| 1 | Efficacy and safety of metformin and sitagliptin based triple antihyperglycemic therapy(STRATEGY):a multicenter,randomized,controlled,non-inferiority clinical trial显示文摘Despite the current guideline's recommendation of a timely stepwise intensification therapy,the 'clinical inertia',termed as the delayed treatment intensification,commonly exists in the real world,which may be partly due to the relatively little substantial evidence and no clear consensus regarding the efficacy and safety of triple oral agents in patients inadequately controlled with dual therapy.In this clinical trial performed in 237 centers in China,5,535 type 2 diabetic patients inadequately controlled by previous therapies were treated with a stable metformin/sitagliptin dual therapy for 20 weeks.The patients who did not reach the glycated hemoglobin A1c(HbA1c) goal were then further randomized into glimepiride,gliclazide,repaglinide,or acarbose group for an additional 24-week triple therapy.A mean HbAlc reduction of 0.85%was observed when sitagliptin was added to the patients inadequately controlled with metformin in 16 weeks.Further HbAlc reductions in the 24-week triple therapy stage were 0.65%in glimepiride group,0.70%in gliclazide group,0.61%in repaglinide group,and 0.45%in acarbose group.The non-inferiority criterion for primary hypotheses was met for gliclazide and repaglinide,but not for acarbose,compared with glimepiride,when added to metformin/sitagliptin dual therapy.The incidences of adverse events(AEs) were 29.2%in the dual therapy stage and30.3%in the triple therapy stage.Metformin/sitagliptin as baseline therapy,with the addition of a third oral antihyperglycemic agent,including glimepiride,gliclazide,repaglinide,or acarbose,was effective,safe and well-tolerated for achieving an HbAlc<7.0%goal in type 2 diabetic patients inadequately controlled with previous therapies.The timely augmentation of up to three oral antihyperglycemic agents is valid and of important clinical benefit to prevent patients from exposure to unnecessarily prolonged hyperglycemia. | Wen Xu Yiming Mu Jiajun Zhao Dalong Zhu Qiuhe Ji Zhiguang Zhou Bin Yao Anhua Mao Samuel S.Engel Bin Zhao Yan Bi Longyi Zeng Xingwu Ran Juming Lu Linong Ji Wenying Yang Weiping Jia Jianping Weng | 2017 | Science China(Life Sciences)2017,60,3: | 19 |
| 2 | A restoration-promoting integrated floating bed and its experimental performance in eutrophication remediation显示文摘Numerous studies on eutrophication remediation have mainly focused on purifying water first, then restoring submerged macrophytes. A restoration-promoting integrated floating bed(RPIFB) was designed to combine the processes of water purification and macrophyte restoration simultaneously. Two outdoor experiments were conducted to evaluate the ecological functions of the RPIFB. Trial 1 was conducted to compare the eutrophication purification among floating bed, gradual-submerging bed(GSB) and RPIFB technologies. The results illustrated that RPIFB has the best purification capacity. Removal efficiencies of RPIFB for TN, TP, NH+ 4-N, NO-3-N, CODCr, Chlorophyll-a and turbidity were 74.45%, 98.31%, 74.71%, 88.81%, 71.42%, 90.17% and 85%, respectively. In trial 2, influences of depth of GSB and photic area in RPIFB on biota were investigated. When the depth of GSB decreased and the photic area of RPIFB grew, the height of Potamogeton crispus Linn. increased, but the biomass of Canna indica Linn. was reduced. The mortalities of Misgurnus anguillicaudatus and Bellamya aeruginosa in each group were all less than 7%. All results indicated that when the RPIFB was embedded into the eutrophic water, the regime shift from phytoplankton-dominated to macrophyte-dominated state could be promoted. | 3 | Schisandra sphenanthera extract(Wuzhi Tablet)protects against chronic-binge and acute alcohol-induced liver injury by regulating the NRF2-ARE pathway in mice显示文摘Alcohol abuse leads to alcoholic liver disease and no effective therapy is currently available.Wuzhi Tablet(WZ), a preparation of extract from Schisandra sphenanthera that is a traditional hepatoprotective herb, exerted a significant protective