups of more than two unpaired values,the Kruskal-Wallis H test was used.Statistical significance was established at P<0.05.RESULTS:The MTT assay results revealed the inhibited rate of GES-1,and indicated that the IC5 of RCderived diterpenoid C and amoxicillin all were 5μg/mL for gastric GES-1 cells.The expression of IL-8 was significantly increased,especially at 12 h time point;and the expression of IL-4 was decreased in H.pyloriinfected GES-1 cells.After H.pylori-infected GES-1 cells were treated with RC-derived diterpenoid C of different concentrations and amoxicillin,the expression of IL-8was decreased at 12,24,48,72 h points(P<0.01),especially in high-concentration diterpenoid C(20μg/mL)group;and the expression of IL-4 was increased,especially in moderate and high-concentration diterpenoid C(10 and 20μg/mL)groups.RC-derived diterpenoid C had the inhibitory effects on H.pylori-induced p65 translocation from cytoplasm into cell nucleus,H.pylori-stimulant IkBαdegradation,the phosphorylation of p65 and IkBα,and the expression of IKKαand IKKβproteins.CONCLUSION:RC-derived diterpenoid C can block NF-κB signal pathway,effectively reducing the secretion of H.pylori-induced proinflammatory cytokine and increasing the secretion of anti-inflammatory cytokine.Xuan Huang Bin Lv Shuo Zhang Qun Dai Bing-Bing Chen Li-Na Meng 2013World Journal of Gastroenterology2013,19,31:10

6Valence band structure of strained Si/(111)Si_(1-x)Ge_x显示文摘The strained Si techique has been widely adopted in the high-speed and high-performance devices and circuits. Based on the valence band E-k relations of strained Si/(111)Si1-xGex, the valence band and hole effective mass along the [111] and [-110] directions were obtained in this work. In comparison with the relaxed Si, the valence band edge degeneracy was partially lifted, and the significant change was observed band structures along the [111] and [-110] directions, as well as in its corresponding hole effective masses with the increasing Ge fraction. 8Reveal training performance mystery between Tensor Flow and PyTorch in the single GPU environment显示文摘Deep learning has gained tremendous success in various fields while training deep neural networks(DNNs)is very compute-intensive,which results in numerous deep learning frameworks that aim to offer better usability and higher performance to deep learning practitioners.Tensor Flow and Py Torch are the two most popular frameworks.Tensor Flow is more promising within the industry context,while Py Torch is more appealing in academia.However,these two frameworks differ much owing to the opposite design philosophy:static vs dynamic computation graph.Tensor Flow is regarded as being more performance-friendly as it has more opportunities to perform optimizations with the full view of the computation graph.However,there are also claims that Py Torch is faster than Tensor Flow sometimes,which confuses the end-users on the choice between them.In this paper,we carry out the analytical and experimental analysis to unravel the mystery of comparison in training speed on single-GPU between Tensor Flow and Py Torch.To ensure that our investigation is as comprehensive as possible,we carefully select seven popular neural networks,which cover computer vision,speech recognition,and natural language processing(NLP).The contributions of this work are two-fold.First,we conduct the detailed benchmarking experiments on Tensor Flow and Py Torch and analyze the reasons for their performance difference.This work provides the guidance for the end-users to choose between these two frameworks.Second,we identify some key factors that affect the performance,which can direct the end-users to write their models more efficiently.Hulin DAI Xuan PENG Xuanhua SHI Ligang HE Qian XIONG Hai JIN 2022Science China(Information Sciences)2022,65,1:6
