Chemical characters and protective effect of Baqi Lingmao formula(巴芪灵猫方) on experimental liver injury显示文摘OBJECTIVE:To investigate the chemical characters of water-extract of Baqi Lingmao formula(巴芪灵猫方,BQLM formula)and its effects on anti-liver injury in model mice and live cells.METHODS:BQLM formula was composed of ten herbal medicines. We determined the contents of alkaloids, saponins, phenolic acids and flavonoid in BQLM formula by UV spectrophotometry. The active components of alkaloids and phenolic acids in BQLM formula were identified by HPLC chromatography. The anti-hepatic injury effects of BQLM formula were investigated with concanavalin A(Con A)-induced hepatitis model of mice, human liver LO2 and Hep G2.2.15 cells.RESULTS:BQLM formula(2 and 10 g/kg,orally)significantly improved the damages of liver tissues and functions caused by Con A in mice, reduced the infiltration of inflammatory cells into liver and inhibited the inflammatory cytokine secretion of interferon-γ,tumor necrosis factor-αand interleukin-6.BQLM formula simultaneously decreased the levels of alanine aminotransferase and aspartate aminotransferase of liver and serum, and recovered the superoxide dismutase and catalase activities of liver to normal levels in Con A-induced hepatic-injury mice. The serum of BQLM formula group stimulated the human liver LO2 cell proliferation in vitro.Further,BQLM formula obviously promoted the proliferation of normal hepatocytes(LO2 cells)and inhibited the hepatocytes death induced by Con A.It also significantly inhibited the proliferation of Hep G2.2.15 cells and decreased the secretion of HBs Ag and HBe Ag in vitro.CONCLUSIONS:BQLM formula has anti-inflammation and anti-hepatitis virus Beffects, and is capable of improving liver injury in vivo and in vitro. | LIU Lulu CHEN Yanxu LIU Lulu LI Man XU Rui QU Wensheng YIN Jiye ZHANG Xin ZHOU Zhenhua GAO Yating NIE Dan SHAN Junjie GAO Yueqiu | 2021 | Journal of Traditional Chinese Medicine2021,41,6: | 0
FvWRKY50 is an important gene that regulates both vegetative growth and reproductive growth in strawberry显示文摘The WRKY transcription factors play important roles in plant growth and resistance, but only a few members have been identified in strawberry. Here we identified a WRKY transcription factor,FvWRKY50,in diploid strawberry which played essential roles in strawberry vegetative growth, and reproductive growth.Knocking out FvWRKY50 by genome editing accelerated flowering time and leaf senescence but delayed anthocyanin accumulation in fruit.Further analysis showed that FvWRKY50 acted as a transcriptional repressor to negatively regulate the expression of flowering-and leaf senescence-related genes, including FvFT2,FvCO,FvFT3,and FvSAUR36.Notably,FvWRKY50 directly upregulated the expression of FvCHI and FvDFR by binding their promoter under normal conditions, but at low temperature FvWRKY50 was phosphorylated by FvMAPK3 and then induced protein degradation by ubiquitination, delaying anthocyanin accumulation. In addition, the homozygous mutant of FvWRKY50 was smaller while the biallelic mutant showed normal size. These new findings provide important clues for us to further reveal the regulatory mechanisms of strawberry growth and fruit ripening. | Yating Chen Liping Liu Qianqian Feng Chuang Liu Yujuan Bao Nan Zhang Ronghui Sun Zhaonan Yin Chuanfei Zhong Yuanhua Wang Qian Li Bingbing Li | 2023 | Horticulture Research2023,10,7: | 0
Synergistic effects of plant hormones on spontaneous late-ripening mutant of'Jinghong'peach detected by transcriptome analysis显示文摘Objectives:Peach(Prunus persica L.)is an ancient fruit tree that originated from China. It is the climacteric fruit belonging to genus Prunus in family Rosaceae.Ethylene, which is produced during ripening, accelerates fruit softening, and therefore peaches cannot be stored for a long time.Materials and Methods:To study the mechanism of fruit late ripening, transcriptome analysis of the fruit of a late-ripening mutant of'Jinghong'peach was performed to identify genes and pathways involved in fruit late ripening. Results:A total of 1805,1511,and 2309 genes were found to be differentially expressed in W2_vs_M1,W3_vs_M2,and W3_vs_M3,respectively.Functional enrichment analysis of the differentially expressed genes showed they were related