| 2 | Mesenchymal stem cells derived from human placenta suppress allogeneic umbilical cord blood lymphocyte proliferation显示文摘ABSTRACTHuman placenta-derived mononuclear cells (MNC) were isolated by a Percoll density gradient and cultured in mesen-chymal stem cell (MSC) maintenance medium. The homogenous layer of adherent cells exhibited a typical fibroblast-like morphology, a large expansive potential, and cell cycle characteristics including a subset of quiescent cells. In vitrodifferentiation assays showed the tripotential differentiation capacity of these cells toward adipogenic, osteogenic andchondrogenic lineages. Flow cytometry analyses and immunocytochemistry stain showed that placental MSC was ahomogeneous cell population devoid of hematopoietic cells, which uniformly expressed CD29, CD44, CD73, CD105,CD166, laminin, fibronectin and vimentin while being negative for expression of CD31, CD34, CD45 and α-smoothmuscle actin. Most importantly, immuno-phenotypic analyses demonstrated that these cells expressed class I majorhistocompatibility complex (MHC-I), but they did not express MHC-II molecules. Additionally these cells could sup-press umbilical cord blood (UCB) lymphocytes proliferation induced by cellular or nonspecific mitogenic stimuli. Thisstrongly implies that they may have potential application in allograft transplantation. | 4 | Association between Ambient Air Pollution and Hospital Emergency Admissions for Respiratory and Cardiovascular Diseases in Beijing: a Time Series Study显示文摘Objective To investigate the association between ambient air pollution and hospital emergency admissions in Beijing. Methods In this study, a semi-parametric generalized additive model(GAM) was used to evaluate the specific influences of air pollutants(PM10, SO2, and NO2) on hospital emergency admissions with different lag structures from 2009 to 2011, the sex and age specific influences of air pollution and the modifying effect of seasons on air pollution to analyze the possible interaction. Results It was found that a 10 μg/m3 increase in concentration of PM10 at lag 03 day, SO2 and NO2 at lag 0 day were associated with an increase of 0.88%, 0.76%, and 1.82% respectively in overall emergency admissions. A 10 μg/m3 increase in concentration of PM10, SO2 and NO2 at lag 5 day were associated with an increase of 1.39%, 1.56%, and 1.18% respectively in cardiovascular disease emergency admissions. For lag 02, a 10 μg/m3 increase in concentration of PM10, SO2 and NO2 were associated with 1.72%, 1.34%, and 2.57% increases respectively in respiratory disease emergency admissions. | 6 | Upregulation of CD81 in trophoblasts induces an imbalance of Treg/Th17 cells by promoting IL-6 expression in preeclampsia显示文摘The disturbance of maternal immune tolerance to a semiallogeneic fetus is recognized as one of the key pathologies of preeclampsia(PE),in which an imbalance between the inflammation-limiting regulatory T cells(Tregs)and the inflammationmediating Th17 cells plays an essential role.Previously,we reported that the abnormal upregulation of tetraspannin CD81 in trophoblast cells(fetal component)participated in the pathogenesis of PE.However,as one of the potential immune regulatory molecules,whether CD81 induces PE by interfering with the balance of the maternal immune system has not yet been clarified.Thus,we investigated the relationship between the upregulation of CD81 in trophoblast cells and the imbalance of Treg and Th17 cells in mothers.Here,we demonstrated that upregulation of CD81 in trophoblast cells was accompanied by a decrease in Treg cells and an increase in Th17 cells in both the basal plate(placental maternal side)and peripheral blood of patients with PE.In vitro culture of naïve T cells with medium from the CD81-overexpressing trophoblast cell line HTR-8 resulted in enhanced differentiation of T cells into Th17 cells and decreased the formation of Tregs,which was dependent on the paracrine signaling of IL-6 in trophocytes,induced by CD81.In a CD81-induced PE rat model,we found a significant shift of T cell differentiation towards Th17 cells,and administration of IL-6 antibody mitigated the PE phenotype and the imbalance of the Treg/Th17 cells.These results define a vital regulatory cascade involving trophocyte-derived CD81,IL-6,and maternal Treg/Th17 cells in the pathogenesis of PE and suggests new therapeutic approaches based on CD81 and IL-6 downregulation to prevent human PE. | Hailin Ding Yimin Dai Yi Lei Zhiyin Wang Dan Liu Ruotian Li Li Shen Ning Gu Mingming Zheng Xiangyu Zhu Guangfeng Zhao Yali Hu | 2019 | Cellular & Molecular Immunology2019,16,3: | 15 |

