1Prevalence and evolution of drug resistance HIV-1 variants in Henan, China显示文摘To understand the prevalence and evolution of drug resistant HIV strains in Henan China after the implementation of free antiretroviral therapy for AIDS patients. 45 drug nave AIDS patients, 118 AIDS patients who received three months antiretroviral therapy and 124 AIDS patients who received six months antiretroviral treatment were recruited in the southern part of Henan province. Information on general condition, antiretroviral medicines, adherence and clinical syndromes were collected by face to face interview. Meanwhile, 14ml EDTA anticoagulant blood was drawn. CD4/CD8 T cell count, viral load and genotypic drug resistance were tested. The rates of clinical improvement were 55.1% and 50.8% respectively three months and six months after antiretroviral therapy. The mean CD4 cell count after antiretroviral therapy was significantly higher than in drug nave patients. The prevalence rate of drug resistant HIV strains were 13. 9%, 45.4% and 62.7% in drug nave patients, three month treatment patients and six month treatment patients, respectively. The number of resistance mutation codons and the frequency of mutations increased significantly with continued antiretroviral therapy. The mutation sites were primarily at the 103, 106 and 215 codons in the three-month treatment group and they increased to 15 codon mutations in the six-month treatment group. From this result, the evolution of drug resistant strains was inferred to begin with the high level NNRTI resistant strain, and then develop low level resistant strains to NRTIs. The HIV strains with high level resistance to NVP and low level resistance to AZT and DDI were highly prevalent because of the AZT+DDI+NVP combination therapy. These HIV strains were also cross resistant to DLV, EFV, DDC and D4T. Poor adherence to therapy was believed to be the main reason for the emergence and prevalence of drug resistant HIV strains. The prevalence of drug resistant HIV strains was increased with the continu- ation of antiretroviral therapy in the southern part of Henan province. 2Risk factors for alcohol-related liver injury in the island population of China: A population-based case-control study显示文摘AIM:To investigate the association of alcohol dose, duration of drinking and obesity with abnormal alcohol-related liver injury indicators, the prevalence of alcohol-related liver injury in the island population of China.METHODS:Randomized multistage stratified cluster sampling from the island population of China was used in the population-based case-control study. Then interview, physical examination, laboratory assessments and ultrasonography were done. RESULTS:Daily alcohol intake ≥ 20 g, duration of drinking ≥ 5 years and obesity were closely related to alcohol-related liver injury (P < 0.05). The odds-ratio (OR) (95% CI) was 1.965 (1.122-3.442), 3.412 (1.789-6.507) and 1.887 (1.261-2.824), respectively. The prevalence rate of alcohol-related liver injury in ≥ 20 g daily alcohol intake group and < 20 g daily alcohol intake group was 37.14% and 12.06%, respectively. The prevalence rate of alcohol-related liver injury in ≥ 5 years drinking group and < 5 years drinking group was 34.44% and 8.53%, respectively. No significant dose-response relation was found between daily alcohol intake and abnormal alcohol-related liver injury indicators as well as between duration of drinking and abnormal alcohol-related liver injury indicators. There was no significant difference in the prevalence of alcohol-related liver injury between beer drinking group and yellow rice wine drinking group, hard liquor drinking group, multiple drinking group.CONCLUSION:The risk threshold of daily alcohol intake is 20 g and duration of drinking inducing alcohol-related liver injury 5 years in the island population of China. 3Breakdown Properties of Nano-SiO2/LDPE Composite显示文摘聚合物纳米复合介质的击穿强度与纳米填充颗粒的质量分数、粒径和表面处理密切相关。以不同表面处理的纳米SiO2颗粒为填料,制备了不同填充质量分数、粒径的纳米SiO2/低密度聚乙烯(low density polyethylene,LDPE)复合介质,测试了其在交流、直流正极性和直流负极性3种不同类型电场下的击穿场强。结果表明:在所研究范围内,填充纳米SiO2颗粒可以提高低密度聚乙烯的击穿场强,并且随着填充质量分数的增加,复合介质的击穿场强有升高的趋势。纳米颗粒填充质量分数相同时,在12~16am粒径下,复合介质的击穿场强有随纳米SiO2粒径的升高而降低的趋势;在7nm小粒径下,可能由于纳米颗粒容易团聚,导致击穿场强较粒径大时要低。另外,纳米SiO2颗粒表面经疏水性处理后,能够有效提高复合介质的击穿场强。LAN Li WU Jiandong JI Zheqiang WANG Qiaohua LI Zhe XIAO Dengming YIN Yi 2012中国电机工程学报2012,32,13:14
