| 4 | Transplantation of UC-MSCs on collagen scaffold activates follicles in dormant ovaries of POF patients with long history of infertility显示文摘Premature ovarian failure(POF) is a refractory disease for clinical treatment with the goal of restoring fertility. In this study,umbilical cord mesenchymal stem cells on a collagen scaffold(collagen/UC-MSCs) can activate primordial follicles in vitro via phosphorylation of FOXO3 a and FOXO1. Transplantation of collagen/UC-MSCs to the ovaries of POF patients rescued overall ovarian function, evidenced by elevated estradiol concentrations, improved follicular development, and increased number of antral follicles. Successful clinical pregnancy was achieved in women with POF after transplantation of collagen/UC-MSCs or UC-MSCs. | 7 | Prediction of Debris-flow Danger Area by Combining Hydrological and Inundation Simulation Methods显示文摘Debris flows have caused serious human casualties and economic losses in the regions strongly affected by the Ms8.0 Wenchuan earthquake of 2008. Debris flow mitigation and risk assessment is a key issue for reconstruction.The existing methods of inundation simulation are based on historical disasters and have no power of prediction.The rain-flood method can not yield detailed flow hydrograph and does not meet the need of inundation simulation. In this paper,the process of water flow was studied by using the Arc-SCS model combined with hydraulic method,and then the debris flow runoff process was calculated using the empirical formula combining the result from Arc-SCS.The peak discharge and runoff duration served as input of inundation simulation. Then,the dangerous area is predicted using kinematic wave method and Manning equation.Taking the debris flow in Huashiban gully in Beichuan County,Sichuan Province,China on 24 Sep.2008 as example,the peak discharge of water flow and debris flow were calculated as 35.52 m3·s-1 and 215.66 m3·s-,with error of 4.15%compared to the measured values.The simulated area of debris-flow deposition was 161,500 m2,vs.the measured area of 144,097 m2,in error of 81.75%.The simulated maximum depth was 12.3 m,consistent with the real maximum depth between 10 and 15 m according to the field survey.The minor error is mainly due to the flow impact on buildings and variations in cross-section configuration.The present methodology can be applied to predict debris flow magnitude and evaluate its risk in other watersheds inthe earthquake area. | CUI Peng | 2011 | Journal of Mountain Science2011,8,1: | 31 |
| 8 | Effects of nitrogen level on yield and quality of japonica soft super rice显示文摘Although studies on the balance between yield and quality of japonica soft super rice are limited, they are crucial for super rice cultivation. In order to investigate the effects of nitrogen application rate on grain yield and rice quality, two japonica soft super rice varieties, Nanjing 9108(NJ 9108) and Nanjing 5055(NJ 5055), were used under seven N levels with the application rates of 0, 150, 187.5, 225, 262.5, 300, and 337.5 kg ha^(–1). With the increasing nitrogen application level, grain yield of both varieties first increased and then decreased. The highest yield was obtained at 300 kg ha^(–1). The milling quality and protein content increased, while the appearance quality, amylose content, gel consistency, cooking/eating quality, and rice flour viscosity decreased. Milling was significantly negatively related with the eating/cooking quality whereas the appearance was significantly positively related with cooking/eating quality. These results suggest that nitrogen level significantly affects the yield and rice quality of japonica soft super rice. | 10 | The Reference Genome Sequence of Scutellaria baicalensis Provides Insights into the Evolution of Wogonin Biosynthesis显示文摘Scutellaria baicalensis Georgi is important in Chinese traditional medicine where preparations of dried roots,'Huang Qin,' are used for liver and lung complaints and as complementary cancer treatments. We report a high-quality reference genome sequence for S. baicalensis where 93% of the 408.14-Mb genome has been assembled into nine pseudochromosomes with a super-N50 of 33.2 Mb. Comparison of this sequence with those of closely related species in the order Lamiales, sesamum indicum and Salvia splendens,revealed that a specialized metabolic pathway for the synthesis of 4'-deoxyflavone bioactives evolved in the genus Scu-tellaria. We found that the gene encoding a specific cinnamate coenzyme A ligase likely obtained its newfunc- tion following recent mutations, and that four genes encoding enzymes in the 4'-deoxyflavone pathway are present as tandem repeats in the genome of S. baicalensis. Further analyses revealed that gene duplications, segmental duplication, gene amplification, and point mutations coupled to gene neo- and subfunctionaliza-tions were involved in the evolution of 4'-deoxyflavone synthesis in the genus Scutellaria. Our study not only provides significant insight into the evolution of specific