to carotenoid biosynthesis, starch and sucrose metabolism plant hormone signal transduction, flavonoid biosynthesis, and photosynthesis. The expression trends of ripening-related genes that encode transcription factors and plant hormone signal transduction-related genes that encode enzymes were similar. Conclusions:It will help to elucidate the transcriptional regulatory network of fruit development in the spontaneous late-ripening mutant of‘Jinghong’peach and provide a theoretical basis for understanding the molecular regulatory mechanism of fruit ripening. | Man Zhang Tingting Du Yarui Yin Hongyan Cao Zhihua Song Mao Ye Yating Liu Yanhong Shen Libin Zhang Qing Yang Dong Meng Junkai Wu | 2022 | Food Quality and Safety2022,6,1: | 0
The clinical effectiveness and safety of using epidermal growth factor, fibroblast growth factor and granulocyte-macrophage colony stimulating factor as therapeutics in acute skin wound healing:a systematic review and meta-analysis显示文摘Background:Promoting wound healing is crucial to restore the vital barrier function of injured skin.Growth factor products including epidermal growth factor(EGF),fibroblast growth factor(FGF)and granulocyte-macrophage colony stimulating factor(GM-CSF)have been used for decades although no systematic evaluation exists regarding their effectiveness and safety issues in treating acute skin wounds. This has resulted in a lack of guidelines and standards for proper application regimes. Therefore, this systematic review and meta-analysis was performed to critically evaluate the effectiveness and safety of these growth factors on skin acute wounds and provide guidelines for application regimes. Methods:We searched PubMed/Medline(1980-2020),Cochrane Library(1980-2020),Cochrane CENTRAL(from establishment to 2020),ClinicalTrials.gov(from establishment to 2020),Chinese Journal Full-text Database(CNKI,1994-2020),China Biology Medicine disc(CBM,1978-2019),Chinese Scientific Journal Database(VIP,1989-2020)andWanfang Database(WFDATA,1980-2019).Randomized controlled trials(RCTs),quasi-RCTs and controlled clinical trials treating patients with acute skin wounds from various causes and with those available growth factors were included. Results:A total of 7573 papers were identified through database searching;229 papers including 281 studies were kept after final screening.Administering growth factors significantly short-ened the healing time of acute skin wounds, including superficial burn injuries[mean differ-ence(MD)=−3.02;95%confidence interval(CI):−3.31∼−2.74;p<0.00001],deep burn injuries(MD=−5.63;95%CI:−7.10∼−4.17;p<0.00001),traumata and surgical wounds(MD=−4.50;95%CI:−5.55∼−3.44;p<0.00001).Growth factors increased the healing rate of acute skin wounds and decreased scar scores. The incidence of adverse reactions was lower in the growth factor treatment group than in the non-growth factor group. Conclusions:The studied growth factors not only are effective and safe for managing acute skin wounds, but also accelerate their healing with no severe adverse reactions. | Yating Wei Jiangfeng Li Yao Huang Xun Lei Lijun Zhang Meifang Yin Jiawen Deng Xiaoyan Wang Xiaobing Fu Jun Wu | 2022 | Burns & Trauma2022,10,1: | 0
Current Status of ADSCs-Enriched Fat Grafts in Plastic Surgery显示文摘Autologous fat grafting is an increasingly popular technique in plastic surgery for volume augmentation and rejuvenation. However, the unpredictability of long-term volume retention limits its clinical application.Various animal studies have documented the positive effects of adipose tissue-derived stem cells(ADSCs)on the acceleration of lipofilling. However, the results have been inconsistent, and there is an insufficient number of high-quality clinical studies to formulate evidence-based recommendations for ADSC-enriched fat grafts. Moreover, related technical standards, such as the final count of harvested ADSCs and the enrichment ratio, have not yet been established. This systematic review included all clinical trials on ADSC-enriched fat grafts in plastic surgery from PubMed in the past 10 years,as well as all registered clinical trials on ClinicalTrials.Gov.To examine the current landscape of ADSCs harvest,we summarize the current applications of ADSCs in the field of plastic surgery and discuss the current barriers to universal clinical use. | Yating YIN Jianhua LI Hao WANG Pingping WANG Wanling ZHENG Qiang LI Peisheng JIN | 2020 | Chinese Journal Of Plastic and Reconstructive Surgery2020,2,4: | 0
