1Generation of GGTA1 biallelic knockout pigs via zinc-finger nucleases and somatic cell nuclear transfer显示文摘Genetically modified pigs are valuable models of human disease and donors of xenotransplanted organs.Conventional gene targeting in pig somatic cells is extremely inefficient.Zinc-finger nuclease(ZFN)technology has been shown to be a powerful tool for efficiently inducing mutations in the genome.However,ZFN-mediated targeting in pigs has rarely been achieved.Here,we used ZFNs to knock out the porcineα-1,3-galactosyl-transferase(GGTA1)gene,which generates Gal epitopes that trigger hyperacute immune rejection in pig-to-human transplantation.Primary pig fibroblasts were transfected with ZFNs targeting the coding region of GGTA1.Eighteen mono-allelic and four biallelic knockout cell clones were obtained after drug selection with efficiencies of 23.4%and 5.2%,respectively.The biallelic cells were used to produce cloned pigs via somatic cell nuclear transfer(SCNT).Three GGTA1 null piglets were born,and one knockout primary fibroblast cell line was established from a cloned fetus.Gal epitopes on GGTA1 null pig cells were completely eliminated from the cell membrane.Functionally,GGTA1 knockout cells were protected from complement-mediated immune attacks when incubated with human serum.This study demonstrated that ZFN is an efficient tool in creating gene-modified pigs.GGTA1 null pigs and GGTA1 null fetal fibroblasts would benefit research and pig-to-human transplantation.BAO Lei CHEN HaiDe JONG UiMyong RIM CholHo LI WenLing LIN XiJuan ZHANG Dan LUO Qiong CUI Chun HUANG HeFeng ZHANG Yan XIAO Lei FU ZhiXin 2014Science China(Life Sciences)2014,57,2:9
2GaN-based substrates and optoelectronic materials and devices显示文摘In order to solve the problems of GaN heteroepitaxy on sapphire substrate,some techniques were explored.Freestanding GaN substrates have been made by hydride vapor phase epitaxy(HVPE),laser lift-off(LLO),and chemical mechanical polishing techniques.Wafer bending and cracking in the HVPE growth were partly settled by pulsed flow modulation method.High-crystal quality was established for 1.2 mm thick GaN substrate by X-ray diffraction measurement,in which the full width of half maximum values were 72,110 arcsec for(102),(002)peaks.A novel micro-size patterned sapphire substrate(PSS)and a nano PSS were also fabricated.High-power vertical structure light emitting diodes(VSLEDs)have been developed by Au–Sn eutectic wafer bonding,homemade micro-area LLO,and light extraction structure preparation.The high-injection-level active region with low temperature GaN sandwiched layers was used for lowefficiency droop.The light output power of VSLED was achieved as 400 mW driven at 350 mA,and the dominant wavelength is about 460 nm.The structures and properties of strain modulated superlattices(SLs)and quantum wells as well as advanced simulation of carriers transport across the electron blocking layer were investigated in laser diodes.The hole concentration was achieved as high as1.6 9 1018cm-3in AlGaN/GaN SLs:Mg by inserting an AlN layer.High-quality AlGaN epilayers and structures were grown by MOCVD.Some device structures of UV LEDs and detectors were demonstrated.The emission wavelength of 262 nm UV LED has been successfully fabricated.At last,high-quality InN and InGaN materials for solar cell were grown by boundary-temperature-controlled epitaxy and growth-temperature-controlled epitaxy.Hall-effect measurement showed a recorded electron mobility of 3,280 cm2/(V s)and a residual electron concentration of 1.47 9 1017cm-3at 300 K.Guoyi Zhang Bo Shen Zhizhong Chen Xiaodong Hu Zhixin Qin Xinqiang Wang Jiejun Wu Tongjun Yu Xiangning Kang Xingxing Fu Wei Yang Zhijian Yang Zhizhao Gan 2014Chinese Science Bulletin2014,59,12:4

