1Lotus seed skin proanthocyanidin extract exhibits potent antioxidant property via activation of the Nrf2-ARE pathway显示文摘Lotus seed is well known as traditional food and medicine,but its skin is usually discarded.Recent studies have shown that lotus seed skin contains a high concentration of proanthocyanidins that have multi-functions,such as antioxidation,anti-inflammation,and anti-cancer effects.In the present study,we aimed to isolate and purify the proanthocyanidins from lotus seed skin by acetone extraction and rotary evaporation,identify their chemical structures by HPLC-MS-MS and NMR,and further investigate the antioxidant properties of the extract purified by macroporous resin (PMR) from lotus seed skin both in vitro and in vivo.The results showed that PMR mainly contained oligomeric proanthocyanidins,especially dimeric procyanidin B1 (PB1),procyanidin B2 and procyanidin B4.Although it had limited ability to directly scavenge radicals in vitro,PMR could significantly enhance the expressions of antioxidant proteins via activation of nuclear factory-related factor 2 (Nrf2)-antioxidant response element (ARE) pathway in HepG2 cells.Molecular data revealed that PB1,a major component in PMR,stabilized Nrf2 by inhibiting the ubiquitination of Nrf2,which led to subsequent activation of the Nrf2-ARE pathway,including the enhancements of Nrf2 nuclear translocation,Nrf2-ARE binding and ARE transcriptional activity.Moreover,the in vivo results in high fat diet-induced mice further verified the powerful antioxidant property of PMR.These results revealed that lotus seed skin is a promising resource for functional food development.Tao Li Qili Li Weiguo Wu Yong Li De-xing Hou Hua Xu Baodong Zheng Shaoxiao Zeng Yang Shan Xiangyang Lu Fangming Deng Si Qin 2019Acta Biochimica et Biophysica Sinica2019,51,1:8
2Influence of calcium content on solid ratio of inclusions in Ca-treated liquid steel显示文摘It has been found that Ca-treated liquid steel can be cast at a lower superheat. To ascertain the reason for the improved castability of Ca-treated liquid steel and to find the optimal range of calcium content, the behavior of calcium in liquid steel was studied in terms of the relationship between the calcium content and solid ratio of inclusions. The relationship between the calcium content and solid ratio of inclusions was obtained by means of the classification of nonmetallic inclusions in solid and liquid steels at casting temperature according to the Al2O3-CaO-SiO2 phase diagram. 3Genetic analysis and molecular mapping of a novel gene for zebra mutation in rice (Oryza sativa L.)显示文摘A novel zebra mutant,zebra-15,derived from the restorer line Jinhui10(Oryza sativa L. ssp. indica) treated by EMS,displayed a distinctive zebra leaf from seedling stage to jointing stage. Its chlorophyll content decreased(55.4%) and the ratio of Chla/Chlb increased(90.2%) significantly in the yellow part of the zebra-15,compared with the wild type. Net photosynthetic rate and fluorescence kinetic parameters showed that the decrease of chlorophyll content significantly influenced the photosynthetic efficiency of the mutant. Genetic analysis of F2 segregation populations derived from the cross of Xinong1A and zebra-15 indicated that the zebra leaf trait is controlled by a single recessive nuclear gene. Ninety-eight out of four hundred and eighty pairs of SSR markers showed the diversity between the Xinong1A and the zebra-15,their F2 population was then used for gene mapping. Zebra-15(Z-15) gene was primarily restricted on the short arm of chromosome 5 by 150 F2 recessive individuals,19.6 cM from marker RM3322 and 6.0 cM from marker RM6082. 