Ultra reliability and massive connectivity provision in integrated internet of military things(IoMT)based on tactical datalink显示文摘One of the major challenges arising in internet of military things(IoMT)is accommodating massive connectivity while providing guaranteed quality of service(QoS)in terms of ultra-high reliability. In this regard, this paper presents a class of code-domain nonorthogonal multiple accesses(NOMAs)for uplink ultra reliable networking of massive IoMT based on tactical datalink such as Link-16 and joint tactical information distribution system(JTIDS).In the considered scenario,a satellite equipped with Nr antennas servers K devices including vehicles, drones, ships, sensors, handset radios, etc.Nonorthogonal coded modulation,a special form of multiple input multiple output(MIMO)-NOMA is proposed. The discussion starts with evaluating the output signal to interference-plus-noise(SINR)of receiver filter, leading to the unveiling of a closed-form expression for overloading systems as the number of users is significantly larger than the number of devices admitted such that massive connectivity is rendered. The expression allows for the development of simple yet successful interference suppression based on power allocation and phase shaping techniques that maximizes the sum rate since it is equivalent to fixed-point programming as can be proved. The proposed design is exemplified by nonlinear modulation schemes such as minimum shift keying(MSK)and Gaussian MSK(GMSK),two pivotal modulation formats in IoMT standards such as Link-16 and JITDS.Numerical results show that near capacity performance is offered.Fortunately, the performance is obtained using simple forward error corrections(FECs)of higher coding rate than existing schemes do,while the transmit power is reduced by 6 dB.The proposed design finds wide applications not only in IoMT but also in deep space communications, where ultra reliability and massive connectivity is a keen concern. | Li Bing Yating Gu Lanke Hu Li Bowen Yang Lihua Jue Wang Yue Yin | 2024 | Defence Technology(防务技术)2024,33,3: | 0
A novel method for objectively, rapidly and accurately evaluating burn depth via near infrared spectroscopy显示文摘The accurate and objective evaluation of burn depth is a significant challenge in burn wound care.Herein,we used near infrared spectroscopy(NIRS)technology to measure the different depth of thermal burns in ex vivo porcine models.Based on the intensity of the spectral signals and the diffuse reflection theory,we extracted the optical parameters involved in functional(total hemoglobin andwater content)and structural(tissue scattered size and scattered particles)features that reflect the changes in burn depth.Next,we applied support vector regression to construct a model including the optical property parameters and the burn depth. Finally,we histologically verified the burn depth data collected via NIRS.The results showed that our inversion model could achieve an average relative error of about 7.63%,while the NIRS technology diagnostic accuracy was in the range of 50μm.For the first time, this novel technique provides physicians with real-time burn depth information objectively and accurately. | Meifang Yin Yongming Li Yongquan Luo Mingzhou Yuan Ubaldo Armato Ilaria Dal Prà Lijun Zhang Dayong Zhang Yating Wei Guang Yang Lixian Huang Pin Wang Jun Wu | 2021 | Burns & Trauma2021,9,1: | 0
Microbially-mediated formation of Ca-Fe carbonates during dissimilatory ferrihydrite reduction:Implications for the origin of sedimentary ankerite显示文摘The origin of sedimentary dolomite has become a long-standing problem in the Earth Sciences. Some carbonate minerals like ankerite have the same crystal structure as dolomite, hence their genesis may provide clues to help solving the dolomite problem. The purpose of this study was to probe whether microbial activity can be involved in the formation of ankerite.Bio-carbonation experiments associated with microbial iron reduction were performed in batch systems with various concentrations of Ca^(2+)(0–20 mmol/L),with a marine iron-reducing bacterium Shewanella