| 3 | Inhibiting effect of antisense oligonucleotides phosphorthioate on gene expression of TIMP-1 in rat liver fibrosis显示文摘AIM To observe the inhibition of antisenseoligonucleotides (asON) phosphorthioate to thetissue inhibitors metalloproteinase-1 (TIMP-1)gene and protein expression in the liver tissue ofimmunologically induced hepatic fibrosis rats.The possibility of reversing hepatic fibrosisthrough gene therapy was observed.METHODS Human serum albumin (HSA) wasused to attack rats, as hepatic fibrosis model, inwhich asONs were used to block the gene andprotein expressing TIMP-1. According to theanalysis of modulator, structure protein, codingseries of TIMP-1 genome, we designed fourdifferent asONs. These asONs were injected intothe hepatic fibrosis models through coccygealvein. The results was observed by RT-PCR formeasuring TIMP-1 mRNA expression,immunohistochemistry and in situ hybridizationfor collagen Ⅰ, Ⅲ, special staining of collagenfiber, and electron microscopic examination.RESULTS Hepatic fibrosis could last within 363days in our modified model. The expressinglevel of TIMP-1 was high during hepatic fibrosisprocess. It has been proved by theimmunohistochemical and the electronmicroscopic examination that the asONphosphorthioate of TIMP-1 could exactly expressin vivo. The effect of colchicine wasdemonstrated to inhibit the expressing level ofmRNA and the content of collagen Ⅰ, Ⅲ in theliver of experimental hepatic fibrosis rats.However, the electron microscopy research andthe pathologic grading of hepatic fibrosisshowed that there was no significant differencebetween the treatment group and the modelgroup (P>0.05).CONCLUSION The experimental rat model ofhepatic fibrosis is one of the preferable modelsto estimate the curative effect of anti-hepaticfibrosis drugs. The asON phosphorthioate ofTIMP-1 could block the gene and proteinexpression of TIMP-1 in the liver of experimentalhepatic fibrosis rats at the mRNA level. It ispossible to reverse hepatic fibrosis, and it isexpected to study a new drug of anti-hepaticfibrosis on the genetic level. | 7 | Contamination of Fungi and Mycotoxins in Foodstuffs in High Risk Area of Esophageal Cancer显示文摘Fungal and mycotoxin contamination has been found in fordstuffs from Cixian County, an area with a high incidence of esophageal carcinoma (Eca). To set a scientific foundation for the prevention of Eca at the etiological level, fungal and mycotoxin contaminations of local foodstuffs in Cixian County were analyzed using classical fungal culture methods and HPLC. From 1990 to 1994, 220 corn/wheat samples and 34 corn samples were studied. As a control, 26 corn samples collected from a relatively low incidence area of Eca in Zanhuang County were analyzed for mycotoxins in 1990. The results showed that fungal contamination in corn and wheat was severe and that several of the predominant contaminating fungi such as Furasium moniliforme, etc. were carcinogenic. HPLC results showed that the detection rate and mean content of the mycotoxin sterigmatocystin (ST) in the mountainous area (5/8; 9. 14 μg/kg) were significantly higher than those in the hilly (0; 0) and plain areas (1/18; 1. 29 μg/kg) as well as in low risk area control samples (3/26; 0. 76 μg/kg). Detection rates of deoxynivalenol (DON) in mountainous and hilly areas (5/8, 4/8 respectively) were slightly higher than that in plain area (8/18), while the mean content of DON in the plain area was significantly higher (90. 45-170. 22 vs 50. 56 and 46. 45 μg/kg).Among the different aflatoxins, AFB1 was detected in samples from the two villages in the plain area for the subsequent two years. The mean content of AFB1 was 0. 0183μg/kg, and the highest level was 0. 0497 μg/kg. No AFB1 was detected in the samples from mountainous and hilly areas. AFG1 was detected in more than half of all the samples, and its concentration ranged from 8. 77 to 46. 51 ng/kg. No AFG2 was found in the samples. | 11 | Furazolidone-based triple and quadruple eradication therapy for Helicobacter pylori infection显示文摘AIM: To evaluate the efficacy of furazolidone-based triple and quadruple therapy in eradicating Helicobacter pylori(H. pylori) in a multi-center randomized controlled trial.METHODS: A total of 720 H. pylori positive patients with duodenal ulcer disease were enrolled at 10 different hospitals in Jiangxi province in China. The patients were randomly assigned to four treatment groups as follows: patients in Groups 1 and 3 received rabeprazole(10 mg), amoxicillin(1000 mg) and furazolidone(100 mg) twice daily for 7 and 10 d, respectively; patients in Groups 2 and 4 received rabeprazole(10 mg), bismuth(220 mg), amoxicillin(1000 mg) and furazolidone(100 mg) twice daily for 7 and 10 d, respectively. The primary outcome measure was H. pylori eradication rate 4 wk after treatment by intention-to-treat and per protocol analysis, while the