2The calcium-dependent kinase OsCPK24 functions in cold stress responses in rice显示文摘Calcium-dependent protein kinases(CPKs)are serine/threonine protein kinases that function in plant stress responses. Although CPKs are recognized as key messengers in signal transduction, the specific roles of CPKs and the molecular mechanisms underlying their activity remain largely unknown. Here, we characterized the function of Os CPK_(24), a cytosol-localized calciumdependent protein kinase in rice. Os CPK_(24) was universally and highly expressed in rice plants and was induced by cold treatment. Whereas Os CPK_(24) knockdown plants exhibited increased sensitivity to cold compared to wild type(WT), Os CPK_(24)-overexpressing plants exhibited increased cold tolerance. Plants overexpressing Os CPK_(24) exhibited increased accumulation of proline(an osmoprotectant) and glutathione(an antioxidant) and maintained a higher GSH/GSSG(reduced glutathione to oxidized glutathione) ratio during cold stress compared to WT. In addition to these effects in response to cold stress, we observed the kinase activity of Os CPK_(24) varied under different calcium concentrations. Further,Os CPK_(24) phosphorylated Os Grx_(10), a glutathionedependent thioltransferase, at rates modulated by changes in calcium concentration. 4Species composition and diversity of Euphausiacea in the East China Sea显示文摘Based on the data of four seasonal oceanographic censuses in the East China Sea (23°30′~33°00′N, 118°30′~128°00′E) in 1997~2000, the species composition and the diversity of Euphausiacea were discussed as well as their relations with environmental variables. Results showed that there were totally 23 different species of Euphausiacea, in which 16 occurred in spring and autumn respectively; 15 were present in summer and only 10 were observed in winter. According to the calculated alternation fraction (R), the species composition showed a clear seasonal alternation with the changes ofseasons. Moreover, the environmental variables had different impacts on the distribution of Euphausiacea in different seasons. The distribution in summer was not significantly related to water temperature and salinity. However, the surface salinity was a major determinant of the distribution in spring. In autumn, both surface and bottom temperatures were influencing factors. The distribution in winter depended on salinity at the surface and 10 m depth as well as the temperature at 10 m depth. Regarding to the seasonal variation of species composition, the variations in spring, summer and autumn were not so significant as those in winter. Except in summer, the species number changed with synchronous water temperature and salinity, as a result of the presence of warm currents in the East China Sea and the habitability of the dominant species. 6Evolution of an Eocene–Oligocene Saline Lake Depositional System and Its Controlling Factors, Jianghan Basin, China显示文摘The Upper Eocene–Lower Oligocene Qianjiang Formation of the Jianghan Basin in central China consists of a 4 700-m-thick lacustrine succession, containing 1 800 m of halite deposits. The maximum thickness of the formation is 4 700 m, and includes 1 800 m of halite. We have identified eight third-order depositional sequences based on pinch-out and onlap stratigraphic patterns in 2-D and 3-D seismic data and well logs. The basin evolved from a deep to shallow under-filled lake during the Eocene–Oligocene interval. The main rock types are dark mudstones, halite, and siltstone/sandstone in the depocenter, and alternating mudstone and gypsum in shallower areas. The vertical succession indicates a strong sedimentary cyclicity. Depositional facies indicate the presence of two lake system types. Halite developed in a saline lake system, whereas clastic sediments were deposited in freshwater lake systems. The alternating sediment types indicate that the basin cycled repeatedly between saline and freshwater lake systems. This cyclicity was caused by availability of accommodation space that was controlled by a combination of climate change, tectonic subsidence and sediment supply; notably, the highest frequency cycles occurred at Milankovitch timescales controlled by the Earth's orbital variations. 7Orbital Forcing and Sea-Level Changes in the Earliest Triassic of the Meishan Section,South China显示文摘The Earliest Triassic Yinkeng Formation is exposed at the well-known Meishan Section,South China,which contains the Global Stratotype of Section and Point(GSSP)for the PermianTriassic boundary(PTB).It records centimeter-scale rhythmic alternations comprised mainly by marlstone and limestone.Seven types of couplet embedded in five types of bundles were recognized based on occurrence and thickness of the lithologic units,suggesting that their formation was controlled by cyclic processes.The various orders of cycles observed correlate well with other Early Triassic counterparts recorded in South China.Here,we present new cyclostratigraphic results based on lithologic thickness and relative carbonate content of the Yinkeng Formation.Power spectra of carbonate content show that the ratio of major wavelengths recognized throughout the formation is similar to that of the 100kyr short eccentricity,33 kyr obliquity,and 21 kyr precession cycles,indicating that astronomical signals are recorded in the Earliest Triassic rhythmic succession.Consistence between pronounced lithologic rhythmicity and sea-level changes obtained from Fischer plots indicates that high-frequency climatic cycles may have driven sea-level changes immediately after the PTB mass extinction.Furthermore,the 4th-order sea-level changes interpreted from the sedimentary record match well with 100kyr short eccentricity component of carbonate content,reflecting that the 100 kyr short eccentricityinduced climate changes may have likely controlled the deposition of 4th-order sequences recognized from rhythmic successions.Shifeng Tian Zhong-Qiang Chen Chunju Huang 2014Journal of Earth Science2014,25,1:6

