2Spatial and temporal variation of particulate matter and gaseous pollutants in 26 cities in China显示文摘O3and PM2.5were introduced into the newly revised air quality standard system in February 2012, representing a milestone in the history of air pollution control, and China's urban air quality will be evaluated using six factors(SO2, NO2, O3, CO, PM2.5and PM10) from the beginning of 2013. To achieve the new air quality standard, it is extremely important to have a primary understanding of the current pollution status in various cities. The spatial and temporal variations of the air pollutants were investigated in 26 pilot cities in China from August 2011 to February 2012, just before the new standard was executed. Hourly averaged SO2, NO2and PM10were observed in 26 cities, and the pollutants O3, CO and PM2.5were measured in 15 of the 26 cities. The concentrations of SO2and CO were much higher in the cities in north China than those in the south. As for O3and NO2, however, there was no significant diference between northern and southern cities. Fine particles were found to account for a large proportion of airborne particles, with the ratio of PM2.5to PM10ranging from 55% to 77%. The concentrations of PM2.5(57.5 μg/m3) and PM10(91.2 μg/m3) were much higher than the values(PM2.5: 11.2 μg/m3; PM10 : 35.6 μg/m3) recommended by the World Health Organization. 3Generation of GGTA1 biallelic knockout pigs via zinc-finger nucleases and somatic cell nuclear transfer显示文摘Genetically modified pigs are valuable models of human disease and donors of xenotransplanted organs.Conventional gene targeting in pig somatic cells is extremely inefficient.Zinc-finger nuclease(ZFN)technology has been shown to be a powerful tool for efficiently inducing mutations in the genome.However,ZFN-mediated targeting in pigs has rarely been achieved.Here,we used ZFNs to knock out the porcineα-1,3-galactosyl-transferase(GGTA1)gene,which generates Gal epitopes that trigger hyperacute immune rejection in pig-to-human transplantation.Primary pig fibroblasts were transfected with ZFNs targeting the coding region of GGTA1.Eighteen mono-allelic and four biallelic knockout cell clones were obtained after drug selection with efficiencies of 23.4%and 5.2%,respectively.The biallelic cells were used to produce cloned pigs via somatic cell nuclear transfer(SCNT).Three GGTA1 null piglets were born,and one knockout primary fibroblast cell line was established from a cloned fetus.Gal epitopes on GGTA1 null pig cells were completely eliminated from the cell membrane.Functionally,GGTA1 knockout cells were protected from complement-mediated immune attacks when incubated with human serum.This study demonstrated that ZFN is an efficient tool in creating gene-modified pigs.GGTA1 null pigs and GGTA1 null fetal fibroblasts would benefit research and pig-to-human transplantation.BAO Lei CHEN HaiDe JONG UiMyong RIM CholHo LI WenLing LIN XiJuan ZHANG Dan LUO Qiong CUI Chun HUANG HeFeng ZHANG Yan XIAO Lei FU ZhiXin 2014Science China(Life Sciences)2014,57,2:9

5Distribution characteristics and its controlling factor of lacustrine high-quality source rocks in the Bozhong sag,Bohai Bay Basin显示文摘Distribution of Paleogene lacustrine high-quality source rocks in the Bozhong sag in Bohai Bay Basin is analyzed through data of geochemistry,geology and well logging,and its differences under the control of climate and tectonics is also well discussed.Distribution characteristics of the high-quality source rocks developed in the saline environment controlled by the climate are quite different from that developed in the rapid subsidence environment controlled by tectonics.The high-quality source rocks in Member 1 of Shahejie Formation developed in the saline environment account for 81.9%of total subsag area,and are distributed widely and extensively.The high-quality source rocks in Member 3 of Shahejie Formation and Member 3 of Dongying Formation is developed in the tectonic subsidence environment,and horizontally,the subsag subsidence rates has a positive correlation with the area proportion of the high-quality source rocks in the sag;vertically,the reduction-oxidation interface of the lake controls the enrichment of highquality source rocks.Controlled by the saline environment and rapid subsidence environment,the highquality source rocks in the Bozhong sag and adjacent areas have three types of development condition:the saline water,the tectonic subsidence,and joint control of the saline water and the tectonic subsidence.The humid climate and low subsidence rate are not favorable for development of high-quality source rocks.Hefeng Sun Lifang Liu Xue Jiang Shengbing Huang Mingyang Geng Shaoping Chen Nan Li Pu Shen 2019Petroleum Research2019,4,3:3