effect against acetaminophen-induced liver injury in our recent studies, but whether WZ can alleviate alcohol-induced toxicity remains unclear. This study aimed to investigate the contribution of WZ to alcohol-induced liver injury by using chronic-binge and acute models of alcohol feeding. The activities of ALT and AST in serum were assessed as well as the level of GSH and the activity of SOD in the liver. The expression of CYP2E1 and proteins in the NRF2-ARE signaling pathway including NRF2, GCLC, GCLM, HO-1 were measured, and the effect of WZ on NRF2 transcriptional activity was determined. We found that both models resulted in liver steatosis accompanied by increased transaminase activities, but that liver injury was significantly attenuated by WZ. WZ administration also inhibited CYP2E1 expression induced by alcohol, and elevated the level of GSH and the activity of SOD in the liver. Moreover, the NRF2-ARE signaling pathway was activated by WZ andthe target genes were all upregulated. Furthermore, WZ significantly activated NRF2 transcriptional activity. | 5 | Hydrothermal activities and seawater acidification in the Late Devonian F-F transition:Evidence from geochemistry of rare earth elements显示文摘Rare-earth elements(REE) in limestones were measured by ICP-MS from the Yangdi section across the Frasnian-Famennian(F-F) transition in Guilin,Guangxi of South China.The averageΣREE contents of measured samples are 33.32 ppm.The average LREE/HREE ratio is 6.35.Data show LREE-enriched REE distribution pattern in the Yangdi section.Most of samples are characterized by positive Eu anomaly with average of 1.13,and there are two maxima of Eu anomaly just below the F-F boundary.Almost all analytical samples show obvious tetrad effects.Remarkable M-shaped tetrad effects are observed in these samples with positive Eu anomaly.The characters of REE and some relative elements(Fe,Mn,Al,etc.) indicate that hydrothermal activities occurred and resulted in seawater acidification in Upper and Lower Kellwasser horizons of the Late Devonian.The changes of marine environment caused by hydrothermal activities,such as seawater acidification,heavy metal pollution,and eutrophication,would destroy the neritic ecosystem and finally result in the F-F mass extinction. | ZENG JianWei XU Ran GONG YiMing | 2011 | Science China Earth Sciences2011,54,4: | 11 |
| 6 | Effect of organic wastes on the plant-microbe remediation for removal of aged PAHs in soils显示文摘The effectiveness of in-situ bioremediation of polycyclic aromatic hydrocarbons(PAHs) may be inhibited by low nutrients and organic carbon.To evaluate the effect of organic wastes on the PAHs removal efficiency of a plant-microbe remediation system,contaminated agricultural soils were amended with different dosages of sewage sludge(SS) and cattle manure(CM) in the presence of alfalfa(Medicago sativa L.) and PAHs-degraders(Bacillus sp.and Flavobacterium sp.).The results indicated that the alfalfa mean biomasses varied from 0.56 to 2.23 g/pot in root dry weight and from 1.80 to 4.88 g/pot in shoot dry weight.Low dose amendments,with rates of SS at 0.1% and CM at 1%,had prominent effects on plant growth and soil PAHs degradation.After 60-day incubation,compared with about 5.6% in the control,25.8% PAHs removal was observed for treatments in the presence of alfalfa and PAHs-degraders;furthermore,when amended with different dosages of SS and CM,the removed PAHs from soils increased by 35.5%-44.9% and 25.5%-42.3%,respectively.In particular,the degradation of high-molecular-weight PAHs was up to 42.4%.Dehydrogenase activities(DH) ranged between 0.41 and 1.83 μg triphenylformazan/(g dry soil·hr) and the numbers of PAHs-degrading microbes(PDM) ranged from 1.14×10 6 to 16.6×10 6 most-probable-number/g dry soil.Further investigation of the underlying microbial mechanism revealed that both DH and PDM were stimulated by the addition of organic wastes and significantly correlated with the removal ratio of PAHs.In conclusion,the effect of organic waste application on soil PAHs removal to a great extent is dependent on the interactional effect of nutrients and dissolved organic matter in organic waste and soil microorganisms. | Jing Zhang Xiangui Lin Weiwei Liu Yiming Wang Jun Zeng Hong Chen | 2012 | Journal of Environmental Sciences2012,24,8: | 11 |