4Prevalence, risk factors, and survival associated with pulmonary hypertension and heart failure among patients with underlying coronary artery disease: a national prospective, multicenter registry study in China显示文摘Background: Coronary artery disease (CAD) is the commonest cause of heart failure (HF), whereas pulmonary hypertension (PH) has not been established or reported in this patient population. Therefore, we assessed the prevalence, risk factors, and survival in CAD-associated HF (CAD-HF) complicated with PH.Methods: Symptomatic CAD-HF patients were continuously enrolled in this prospective, multicenter registry study. Echocardiography, coronary arteriography, left and right heart catheterization (RHC), and other baseline clinical data were recorded. Patients were followed up and their survival was recorded.Results: One hundred and eighty-two CAD-HF patients were enrolled, including 142 with HF with a preserved ejection fraction (heart failure with preserved ejection fraction [HFpEF];left ventricular ejection fraction [LVEF] ≥50%) and 40 with a reduced ejection fraction (heart failure with reduced ejection fraction [HFrEF];LVEF < 50%). PH was diagnosed with RHC in 77.5% of patients. Patients with PH showed worse hemodynamic parameters and higher mortality. HFrEF-PH patients had worse survival than HFpEF-PH patients. CAD-HF patients with an enlarged left ventricular end-diastolic diameter and reduced hemoglobin were at higher risk of PH. Nitrate treatment reduced the risk of PH. Elevated creatinine and mean pulmonary arterial pressure (mPAP), diastolic pressure gradient (DPG) ≥7 mmHg, and previous myocardial infarction (MI) entailed a higher risk of mortality in CAD-HF patients with PH.Conclusions: PH is common in CAD-HF and worsens the hemodynamics and survival in these patients. Left ventricle enlargement and anemia increase the risk of PH in CAD-HF. Patients may benefit from nitrate medications. 5Global perspectives and future research directions for the phytoremediation of heavy metal-contaminated soil:A knowledge mapping analysis from 2001 to 2020显示文摘In total,9,552 documents were extracted from the Web of Science Core Collection and subjected to knowledge mapping and visualization analysis for the field of phytoremediation of HM-contaminated soil(PHMCS)with CiteSpace 5.7 R3 software.The results showed that(1)the number of publications increased linearly over the studied period.The top 10 countries/regions,institutions and authors contributing to this field were exhibited.(2)Keyword co-occurrence cluster analysis revealed a total of 8 clusters,including“Bioremediation,”“Arsenic,”“Biochar,”“Oxidative stress,”“Hyperaccumulation,”“EDTA,”“Arbuscular mycorrhizal fungi,”and“Environmental pollution”clusters(3)In total,334 keyword bursts were obtained,and the 25 strongest,longest duration,and newest keyboard bursts were analyzed in depth.The strongest keyword burst test showed that the hottest keywords could be divided into 7 groups,i.e.,“Plant bioremediation materials,”“HM types,”“Chelating amendments,”“Other improved strategies,”“Bioremediation characteristics,”“Risk assessment,”and“Other.”Almost half of the newest topics had emerged in the past 3 years,including“biochar,”“drought,”“health risk assessment,”“electrokinetic remediation,”“nanoparticle,”and“intercropping.”(4)In total,9 knowledge base clusters were obtained in this study.The studies of Ali et al.(2013),Blaylock et al.(1997),Huang et al.(1997),van der Ent et al.(2013),Salt et al.(1995),and Salt(1998),which had both high frequencies and the strongest burst scores,have had the most profound effects on PHMCS research.Finally,future research directions were proposed.Kehui Liu Xiaojin Guan Chunming Li Keyi Zhao Xiaohua Yang Rongxin Fu Yi Li Fangming Yu 2022Frontiers of Environmental Science & Engineering2022,16,6:4