9Intergranular corrosion behavior and mechanism of the stabilized ultra-pure 430LX ferritic stainless steel显示文摘Intergranular corrosion(IGC) behavior of the stabilized ultra-pure 430 LX ferritic stainless steel(FSS) was investigated by using double loop electrochemical potentiokinetic reactivation(DL-EPR) and oxalic acid etch tests to measure the susceptibility of specimens given a two-step heat treatment. The results reveal that IGC occurs in the specimens aged at the temperature range of 600–750℃ for a short time. The aging time that is required to cause IGC decreases with the increase of aging temperature. A longer aging treatment can reduce the susceptibility to IGC. The microstructural observation shows that M(23)C6 precipitates form along the grain boundaries, leading to the formation of Cr-depleted zones. The presence of Cr-depleted zones results in the susceptibility to IGC. 11Prevalence and Determinants of High-risk HPV Infection among 11549 Women from an Opportunistic Screening in Hubei Province显示文摘High-risk human papillomavirus (hrHPV) infection plays an important role in the development of cervical intraepithelial neoplasia and cervical cancer.A total of 11 549 women were enrolled from the Maternal and Child Health Hospital of Hubei Province.Each participant accepted hrHPV testing and completed a self^administered questionnaire about basic information and potential risk factors.The univariable and multivariable logistic regression model was used to explore the associations between variants and hrHPV infection.Our results showed that hrHPV prevalence was 16.09% in Hubei Province,among which,hrHPV was more likely to be positive in women aged 51 years or above (OR=1.65,95% CI:1.28-2.14),and in women who had symptoms of bleeding after intercourse (OR=1.32,95% CI:1.17-1.50),had first sexual intercourse at the age of 18 years or below (OR=1.33,95% CI:1.07-1.64),had at least three male sexual partners (OR=2.50,95% CI:2.07-3.03),and who had been diagnosed with sexually transmitted infections (OR=1.50,95% CI:1.12-2.03).Married women (OR=0.66,95% CI:0.55-0.78) and women who frequently used condoms (OR=0.75,95% CI:0.67-0.84) had a relatively lower hrHPV prevalence.This study confirms that hrHPV infection was associated with age,marital status,symptoms of intercourse bleeding,history of sexually transmitted infections,and sex-related behaviors.Above all,this study provides a baseline database prior to obtaining vaccinations for dynamic tracking of the changes in hrHPV prevalence.Quan-fu MA Yu-lin GUO Han GAO Bin YAN Xuan DAI Meng XU Yu-jing XIONG Qiu-zi PENG Ying WANG Miao ZOU Xu-feng WU 2019Current Medical Science2019,39,4:5
12The electromagnetic response of composition-regulated honeycomb structural materials used for broadband microwave absorption显示文摘The undesirable absorption bandwidth is the main limitation for the application of microwave absorbing materials.Noting that the design of structure and composition of absorbing coating allows it to obtain a satisfying absorption bandwidth and to avoid weight increasing,honeycomb and its sandwich structures coated by various absorbant have been developed and measured using an arch measurement system over2-18 GHz.To further understand the absorption mechanisms of honeycomb structural materials,simulation based on finite element method and transmission line theory is performed to analyze the different responses to the applied microwave between honeycomb structural materials and uniform-medium slab materials.The proper composition design allows control over the microwave loss mechanisms,which optimize the coating to possess both of the dielectric and magnetic advantages.For the honeycomb structural materials,the magnetic resonation is found for the first time,which is resulting from the periodic honeycomb walls on both sides of the dielectric cavity(consisting of the aramid paper interlayer and vacuum hexagonal prisms).In addition,the conversion of structure from solid to honeycomb brings about changes in impedance,propagating path,effective wavelength,effective attenuation area,microwave phases,and inductive coupling effect,etc.,which result in a better microwave absorbing performance.The honeycomb sandwich structure coated by CIP/CB/EP with the weight ratio of 4:0.03:1 shows the absorption bandwidth of~9.8 GHz for reflection loss(RL)lower than-10 dB which covers part of the S and C bands and almost the whole X and Ku bands.Huifang Pang Yuping Duan Xuhao Dai Lingxi Huang Xuan Yang Tuo Zhang Xiaoji Liu 2021Journal of Materials Science & Technology2021,,29:4