| 8 | Pain management of hemiplegic shoulder pain post stroke in patients from Nanjing,China显示文摘We selected 106 hemiplegic patients with shoulder pain hospitalized after stroke from three hospitals in Nanjing,China between February 2007 and January 2012.All patients had complete clinical data sets and accounted for 45.5%of the inpatients because of stroke.Results showed that the number of patients with hemiplegic shoulder pain post stroke increased yearly,attacking mainly males 50–69 years of age.Of 106 patients,there were 60 cases(56.6%)of adhesive capsulitis,19(17.9%)of shoulder subluxation,14(13.2%)of complex regional pain syndrome,and 13(12.6%)of central pain.The main symptoms were shoulder pain(100%),limit of shoulder mobility(98.1%),and adhesion of the scapula(56.6%).MRI of the shoulder showed tendon and ligament lesions(57.1%)and rotator cuff tear(38.1%).53.8%of central pain was related to the thalamus,in addition to the basal ganglia,brain stem,and cerebellopontine angle.Shoulder pain,upper limb motor function and function independence were significantly improved after comprehensive rehabilitation.In particular,electroacupuncture based on basic physical therapy exhibited efficacy on shoulder subluxation and complex regional pain syndrome.Multiple linear regression results showed a negative relationship of efficacy of pain management with the attack period of shoulder pain involvement of the posterior limb of the internal capsule,and duration between onset and rehabilitation treatment,but a positive correlation with pain-related education,pain regression period,and pain diagnosis. | Yi Zhu Bin Su Ning Li Hongzhu Jin | 2013 | Neural Regeneration Research2013,8,25: | 14 |

On behalf of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology(CSCO) and the China Actionable Genome Consortium(CAGC), the present expert group hereby proposes advisory guidelines on clinical applications of NGS technology for the analysis of cancer driver genes for precision cancer therapy. This group comprises an assembly of laboratory cancer geneticists, clinical oncologists, bioinformaticians,pathologists, and other professionals. After multiple rounds of discussions and revisions, the expert group has reached a preliminary consensus on the need of NGS in clinical diagnosis, its regulation, and compliance standards in clinical sample collection. | 12 | Deep learning applied to two-dimensional color Doppler flow imaging ultrasound images significantly improves diagnostic performance in the classification of breast masses:a multicenter study显示文摘Background:The current deep learning diagnosis of breast masses is mainly reflected by the diagnosis of benign and malignant lesions.In China,breast masses are divided into four categories according to the treatment method:inflammatory masses,adenosis,benign tumors,and malignant tumors.These categorizations are important for guiding clinical treatment.In this study,we aimed to develop a convolutional neural network(CNN)for classification of these four breast mass types using ultrasound(US)images.Methods:Taking breast biopsy or pathological examinations as the reference standard,CNNs were used to establish models for the four-way classification of 3623 breast cancer patients from 13 centers.The patients were randomly divided into training and test groups(n=1810 vs.n=1813).Separate models were created for two-dimensional(2D)images only,2D and color Doppler flow imaging(2D-CDFI),and 2D-CDFI and pulsed wave Doppler(2D-CDFI-PW)images.The performance of these three models was compared using sensitivity,specificity,area under receiver operating characteristic curve(AUC),positive(PPV)and negative predictive values(NPV),positive(LR+)and negative likelihood ratios(LR-),and the performance of the 2D model was further compared between masses of different sizes with above statistical indicators,between images from different hospitals with AUC,and with the performance of 37 radiologists.Results:The accuracies of the 2D,2D-CDFI,and 2D-CDFI-PW models on the test set were 87.9%,89.2%,and 88.7%,respectively.The AUCs for classification of benign tumors,malignant tumors,inflammatory masses,and adenosis were 0.90,0.91,0.90,and 0.89,respectively(95%confidence intervals[CIs],0.87-0.91,0.89-0.92,0.87-0.91,and 0.86-0.90).The 2D-CDFI model showed better