| 2 | Development of a novel RNAi therapy:Engineered miR-31 exosomes promoted the healing of diabetic wounds显示文摘Rationale:Chronic wounds associated with diabetes exact a heavy burden on individuals and society and do not have a specific treatment.Exosome therapy is an extension of stem cell therapy,and RNA interference(RNAi)-based therapy is a type of advanced precision therapy.Based on the discovery of chronic wound-related genes in diabetes,we combined exosome therapy and RNAi therapy through an engineering approach for the treatment of diabetic chronic wounds.Methods:We combined exosome therapy and RNAi therapy to establish a precision therapy for diabetes-associated wounds via an engineered exosome approach.Results:First,chronic diabetic wounds express low levels of miR-31-5p compared with nondiabetic wounds,and an miR-31-5p mimic was shown to be effective in promoting the proliferation and migration of three wound-related cell types in vitro.Second,bioinformatics analysis,luciferase reporter assays and western blotting suggested that miR-31-5p promoted angiogenesis,fibrogenesis and reepithelization by inhibiting factor-inhibiting HIF-1(HIF1AN,also named FIH)and epithelial membrane protein-1(EMP-1).Third,engineered miR-31 exosomes were generated as a miR-31-5p RNAi therapeutic agent.In vivo,the engineered miR-31 exosomes promoted diabetic wound healing by enhancing angiogenesis,fibrogenesis and reepithelization.Conclusion:Engineered miR-31 exosomes are an ideal disease pathophysiology-initiated RNAi therapeutic agent for diabetic wounds. | Jinghuan Huang Muyu Yu Wenjing Yin Bo Liang Ang Li Jingfeng Li Xiaolin Li Shichang Zhao Fang Liu | 2021 | Bioactive Materials2021,6,9: | 3 |
| 3 | Grain Growth Behavior of 5Cr21Mn9Ni4N Steel Micro-Alloyed by Rare Earth显示文摘Grain growth behavior of 5Cr21Mn9Ni4NRE steel was experimentally studied at various solid solution treatment temperatures and holding for different times. The experimental results show that the 5Cr21Mn9Ni4NRE steel has the feature of sharp austenite grain coarsening after solid solution treatment at the temperature above 1150 ℃. RE added in the steel has the benefit to restrain grain growth and increase grain growth activation energy. Grain growth mechanism is self-diffusion controlling process. The dispersed carbo-nitride in the steel greatly restrains grain growth. Solid solution treatment at the temperature below 1100 ℃ leads to little increase of grain size and little decrease of Rockwell hardness for mass carbo-nitride undissolved, whereas solid solution treatment at the temperature above 1150 ℃ leads to rapid grain coarsening and Rockwell hardness decrease. | 7 | Conspicuous cruciform silk decorations deflect avian predator attacks显示文摘Although camouflage as an effective antipredator defense strategy is widespread across animals,highly conspicuous color patterning is not uncommon either.Many orb-web spiders adorn their webs with extra bright white silk.These conspicuous decorations are hypothesized to deter predators by warning the presence of sticky webs,camouflaging spiders,acting as a decoy,or intimidating predators by their apparent size.The decorations may also deflect predator attacks from spiders.However,empirical evidence for this deflection function remains limited.Here,we tested this hypothesis using the X-shaped silk cruciform decorations built by females of Argiope minuta.We employed visual modeling to quantify the conspicuousness of spiders and decorations from a perspective of avian predators.Then,we determined actual predation risk on spiders using naïve chicks as predators.Spider bodies and decorations were conspicuous against natural backgrounds to the avian visual systems.Chicks attacked the spider main bodies significantly less frequently on the decorated webs than on the undecorated webs,thus reducing predation risk.When both spiders and decorations were present,chicks also attacked the spider main bodies and their legs or decorations,and not randomly:they attacked the legs or decorations sooner and more frequently than they attacked the main bodies,independent of the ratio of the surface area between the decoration and spider size.Despite the increase in detectability,incorporating a conspicuous cruciform decoration to the web effectively defends the spider by diverting the attack toward the decoration or leg,but not by camouflaging or intimidating,thus,supporting the deflection hypothesis. | Bingjun WANG Long YU Nina MA Zengtao ZHANG Deyong GONG Rui LIU Daiqin LI Shichang ZHANG | 2022 | Integrative Zoology2022,17,5: | 1 |

