| 1 | Level of circulating PD-L1 expression in patients with advanced gastric cancer and its clinical implications显示文摘Objective:The programmed cell death-1 receptor/programmed cell death-1 ligand(PD-1/PD-L1) pathway plays a crucial role in tumor evasion from host immunity.This study was designed to evaluate the association between circulating PD-L1 expression and prognosis in patients with advanced gastric cancer.Methods:Totally 80 advanced gastric cancer patients and 40 health controls from Beijing Cancer Hospital were enrolled in the present study.Circulating PD-L1 expression was tested by enzymelinked immunosorbent assay(ELISA).The associations between the expression level of PD-L1 and clinicopathological features and prognosis were analyzed statistically.Results:Expression of PD-L1 in advanced gastric cancer patients was significantly up-regulated compared with health people(P=0.006).The expression of PD-L1 was significantly correlated with differentiation and lymph node metastasis(P=0.026 and P=0.041,respectively).Although we didn't find significant difference in all advanced gastric cancer patients with different PD-L1 expression,the adenocarcinoma patients with higher up-regulated PD-L1 expression had much better prognosis than low expression patients(65.6% vs.44.7%,P=0.028).Conclusions:PD-L1 was elevated in advance gastric cancer patients and may play an important role in tumor immune evasion and patients prognosis. | Zhixue Zheng Zhaode Bu Xijuan Liu Lianhai Zhang Ziyu Li Aiwen Wu Xiaojiang Wu Xiaojing Cheng Xiaofang Xing Hong Du Xiaohong Wang Ying Hu Jiafu Ji | 2014 | Chinese Journal of Cancer Research2014,26,1: | 37 |

| 3 | LGR5 is a promising biomarker for patients with stage Ⅰ and Ⅱ gastric cancer显示文摘Objective: To investigate Leucine-rich repeat-containing G protein-coupled receptor 5 (LGR5) expressions in gastric cancer and to evaluate its clinical significance. Methods: LGR5 expression was assessed by immunohistochemistry in 257 gastric cancer patients after surgery. The relationships between LGR5 expression and clinicopathological features and patients prognosis were statistically analyzed. Results: The expression of LGR5 was significantly higher in gastric cancers as a cancer stem cell marker than in adjacent normal tissues (P<0.001), and more frequently in patients with intestinal type, well-moderate differentiation and stage Ⅰ and Ⅱ (P<0.05). Although we found gastric cancer patients with LGR5 positive expression had a poorer prognosis, it didn't meet statistical significance (P>0.05). LGR5 negative expression was significantly related to the favorable overall survival in stage Ⅰ and Ⅱ gastric cancer patients (P<0.05). Furthermore, patients with high LGR5 expression tended to be more likely to get progression and have poorer progress-free survival (P<0.05). Multivariate Cox regression analysis revealed that LGR5 expression was an independent factor of overall survival for the patients with stage Ⅰ and Ⅱ gastric cancer (P<0.05). | 4 | Downstaging and resection of hepatocellular carcinoma in patients with extrahepatic metastases after stereotactic therapy显示文摘Background:A combination of tyrosine kinase inhibitors(TKIs)and anti-PD-1 antibodies with local regional therapy has elicited yield substantial clinical benefits in patients who have hepatocellular carcinoma(HCC)with extrahepatic metastases.Using this treatment strategy to convert HCC patients with extrahepatic metastases from unresectable to resectable has not yet been reported.Methods:Consecutive hepatocellular carcinoma patients with extrahepatic metastases who received first-line therapy with a combination of TKIs and anti-PD-1 antibodies and at least one local regional therapy were analysed.Results:Nine patients with localized disease who received first-line systemic therapy were enrolled.At baseline,all of them had oligometastatic disease,namely,Barcelona Clinic Liver Cancer stage C(or Chinese Liver Cancer stage IIIB).The most common treatment administered was lenvatinib plus anti-PD-1 antibody and transarterial chemoembolization,and the median time span from systemic therapy to surgery was 3.2(IQR,2.8-6.2)months.Three patients achieved a pathological complete response.Six patients underwent laparoscopic surgery,and the other 3 patients underwent open surgery.After a median follow-up of 10.2(IQR,8.6-20.0)months,7 patients survived without disease recurrence,and 2 experienced tumour recurrence.All patients had any-grade AEs,and 55.6%of the patients experienced grade 3 AEs.Fatigue was the most common AE,followed by elevated aminotransferase levels and hypertension.Conclusions:Stereotactic therapy is a feasible conversion therapy for HCC patients with extrahepatic metastases to become resectable.This is the first study to analyse therapeutic outcomes of patients receiving these therapies for HCC with extrahepatic metastases. | Xiaobo Yang Haifeng Xu Bangyou Zuo Xu Yang Jin Bian Junyu Long Dongxu Wang Junwei Zhang Cong Ning Yanyu Wang Ziyu Xun Yunchao Wang Xin Lu Yilei Mao Xinting Sang Haitao Zhao | 2021 | Hepatobiliary Surgery and Nutrition2021,10,4: | 17 |