accuracy(89.2%)on the test set than the 2D(87.9%)and 2D-CDFI-PW(88.7%)models.The 2D model showed accuracy of 81.7%on breast masses≤1 cm and 82.3%on breast masses>1 cm;there was a significant difference between the two groups(P<0.001).The accuracy of the CNN classifications for the test set(89.2%)was significantly higher than that of all the radiologists(30%).Conclusions:The CNN may have high accuracy for classification of US images of breast masses and perform significantly better than human radiologists.Trial registration:Chictr.org,ChiCTR1900021375;http://gffzz62a03ee25e6f4a77hf0vkfoc99nwx69np.ffgz.tsg.suse.edu.cn/showproj.aspx?proj=33139. | Teng-Fei Yu Wen He Cong-Gui Gan Ming-Chang Zhao Qiang Zhu Wei Zhang Hui Wang Yu-Kun Luo Fang Nie Li-Jun Yuan Yong Wang Yan-Li Guo Jian-Jun Yuan Li-Tao Ruan Yi-Cheng Wang Rui-Fang Zhang Hong-Xia Zhang Bin Ning Hai-Man Song Shuai Zheng Yi Li Yang Guang | 2021 | Chinese Medical Journal2021,,4: | 11 |

| 15 | Crosstalk between the Akt/mTORC1 and NF-kB signaling pathways promotes hypoxia-induced pulmonary hypertension by increasing DPP4 expression in PASMCs显示文摘Abnormal wound healing by pulmonary artery smooth muscle cells(PASMCs)promotes vascular remodeling in hypoxia-induced pulmonary hypertension(HPH).Increasing evidence shows that both the mammalian target of rapamycin complex 1(mTORCI)and nuclear factor-kappa B(NF-kB)are Involved in the development of HPH.In this study,we explored the crosstalk between mTORCI and NF-kB in PASMCs cultured under hypoxic condition and in a rat model of hypoxia-induced pulmonary hypertension(HPH).We showed that hypoxia promoted wound healing of PASMCs,which was dose-dependently blocked by the mTORCI inhibitor rapamycin(5-20 nM).In PASMCs,hypoxia activated mTORCI,which in turn promoted the phosphorylation of NF-k B.Molecular docking revealed that mTOR interacted with IkB kinases(IKKs)and that was validated by immunoprecipitation.In vitro kinase assays and mass spectrometry demonstrated that mTOR phosphorylated IKKa and IKKp separately.Inhibition of mTORCI decreased the level of phosphorylated IKKa/p,thus reducing the phosphorylation and transcriptional activity of NF-kB.Bioinformatics study revealed that dipeptidyl peptidase-4(DPP4)was a target gene of NF-kB;DPP4 inhibitor,sitagliptin(10-500 pM)effectively inhibited the abnormal wound healing of PASMCs under hypoxic condition.In the rat model of HPH,we showed that NF-kB activation(at 3 weeks)was preceded by mTOR signaling activation(after 1 or 2 weeks)in lungs,and administration of sitagliptin(1-5 mg/kg every day,ig)produced preventive effects against the development of HPH.In conclusion,hypoxia activates the crosstalk between mTORCI and NF-kB,and increased DPP4 expression in PASMCs that leads to vascular remodeling.Sitagliptin,a DPP4 inhibitor,exerts preventive effect against HPH. | Ying Li Li Yang Liang Dong Zhi-wei Yang Jing Zhang Sheng-li Zhang Meng-jie Niu Jing-wen Xia Yi Gong Ning Zhu Xiu-juan Zhang Yuan-yuan Zhang Xiao-min Wei You-zhi Zhang Peng Zhang Sheng-qing Li | 2019 | Acta Pharmacologica Sinica2019,40,10: | 10 |
| 16 | Diagnostic value of 5 serum biomarkers for hepatocellular carcinoma with different epidemiological backgrounds:A large-scale,retrospective study显示文摘Objective:Hepatocellular carcinoma(HCC)is a lethal global disease that requires an accurate diagnosis.We assessed the potential of 5 serum biomarkers(AFP,AFU,GGT-II,GPC3,and HGF)in the diagnosis of HCC.Methods:In this retrospective study,we measured the serum levels of each biomarker using ELISAs in 921 participants,including 298 patients with HCC,154 patients with chronic hepatitis(CH),122 patients with liver cirrhosis(LC),and 347 healthy controls from 3 hospitals.Patients negative for hepatitis B surface antigen and hepatitis C antibody(called'NBNC-HCC')and patients positive for the above indices(called'HBV-HCC and HCV-HCC')were enrolled.The selected diagnostic model was constructed using a training cohort(n=468),and a validation cohort(n=453)was used to validate our results.Receiver operating characteristic analysis was used to evaluate the diagnostic accuracy.Results:Theα-L-fucosidase(AFU)/α-fetoprotein(AFP)combination was best able to distinguish NBNC-HCC[area under the curve:0.986(95%confidence interval:0.958–0.997),sensitivity:92.6%,specificity:98.9%]from healthy controls in the test cohort.For screening populations at risk of developing HCC(CH and LC),the AFP/AFU combination improved the diagnostic specificity