flavone biosynthetic pathways in the mint family, Lamiaceae, but also will facilitate the development of tools for enhancing bioactive productivity by metabolic engineering in microbes or by molecular breeding in plants. | 11 | The first all-season sample set for mapping global land cover with Landsat-8 data显示文摘We report the world's first all-season training and validation sample sets for global land cover classification with Landsat-8 data.Prior to this,such samples were only available at a single date primarily from the growing season.It is unknown how much limitation such a single-date sample has to mapping global land cover in other seasons of the year.To answer this question,we selected available Landsat-8 images from four seasons and collected training and validation samples from them.We compared the performances of training samples in different seasons using Random Forest algorithm.We found that the use of training samples from any individual season would result in the best overall classification accuracy when validated by samples in the same season.The global overall accuracy from combined best seasonal results was 67.2% when classifying the 11 Level-1 classes in the Finer Resolution Observation and Monitoring of Global Land Cover(FROM-GLC) classification system.The use of training samples from all seasons(named all-season training sample set hereafter) produced an overall accuracy of 67.0%.We also tested classification within 10° latitude 60° longitude zones using all-season training subsample within each zone and obtained an overall accuracy of 70.2%.This indicates that properly grouped subsamples in space can help improve classification accuracies.All the results in this study seem to suggest that it is possible to use an all-season training sample set to reach global optimality with universal applicability in classifying images acquired at any time of a year for global land cover mapping. | Congcong Li Peng Gong Jie Wang Zhiliang Zhu Gregory S. | 12 | Characteristics and triggering mechanism of Xinmo landslide on 24 June 2017 in Sichuan, China显示文摘At 5: 39 AM on 24 June 2017, a huge landslide-debris avalanche occurred on Fugui Mountain at Xinmo village, Diexi town, Maoxian county, Sichuan province, China. The debris blocked the Songpinggou River for about 2 km, resulting in a heavy loss of both human lives and properties(10 deaths, 3 injuries, 73 missing, and 103 houses completely destroyed). The objectives of this paper are to understand the overall process and triggering factors of this landslide and to explore the affecting factors for its long term evolution before failure. Post event surveys were carried out the day after the landslide occurrence. Information was gathered from literature and on-site investigation and measurement. Topography, landforms, lithology, geological setting, earthquake history, meteorological and hydrological data of the area were analysed. Aerial photographs and other remote sensing information were used for evaluation and discussion. Eye witnesses also provided a lot of helpful information for us to understand the process of initiation, development and deposition. The depositional characteristics of the moving material as well as the traces of the movement,the structural features of the main scarp and the seismic waves induced by the slide are presented and discussed in detail in this paper. The results show that the mechanism of the landslide is a sudden rupture of the main block caused by the instability of a secondary block at a higher position. After the initiation, the failed rock mass at higher position overloaded the main block at the lower elevation and collapsed in tandem. Fragmentation of the rock mass occurred later, thus forming a debris avalanche with high mobility. | 14 | Effects of slow or controlled release fertilizer types and fertilization modes on yield and quality of rice显示文摘There is limited information about the influence of slow or controlled release fertilizer(S/CRF) on rice yield and quality. In this study, japonica rice cultivar Nanjing 9108 was used to study the effects of three different S/CRFs(polymer-coated urea(PCU), sulfur-coated urea(SCU), and urea formaldehyde(UF)) and two fertilization modes(both S/CRF and common urea(CU) as basal fertilizer, S/CRF as basal and CU as tillering fertilizer) on rice yield and quality. CU only was applied separately as control(CK). Results showed that, rice grain yield, chalky kernel rate, chalky area, overall chalkiness, and the content of gliadin, glutenin, and protein, all showed the trends of UF>PCU>SCU within the same fertilization mode, and showed the trends of S/CRF as basal and CU as tillering fertilizer>both S/CRF and CU as basal fertilizer within the same type of S/CRF. In contrast, the contents of amylose, amylopectin, and starch, as well as taste value, and peak and hot viscosity showed trends of SCU>PCU>UF, and the trends of both S/CRF and CU as basal fertilizer>S/CRF as basal and CU as tillering fertilizer. Among S/CRF treatments and