10Study on abundance variation of pteropods in the East China Sea显示文摘On the basis of the census in four seasons from 1997 to 2000 in the East China Sea (23°30′ ̄33°N, 118°30′ ̄128°E), the horizontal distribution of pteropods, the seasonal variations of their abundance and dynamic mechanisms of these variations were approached with quantitative and qualitative methods. Results showed that the total abundance of pteropods varied clearly with the seasons in an order of decrease of autumn (108×10-2 ind./m3), summer (33×10-2 ind./m3), winter(28×10-2 ind./m3) and spring (10×10-2 ind./m3). Pteropods were not observed in the north nearshore of the East China Sea (29°30′ ̄33°N, 122°30′ ̄125°E)in winter and spring. In all seasons, the peteropod abundance was higher in the south than that in the north and higher in the offshore than that in the nearshore. The major species influencing the peteropod abundance were Creseis acicula in spring, Creseis virgula in summer, Desmopterus papilio in autumn and Limacina trochiformis in winter. The variation of pteropod abundance showed a prominent linear correlation with the surface water temperature and 10 m water temperature, but no significant correlation with both the bottom water temperature and salinity. In summer, the high-abundance distribution area of pteropods was located in the place where the Taiwan Warm Current branchedfromtheKuroshio. Inautumn, thedistributionareawithanabundancehigherthan250×10-2ind./m3expanded northward along with the Taiwan Warm Current within the mixing area where the Changjiang freshwater, the Fujian—Zhejiang coastal water and the Huanghai Sea Cold Water met together. 13Identification of late-stage pollen-specific promoters for construction of pollen-inactivation system in rice显示文摘Large-scale production of male sterile seeds can be achieved by introducing a fertility-restoration gene linked with a pollen-killer gene into a recessive male sterile mutant.We attempted to construct this system in rice by using a late-stage pollen-specific(LSP)promoter driving the expression of maizeα-amylase gene ZM-AA1.To obtain such promoters in rice,we conducted comparative RNA-seq analysis of mature pollen with meiosis anther,and compared this with the transcriptomic data of various tissues in the Rice Expression Database,resulting in 269 candidate LSP genes.Initial test of nine LSP genes showed that only the most active OsLSP3 promoter could drive ZM-AA1 to disrupt pollen.We then analyzed an additional 22 LSP genes and found 12 genes stronger than OsLSP3 in late-stage anthers.The promoters of OsLSP5 and OsLSP6 showing higher expression than OsLSP3 at stages 11 and 12 could drive ZM-AA1 to inactivate pollen,while the promoter of OsLSP4 showing higher expression at stage 12 only could not drive ZM-AA1 to disrupt pollen,suggesting that strong promoter activity at stage 11 was critical for pollen inactivation.The strong pollen-specific promoters identified in this study provided valuable tools for genetic engineering of rice male sterile system for hybrid rice production.Menglong Wang Wei Yan Xiaoqun Peng Zhufeng Chen Chunjue Xu Jianxin Wu Xing Wang Deng Xiaoyan Tang 2020Journal of Integrative Plant Biology2020,62,8:4
14GDSL esterase/lipases OsGELP34 and OsGELP110/OsGELP115 are essential for rice pollen development显示文摘Pollen exine contains complex biopolymers of aliphatic lipids and phenolics.Abnormal development of pollen exine often leads to plant sterility.Molecular mechanisms regulating exine formation have been studied extensively but remain ambiguous.Here we report the analyses of three GDSL esterase/lipase protein genes,OsGELP34,OsGELP110,and OsGELP115,for rice exine for-mation.OsGELP34 was identified by cloning of a male sterile mutant gene.OsGELP34 encodes an endoplasmic reticulum protein and was mainly expressed in anthers during pollen exine formation.osgelp34 mutant displayed abnormal exine and altered expression of a number of key genes required for pollen development.OsGELP110 was previously identified as a gene differentially expressed in meiotic anthers.OsGELP110 was most homologous to OsGELP115,and the two genes showed similar gene expression patterns.Both OsGELP110 and OsGELP115 proteins were localized in peroxisomes.Individual knockout of OsGELP110 and OsGELP115 did not affect the plant fertility,but double knockout of both genes altered the exine structure and rendered the plant male sterile.OsGELP34 is distant from OsGELP110 and OsGELP115 in sequence,and osgelp34 and osgelp110/osgelp115 mutants were different in anther morphology despite both were male sterile.These results suggested that OsGELP34 and OsGELP110/OsGELP115 catalyze different compounds for pollen exine development.Huihui Zhang Menglong Wang Yiqi Li Wei Yan Zhenyi Chang Haoling Ni Zhufeng Chen Jianxin Wu Chunjue Xu Xing Wang Deng Xiaoyan Tang 2020Journal of Integrative Plant Biology2020,62,10:4