for early-stage HCC[area under the curve:0.776(0.712–0.831),sensitivity:52.5%,specificity:91.6%in the test group].In all-stage HBV-HCC and HCV-HCC,AFU was also the best candidate biomarker combined with AFP[area under the curve:0.835(0.784–0.877),sensitivity 69.1%,specificity:87.4%in the test group].All results were verified in the validation group.Conclusions:The AFP/AFU combination could be used to identify NBNC-HCC from healthy controls and hepatitis-related HCC from at-risk patients. | Dongming Liu Yi Luo Lu Chen Liwei Chen Duo Zuo Yueguo Li Xiaofang Zhang Jing Wu Qing Xi Guangtao Li Lisha Qi Xiaofen Yue Xiehua Zhang Zhuoyu Sun Ning Zhang Tianqiang Song Wei Lu Hua Guo | 2021 | Cancer Biology & Medicine2021,18,1: | 10 |

Net Energy Requirement of Finishing Pigs Maximizing Performance and Carcass Quality Fed Low Crude Protein Diets Supplemented with Crystalline Amino Acids显示文摘Two experiments were conducted to investigate the effects of net energy (NE) level on the performance and carcass traits of finishing pigs fed low crude protein (CP) diets supplemented with crystalline amino acids (CAA). A total of 216 (Exp.1) and 360 (Exp.2) barrows (Yorkshire×Landrace×Duroc) were allotted to one of six treatments (n=6). The experimental diets were based on corn and soybean meal, and consisted of a highCP diet that contained approximately 16% CP and 2.50 Mcal/kg of NE as well as five lowCP diets in which the CP level of the diet was reduced by approximately four percentage units but was supplemented with crystalline lysine, methionine, threonine, and tryptophan. The low CP diets provided 2.64, 2.58, 2.50, 2.42, or 2.36 Mcal/kg in Exp.1 as well as 2.45, 2.40, 2.35, 2.30, or 2.25 Mcal/kg in Exp.2. In Exp.1, a linear (P=0.03) increase in weight gain was observed with decreasing NE level while the ratio of gain to feed was unaffected (P>0.05) by NE level. There was a linear increase (P=0.01) in the percentage of fatfree lean and a linear decrease (P=0.03) in the percentage of total fat with decreasing NE levels. In Exp.2, a significant quadratic (P=0.03) effect of NE level was observed for weight gain. The ratio of gain to feed demonstrated a significant (P<0.01) quadratic effect with pigs fed 2.35 and 2.40 Mcal/kg of NE. Pigs fed the diet containing 2.40 Mcal/kg of NE had the lowest percentage of total fat (36.95%) and the highest percentage of fatfree lean (49.36%). The overall results of these experiments indicate that feeding either a surplus or a deficiency of NE is detrimental to both pig performance and carcass composition when low CP diets supplemented with CAA are fed. | 20 | Association Between Serum Uric Acid and Prevalence of Type 2 Diabetes Diagnosed using HbA1c Criteria Among Chinese Adults in Qingdao,China显示文摘Objective To determine whether elevated serum uric acid(UA)levels are associated with type 2 diabetes diagnosed using Hb A1 c levels among Chinese adults.Methods We conducted two population-based cross-sectional studies in Qingdao in China in 2006 and 2009.A total of 6894(39.4% men)subjects aged 35-74 years were included in the data analysis.Newly diagnosed diabetes was defined as Hb A1 c level of ≥6.5%,and prediabetes was classified as Hb A1 c level between 5.7% and 6.4% according to the International Diabetes Federation criteria.Multivariate logistic regression was employed to assess the association between UA and prevalence of type 2 diabetes defined using Glycated hemoglobin A1c(Hb A1 c levels.Results Subjects with prediabetes had higher UA levels than those with normal glucose tolerance,newly diagnosed diabetes,and known diabetes,with corresponding values of 325.1(82.5)μmol/L,310.9(84.2)μmol/L,291.3(81.7)μmol/L,305.2(83.6)μmol/L,respectively(P<0.001 for all comparisons).Binary logistic regression analysis showed that UA was a possible predictor for the prevalence of type 2 diabetes diagnosed using Hb A1 c levels,and the second quartile of UA levels had a higher odds ratio(OR:4.088;95% CI:2.900-5.765)for Hb A1 c than the other quartiles after adjusting for age,body mass index,sex,marital status,education,income,alcohol consumption,smoking,and cardiometabolic parameters.Conclusion Serum UA is significantly associated with type 2 diabetes diagnosed using Hb A1 c levels,independent of other cardiometabolic parameters. | XUE Bai TAN Ji Bin NING Feng SUN Jian Ping ZHANG Ke Yi LIU Li WANG Shao Jie ZHANG Dong Feng QIAO Qing PANG Zeng Chang | 2015 | Biomedical and Environmental Sciences2015,28,12: | 9 |