fertilization modes, taste values of cooked rice were positively correlated with amylose, amylopectin, and starch contents, as well as gel consistency, peak viscosity, hot viscosity, and cool viscosity, while negatively correlated with globulin, gliadin, glutenin, and protein contents. The types of S/CRF and fertilization modes are important for improving rice yield and quality. | 15 | PERSPECTIVES IN MECHANICS OF HETEROGENEOUS SOLIDS显示文摘The Microand Nano-mechanics Working Group of the Chinese Society of Theoretical and Applied Mechanics organized a forum to discuss the perspectives,trends,and directions in mechanics of heterogeneous materials in January 2010.The international journal,Acta Mechanica Solida Sinica,is devoted to all fields of solid mechanics and relevant disciplines in science,technology,and engineering,with a balanced coverage on analytical,experimental,numerical and applied investigations.On the occasion of the 30 th anniversary of Acta Mechanica Solida Sinica,its editor-in-chief,Professor Q.S.Zheng invited some of the forum participants to review the state-of-the-art of mechanics of heterogeneous solids,with a particular emphasis on the recent research development results of Chinese scientists.Their reviews are organized into five research areas as reported in different sections of this paper.§I firstly brings in focus on microand nano-mechanics,with regards to several selective topics,including multiscale coupled models and computational methods,nanocrystal superlattices,surface effects,micromechanical damage mechanics,and microstructural evolution of metals and shape memory alloys.§II shows discussions on multifield coupled mechanical phenomena,e.g.,multi-fields actuations of liquid crystal polymer networks,mechanical behavior of materials under radiations,and micromechanics of heterogeneous materials.In §III,we mainly address the multiscale mechanics of biological nanocomposites,biological adhesive surface mechanics,wetting and dewetting phenomena on microstructured solid surfaces.The phononic crystals and manipulation of elastic waves were elaborated in §IV.Finally,we conclude with a series of perspectives on solid mechanics.This review will set a primary goal of future science research and engineering application on solid mechanics with the effort of social and economic development. | C.Q. | 17 | Soil Bacterial Communities Under Different Long-Term Fertilization Regimes in Three Locations Across the Black Soil Region of Northeast China显示文摘Although soil bacteria play critical roles in agro-ecosystems, the knowledge of their response to long-term fertilization across the black soil region of Northeast China is limited. In this study, we sequenced 16 S rRNA genes to assess the effects of four long-term fertilization regimes—non-fertilization(NoF), chemical fertilizer(CF), manure(M), and chemical fertilizer plus manure(CFM)—on soil properties and bacterial communities in three locations, the northern, middle, and southern parts, across the black soil region. Results showed that the influence of fertilization regimes on soil properties varied significantly among the three locations. Manure fertilization significantly increased microbial biomass carbon and relative abundance of copiotrophic bacteria. Principal component analysis(PCA)showed that the total bacterial communities were separated into three groups according to the sampling location despite long-term fertilization, and that soil pH was the most important factor in shifting bacterial communities. In addition, similar fertilization regimes resulted in different influences on bacterial community composition, and the most influential soil properties varied among the three locations. | 19 | Calcium channel blockers reduce severe fever with thrombocytopenia syndrome virus (SFTSV) related fatality显示文摘Severe fever with thrombocytopenia syndrome (SFTS),an emerging tick-borne infectious disease caused by a novel phlebovirus (SFTS virus, SFTSV), was listed among the top 10 priority infectious diseases by the World Health Organization due to its high fatality of 12%-50% and possibility of pandemic transmission. Currently, effective anti-SFTSV intervention remains unavailable. Here, by screening a library of FDA-approved drugs, we found that benidipine hydrochloride, a calcium channel blocker (CCB), inhibited SFTSV replication in vitro. Benidipine hydrochloride was revealed to inhibit virus infection through impairing virus internalization and genome replication. Further experiments showed that a broad panel of CCBs, including nifedipine, inhibited SFTSV infection. The anti-SFTSV effect of these two CCBs was further an a lyzed in a humanized mouse model in which CCB treatment resulted in reduced viral load and decreased fatality rate. Importantly, by performing a retrospective clinical investigation on a large cohort of 2087 SFTS patients, we revealed that nifedipine administration enhaneed virus clearance, improved clinical recovery, and remarkably reduced the case fatality rate by >5-fold.