4Effects of mowing and nitrogen addition on soil respiration in three patches in an oldfield grassland in Inner Mongolia显示文摘Aims Vegetation type is important in determining variations in soil carbon(C)efflux under grassland managements.This study was conducted to examine the effects of mowing and nitrogen(N)addition on soil respiration and their dependences upon vegetation types in an oldfield grassland of northern China.Methods Soil respiration,temperature,moisture and aboveground net primary productivity(ANPP)and belowground net primary productivity(BNPP)were examined in response to mowing and N addition among the three patches dominated by different species(named as grass,forb and mixed patches,respectively)in the growing seasons(May–October)from 2006 to 2008.Important Findings Across the 3 years,soil respiration in the grass patch was greater than those in the forb and mixed patches,which could have been ascribed to the higher soil moisture(SM)in the grass patch.Mowing had no impact on soil respiration due to unaltered SM and plant growth.Soil respiration was stimulated by 6.53%under N addition,and the enhancement was statistically significant in 2006 but not in 2007 or 2008 because of the limited water availability in the later 2 years.There were no interactive effects between mowing and N addition on soil respiration.Soil respiration showed positive dependence upon SM,ANPP and BNPP across plots.The results suggest that soil water availability and plant growth could be the primary factors in controlling the temporal and spatial variations in soil respiration and its response to different treatments.Our observations indicate that grassland managements(i.e.mowing for hay once a year)may have little influence on soil respiration of the oldfield grassland in northern China.Yi Han Zhe Zhang Changhui Wang Fenghe Jiang Jianyang Xia 2012Journal of Plant Ecology2012,5,2:14
5Conductive,Stretchable,and Self-healing Ionic Gel Based on Dynamic Covalent Bonds and Electrostatic Interaction显示文摘Integrating multiple functions into one gel that can be widely applied to electronic devices as well as chemical and biomedical engineering remains a big challenge.Here,a multifunctional ionic liquid/dynamic covalent bonds (ionic/DCB) type gel was designed and synthesized via one-pot polymerization.With the assistance of electrostatic interaction provided by the imidazolium cations of IL and the reversible DCB of boronic ester,as-prepared ionic/DCB gel showed good stretchable properties and high ionic conductivity at ambient conditions.In addition,the electrostatic interaction between imidazolium cations and sulfonate anions and the reversible DCB led to enhanced chain mobility and thereby excellent self-healing properties.Particularly,sulfonate anions in ionic/DCB gel could alleviate the migration of electronegative polysulfide and promote the transportation of electropositive lithium ion in lithium-sulfur battery system.Therefore,this work provides a new insight to promote the current research on self-healing gels,hopefully expanding their applications in electronic devices.Yi Sun Yong-Yuan Ren Qi Li Rong-Wei Shi Yin Hu Jiang-Na Guo Zhe Sun Feng Yan 2019Chinese Journal of Polymer Science2019,37,11:9