secondary outcome measures were symptom and sign changes at the end of treatment and 4 wk after the end of treatment, as well as the proportion of patients who developed adverse events.RESULTS: The demographic data of the four groups were not significantly different. Overall, 666 patients completed the scheme and were re-assessed with the 13C-urea breath test. The intention-to-treat analysis of the H. pylori eradication rates in Groups 1, 2, 3 and 4 were 74.44%, 82.78%, 78.89% and 86.11%, respectively. The H. pylori eradication rate in Group 4 was significantly higher than that in Group 1. According tothe per protocol analysis, the H. pylori eradication rates in Groups 1, 2, 3 and 4 were 81.21%, 89.22%, 85.54% and 92.26%, respectively. The H. pylori eradication rate in Group 4 was significantly higher than that in Group 1. The number of adverse events was 15(8.3%), 16(8.9%), 15(8.3%) and 17(9.4%) in Groups 1, 2, 3 and 4, respectively, including dizziness, vomiting, diarrhea, nausea, skin rash, itchy skin, and malaise. | 16 | Controlling factors on the submarine canyon system: A case study of the Central Canyon System in the Qiongdongnan Basin, northern South China Sea显示文摘Based on an integrated analysis of high-resolution 2D/3D seismic data and drilling results, this study analyzes the tectonicsedimentary evolution of the Qiongdongnan Basin(QDNB) since the late Miocene, and discusses the controlling factors on the formation and development of the Central Canyon System(CCS). The sediment failures caused by the relative sea level falling might have discharged deposits from the slope to the canyon. The two suits of the infillings, i.e., turbidites and mass transport complex(MTC), were derived from the northwestern source and northern source, respectively. The sediment supplies, which differ significantly among different areas, might have led to the variations observed in the internal architectures. Tectonic transformation around 11.6 Ma had provided the tectonic setting for the CCS and formed an axial sub-basin in the central part of the Changchang Depression, which could be called the rudiment of the CCS. The tectonic activity of the Red River Fault(RRF) at about 5.7 Ma might have strengthened the hydrodynamics of the deposits at the junction of the Yinggehai Basin(YGHB) and the QDNB to trigger a high-energy turbidity current. The MTC from the northern continental slope system might have been constrained by the Southern Uplift, functioning as a barrier for the infillings of the CCS. Thanks to a sufficient sediment supply during the Holocene period and the paleo-seafloor morphology, the relief of modern central canyon with the starving landform in the eastern Changchang Depression might have been accentuated by deposition of sediments and vertical growth along the canyon flanks, where collapse deposits were widely developed. Corresponding to the segmentation of the CCS, the forming mechanisms of the canyon between the three segments would be different. The turbidite channel in the head area had likely been triggered by the abundant sediment supply from the northwestern source together with the fault activity at about 5.7 Ma of the RRF. The formation and evolution of the canyon in the western segment were caused by combined effects of the turbidite channel from the northwestern source, the MTC from the northern continental slope, and the paleo-seafloor geomorphology. | 17 | Regulatory effect and mechanism of gastrin and its antagonists on colorectal carcinoma显示文摘AIM To explore the effect and mechanism ofgastrin and its antagonists proglumide and somatostatin on coIorectal carcinoma and their clinical significance.METHODS A model of transplanted human colonic carcinoma was established from SW480cell line in gymnomouse body. The volume andweight of transplanted carcinoma was observedunder the effect of pentagatrin (PG), proglumide (PGL) and octapeptide somotostatin (SMS201-995, SMS). The cAMP content ot carcinoma cell was determined by redioimmunoassay and the DNA, protein content and cell cycle were determined by flow-cytometry. The amount of viabIe cells was determined by MTT colorimetric analysis, lP3 content was determined by radioimmunoassay, Ca2+ concentration in cell by fluorometry and PKC activity by isotopic enzymolysis. The expression of gastrin, c-myc,C-fos and rasP21 in 48 cases of colorectal carcinoma tissue was detected by the immunocytochemistry SP method. Argyrophilianucleolar organizer regions was determined withargyrophilia stain.RESULTS The volume, weight, cAMP, DNAand protein content in carcinoma cell, cellamount and proliferation index of S and G2Mphase in PG group were all significantly higher than those of control group. When PG was at theconcentration of 25 mg/ L, the amount of viablecells, lP3 content and Ca2+ concentration in celland membrane PKC activity in PG group weresigniticantly higher than those in control group;when PGL was at a concentration ot 32 mg/L,they dropped to the lowest level in PG (25 mg/L)+ PGL group, but without significant differencefrom the control group. The positive expression rate of gastrin, c-myc, c-fos and rasp21 in carcinoma tissue was 39 .6%, 54 .2%, 47. 