13Linkage with cathepsin B-sensitive dipeptide promotes the in vitro and in vivo anticancer activity of PEGylated tumor necrosis factor-alpha(TNF-α) against murine fibrosarcoma显示文摘To improve the pharmacological profile of tumor necrosis factor alpha(TNF-α),we have synthesized a new PEGylated prodrug,PEG-vcTNF-α,using a cathepsin B-sensitive dipeptide(valine-citrulline,vc) to link branched PEG and TNF-α.PEG-modified TNF-α without the dipeptide linker(PEG-TNF-α) and unconjugated TNF-α were also tested as controls.It was found for the first time that TNF-α released from PEG-vcTNF-α was specifically dependent on the presence of cathepsin B.PEG-vcTNF-α induced higher cytotoxicity and greater apoptosis against L929 murine fibrosarcoma cells than PEG-TNF-α.Reversal of these effects by a cathepsin-B inhibitor confirmed that these effects were mediated by cathepsin B-specific release of TNF-α.In vivo pharmacokinetics studies demonstrated that the plasma stability of PEG-vcTNF-α was significantly increased compared to TNF-α.Finally,the improved anticancer efficacy of PEG-vcTNF-α and the distinct activities among the three formulations confirmed the positive contribution of both PEGylation and the dipeptide linkage to the improved drug-like properties of PEG-vcTNF-α.The results here indicate that linking proteins and PEG via the cathepsin B-sensitive dipeptide may be a promising strategy for developing protein therapeutics.DAI ChuanYun FU Ya LI Biao WANG YiGuang ZHANG Xuan WANG JianCheng ZHANG Qiang 2011Science China(Life Sciences)2011,54,2:4
14A VLIW Architecture Stream Cryptographic Processor for Information Security显示文摘As an important branch of information security algorithms,the efficient and flexible implementation of stream ciphers is vital.Existing implementation methods,such as FPGA,GPP and ASIC,provide a good support,but they could not achieve a better tradeoff between high speed processing and high flexibility.ASIC has fast processing speed,but its flexibility is poor,GPP has high flexibility,but the processing speed is slow,FPGA has high flexibility and processing speed,but the resource utilization is very low.This paper studies a stream cryptographic processor which can efficiently and flexibly implement a variety of stream cipher algorithms.By analyzing the structure model,processing characteristics and storage characteristics of stream ciphers,a reconfigurable stream cryptographic processor with special instructions based on VLIW is presented,which has separate/cluster storage structure and is oriented to stream cipher operations.The proposed instruction structure can effectively support stream cipher processing with multiple data bit widths,parallelism among stream cipher processing with different data bit widths,and parallelism among branch control and stream cipher processing with high instruction level parallelism;the designed separate/clustered special bit registers and general register heaps,key register heaps can satisfy cryptographic requirements.So the proposed processor not only flexibly accomplishes the combination of multiple basic stream cipher operations to finish stream cipher algorithms.It has been implemented with 0.18μm CMOS technology,the test results show that the frequency can reach 200 MHz,and power consumption is 310 mw.Ten kinds of stream ciphers were realized in the processor.The key stream generation throughput of Grain-80,W7,MICKEY,ACHTERBAHN and Shrink algorithm is 100 Mbps,66.67 Mbps,66.67 Mbps,50 Mbps and 800 Mbps,respectively.The test result shows that the processor presented can achieve good tradeoff between high performance and flexibility of stream ciphers.Longmei Nan Xuan Yang Xiaoyang Zeng Wei Li Yiran Du Zibin Dai Lin Chen 2019China Communications2019,16,6:4
15Effect of Tai Chi and Resistance Training on Cancer-Related Fatigue and Quality of Life in Middle-Aged and Elderly Cancer Patients显示文摘Objective:To study the effect of Tai Chi(TC)and resistance training(RT)with different intensity on the cancer-related fatigue(CRF)and quality of life(QoL)of middle-aged and elderly cancer patients.Methods:Totally 120 cancer patients were enrolled and randomly assigned to 4 groups by a random number table,including TC group,high-intensity 60%one repetition maximum(1-RM)RT group(HIRT),low-intensity(30%1-RM)RT group(LIRT)and control group,30 patients in each group.Participants in the TC group received 24-form simplified Yang-style TC training at a frequency of 40 min per day,3 days per week for 12 weeks.Patients in the two RT groups received 10 sessions,6 designated movements per day,3 days per week for 12 weeks.The 1-RM of 6 muscle groups,fat mass(FM),lean body mass(LBM),along with the scores of Brief Fatigue Inventory(BFI),QoL questionnaire for Chinese cancer patients receiving chemobiotherapy(QLQ-CCC),Generalized Anxiety Disorder-7(GAD-7),Patient Health Questionnaire-9(PHQ-9)and Pittsburgh Sleep Quality