| 5 | Depth of tumor invasion and tumor-occupied portions of stomach are predictive factors of intra-abdominal metastasis显示文摘Objective:Diagnostic laparoscopy is recommended for the pretherapeutic staging of gastric cancer to detect any unexpected or unconfirmed intra-abdominal metastasis.The aim of this study was to evaluate the role and indications of diagnostic laparoscopy in the detection of intra-abdominal metastasis.Methods:Standard diagnostic laparoscopy with peritoneal cytology examination was performed prospectively on patients who were clinically diagnosed with primary local advanced gastric cancer(c T≥2M0).We calculated the rate of intra-abdominal metastases identified by diagnostic laparoscopy,and examined the relationship between peritoneal dissemination(P)and cytology results(CY).Split-sample method was applied to find clinical risk factors for intra-abdominal metastasis.Multivariate logistic regression analysis and receiver-operator characteristic(ROC)analysis were performed in training set to find out risk factors of intra-abdominal metastasis,and then validate it in testing set.Results:Out of 249 c M0 patients,51(20.5%)patients with intra-abdominal metastasis were identified by diagnostic laparoscopy,including 20(8.0%)P1CY1,17(6.8%)P0CY1 and 14(5.6%)P1CY0 patients.In the training set,multivariate logistic regression analysis and ROC analysis showed that the depth of tumor invasion on computer tomography(CT)scan≥21 mm and tumor-occupied≥2 portions of stomach are predictive factors of metastasis.In the testing set,when diagnostic laparoscopy was performed on patients who had one or two of these risk factors,the sensitivity and positive predictive value for detecting intra-abdominal metastasis were 90.0%and32.1%,respectively.Conclusions:According to our results,depth of tumor invasion and tumor-occupied portions of stomach are predictive factors of intra-abdominal metastasis. | Ziyu Li Zhemin Li Shuqin Jia Zhaode Bu Lianhai Zhang Xiaojiang Wu Shuangxi Li Fei Shan Xin Ji Jiafu Ji | 2017 | Chinese Journal of Cancer Research2017,29,2: | 14 |
| 6 | Inhibitory effect of blue honeysuckle extract on high-fat-diet-induced fatty liver in mice显示文摘Blue honeysuckle is rich in polyphenols, and recently receiving attention because of its potential antioxidant and anti-inflammatory properties. Nonalcoholic fatty liver disease(NAFLD) is the leading cause of chronic liver disease that develops hepatic inflammation and metabolic syndrome. The present study aims to study the effect of blue honeysuckle extract(BHE) on fat deposition and hepatic lipid peroxidation in a high-fat-diet(HFD)-induced mouse model. Mice were fed a normal diet(ND) or a HFD containing 0.5% or 1% of BHE or not for 45 d. Liver sections were stained by hematoxylin-eosin staining.Serum lipids were measured by a clinical analyzer. Insulin was examined by ELISA, and hepatic proteins were detected by Western blotting. Dietary supplementation of BHE dose-dependently suppressed HFD-induced obesity and hepatic fat deposition. | 7 | Construction and external validation of a nomogram that predicts lymph node metastasis in early gastric cancer patients using preoperative parameters显示文摘Objective: To create a nomogram to predict the incidence of lymph node metastasis(LNM) in early gastric cancer(EGC) patients and to externally validate the nomogram.Methods: To construct the nomogram,we retrospectively analyzed a primary cohort of 272 EGC patients.Univariate analysis and a binary logistic regression were performed.A nomogram predicting the incidence of LNM in EGC patients was created.The discrimination ability of the nomogram was measured using the concordance index(c-index),and the nomogram was also calibrated.Then,another prospective cohort of 81 patients was analyzed to validate the nomogram.Results: In the primary cohort,LNM was pathologically confirmed in 37(13.6%) patients.In multivariate analysis,the presence of an ulcer,the maximum lesion diameter observed via gastroscopy,the thickness of the lesion observed via endoscopic ultrasonography,and the presence of enlarged lymph nodes on computed tomography(CT) were independent risk factors for LNM.A nomogram was then created based on the regression model with the c-index of 0.905,and the calibration curve of the nomogram fell approximately on the ideal 45-degree line.The cut-off score of the nomogram was 110,and the sensitivity,specificity,positive predictive and negative predictive values of the nomogram in the primary cohort were 81.1%,86.0%,47.6% and 96.7%,respectively,and in the prospective validation cohort were 75.0%,91.0%,60.0% and 95.5%,respectively.The calibration curve of the external validation cohort was almost on the 45-degree line.Conclusions: We developed an effective nomogram predicting the incidence of LNM for EGC patients. | Yinan Zhang Yiqiang Liu Ji Zhang Xiaojiang Wu Xin Ji Tao Fu Ziyu Li Qi Wu Zhaode Bu Jiafu Ji | 2018 | Chinese Journal of Cancer Research2018,30,6: | 12 |