6Morphological and physiological analysis of narrow and striped leaf 1(nsl1) mutant of rice(Oryza sativa L.) and the gene mapping显示文摘The shape and color of rice leaves are important agronomic traits that directly influence the proportion of sunlight energy utilization and ultimately affect the yield and quality.A new mutant exhibiting stable inheritance was identified as derived from ethyl methane sulfonate(EMS)-treated restorer Jinhui 10,tentatively named as narrow and striped leaf 1(nsl1).The nsl1 displayed pale white leaves at the seeding stage and then white striped leaves in parallel to the main vein at the jointing stage.Meanwhile,its leaf blades are significantly narrower than the control group of Jinhui 10.The chloroplast structures of cells in the white striped area of the nsl1 mutant break down,and the photosynthetic pigments are significantly lower than that of the wild type.Moreover,fluorescence parameters,such as F0,F v/F m,U psII,qp,and ETR,in the nsl1 mutant are significantly lower than those of the wild type,and the photosynthetic efficiency is also significantly decreased.These changes in leaf color and shape,together with physiological changes in the nsl1,result in smaller plant height and a decrease in the most important agronomic traits,such as the number of grains per panicle,grain weight,etc.Genetic analysis shows that the narrow and striped traits of the nsl1 mutant are controlled by a single recessive nuclear gene,which is located between InDel 16and InDel 12 in chromosome 3.The physical distance is204 kb.So far,no similar genes of such leaf color and shape in this area have been reported.This study has laid a solid foundation for the gene cloning and function analysis of NSL1.Fangming Zhao Yong Zhang Yanru Wu Tongming Wang Ling Ma Zhenglin Yang Xia Wei Xianchun Sang Yinghua Ling Nan Wang Changwei Zhang Guanghua He 2014Chinese Science Bulletin2014,59,9:3
7Deficient Rnf43 potentiates hyperactive Kras-mediated pancreatic preneoplasia initiation and malignant transformation显示文摘Background:Largely due to incidental detection,asymptomatic pancreatic cystic le-sions(PCLs)have become prevalent in recent years.Among them,intraductal papillary mucinous neoplasm(IPMN)infrequently advances to pancreatic ductal adenocarcinoma(PDAC).Conservative surveillance versus surgical intervention is a difficult clinical decision for both caregivers and PCL patients.Because RNF43 loss-of-function mutations and KRAS gain-of-function mutations concur in a subset of IPMN and PDAC,their biological significance and therapeutic potential should be elucidated.Methods:Pancreatic Rnf43 knockout and Kras activated mice(Rnf43^(−/−);Kras^(G12D))were generated to evaluate their clinical significance in pancreatic pre-neoplastic initiation and malignant transformation.Results:Loss of Rnf43 potentiated the occurrence and severity of IPMN and PDAC in oncogenic Kras mice.The Wnt/β-catenin signaling pathway was activated in pan-creatic Kras^(G12D)and Rnf43 knockout mice and the PORCN inhibitor LGK974 blocked pancreatic IPMN initiation and progression to PDAC accordingly.Conclusions:Rnf43 is a tumor suppressor in the prevention of pancreatic malignant transformation.This genetically reconstituted autochthonous pancreatic Rnf43^(−/−);Kras^(G12D)preclinical cancer model recapitulates the pathological process from pancreatic cyst to cancer in humans and can be treated with inhibitors of Wnt/β-catenin signaling.Since the presence of RNF43 and KRAS mutations in IPMNs predicts future development of advanced neoplasia from PCLs,patients with these genetic anomalies warrant surveillance,surgery,and/or targeted therapeutics such as Wnt/β-catenin inhibitors.Xian Zhou Zhichao Sun Mengdi Zhang Xiaoyu Qu Shuhui Yang Lianmei Wang Yanling Jing Li Li Weiwei Deng Fangming Liu Jin Di Jie Chen Jian Wu Hongbing Zhang 2022Animal Models and Experimental Medicine2022,5,1:3