Index(PSQI)were measured before and after training.The adverse effect was also observed.Results:After 12-week intervention,patients in both TC and RT groups showed significant improvements in CRF and QLQ-CCC compared to pre-treatment(P<0.05).Compared with the LIRT and TC groups,patients in the HIRT group improved more significantly in increasing muscle strength and LBM,and reducing in FM(P<0.05).Patients in the TC group significantly increased in lower limb muscle strength compared with the LIRT group(P<0.05).In addition,patients in the TC group showed more significant improvements in scores of GAD-7,PHQ-9 and PSQI than 2 RT groups(P<0.05).Conclusions:TC and RT,both low-and high-intensity training,can significantly increase muscle strength,reduce CRF and improve QoL in the middle-aged and elderly cancer patients.TC has a better effect than RT in terms of sleep quality and mental health.The long-term application is needed to substantiate the effect of TC as an alternative exercise in cancer patients.CHENG Duan WANG Xuan HU Jie DAI Ling-li LV Ying FENG Hui ZHANG Yan GUO Yan WANG Lei 2021Chinese Journal of Integrative Medicine2021,27,4:3
16Endoplasmic reticulum stress-induced apoptosis in intestinal epithelial cells:a feed-back regulation by mechanistic target of rapamycin complex 1(mTORC1)显示文摘Background: Endoplasmic reticulum(ER) stress is associated with multiple pathological processes of intestinal diseases. Despite a critical role of mechanistic target of rapamycin complex 1(m TORC1) in regulating cellular stress response, the crosstalk between m TORC1 and ER stress signaling and its contribution to the intestinal barrier function is unknown.Results: In the present study, we showed that intestinal epithelial cells(IEC-6) incubated with tunicamycin led to caspase-3-dependent apoptotic cell death. The induction of cell death was accompanied by activation of unfolded protein response as evidenced by increased protein levels for Bi P, p-IRE1α, p-e IF2α, p-JNK, and CHOP. Further study demonstrated that tunicamycin-induced cell death was enhanced by rapamycin, a specific inhibitor of m TORC1.Consistently, tunicamycin decreased transepithelial electrical resistance(TEER) and increased permeability of the cells. These effects of tunicamycin were exacerbated by m TORC1 inhibitor.Conclusions: Taken together, the data presented here identified a previously unknown crosstalk between an unfold protein response and m TORC1 signaling in the intestinal epithelium. 17Effects of Granule Dendrobii on chronic atrophic gastritis induced by N-methyl-N'-nitro-N-nitrosoguanidine in rats显示文摘BACKGROUND Dendrobium officinale is an herb of Traditional Chinese Medicine(TCM)commonly used for treating stomach diseases.One formula of Granule Dendrobii(GD)consists of Dendrobium officinale and American Ginseng(Radix Panacis quinquefolii),and is a potent TCM product in China.Whether treatment with GD can promote gastric acid secretion and alleviate gastric gland atrophy in chronic atrophic gastritis(CAG)requires verification.AIM To determine the effect of GD treatment on CAG and its potential cellular mechanism.METHODS A CAG model was induced by feeding rats N-methyl-N'-nitro-N-nitrosoguanidine(MNNG)for 12 wk.After oral administration of low,moderate,and high doses of GD in CAG rats for 8 wk,its effects on body weight,gastric mucosa histology,mucosal atrophy,intestinal metaplasia,immunohistochemical staining of proliferating cell nuclear antigen(PCNA)and B-cell lymphoma-2,and hemoglobin and red blood cells were examined.RESULTS The body weights of MNNG-induced CAG model rats before treatment(143.5±14.26 g)were significantly lower than that of healthy rats(220.2±31.20 g,P<0.01).At the 8th week of treatment,the body weights of rats in the low-,moderate-,and high-dose groups of GD(220.1±36.62 g)were significantly higher than those in the untreated group(173.3±28.09 g,all P<0.01).The level of inflammation in gastric tissue of the high-dose group(1.68±0.54)was significantly reduced(P<0.01)compared with that of the untreated group(3.00±0.00,P<0.05).The number and thickness of gastric glands in the high-dose group(31.50±6.07/mm,306.4±49.32μm)were significantly higher than those in the untreated group(26.86±6.41/mm,244.3±51.82μm,respectively,P<0.01 and P<0.05),indicating improved atrophy of gastric mucosa.The areas of intestinal metaplasia were significantly lower in the high-dose group(1.74%±1.13%),medium-dose