| 8 | Dietary protocatechuic acid ameliorates inflammation and up-regulates intestinal tight junction proteins by modulating gut microbiota in LPS-challenged piglets显示文摘Background: Weaning is one of the major factors that cause stress and intestinal disease in piglets. Protocatechuic acid(PCA) is an active plant phenolic acid which exists in Chinese herb, Duzhong(Eucommia ulmoides Oliver), and is also considered as the main bioactive metabolite of polyphenol against oxidative stress and inflammation. This study aimed to investigate the effect of PCA on growth performance, intestinal barrier function, and gut microbiota in a weaned piglet model challenged with lipopolysaccharide(LPS).Methods: Thirty-six piglets(Pig Improvement Company line 337 × C48, 28 d of age, 8.87 kg ± 0.11 kg BW) were randomly allocated into 3 treatments and fed with a basal diet(CTL), a diet added 50 mg/kg of aureomycin(AUR), or a diet supplemented with 4000 mg/kg of PCA, respectively. The piglets were challenged with LPS(10 μg/kg BW) on d 14 and d 21 by intraperitoneal injection during the 21-d experiment. Animals(n = 6 from each group) were sacrificed after being anesthetized by sodium pentobarbital at 2 h after the last injection of LPS. The serum was collected for antioxidant indices and inflammatory cytokines analysis, the ileum was harvested for detecting mRNA and protein levels of tight junction proteins by PCR and immunohistochemical staining, and the cecum chyme was collected for intestinal flora analysis using 16 S rRNA gene sequencing.Results: Dietary supplementation of PCA or AUR significantly increased the expression of tight junction proteins including ZO-1 and claudin-1 in intestinal mucosa, and decreased the serum levels of thiobarbituric acid reactive substances(TBARS) and IL-6, as compared with CTL group. In addition, PCA also decreased the serum levels of IL-2 and TNF-α(P < 0.05). Analysis of gut microbiota indicated that PCA increased the Firmicutes/Bacteroidetes ratio(P < 0.05). | 9 | Prognostic role of lymph node metastasis in early gastric cancer显示文摘Objective: To clarify the relationship between clinicopathological features and lymph node metastasis and to propose the potential indications of lymph node metastasis for prognosis in early gastric cancer(EGC) patients. Methods: We retrospectively observed 226 EGC patients with lymph node resection, and analyzed the associations between lymph node metastasis and clinicopathological parameters using the chi-square test in univariate analysis and logistic regression analysis in multivariate analysis. Overall survival analysis was determined using the Kaplan-Meier and log-rank test. We conducted multivariate prognosis analysis using the Cox proportional hazards model.Results: Of all the EGC patients, 7.5%(17/226) were histologically shown to have lymph node metastasis. The differentiation, lymphovascular invasion and depth of invasion were independent risk factors for lymph node metastasis in EGC. The 5- and 10-year survival rates were significantly lower in patients with lymph node metastasis than in those without and the patients also had shorter progress-free survival time. Lymph node metastasis and tumor size were independent prognostic factors for EGC. The status of the lymph nodes was a significant factor in predicting recurrence or metastasis after surgery. | 10 | Oxaliplatin plus S-1 or capecitabine as neoadjuvant or adjuvant chemotherapy for locally advanced gastric cancer with D2lymphadenectomy: 5-year follow-up results of a phase II-III randomized trial显示文摘Objective: To compare the effect of neoadjuvant chemotherapy(NACT) with adjuvant chemotherapy(ACT)using oxaliplatin plus S-1(SOX) or capecitabine(CapeOX) on gastric cancer patients with D2 lymphadenectomy.Methods: This was a two-by-two factorial randomized phase II-III trial, and registered on ISRCTN registry(No. ISRCTN12206108). Locally advanced gastric cancer patients were randomized to neoadjuvant SOX,neoadjuvant CapeOX, adjuvant SOX, or adjuvant CapeOX arms. Primary analysis was performed on an intentionto-treat(ITT) basis using overall survival(OS) as primary endpoint.Results: This trial started in September 2011 and closed in December 2012 with 100 patients enrolled.Treatment completion rate was 56%, 52%, 38% and 30% in the four arms, respectively. NACT group had fewer dropouts due to unacceptable toxicity(P=0.042). Surgical complication rate did not differ by the four groups(P=0.986). No survival significant difference was found comparing