8Identification of QTL for kernel number-related traits in a rice chromosome segment substitution line and fine mapping of qSP1显示文摘A chromosome segment substitution line (CSSL) is a powerful tool for combining quantitative trait locus (QTL) mapping with the pyramiding of desirable alleles. The rice CSSL Z1364 with increased kernel number was identified in a BC3F8 population derived from a cross of Nipponbare as the recipient with Xihui 18 as the donor parent. Z1364 carried three substitution segments distributed on chromosomes 1, 6, and 8. The mean substitution length was 1.19 Mb. Of 17 QTL identified on the substitution segments, qSP1 for spikelets per panicle, qSSD1 for seed-set density, and qNSB1 for number of secondary branches explained respectively 57.34%, 87.7%, and 49.44% of the corresponding phenotypic variance and were all linked to RM6777. Chi-square analysis showed that the increased kernel number in Z1364 was inherited recessively by a single gene. By fine mapping, qSP1 was delimited to a 50-kb region on the short arm of chromosome 1. Based on DNA sequence, a previously uncharacterized rice homolog of Arabidopsis thaliana AT4G32551 was identified as a candidate gene for qSP1 in which mutation increases the number of spikelets and kernels in Z1364. qSP1 was expressed in all tissues, but particularly in 1-cm panicles. The expression levels of OsMADS22, GN1A, and DST were upregulated and those of LAX2, GNP1, and GHD7 were downregulated in Nipponbare. 10The establishment of polypeptide PSMA-targeted chimericantigen receptor-engineered natural killer cells forcastration-resistant prostate cancer and the induction of ferroptosis-related cell death显示文摘Background:The mortality of castration-resistant prostate cancer(CRPC)is high due to lack of an effective treatment.Chimeric antigen receptor(CAR)-based therapy is a promising immunotherapeutic strategy.Here,we aimed to design a novel CAR-natural killer(NK)cells with a clinically significant tumoricidal effect on CRPC.Methods:We constructed novel CAR-NK92MI cells with a CD244-based recombinant lentiviral vector.Different intracellular segments(CD244,NKG2D,or CD3ζ)were screened to identify the best candidate according to cell lysis assay and CD107a expression levels.To enhance the affinity of the CAR to the tumor antigen,we compared an antibody specific for prostate-specific membrane antigen(anti-PSMA)with PSMA-targeted polypeptide(p-PSMA),which was screened by phage display combinatorial library.Then,CAR-NK92MI cells with both a high affinity for PSMA and a strong tumoricidal capacity were generated.In addition,we verified their tumor-killing effect in vitro and in vivo.The release of cytokine by NK92MI cells was compared with that by CAR-NK92MI cells through flow cytometry and enzyme-linked immunosorbent assay.Moreover,ferroptosis-related cell death was explored as a possible underlying mechanism.Results:Three different CAR intracellular regions CAR1(CD244),CAR2(CD244,NKG2D)and CAR3(CD244,NKG2D,and CD3ζ)were constructed.CAR2 was chosen to confer a stronger tumoricidal ability on CAR-NK92MI cells.Compared with anti-PSMA,p-PSMA exhibited enhanced affinity for the tumor antigen.Thus,p-PSMA-CAR-NK92MI cells,which expressed CAR with a polypeptide-based antigen-binding region,an intracellular CD244 and a NKG2D costimulatory domain,were generated.They could selectively and successfully kill PSMA+target cells and exhibited specific lysis rate of 73.19%for PSMA-positive C4-2 cells and 33.04%for PSMA-negative PC3 cells.Additionally,p-PSMA-CAR-NK92MI cells had significantly higher concentrations of IFN-γ,TNF-αand granzyme B than NK92MI cells.In a CRPC cancer xenograft model,p-PSMA-CAR-NK92MI cells significantly inhibited tumor growth and exerted a more consistent killing effect than NK92MI cells.Moreover,ferroptosis is a potential mechanism through which CAR-NK92MI cells attack cancer cells,and is triggered by IFN-γ.Conclusions:p-PSMA-CAR-NK92MI cells can effectively kill CRPCPSMA+cells in vitro and in vivo.This strategy may provide additional treatment options for patients with CRPC.Liyuan Wu Fei Liu Le Yin Fangming Wang Hui Shi Qinxin Zhao Feiya Yang Dong Chen Xiying Dong Yuchun Gu Nianzeng Xing 2022Cancer Communications2022,42,8:2