group(1.81%±0.66%)and low-dose group(2.36%±1.08%)than in the untreated group(3.91%±0.96%,all P<0.01).The expression of PCNA in high-dose group was significantly reduced compared with that in untreated group(P<0.01).Hemoglobin level in the high-dose group(145.3±5.90 g/L),medium-dose group(139.3±5.71 g/L)and low-dose group(137.5±7.56 g/L)was markedly increased compared with the untreated group(132.1±7.76 g/L;P<0.01 or P<0.05).CONCLUSION Treatment with GD for 8 wk demonstrate that GD is effective in the treatment of CAG in the MNNG model by improving the histopathology of gastric mucosa,reversing gastric atrophy and intestinal metaplasia,and alleviating gastric inflammation.Yue Wu Yu Li Xiao-Ming Jin Guan-Hai Dai Xuan Chen Ye-Ling Tong Ze-Ming Ren Yu Chen Xiao-MinXue Ren-Zhao Wu 2022World Journal of Gastroenterology2022,28,32:3

19The Agpat4/LPA axis in colorectal cancer cells regulates antitumor responses via p38/p65 signaling in macrophages显示文摘Lipid metabolic reprogramming plays an essential role in regulating the progression of colorectal cancer(CRC).However,the effect of lysophosphatidic acid(LPA)metabolism on CRC development is incompletely characterized.Here,we compared the mRNA levels of human CRC tissues to those of paracarcinoma tissues and focused on the notably enriched LPA metabolic pathways.We identified and verified that 1-acylglycerol-3-phosphate O-acyltransferase 4(Agpat4)was aberrantly expressed in CRC tissues and predicted poor survival in CRC patients.Manipulating Agpat4 expression in CRC cells did not affect the growth or migration of CRC cells in vitro,whereas Agpat4 silencing suppressed CRC cell growth in subcutaneous and peritoneal xenograft models.Mechanistically,Agpat4 silencing-induced LPA release from CRC cells and polarized macrophages to an M1-like phenotype through LPA receptors 1 and 3.This M1 activation,characterized by elevated p38/p65 signaling and increased proinflammatory cytokines,promoted the infiltration and activation of CD4+and CD8+T cells in the tumor microenvironment.Modulation of the Agpat4/LPA/p38/p65 axis regulated macrophage polarization,T-cell activity and CRC progression.Notably,combined therapy with LPA and regular chemotherapy drugs synergistically suppressed CRC development.Taken together,our results showed that the Agpat4/LPA axis in CRC cells regulated p38/p65 signaling-dependent macrophage polarization,T-cell activation,and CRC progression.The Agpat4/LPA/p38/p65 axis might represent a potential target for therapy in the clinic.Dapeng Zhang Rongchen Shi Wei Xiang Xia Kang Bo Tang Chuan Li Linfeng Gao Xuan Zhang Lili Zhang Rongyang Dai Hongming Miao 2020Signal Transduction and Targeted Therapy2020,5,1:3
20A drug-loaded composite coating to improve osteogenic and antibacterial properties of Zn-1Mg porous scaffolds as biodegradable bone implants显示文摘Zinc(Zn)alloy porous scaffolds produced by additive manufacturing own customizable structures and biodegradable functions,having a great application potential for repairing bone defect.In this work,a hydroxyapatite(HA)/polydopamine(PDA)composite coating was constructed on the surface of Zn-1Mg porous scaffolds fabricated by laser powder bed fusion,and was loaded with a bioactive factor BMP2 and an antibacterial drug vancomycin.The microstructure,degradation behavior,biocompatibility,antibacterial performance and osteogenic activities were systematically investigated.Compared with as-built Zn-1Mg scaffolds,the rapid increase of Zn2+,which resulted to the deteriorated cell viability and osteogenic differentiation,was inhibited due to the physical barrier of the composite coating.In vitro cellular and bacterial assay indicated that the loaded BMP2 and vancomycin considerably enhanced the cytocompatibility and antibacterial performance.Significantly improved osteogenic and antibacterial functions were also observed according to in vivo implantation in the lateral femoral condyle of rats.The design,influence and mechanism of the composite coating were discussed accordingly.It was concluded that the additively manufactured Zn-1Mg porous scaffolds together with the composite coating could modulate biodegradable performance and contribute to effective promotion of bone recovery and antibacterial function.Zhenbao Zhang Aobo Liu Jiadong Fan Menglin Wang Jiabao Dai Xiang Jin Huanze Deng Xuan Wang Yijie Liang Haixia Li Yantao Zhao Peng Wen Yanfeng Li 2023Bioactive Materials2023,,9:2