6Fedora-type magnetic compression anastomosis device for intestinal anastomosis显示文摘BACKGROUND Although previous studies have confirmed the feasibility of magnetic compression anastomosis(MCA),there is still a risk of long-term anastomotic stenosis.For traditional MCA devices,a large device is associated with great pressure,and eventually increased leakage.AIM To develop a novel MCA device to simultaneously meet the requirements of pressure and size.METHODS Traditional nummular MCA devices of all possible sizes were used to conduct ileac anastomosis in rats.The mean(±SD)circumference of the ileum was 13.34±0.12 mm.Based on short-and long-term follow-up results,we determined the appropriate pressure range and minimum size.Thereafter,we introduced a novel“fedora-type”MCA device,which entailed the use of a nummular magnet with a larger sheet metal.RESULTS With traditional MCA devices,the anastomoses experienced stenosis and even closure during the long-term follow-up when the anastomat was smaller thanΦ5 mm.However,the risk of leakage increased when it was larger thanΦ4 mm.On comparison of the different designs,it was found that the“fedora-type”MCA device should be composed of aΦ4-mm nummular magnet with aΦ6-mm sheet metal.CONCLUSION The diameter of the MCA device should be greater than 120%of the enteric diameter.The novel“fedora-type”MCA device controls the pressure and optimizes the size.Huan Chen Tao Ma Yue Wang Hao-Yang Zhu Zhe Feng Rong-Qian Wu Yi Lv Ding-Hui Dong 2020World Journal of Gastroenterology2020,26,42:7
7Characterization of inflammatory factor-induced changes in mesenchymal stem cell exosomes and sequencing analysis of exosomal microRNAs显示文摘BACKGROUND Treatments utilizing stems cells often require stem cells to be exposed to inflammatory environments,but the effects of such environments are unknown.AIM To examine the effects of inflammatory cytokines on the morphology and quantity of mesenchymal stem cell exosomes(MSCs-exo)as well as the differential expression of microRNAs(miRNAs)in the exosomes.METHODS MSCs were isolated from human umbilical tissue by enzymatic digestion.Exosomes were then collected after a 48-h incubation period in a serum-free medium with one of the following the inflammatory cytokines:None(control),vascular cell adhesion molecule-1(VCAM-1),tumor necrosis factor(TNF)α,and interleukin(IL)6.The morphology and quantity of each group of MSC exosomes were observed and measured.The miRNAs in MSCs-exo were sequenced.We compared the sequenced data with the miRBase and other non-coding databases in order to detect differentially expressed miRNAs and explore their target genes and regulatory mechanisms.In vitro tube formation assays and Western blot were performed in endothelial cells which were used to assess the angiogenic potential of MSCs-exo after inflammatory cytokine stimulation.RESULTS MSCs-exo were numerous,small,and regularly shaped in the VCAM-1 group.TNFαstimulated MSCs to secrete larger and irregular exosomes.IL6 led to a reduced quantity of MSCs-exo.Compared to the control group,the TNFαand IL6 groups had more downregulated differentially expressed miRNAs,particularly angiogenesis-related miRNAs.The angiogenic potential of MSCs-exo declined after IL6 stimulation.CONCLUSION TNFαand IL6 may influence the expression of miRNAs that down-regulate the PI3K-AKT,MAPK,and VEGF signaling pathways;particularly,IL6 significantly down-regulates the PI3K-AKT signaling pathway.Overall,inflammatory cytokines may lead to changes in exosomal miRNAs that abnormally impact cellular components,molecular function,and biological processes.Chen Huang Wen-Feng Luo Yu-Feng Ye Li Lin Zhe Wang Ming-Hua Luo Qi-De Song Xue-Ping He Han-Wei Chen Yi Kong Yu-Kuan Tang 2019World Journal of Stem Cells2019,11,10:7