15Astronomical time scale for the lower Doushantuo Formation of early Ediacaran,South China显示文摘Nearly 90% of the Ediacaran Period(635–541 Ma) of the Neoproterozoic is represented by the Doushantuo Formation(DST Fm) in South China.Its lowest Member I is a 3.7 m-thick cap carbonate deposited at the termination of the Cryogenian Marinoan glaciation.The DST Fm consists of alternating organic-rich black shale and thinly bedded dolostone, and it contains some of the oldest records of multicellular life and three pronounced negative carbon isotope excursions.The Jiulongwan(JLW) section is a well-studied reference section for these Ediacaran events.Spectral analysis of geochemical data through the lower DST Fm(22.3 m) shows 27 predominant ~90 cm sedimentary cycles that correspond to 405-ka long eccentricity cycles.The power spectra of the 405-ka tuned Ca and Fe/Ti series show significant peaks at ~1.2-Ma, 405-ka, 133-ka, 128-ka, 100-ka, 82-ka, ~31-ka and 29-ka periods, respectively.A 11.16 Malong astronomical time scale has been constructed for the lower DST Fm and provide a duration of 1.6 Ma for the cap carbonate(Member Ⅰ) based on the 405-ka long eccentricity cycle tuning.Using the U-Pb age of 635.2 ± 0.6 Ma for the volcanic ash bed at the Member Ⅰ/Ⅱboundary, we proposed a 636.8 Ma age for the base of the DST Fm.These ages and astronomical timescale provide important new constraints on the subdivision of Ediacaran strata, and have implications for understanding the character of the first negative δ^(13)C excursion(EN1).Orbital forcing may have been played an important role for the climate changes and the evolution of Ediacaran multi-cellular life and the carbon cycle variations.Yu Sui Chunju Huang Rui Zhang Zhixiang Wang James Ogg David B.Kemp 2018Science Bulletin2018,63,22:4
16Climate change response to astronomical forcing during the Oligocene-Miocene transition in the equatorial Atlantic(ODP Site 926)显示文摘The Oligocene-Miocene transition period was characterized by a decrease in global CO2 levels, expansion of polar ice sheet, fall in global sea-level, etc. However, the reasons for, and mechanisms of, this global, extreme-cold climate change event(Mi-1) still remain controversial. Our samples from the core of the Ocean Drilling Program(ODP) Leg 154, Site 926, located in the equatorial Atlantic, mainly consist of light-gray, nannofossil chalk with foraminifers interbedded with greenish-gray, clayey, nannofossil chalk sediments. Color variation from light-gray layers(up to 80% carbonate content) to dark layers(60% carbonate content) was observed to occur cyclically at the meter scale. Therefore, we chose color reflectance lightness(L*) data as the paleoclimate proxy on which to perform cyclostratigraphic analysis because it could reflect carbonate content changes. Based on the recognition of the 405 kyr long eccentricity and 40 kyr obliquity cycles of the L* series, we tuned the series to establish an absolute astronomical time scale using the published age of the Oligocene-Miocene boundary(OMB) as the anchor for an absolute age control point. The power spectra of the tuned L* series showed that the long eccentricity signals became significantly weak, while the obliquity signals became strong, from the Late Oligocene to the Early Miocene. The 405 kyr long eccentricity minimum coincided with the 1.2 Myr obliquity node at the OMB, and similar convergences might be closely related to other extreme-cold events in Earth’s history. In addition, the sedimentation accumulation rate, oxygen isotopes of benthonic foraminifers, and rodents’ per-taxon turnover rate from Central Spain showed the same 2 Myr cyclicity, which indicates the significant influence of Earth-orbital forcing on the Earth system and ecological evolution on the million-year time scale.ZOU ZhuoYan HUANG ChunJu LI MingSong ZHANG Yang 2016Science China Earth Sciences2016,59,8:3
17Oxidized mitochondrial DNA sensing by STING signaling promotes the antitumor effect of an irradiated immunogenic cancer cell vaccine显示文摘Exposure to ionizing radiation,a physical treatment that inactivates live tumor cells,has been extensively applied to enhance the antitumor responses induced by cancer cell vaccines in both animal research and human clinical trials.However,the mechanisms by which irradiated cells function as immunogenic tumor vaccines and induce effective antitumor responses have not been fully explored.Here,we demonstrate that oxidized mitochondrial DNA(mtDNA)and stimulator of interferon genes(STING)signaling play a key roles in the enhanced antitumor effect achieved with an irradiated tumor cell vaccine.Elevations in ROS and oxidized mtDNA 8-OHG content could be induced in irradiated tumor cells.Oxidized mtDNA derived from irradiated tumor cells gained access to the cytosol of dendritic cells(DCs).Oxidized mtDNA,as a DAMP or adjuvant,activated the STING-TBK1-IRF3-IFN-β pathway in DCs,which subsequently cross-presented irradiated tumor cell-derived antigens to CD8^(+)T cells and elicited antitumor immunity.The results of our study provide insight into the mechanism by which an irradiated cell vaccine mediates antitumor immunity,which may have implications for new strategies to improve the efficacy of irradiated vaccines.Chunju Fang Fei Mo Li Liu Jing Du Min Luo Ke Men Feifei Na Wei Wang Hanshuo Yang Xiawei Wei 2021Cellular & Molecular Immunology2021,18,9:2