13Mechanism study of reduction of CO_2 into formic acid by in-situ hydrogen produced from water splitting with Zn: Zn/ZnO interface autocatalytic role显示文摘We have previously developed a new process of highly efficient conversion of CO_2 and water into formic acid with metallic Zn without the addition of catalyst, however, its mechanism is not clear, particularly in the catalytic role of Zn/ZnO interface. Herein, the autocatalytic role of Zn/ZnO interface formed in situ during the reduction of CO_2 into formic acid with Zn in water was studied by combining high resolution transmission electron microscopy(HRTEM), X-ray diffraction(XRD) and X-ray photoelectron spectroscopy(XPS) techniques and experimental data. The electron microscope results show that possible defects or dislocations formed on Zn/ZnO interface, in which plays a key role for Zn H-formation. Further XPS analyses indicate that oxygen vacancies on Zn/ZnO interface increased at short reaction times(less than 10 min). 14Over 50,000 Metagenomically Assembled Draft Genomes for the Human Oral Microbiome Reveal New Taxa显示文摘The oral cavity of each person is home to hundreds of bacterial species.While taxa for oral diseases have been studied using culture-based characterization as well as amplicon sequencing,metagenomic and genomic information remains scarce compared to the fecal microbiome.Here,using metagenomic shotgun data for 3346 oral metagenomic samples together with 808 published samples,we obtain 56,213 metagenome-assembled genomes(MAGs),and more than 64%of the 3589 species-level genome bins(SGBs)contain no publicly available genomes.The resulting genome collection is representative of samples around the world and contains many genomes from candidate phyla radiation(CPR)that lack monoculture.Also,it enables the discovery of new taxa such as a genus Candidatus Bgiplasma within the family Acholeplasmataceae.Large-scale metagenomic data from massive samples also allow the assembly of strains from important oral taxa such as Porphyromonas and Neisseria.The oral microbes encode genes that could potentially metabolize drugs.Apart from these findings,a strongly male-enriched Campylobacter species was identified.Oral samples would be more user-friendly collected than fecal samples and have the potential for disease diagnosis.Thus,these data lay down a genomic framework for future inquiries of the human oral microbiome.Jie Zhu Liu Tian Peishan Chen Mo Han Liju Song Xin Tong Xiaohuan Sun Fangming Yang Zhipeng Lin Xing Liu Chuan Liu Xiaohan Wang Yuxiang Lin Kaiye Cai Yong Hou Xun Xu Huanming Yang Jian Wang Karsten Kristiansen Liang Xiao Tao Zhang Huijue Jia Zhuye Jie 2022Genomics, Proteomics & Bioinformatics2022,20,2:1

18Comparative risk of acute kidney injury among cancer patients treated with immune checkpoint inhibitors显示文摘With the development and introduction of immune checkpoint inhibitors(ICIs)in cancer patients,immune-related side effects have increasingly attracted attention.However,the risks of immune-related renal toxicity are poorly characterized.In this study,we performed a network meta-analysis(NMA)of ICI-related randomized clinical trials(RCTs)to elucidate the comparative risk of acute kidney injury(AKI)in cancer patients receiving different ICIs.We also sought to identify other factors potentially affecting the risk of AKI.PubMed and EMBASE were searched for peer-reviewed trial reports published between January 2000 and May 2021.Eligible studies were RCTs studying ICIs in cancer patients and reporting AKI data.We performed a frequentist NMA to evaluate the risk ratios for grade 1-5 and grade 3-5 AKI between the treatment groups.We also assessed the absolute incidence of AKI in the ICI-containing arm using traditional direct meta-analysis.Once significant heterogeneity was detected in a traditional direct meta-analysis,multivariable meta-regression analysis was applied to identify factors that significantly affected the absolute incidence of AKI.A total of 85 RCTs were included in this study.In the NMA for the risk of grade 1-5 and 3-5 AKI,ipilimumab showed a significantly higher risk than avelumab and durvalumab,whereas 1 mg/kg nivolumab plus 3 mg/kg ipilimumab(N1I3)showed a significantly higher risk than other groups.In terms of treatment ranking,durvalumab±low-dose tremelimumab and