8Characteristics of fecal microbial communities in patients with non-anastomotic biliary strictures after liver transplantation显示文摘AIM To explore the possible relationship between fecal microbial communities and non-anastomotic stricture(NAS) after liver transplantation(LT).METHODS A total of 30 subjects including 10 patients with NAS, 10 patients with no complications after LT, and 10 nonLT healthy individuals were enrolled. Fecal microbial communities were assessed by the 16 S r RNA gene sequencing technology.RESULTS Different from the uncomplicated and healthy groups, unbalanced fecal bacterium ratio existed in patients with NAS after LT. The results showed that NAS patients were associated with a decrease of Firmicutes and Bacteroidetes and an increase of Proteobacteria at the phylum level, with the proportion-ratio imbalance between potential pathogenic families including Enterococcaceae, Streptococcaceae, Enterobacteriaceae, Pseudomonadaceae and dominant families including Bacteroidaceae. 10Effect of Mg Addition on Nucleation of Intra-granular Acicular Ferrite in Al-killed Low Carbon Steel显示文摘To verify the formation behaviors and mechanisms of intra-granular acicular ferrite( IAF) grains nucleated by Mg-Al-O in low carbon steel,the steels containing different Mg contents were refined in a vacuum induction furnace. The effect of Mg addition on the formation of IAF structure in Al-killed low carbon steel was investigated by optical microscope( OM) and scanning electron microscope with energy dispersive X-ray spectroscope( SEM-EDX). It reveals that the IAFs are only detected in Mg-added steels,and the volume fraction of IAF increases with the Mg concentration from 8 × 10^(-6) to 26 × 10^(-6). It shows that not only the MgO-Al_2O_3-MnS and MgO-Al_2O_3-P_2O_5 particles are the effective nucleation sites for IAF,but also the pure MgO·Al_2O_3 phase can promote the ferrite nucleation. A Mn-depletion zone( MDZ) is characterized adjacent to the MgO-Al_2O_3-MnS,which is believed to be one of the possible mechanisms to explain the IAF nucleation. The MDZ around the MgO-Al_2O_3-MnS inclusion would be induced by the Mn S precipitation on the inclusion. 11Metformin activates chaperone-mediated autophagy and improves disease pathologies in an Alzheimer disease mouse model显示文摘Chaperone-mediated autophagy(CMA)is a lysosome-dependent selective degradation pathway implicated in the pathogenesis of cancer and neurodegenerative diseases.However,the mechanisms that regulate CMA are not fully understood.Here,using unbiased drug screening approaches,we discover Metformin,a drug that is commonly the first medication prescribed for type 2 diabetes,can induce CMA.We delineate the mechanism of CMA induction by Metformin to be via activation of TAK1-IKKα/β signaling that leads to phosphorylation of Ser85 of the key mediator of CMA,Hsc70,and its activation.Notably,we find that amyloid-beta precursor protein(APP)is a CMA substrate and that it binds to Hsc70 in an IKKα/β-dependent manner.The inhibition of CMA-mediated degradation of APP enhances its cytotoxicity.Importantly,we find that in the APP/PS1 mouse model of Alzheimer's disease(AD),activation of CMA by Hsc70 overexpression or Metformin potently reduces the accumulated brain Aβplaque levels and reverses the molecular and behavioral AD phenotypes.Our study elucidates a novel mechanism of CMA regulation via Metformin-TAK1-IKKα/β-Hsc70 signaling and suggests Metformin as a new activator of CMA for diseases,such as AD,where such therapeutic intervention could be beneficial.Xiaoyan Xu Yaqin Sun Xufeng Cen Bing Shan Qingwei Zhao Tingxue Xie Zhe Wang Tingjun Hou Yu Xue Mengmeng Zhang Di Peng Qiming Sun Cong Yi Ayaz Najafov Hongguang Xia 2021Protein & Cell2021,12,10:5

14Leptin activates the JAK/STAT pathway to promote angiogenesis in RF/6A cells in vitro显示文摘AIM: To investigate the effect of leptin on the angiogenesis of RF/6 A cells(monkey retinal choroidal endothelial cells) in vitro and test the cellular signaling in the mechanism.METHODS: RF/6 A cells were cultured in vitro and randomly divided into four groups: normal control, with leptin at 50, 100, 200 ng/m L for cell counting kit-8(CCK8). RF/6 A cell proliferation and migration were examined by Transwell assays, while RF/6 A cell tube formation by Matrigel assay. JAK2, p-JAK2, STAT3, and p-STAT3 protein expression was measured by Western blotting. Cells were then divided into the following treatment groups: control, 100 ng/m L leptin and AG-490(100 ng/m L leptin+10 μmol/L AG-490) for examinations of RF/6 A cellular behaviour again. Analysis of differences was carried out using one-way ANOVA and least significant difference(LSD).RESULTS: RF/6 A cell proliferation, migration and cell tube formation were promoted significantly by leptin in a dose-dependent manner(P<0.05). Western blotting showed that leptin up-regulated p-JAK2 and p-STAT3 expression levels. Treatment with the JAK/STAT pathway inhibitor, AG-490, decreased leptin-induced p-JAK2 and p-STAT3 expression, and inhibited cell proliferation, migration and cell tube formation induced by leptin(P<0.05). 