piezotolerans WP3 as the reaction mediator, and with lactate and ferrihydrite as the respective electron donor and acceptor. Our biomineralization data showed that Ca-amendments expedited microbially-mediated ferrihydrite reduction by enhancing the adhesion between WP3 cells and ferrihydrite particles. After bioreduction, siderite occurred as the principal secondary mineral in the Ca-free systems.Instead,Ca-Fe carbonates were formed when Ca^(2+)ions were present. The CaCO_(3) content of microbially-induced Ca-Fe carbonates was positively correlated with the initial Ca2+concentration.The Ca-Fe carbonate phase produced in the 20 mmol/L Ca-amended biosystems had a chemical formula of Ca_(0.8)Fe_(1.2)(CO_(3))_(2),which is close to the theoretical composition of ankerite. This ankeritelike phase was nanometric in size and spherical,Ca-Fe disordered, and structurally defective. Our simulated diagenesis experiments further demonstrated that the resulting ankerite-like phase could be converted into ordered ankerite under hydrothermal conditions. We introduced the term“proto-ankerite”to define the Ca-Fe phases that possess near-ankerite stoichiometry but disordered cation arrangement. On the basis of the present study,we proposed herein that microbial activity is an important contributor to the genesis of sedimentary ankerite by providing the metastable Ca-Fe carbonate precursors. | Deng LIU Jinpeng CAO Shanshan YANG Yating YIN Pengcong WANG Dominic PAPINEAU Hongmei WANG Xuan QIU Genming LUO Zongmin ZHU Fengping WANG | 2024 | Science China Earth Sciences2024,67,1: | 0
Experimental Investigation on Local Behaviors of PEMFC with Segmented Cell显示文摘When designing a cell stack and developing an operational strategy for proton exchange membrane fuel cell,it is critical to characterize the local current, water and heat.To measure distributions of current density, relative humidity and temperature for both anode and cathode simultaneously along the straight parallel flow channels, this paper uses a segmented tool based on the multilayered printed circuit board flow field plates with embedded sensors. In this study, two kinds of experimental operations of fuel cell reactants are carried out for comparison:the co-flow operation with identical gas flow direction of hydrogen and air and the counter-flow operation with opposite gas flow directions. The detected relative humidity(RH)distributions of both anode and cathode indicate that the asymmetry of RH distribution at two sides of the membrane in counter-flow operation is better at holding water inside the fuel cell compared with the co-flow operation. The in situ measured performance distributions show that segments around the middle of the fuel cell contribute the highest current in counter-flow operation, while for co-flow operation, the current peak locates near the outlet of reactants. | Cong Yin Yan Gao Kai Li Yating Song Hao Tang | 2021 | Automotive Innovation2021,4,2: | 0
CircUCP2 promotes the tumor progression of non-small cell lungcancer through the miR-149/UCP2 pathway显示文摘Non-small cell lung cancer(NSCLC)is a highly lethal cancer, and better treatments are urgently needed. Many studies have implicated circular RNAs(circRNAs)in the progression of multiple malignant tumors.Nonetheless, the functions of circRNAs in NSCLC remain unclear. To study new targets for the treatment of NSCLC,circRNA expression profiling was performed on NSCLC tissues and para-carcinoma nonmalignant tissues.RNA was isolated and used for circRNA sequencing.Biological studies were performed in vitro and in vivo to determine the functions of circRNAs in NSCLC,including their functions in cell proliferation and migration. How circRNAs function in NSCLC was explored to clarify the underlying regulatory mechanisms. We found that circUCP2 was upregulated in NSCLC tissues compared with neighboring nonmalignant tissues.circUCP2 promoted the proliferation and metastasis of NSCLC cells.circUCP2 promoted NSCLC progression by sponging miR-149 and upregulating UCP2.The circUCP2/miR-149/UCP2 axis accelerates the progression of NSCLC,and circUCP2 may therefore be a novel diagnostic biomarker for the progression of NSCLC. | WEI DU FANG YIN YATING ZHONG MINJIE LUO ZHEN WANG PENG LIN QING LIU HAN YANG | 2023 | Oncology Research2023,31,6: | 0