7Dietary fibers with low hydration properties exacerbate diarrhea and impair intestinal health and nutrient digestibility in weaned piglets显示文摘Background:This study aimed to investigate the hydration properties of different-source fibrous materials by com-paring their water-binding capacity(WBC),water swelling capacity(WSC),viscosity,and in vivo effects of selected samples on growth performance,nutrient digestibility,diarrhea,and intestinal health in weaned piglets.Methods:A total of 13 commercially available fibrous materials were first compared in chemical composition and in vitro hydration property.Subsequently,40 weaned piglets were randomized to five experimental dietary groups(8 piglets per group):control diet(a basal diet without dietary fiber,CON),basal diet supplemented with 5%microcrys-talline cellulose(MCC),5%wheat bran(WB),5%Moringa oleifera leaf powder(MOLP),or 5%sugar beet pulp(SBP),fol-lowed by analyzing their growth performance and diarrhea rate in a 28-d experiment.After the feeding experiment,anaesthetized piglets were killed,and their intestinal and colon content or plasma samples were analyzed in nutrient digestibility,intestinal morphology,intestinal barrier,short-chain fatty acids(SCFAs),and bacterial population.Results:In vitro studies showed low hydration properties for WB and MCC,while medium hydration properties for MOLP and SBP.In vivo studies indicated that compared with medium hydration property groups,low hydration prop-erty groups showed(1)exacerbated diarrhea,impaired intestinal health,and reduced apparent fecal digestibility of dry matter,gross energy,acid detergent fiber,and neutral detergent fiber;(2)decreased SCFAs concentration and rela-tive levels of Lactobacillus and Bifidobacterium,but increased levels of Escherichia coli and Brachyspira hyodysenteriae in colon contents.Additionally,SBP showed optimal performance in reducing diarrhea and increasing SCFAs produc-tion.Correlation analysis revealed a positive correlation of fiber hydration properties with in vitro SCFAs production,and diarrhea index and nutrient digestibility were negatively and positively correlated with SCFAs levels in the colon contents of weaned piglets,respectively.Conclusions:Different-source dietary fibers varied in their hydration properties and impacts on diarrhea,microbial composition and SCFAs production in weaned piglets.WB and MCC could exacerbate diarrhea and impair nutrient digestibility,probably because their low hydration properties were detrimental to gut microbial homeostasis and fermentation.Our findings provide new ideas for rational use of fiber resources in weaned piglets.Shuangbo Huang Zhijuan Cui Xiangyu Hao Chuanhui Cheng Jianzhao Chen Deyuan Wu Hefeng Luo Jinping Deng Chengquan Tan 2023Journal of Animal Science and Biotechnology2023,14,2:1

9Intrauterine hyperglycemia impairs endometrial receptivity via up-regulating SGK1 in diabetes显示文摘Diabetes is a complex metabolic disorder which can adversely affect reproductive function. SGK1 is found to be up-regulated in multiple tissues of diabetic patients. However, the effects of diabetes on endometrial SGK1 expression and endometrial receptivity remain unknown. In this study, we established a streptozotocin-induced diabetic mouse model and observed reduced implantation sites, retarded development of pinopodes, increased SGK1, and aberrant expression of LIF and MUC1 in the endometrial epithelium. We injected the uterine lumen of normal mice with high-glucose solution and cultured endometrial cells in high-glucose medium to mimic intrauterine hyperglycemia. Both studies provided compelling evidence that hyperglycemia could lead to diminished embryo implantation and dysregulated SGK1, LIF and MUC1. Additionally, through over-expression of SGK1 in vivo and in vitro, we found that enhanced SGK1 also decreased LIF expression, increased MUC1 expression, and attenuated embryo implantation rate. We further identified that hyperglycemia-activated SMAD2/3 might be responsible for the enhancement of SGK1 and verified directly the interaction between SMAD3 and corresponding SMAD binding elements within SGK1 promoter. Taken together, our study confirmed the association between diabetes-related hyperglycemia and endometrial receptivity defects. 13Dietary adenosine supplementation improves placental angiogenesis in IUGR piglets by up-regulating adenosine A2a receptor显示文摘Abnormal placental angiogenesis is associated with the occurrence of intrauterine growth restriction(IUGR)in piglets,and effective treatment strategies against this occurrence remain to be explored.Adenosine has been reported to play an important role in angiogenesis,but its role in placental angio-genesis is still unknown.Here,we investigated the effect of dietary adenosine supplementation on IUGR occurrence in piglets by analyzing the role of adenosine in placental angiogenesis for Normal and IUGR piglets.Specifically,88 sows were allotted to 2 treatments(n=44)and fed a basal diet supplemented with 0%or 0.1%of adenosine from day 65 of gestation until farrowing,followed by collecting the placental samples of Normal and IUGR piglets,and recording their characteristics.The results showed that adenosine supplementation increased the mean birth weight of piglets(P<0.05)and placental efficiency(P<0.05),while decreasing the IUGR