| 7 | Devonian Frasnian-Famennian transitional event deposits of Guangxi, South China and their possible tsunami origin显示文摘A suite of event deposits, isochronous and interrelated in origin, occurs widely near the Frasnian- Famennian boundary in Guangxi, South China. It is mainly distributed in the facies areas of the platform-margin slope and inter-platform rift-trough. The rudstone or calcirudite occur in the Liujing section of Hengxian and Luoxiu and Baqi sections of Xiangzhou in the facies area of platform-margin slope. The turbidites are observed in the sections of Yangdi and Baisha of Guilin, Du'an of Debao, Nandong and Sanli of Wuxuan, Xiangtian, Ma'anshan of Xiangzhou, Nayi of Chongzuo, Yunpan of Shanglin in the facies area of inter-platform rift-trough. The massive homogenites occur in sections of Mangchang, Luofu and Road from Nandan to Tian'e and Ma'anshan etc in the facies area of inter-platform rift-trough. Herein event deposits can be correlated in stratohorizon to the turbidite in the bottom of the Lower triangularis Zone in Hony railroad cut of Belgium, Devils Gate of Nevada, USA, Atrous of Morocco, South Urals and Fore-Kolyma of northeastern Siberia of Russia, and erosional discontinuities and brecciation in the Frasnian-Famennian boundary in South Polish-Moravian shelf. | 10 | Coupling relationships between brachiopods and Girvanella during the Late Devonian F-F transition in Guilin, South China显示文摘The abundance curves derived from area proportions of brachiopods and Girvanella in the thin sections from the Yangdi section, South China, exhibits complex relationships during the Late Devonian Frasnian-Famennian transition. The living activity of brachiopods such as grazing and borrowing did great damage to the growth of Girvanella. However, there was more to just a mere a survival competing relationship between them, Girvanella actually improved marine environments by oxygenating the dysoxic ambient sea-water through photosynthesis in the lower Kellwasser Horizon. Profited from this improvement brachiopods' abundance increased subsequently and suppressed Girvanella again. Nonetheless, without Girvanella's photosynthesis, brachiopods were wiped out by the farther anoxic environments in the upper Kellwasser Horizon. | 11 | A new kind of magnetic targeting induction heating drug carrier and its physical and biological properties显示文摘Nano-carbon and iron composite―carbon-coated iron nanoparticles (CCINs) produced by carbon arc method can be used as a new kind of magnetic targeting induction heating drug carrier for cancer therapy. The structure and morphology of CCINs are studied by X-ray diffraction (XRD) and transmission electron microscope (TEM). Mossbauer spectra of these nanoparticles show that they contain only iron and carbon, without ferric carbide and ferric oxide. CCINs can be used as the magnetic drug carrier, with the effect of targeting magnetic induction heating in its inner core and higher drug adsorption in its nano-carbon shell outside because of its high specific surface area. CCINs can absorb Epirubicin (EPI) of 160 μg/mg measured by an optical spectrometer. In acute toxicity experiment with mice, the median lethal dose (LD50) of EPI is 16.9 mg/kg, while that of EPI-CCINs mixture is 20.7 mg/kg and none of the mice died after pure CCINs medication. The results show that pure CCINs belong to non-toxic grade and EPI delivery in mixture with CCINs can reduce its acute toxicity in mice. The magnetic properties of CCINs and their magnetic induction heating are investigated. The iron nanoparticle in its inner core has better magnetism with a good effect on targeting magnetic induction heating. When the CCINs are mixed with physiological salt water and are injected uniformly in pig's liver, the temperature goes up to 48℃. While in the case that CCINs are filled in a certain section of pig's liver, the temperature goes up to 52℃. In both cases the temperature is high enough to kill the cancer cell.
