2Hepatocellular carcinoma in viral and autoimmune liver diseases:Role of CD4+CD25+Foxp3+regulatory T cells in the immune microenvironment显示文摘More than 90%of cases of hepatocellular carcinoma(HCC)occurs in patients with cirrhosis,of which hepatitis B virus and hepatitis C virus are the leading causes,while the tumor less frequently arises in autoimmune liver diseases.Advances in understanding tumor immunity have led to a major shift in the treatment of HCC,with the emergence of immunotherapy where therapeutic agents are used to target immune cells rather than cancer cells.Regulatory T cells(Tregs)are the most abundant suppressive cells in the tumor microenvironment and their presence has been correlated with tumor progression,invasiveness,as well as metastasis.Tregs are characterized by the expression of the transcription factor Foxp3 and various mechanisms ranging from cell-to-cell contact to secretion of inhibitory molecules have been implicated in their function.Notably,Tregs amply express checkpoint molecules such as cytotoxic T lymphocyte-associated antigen 4 and programmed cell-death 1 receptor and therefore represent a direct target of immune checkpoint inhibitor(ICI)immunotherapy.Taking into consideration the critical role of Tregs in maintenance of immune homeostasis as well as avoidance of autoimmunity,it is plausible that targeting of Tregs by ICI immunotherapy results in the development of immune-related adverse events(irAEs).Since the use of ICI becomes common in oncology,with an increasing number of new ICI currently under clinical trials for cancer treatment,the occurrence of irAEs is expected to dramatically rise.Herein,we review the current literature focusing on the role of Tregs in HCC evolution taking into account their opposite etiological function in viral and autoimmune chronic liver disease,and we discuss their involvement in irAEs due to the new immunotherapies.Alessandro Granito Luigi Muratori Claudine Lalanne Chiara Quarneti Silvia Ferri Marcello Guidi Marco Lenzi Paolo Muratori 2021World Journal of Gastroenterology2021,27,22:11

5αV integrin:A new gastrin target in human pancreatic cancer cells显示文摘AIM:To analyse αV integrin expression induced by gas-trin in pancreatic cancer models. METHODS:αV integrin mRNA expression in human pan-creatic cancer cells was analysed using a 'cancer genes' array and confirmed by real-time reverse transcription-polymerase chain reaction (PCR). Western blotting and semi-quantitative immunohistochemistry were used to examine protein levels in human pancreatic cancer cell lines and pancreatic tissues, respectively. The role of αV integrin on gastrin-induced cell adhesion was examined using blocking anti-αV integrin monoclonal antibodies. Adherent cells were quantified by staining with crystal violet.RESULTS: Using a 'cancer genes' array we identified αV integrin as a new gastrin target gene in human pancreatic cancer cells. A quantitative real-time PCR approach was used to confirm αV integrin gene expression. We also demonstrate that Src family kinases and the PI 3-kinase, two signalling pathways specifically activated by the CCK-2 receptor (CCK2R), are involved in gastrin-mediated αV integrin expression. In contrast, inhibition of the ERK pathway was without any effect on αV integrin expression induced by gastrin. Our results also show that gastrin modulates cell adhesion via α V integrins. Indeed, in vitro adhesion assays performed on fibronectin show that gastrin significantly increases adhesion of pancreatic cancer cells. The use of blocking anti-αV integrin monoclonal antibodies completely reversed the increase in cell-substrate adhesion induced by gastrin. In addition, we showed in vivo that the targeted CCK2R expression in the pancreas of Elas-CCK2 mice, leads to the overexpression of αV integrin. This process may contribute to pancreatic tumour development observed in these transgenic animals. 8抗生素对细枝木麻黄愈伤组织再生及农杆菌生长的影响显示文摘以细枝木麻黄3个无性系水培苗作为材料,研究了硫酸卡那霉素(kanamycin sulfate,Km)对细枝木麻黄愈伤组织诱导、增殖和芽分化的影响,以及头孢霉素(cefotaxime,Cef)、羧苄青霉素(carbenicillin,Cab)和氨苄青霉素(ampicillin,Amp)对农杆菌的抑制效果。结果表明,Km对细枝木麻黄愈伤组织诱导适宜的选择压力是20mg/L,Km选择性筛选愈伤组织增殖的适宜质量浓度为40mg/L,愈伤组织芽分化的选择压力是40~50mg/L。在抗生素抑菌效果方面,Cef质量浓度为200mg/L、Cab300mg/L和Amp500mg/L能完全抑制农杆菌的生长。综合比较,对细枝木麻黄遗传转化研究最理想的抑菌剂是Cef,使用质量浓度低且能有效抑制根癌农杆菌的生长。姜清彬 仲崇禄 曾炳山 张勇 Didier Bogusz Claudine Franche 2010东北林业大学学报2010,38,9:2

12Epigenetic mechanisms involved in developmental nutritional programming显示文摘The ways in which epigenetic modifications fix the effects of early environmental events,ensuring sustained responses to transient stimuli,which result in modified gene expression patterns and phenotypes later in life,is a topic of considerable interest.This review focuses on recently discovered mechanisms and calls into question prevailing views about the dynamics,position and functions of epigenetic marks.Most epigenetic studies have addressed the long-term effects on a small number of epigenetic marks,at the global or individual gene level,of environmental stressors in humans and animal models.In parallel,increasing numbers of studies based on high-throughput technologies and focusing on humans and mice have revealed additional complexity in epigenetic processes,by highlighting the importance of crosstalk between the different epigenetic marks.A number of studies focusing on the developmental origin of health and disease and metabolic programming have identified links between early nutrition,epigenetic processes and long-term illness.The existence of a self-propagating epigenetic cycle has been demonstrated.Moreover,recent studies demonstrate an obvious sexual dimorphism both for programming trajectories and in response to the same environmental insult.Despite recent progress,we are still far from understanding how,when and where environmental stressors disturb key epigenetic mechanisms.Thus,identifying the original key marks and their changes throughout development during an individual's lifetime or over several generations remains a challenging issue.Anne Gabory Linda Attig Claudine Junien 2011World Journal of Diabetes2011,2,10:2