4Applications of single-cell technology on bacterial analysis显示文摘Background: Traditionally,scientists studied microbiology through the manner of batch cultures,to conclude the dynamics or outputs by averaging all individuals.However,as the researches go further,the heterogeneities among the individuals have been proven to be crucial for the population dynamics and fates.Results:Due to the limit of technology,single-cell analysis methods were not widely used to decipher the inherent connections between individual cells and populations.Since the early decades of this century,the rapid development of microfluidics,fluorescent labelling,next-generation sequencing,and high-resolution microscopy have speeded up the development of single-cell technologies and further facilitated the applications of these technologies on bacterial analysis.Conclusions:In this review,we summarized the recent processes of single-cell technologies applied in bacterial analysis in terms of intracellular characteristics,cell physiology dynamics,and group behaviors,and discussed how single-cell technologies could be more applicable for future bacterial researches.Zhixin Ma Pan MChu Yingtong Su Yue Yu Hui Wen Xiongfei Fu Shuqiang Huang 2019Quantitative Biology2019,7,3:2

7Molecular insights of nanozymes from design to catalytic mechanism显示文摘Emerging as cost-effective potential alternatives to natural enzymes,nanozymes have attracted increasing interest in broad fields.To exploit the in-depth potential of nanozymes,rational structural engineering and explicit catalytic mechanisms at the molecular scale are required.Recently,impressive progress has been made in mimicking the characteristics of natural enzymes by constructing metal active sites,binding pockets,scaffolds,and delicate allosteric regulation.Ingenious in-depth studies have been conducted with advances in structural characterization and theoretical calculations,unveiling the“black box”of nanozymecatalytic mechanisms.This review introduces the state-of-art synthesis strategies by learning from the natural enzyme counterparts and summarizes the general overview of the nanozyme mechanism with a particular emphasis on the adsorbed intermediates and descriptors that predict the nanozyme activity.The emerging activity assessment methodology that illustrates the relationship between electrochemical oxygen reduction and enzymatic oxygen reduction is discussed with up-to-date advances.Future opportunities and challenges are presented in the end to spark more profound work and attract more researchers from various backgrounds to the flourishing field of nanozymes.Yuan Xu Zhixin Zhou Nankai Deng Kangchun Fu Caixia Zhu Qing Hong Yanfei Shen Songqin Liu Yuanjian Zhang 2023Science China Chemistry2023,66,5:1
8Hollow Bio-derived Polymer Nanospheres with Ordered Mesopores for Sodium-Ion Battery显示文摘Bio-inspired hierarchical self-assembly provides elegant and powerful bottom-up strategies for the creation of complex materials.However,the current self-assembly approaches for natural bio-compounds often result in materials with limited diversity and complexity in architecture as well as microstructure.Here,we develop a novel coordination polymerization-driven hierarchical assembly of micelle strategy,using phytic acid-based natural compounds as an example,for the spatially controlled fabrication of metal coordination bio-derived polymers.The resultant ferric phytate polymer nanospheres feature hollow architecture,ordered meso-channels of^12 nm,high surface area of 401 m2 g−1,and large pore volume of 0.53 cm3 g−1.As an advanced anode material,this bio-derivative polymer delivers a remarkable reversible capacity of 540 mAh g−1 at 50 mA g−1,good rate capability,and cycling stability for sodium-ion batteries.This study holds great potential of the design of new complex bio-materials with supramolecular chemistry.Yan Ai Yuxiu You Facai Wei Xiaolin Jiang Zhuolei Han Jing Cui Hao Luo Yucen Li Zhixin Xu Shunqi Xu Jun Yang Qinye Bao Chengbin Jing Jianwei Fu Jiangong Cheng Shaohua Liu 2020Nano-Micro Letters2020,12,3:1

16Ecological significance and risks of mineral licks to mammals in a nature reserve on the Eastern Qinghai-Tibet Plateau显示文摘Mineral licks are important components of terrestrial ecosystems that have special value to the maintenance of fluid balance of mammals,by providing them with essential mineral nutrients such as sodium,calcium,and magnesium.In the Anzihe Nature Reserve,southwest China,seven species of ungulates and one species of primates were frequently observed visiting the mineral licks in high densities.As a consequence,mineral licks have attracted illegal hunters and facilitated the spread of parasites and diseases,leading to a depletion of local populations.Therefore,we suggest that regions with abundant mineral licks should be designated as key protected areas in the Anzihe Nature Reserve and relevant zones for animal health surveillance.Xingcheng He Zhixin Wen Yujin Wang Anderson Feijó Qiang Fu Jianghong Ran 2022Ecosystem Health and Sustainability2022,8,1:0