NACT with ACT(P=0.664; 5-year-OS: 70% vs.74% respectively), nor between the SOX and CapeOX groups(P=0.252; 5-year-OS: 78% vs. 66% respectively).Subgroup analysis showed SOX significantly improved survival in patients with diffuse type(P=0.048).Conclusions: No significant survival difference was found between NACT and ACT. SOX and CapeOX had good safety and efficacy as neoadjuvant regimens. | 11 | Soil organic carbon and nitrogen content of density fractions and effect of meadow degradation to soil carbon and nitrogen of fractions in alpine Kobresia meadow显示文摘This research was conducted on the non-disturbed native alpine Kobresia meadow(YF) and the severely degraded meadow(SDL) of Dari County of Qinghai Province.By a density fractionation approach,each soil sample was divided into two fractions:light fraction(LF) and heavy fraction(HF).The obtained fractions were analyzed for organic carbon(OC) and nitrogen(N) concentrations.The results showed:(1) the OC concentration in HF and LF was 3.84% and 28.63% respectively while the nitrogen concentration in HF and LF was 0.362% and 1.192% respectively in 0-10 cm depth.C:N ratio was 10.6 in HF and 23.8 in LF respectively.(2) As far as the ratio of OC in given fraction to that in gross sample was concerned,dominance of OC in HF was obvious in the whole soil profile.OC in HF increased from 78.95% to 90.33%,while OC in LF decreased from 21.05% to 9.68% with depths.(3) Soil total OC amounted to 47.47 in YF while 17.63 g.kg-1 in SDL,in which the OC content in HF decreased from 37.31 to 16.01 g.kg-1 while OC content in LF decreased from 10.01 to 1.62 g.kg-1.In other words,results of OC and N content show meadow degradation led to the loss of 57% OC in HF and 84% OC in LF from originally native ecosystem on alpine meadow.In addition,meadow degradation led to the loss of 43% N in HF and 79% N in LF from originally native ecosystem on alpine meadow.(4) The main reason for loss of C and N in LF during meadow degradation was not attributed to the decrease of OC and N concentration in LF and LF,but to the decrease in LF dry weight.Loss of N was far lower than loss of C in HF.This may suggest that there is difference in protection mode of C and N in HF. | WANG WenYing WANG QiJi LU ZiYu | 2009 | Science China Earth Sciences2009,52,5: | 10 |

| 13 | Diode-pumped power scalable Kerr-lens mode-locked Yb:CYA laser显示文摘Stable 68 fs pulses with the average power of 1.5 W is directly generated from a multimode diode-pumped Kerr-lens mode-locked Yb:CYA laser by separating the gain medium and Kerr medium. The repetition rate is about 50 MHz, resulting in a single pulse energy of 30 n J and a peak power of 0.44 MW. To the best of our knowledge, this is the highest single pulse energy ever produced from a mode-locked Yb:CYA oscillator. Our experimental results show that Yb:CYA crystal is an excellent candidate for multiwatt, sub-100 fs pulse generation in diode-pumped all-solid-state lasers. | 16 | Highly stable lead-free Cs3Bi2I9 perovskite nanoplates for photodetection applications显示文摘Lead halide perovskites have received tremendous attentions recently for their excellent properties such as high light absorption coefficient and long charge carrier diffusion length. However, the stability issues and the existence of toxic lead cations have largely limited their applications in optoelectronic area. Herein, we report the synthesis and investigation of highly stable and lead-free Cs3Bi2I9 perovskite nanoplates for visible light photodetection applications. The Cs3Bi2I9 nanoplates were synthesized through a facile solution-processed method, which is also applicable to various substrates. The achieved nanoplates present very good crystal quality and exhibit excellent long-term stability even exposed in moist air for several months. Photodetectors were constructed based on these high-quality perovskite nanoplates for the first time, and display a maximum photoresponsivity of 33.1 mA/W under the illumination of 450 nm laser, which is six times higher than the solution-synthesized CH3NH3PbI3 nanowire photodetectors. The specific detectivity of these devices can reach up to 10^10 Jones. Additionally, the devices exhibit fast rise and decay time of 10.2 and 37.2 ms, respectively, and highly stable photoswitching behavior with their photoresponse well retaining under alternating light and darkness. The triaxial yarns in micromechanical representative unit cell(RUC) were idealized as curved beams with a path depicted using the sinusoidal shape functions, and the tensile and shear moduli of TWF composites were derived by means of the strain energy approach founded on micromechanics. In order to validate the new models, the predictions were compared with the experimental data from literature. It was shown that the predictions from the new model agree well with the experimental results.