The results indicate that oxidation exponent n and oxidation activation energy are increased, and oxidation velocity constant k_p is decreased when 0.2% RE is added in 5Cr21Mn9Ni4N steel. The addition of RE elements does not alter phase constitution of oxidation scale, however it improves the configuration of oxidation scale, and increases thermal stability and adhesivity of oxidation scale, which results in the raise of oxidation resistance of 5Cr21Mn9Ni4N steel at high temperature. The oxidation scale constitutes of refractory steel transfer from manganic oxide mostly to ferric oxide mostly with the increase of temperature, which leads to descend of compactness and desquamation resistance of oxidation scale. | 19 | Capturing the hierarchically assorted modules of protein–protein interactions in the organized nucleome显示文摘Nuclear proteins are major constituents and key regulators of nucleome topological organization and manipulators of nuclear events.To decipher the global connectivity of nuclear proteins and the hierarchically organized modules of their interactions,we conducted two rounds of cross-linking mass spectrometry(XL-MS)analysis,one of which followed a quantitative double chemical cross-linking mass spectrometry(in vivo qXL-MS)workflow,and identified 24,140 unique crosslinks in total from the nuclei of soybean seedlings.This in vivo quantitative interactomics enabled the identification of 5340 crosslinks that can be converted into 1297 nuclear protein–protein interactions(PPIs),1220(94%)of which were non-confirmative(or novel)nuclear PPIs compared with those in repositories.There were 250 and 26 novel interactors of histones and the nucleolar box C/D small nucleolar ribonucleoprotein complex,respectively.Modulomic analysis of orthologous Arabidopsis PPIs produced 27 and 24 master nuclear PPI modules(NPIMs)that contain the condensate-forming protein(s)and the intrinsically disordered region–containing proteins,respectively.These NPIMs successfully captured previously reported nuclear protein complexes and nuclear bodies in the nucleus.Surprisingly,these NPIMs were hierarchically assorted into four higher-order communities in a nucleomic graph,including genome and nucleolus communities.This combinatorial pipeline of 4C quantitative interactomics and PPI network modularization revealed 17 ethylene-specific module variants that participate in a broad range of nuclear events.The pipeline was able to capture both nuclear protein complexes and nuclear bodies,construct the topological architectures of PPI modules and module variants in the nucleome,and probably map the protein compositions of biomolecular condensates. | Shuaijian Dai Shichang Liu Chen Zhou Fengchao Yu Guang Zhu Wenhao Zhang Haiteng Deng Al Burlingame Weichuan Yu Tingliang Wang Ning Li | 2023 | Molecular Plant2023,16,5: | 0 |
| 20 | Impact of the Shrinkage of Arctic Sea Ice on Eurasian Snow Cover Changes in 1979-2021显示文摘Recent research has shown that snow cover induces extreme wintertime cooling and has detrimental impacts.Although the dramatic loss of Arctic sea ice certainly has contributed to a more extreme climate,the mechanism connecting sea-ice loss to extensive snow cover is still up for debate.In this study,a significant relationship between sea ice concentration(SIC)in the Barents-Kara(B-K)seas in November and snow cover extent over Eurasia in winter(November-January)has been found based in observational datasets and through numerical experiments.The reduction in B-K sea ice gives rise to a negative phase of Arctic Oscillation(AO),a deepened East Asia trough,and a shallow trough over Europe.These circulation anomalies lead to colder-than-normal Eurasian mid-latitude temperatures,providing favorable conditions for snowfall.In addition,two prominent cyclonic anomalies near Europe and Lake Baikal affect moisture transport and its divergence,which results in increased precipitation due to moisture advection and wind convergence.Furthermore,anomalous E-P flux shows that amplified upward propagating waves associated with the low SIC could contribute to the weakening of the polar vortex and southward breakouts of cold air.This work may be helpful for further understanding and predicting the snowfall conditions in the middle latitudes. | Qian YANG Shichang KANG Haipeng YU Yaoxian YANG | 2023 | Advances in Atmospheric Sciences2023,40,12: | 0 |