17Two-Step Preparation of Hierarchical Porous Carbon Materials Derived from Tannin for Use as an Electrode Material for Supercapacitors显示文摘The development and utilization of biomass and agroforestry processing byproducts for high-value applications have been an important topic in the field of renewable materials research.Based on this,a two-step microwave hydrothermal pre-carbonization and KOH activation method was proposed to synthesize tannin-based activated carbons with a high specific surface area,hierarchical pore structure,and good electrochemical performance.The microstructure,texture properties,and physicochemical characteristics were investigated.The results show that the prepared tannin-based activated carbons presented a hierarchical pore structure(micro-and mesopores)with a specific surface area as high as 997.46 m^(3) g^(−1).The electrochemical analysis shows that the tannin-based activated carbons have good wettability and charge transfer rates.Under the three-electrode system with 6 M KOH as the electrolyte,the active material TAC_(600-4) had a maximum specific capacitance of 171 F g^(−1)at 0.5 A g^(−1).As the current density increases to 10 A g^(−1),the specific capacitance can still be maintained at 149 F g^(−1),indicating a good rate capability.Therefore,the specific surface area and pore size of tannin-based activated carbons can be effectively adjusted by the alkali/carbon ratio,making it a promising supercapacitor electrode material and providing a new method for the high-value development of tannins in the field of electrochemical energy storage.Jianping Deng Qianqian Zhang Yuling Lan Lingcong Luo Zimin Dai Zhonghang Lin Zhixin Lu Jiancheng Yuan Yiqi Fu Lu Luo Weigang Zhao 2023Journal of Renewable Materials2023,11,6:0

19A simple and visualized method to screen for effective siRNAs by using green fluorescence protein (GFP) as a reporter显示文摘To screen for effective small interference RNA(siRNA),a simple and visualized method was developed using the green fluorescence protein(GFP)as a reporter.Candidate siRNAs targeting macrophage migration inhibition factor genes(MIF)were identified.By using the pEGFP-N3 vector,the MIF-GFP expression plasmid,pEGFP-MIF,was constructed with the same Kozak consensus translation initiation site and start code ATG for the MIF-EGFP coding sequence.Based on the siRNA expression vector pSilencer-4.1,3 candidate MIF siRNA expression plasmids were constructed and co-transfected with the pEGFP-MIF into the HEK293 cells,respectively.The GFP expression in HEK293 cells could be viewed by fluorescence microscopy and the MIF mRNA expressions were determined by real-time quantitative PCR.The 3 candidate MIF siRNA expression plasmids were also co-transfected with the MIF expression plasmid into the HEK293 cells,respectively,and the MIF mRNA expressions were determined by real-time quantitative PCR.The results show that the down-regulated expression of the MIF mRNA was consistent with the GFP expression and the same effective MIF siRNAs were screened by using the pEGFP-MIF or MIF expression plasmid with the candidate MIF siRNAs expression plasmids.Therefore,by using the GFP as a reporter,a useful method was provided to screen for effective siRNAs targeting specific genes co-expressed with the GFP.This may be a good strategy for screening for effective siRNAs targeting different genes.Zhixin SHAN Qiuxiong LIN Yongheng FU Chunyu DENG Xiaohong LI Xiyong YU 2008Frontiers in Biology2008,3,4:0
20Isolation and Identification of Pathogenic Bacteria Causing Bovine Subclinical Mastitis in Eastern Hebei Province显示文摘In this study, the milk samples of 1 021 cows in eight dairy farms in Eastern Hebei Province were collected and detected with LMT reagent and somatic cell count for subclinical mastitis. Pathogenic bacteria in subclinical mastitis positive milk samples were isolated and identified.The results showed that 60.63%(619/1 021) of the sampled cows were diagnosed with subclinical mastitis, and mixed infections accounted for 88.21%(546/619) of the cases. In addition, 82 strains of 14 species were isolated from the subclinical mastitis positive milk samples, including 36 strains of Staphylococcus(43.90%), 33 strains of Streptococcus(40.24%), 8 strains of Enterobacteriaceae(9.76%) and 5 strains of Corynebacterium(6.10%), respectively.