9% and54 .2% resPectively and significantly higher than that in mucosa 3 cm and 6 cm adjacent tocarcinoma tissue and normal colorectal mucosa.The positive expression rate of gastrin of highlydifferentiated adenocarcinoma group was significantly higher than that of poorly differentiated and mucinous adenocarcinoma groups. The AgNORs count of carcinoma tissue was significantly higher than that in mucosa 3 cm and 6 cm adjacent to carcinoma tissue and normal colorectal mucosa; and the positive expression of c-myc and c-fos and the AgNORs count in gastrin-positive group was significantly higher than those in gastrin-negative group.CONCLUSION Pentagastrin has a promoting effect on the growth of transplanted human colonic carcinoma from SW480 cell line. PGL hasno obvious effect on the growth of human colonic carcinoma SW480 cell line, but couldinhibit the growth-promoting effect of PG on transplanted carcinoma. Somatostatin can not only inhibit the growth of transplanted human colonic carcinoma from SW480 cell line directlybut also depress the growth-promoting effect ofgastrin on the transplanted carcinoma. Somecolorectal carcinoma cells can produce and secrete gastrin through autocrine, highly-differentiated adenocarcinoma express the highest level gastrin. Endogenous gastrin can stimulate the cell division and proliferation of carcinoma cell and promote the growth of colorectal carcinoma regulating the expression of oncogene omyc, c-fos. | 18 | Comparative Analysis on Chemical Components and Sensory Quality of Aging Flue-Cured Tobacco from Four Main Tobacco Areas of China显示文摘Complicated changes are happening during the aging process of flue-cured tobacco (FCT) and understanding of the changing components is of great significance in improving the quality,shortening aging time,and enhancing production and economic efficiency in tobacco industry.The aging samples of FCT from four main producing areas of China,i.e.,Yunnan,Sichuan,Hubei,and Dongbei,were used to study the changing rule of the components such as alkali,acid,and carbohydrate as well as the aroma and their relationship with sensory quality;and based on the correlation among the components and the sensory quality index,multivariable models were established to predict the aging quality.The results showed that the sensory quality of FCT increased steadily during the aging time,and at the same time,the acidic components (total organic acids,volatile acids) increased gradually,while the alkaline substances (nicotine,volatile alkali),carbohydrate (total sugars,reducing sugar) and pH values showed a downward trend.Correlation analysis found that the sensory quality and pH values were negatively correlated (P<0.05),while the sensory quality with total organic acids and aroma were positively correlated.The optimal model for predicting the quality of FCT was y=56.75-0.63x12+50.09x2-13.27x22,(y:sensory quality;x1:pH;x2:total organic acids).The average predicating error was 1.04% with maximum predicating error of 2.89% and the accuracy rate of above 97%. | SUN Jing-guo HE Jie-wang WU Feng-guang TU Shu-xing YAN Tie-jun SI Hui XIE Hao | 2011 | Agricultural Sciences in China2011,10,8: | 20 |

| 20 | Soil pH impact on microbial tetraether lipids and terrestrial input index (BIT) in China显示文摘The distributions of microbial tetraether lipids i.e. glycerol dialkyl glycerol tetraethers (GDGTs) were investigated in a variety of Chinese soils featured by a wide range of pH values in different climate zones. The bacterial branched GDGTs and archaeal isoprenoid GDGTs were found to be widely distributed in Chinese soils and their relative abundance was controlled primarily by soil pH. The bacterial bGDGTs usually dominate over archaeal iGDGTs in abundance when the soil pH is below 8.0. However archaeal iGDGTs become dominant and the abundance of halophilic archaea characterized by GDGT V without cyclopentyl ring increases dramatically in the alkaline soil with pH greater than 8.0. The relative abundance of archaeal iGDGTs to bacterial bGDGTs was defined here as RAI index. The RAI index decreases with elevated soil pH and a linear correlation is found between them indicating that the relative abundance of archaeal iGDGTs to bacterial bGDGTs increases with increased pH. The RAI index thus could be a novel proxy of the pH values of paleosol. The terrestrial input index BIT (Branched and Isoprenoid Tetraethers Index) decreases with increased soil pH suggesting that soil pH should be taken into consideration when the terrestrial contribution to marine sediments is estimated by BIT index.