avelumab were consistently among the top three safest treatments for grade 1-5 or 3-5 AKI,whereas N1I3,ipilimumab and tremelimumab were consistently among the top three treatments with the highest risk for grade 1-5 or 3-5 AKI.Compared with other cancers,renal cell carcinoma and urothelial carcinoma showed a significantly higher risk of AKI.The incidence of AKI was significantly higher with ICI+chemotherapy than with ICI monotherapy.In this NMA involving largescale up-to-date ICI trials,we demonstrated the comparative safety of existing ICI drugs for grade 1-5 and grade 3-5 AKI.Based on data from the ICI arms of these trials,we also revealed several potential risk factors for immune-related AKI,including tumor type and treatment paradigm.Fei Liu ZixianWang Xiaofan Li Zhen Zhang Yue Yang Junquan Chen Dinghua Chen Lingling Wu Xiangyu Liu Sujun Han Fangming Wang Wasilijiang Wahafu Yibo Gao Shancheng Ren Nianzeng Xing Guangyan Cai Xiangmei Chen 2023Cancer Communications2023,43,2:1
19The enhanced genomic 6mA metabolism contributes to the proliferation and migration of TSCC cells显示文摘In contrast to the well-established genomic 5-methylcytosine(5mC),the existence of N^(6)-methyladenine(6 mA)in eukaryotic genomes was discovered only recently.Initial studies found that it was actively regulated in cancer cells,suggesting its involvement in the process of carcinogenesis.However,the contribution of 6 mA in tongue squamous cell carcinoma(TSCC)still remains uncharacterized.In this study,a pan-cancer type analysis was first performed,which revealed enhanced 6mA metabolism in diverse cancer types.The study was then focused on the regulation of 6 mA metabolism,as well as its effects on TSCC cells.To these aspects,genome 6mA level was found greatly increased in TSCC tissues and cultured cells.By knocking down 6mA methylases N6AMT1 and METTL4,the level of genomic 6mA was decreased in TSCC cells.This led to suppressed colony formation and cell migration.By contrast,knockdown of 6mA demethylase ALKBH1 resulted in an increased 6 mA level,enhanced colony formation,and cell migration.Further study suggested that regulation of the NF-κB pathway might contribute to the enhanced migration of TSCC cells.Therefore,in the case of TSCC,we have shown that genomic 6mA modification is involved in the proliferation and migration of cancer cells.Lei Xi Ying Yang Ying Xu Fangming Zhang Jinghui Li Xiyang Liu Zhenxi Zhang Quan Du 2022International Journal of Oral Science2022,14,1:0
20SARS-CoV-2-specific immune response in COVID-19 convalescent individuals显示文摘We collected blood from coronavirus disease 2019(COVID-19)convalescent individuals and investigated SARS-CoV-2-specific humoral and cellular immunity in these discharged patients.Follow-up analysis in a cohort of 171 patients at 4-11 months after the onset revealed high levels of IgG antibodies.A total of 78.1%(164/210)of the specimens tested positive for neutralizing antibody(NAb).SARS-CoV-2 antigen peptide pools-stimulated-IL-2 and-IFN-y response can distinguish COVID-19 convalescent individuals from healthy donors.Interestingly,NAb survival was significantly affected by the antigen peptide pools-stimulated-IL-2 response,-IL-8 response,and-IFN-y response.The antigen peptide pools-activated CD8+T cell counts were correlated with NAb.The antigen peptide pools-activated natural killer(NK)cell counts in convalescent individuals were correlated with NAb and disease severity.Our data suggested that the development of NAb is associated with the activation of T cells and NK cells.Our work provides a basis for further analysis of the protective immunity to SARS-CoV-2 and for understanding the pathogenesis of COVID-19.It also has implications for the development of an effective vaccine for SARS-CoV-2 infection.Yunbao Pan Xianghu Jiang Liu Yang Liangjun Chen Xiaojiao Zeng Guohong Liu Yueting Tang Chungen Qian Xinming Wang Fangming Cheng Jun Lin Xinghuan Wang Yirong Li 2021Signal Transduction and Targeted Therapy2021,6,8:0