15Nanoporous titanium implant surface promotes osteogenesis by suppressing osteoclastogenesis via integrin β1/FAKpY397/MAPK pathway显示文摘Macrophages and osteoclasts are both derived from monocyte/macrophage lineage,which plays as the osteoclastic part of bone metabolism.Although they are regulated by bone implant surface nanoarchitecture and involved in osseointegration,the beneath mechanism has not been simultaneously analyzed in a given surface model and their communication with osteoblasts is also blurring.Here,the effect of implant surface topography on monocyte/macrophage lineage osteoclastogenesis and the subsequent effect on osteogenesis are systematically investigated.The nanoporous surface is fabricated on titanium implant by etching and anodizing to get the nanotubes structure.The early bone formation around implant is significantly accelerated by the nanoporous surface in vivo.Meanwhile,the macrophage recruitment and osteoclast formation are increased and decreased respectively.Mechanistically,the integrin mediated FAK phosphorylation and its downstream MAPK pathway(p-p38)are significantly downregulated by the nanoporous surface,which account for the inhibition of osteoclastogenesis.In addition,the nanoporous surface can alleviate the inhibition of osteoclasts on osteogenesis by changing the secretion of clastokines,and accelerate bone regeneration by macrophage cytokine profiles.In conclusion,these data indicate that physical topography of implant surface is a critical factor modulating monocyte/macrophage lineage commitment,which provides theoretical guidance and mechanism basis for promoting osseointegration by coupling the osteogenesis and osteoclastogenesis.Yide He Zhe Li Xin Ding Boya Xu Jinjin Wang Yi Li Fanghao Chen Fanhui Meng Wen Song Yumei Zhang 2022Bioactive Materials2022,7,2:4
16Overexpression CPT1A reduces lipid accumulation via PPARα/CD36 axis to suppress the cell proliferation in ccRCC显示文摘Clear cell renal carcinoma(ccRCC)is histologically defined by its cytoplasmic lipid deposits.Lipid metabolism disorder largely increases the risk of ccRCC.In this study,we aimed to investigate the biological functions and molecular mechanisms of carnitine palmitoyl transferase 1A(CPT1A)in ccRCC.Our results showed that CPT1A is decreased in ccRCC clinical samples and cell lines compared with that in normal samples.Lentivirus overexpressing CPT1A was used to investigate the neoplastic phenotypes of ccRCC,and the results showed that lipid accumulation and tumor growth are attenuated both in vitro and in vivo.In addition,CPT1A prevents cholesterol uptake and lipid accumulation by increasing the peroxisome proliferator-activated receptorα(PPARα)level through regulation of Class B scavenger receptor type 1(SRB1)and cluster of differentiation 36(CD36).Furthermore,PI3K/Akt signaling pathway promotes tumor cell proliferation in ccRCC,which is related to the enhanced expression of CD36.Functionally,weakened CPT1A expression is critical for lipid accumulation to promote ccRCC development.Collectively,our research unveiled a novel function of CPT1A in lipid metabolism via PPARα/CD36 axis,which provides a new theoretical explanation for the pathogenesis of ccRCC.Targeting CPT1A may be a potential therapeutic strategy to treat ccRCC.Hui Yang Hongbo Zhao Zhongkun Ren Xiaojia Yi Qiao Zhang Zhe Yang Yingmin Kuang Yuechun Zhu 2022Acta Biochimica et Biophysica Sinica2022,54,2:4