piglet rate(P<0.05).Expectedly,the placenta for IUGR neonates showed a down-regulated vascular density(P<0.05)and angiogenesis as evidenced by the expression level of vascular cell adhesion molecule-1(VCAM1)(P<0.05).Notably,dietary adenosine supplementation promoted angiogenesis(P<0.05)both in the Normal and IUGR placenta.More importantly,the expression level of adenosine A2a receptor(ADORA2A)was lower(P<0.05)in the IUGR placenta than in Normal placenta,whereas adenosine treatment could significantly increase ADORA2A expression,and also had an interaction effect between factors IUGR and Ado.Collectively,placentae for IUGR piglets showed impaired angiogenesis and down-regulated expression level of ADORA2A,while dietary adenosine supplementation could activate ADORA2A expression,improve the placental angio-genesis,and ultimately decrease the occurrence of IUGR in piglets.Zifang Wu Jiawei Nie Deyuan Wu Shuangbo Huang Jianzhao Chen Huajin Liang Xiangyu Hao Li Feng Hefeng Luo Chengquan Tan 2023Animal Nutrition2023,,2:0
14Comprehensive analysis of long noncoding RNAs and mRNAs expression profiles and functional networks during chondrogenic differentiation of murine ATDC5 cells显示文摘Chondrogenic differentiation is a coordinated biological process orchestrated by various cell signaling pathways,involving complex pathways regulated at both transcriptional and post-transcriptional levels.Long noncoding RNAs(lncRNAs)are emerging as important regulators in the modulation of multiple cell processes.However,the potential roles of lncRNAs and their regulatory mechanisms in chondrogenic differentiation remain largely unclear.In this study,microarray was performed to detect the expression profiles of lncRNAs and messenger RNAs(mRNAs)during chondrogenic differentiation of murine chondrogenic cell line ATDC5.Gene ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway analysis were performed to explore their functions.Coding-noncoding co-expression(CNC)and competing endogenous RNA(ceRNA)networks were also constructed with bioinformatics methods.The results revealed that 1009 lncRNAs and 1206 mRNAs were differentially regulated during chondrogenic differentiation.GO and KEGG pathway analysis indicated that the principal functions of the transcripts were associated with system development and extracellular matrix-receptor interaction,TGF-βsignaling,and PI3K-Akt signaling pathways.The CNC network showed that lncRNA AK136902 was positively correlated with prostaglandin F receptor(FP).The ceRNA network covered 3 lncRNAs,121 miRNAs and 241 edges.The upregulated lncRNA AK136902,AK016344,and ENSMUST00000180767 might promote chondrogenic differentiation by acting as ceRNAs.Knockdown of lncRNA AK136902 could inhibit the mRNA expression of FP and other chondrogenic related genes,including Aggrecan and Col2a1 during chondrogenic differentiation.Our results provide a new perspective on the modulation of lncRNAs during chondrogenic differentiation.Wei Wang Yu Ding Yanhua Xu Hefeng Yang Wenjing Liu Haixia Wang Ceshi Chen Rong Liu Song Li 2019Acta Biochimica et Biophysica Sinica2019,51,8:0
15An alternative splicing variant of mineralocorticoid receptor discovered in preeclampsia tissues and its effect on endothelial dysfunction显示文摘The pathophysiology of preeclampsia(PE)remains unclear.PE spiral artery remodeling dysfunction and PE offspring cardiovascular future development has been a worldwide concern.We collected placental and umbilical artery samples from normotensive and PE pregnancies.Mineralocorticoid receptor(MR)and its alternative splicing variant(ASV)expression and their biological effects on PE were examined.An MR ASV was found to be highly expressed in all PE samples and slightly expressed in about half of the normotensive samples(umbilical artery,~57.58%;placenta,~36.84%).The MR ASV expression was positively associated with blood pressure in both groups.The MR ASV protein changed the aldosterone-induced expression pattern of MR target genes related to ion exchanges and cell signaling pathways.The MR ASV can also impair the proliferation,migration,and tube formation ability of endothelial cells.These findings indicate that MR ASV in PE placenta plays a pathogenic role in PE pathophysiology,especially in endothelial dysfunction,and the existence of the MR ASV in PE umbilical artery provides a new direction in the study of PE offspring with increased risk of cardiovascular diseases.Mengxi Guo Chengliang Zhou Gufeng Xu Lin Tang Yechun Ruan Ying Yu Xianhua Lin Dandan Wu Hao Chen Priscilla Yu Luyang Jin Yinyu Wang Yimei Wu Kamran Ullah Tanzil Ur Rahman Xinmei Liu Jianzhong Sheng Hsiao-Chang Chan Hefeng Huang 2020Science China(Life Sciences)2020,63,3:0
16Performance evaluation of best universities in China's Mainland based on ESI显示文摘The research performance of top universities in China is revealed in this study and analyzed using the essential science indicators(ESI) database.Different types of bibliometric indicators are applied in this study,including publication output and impact indicators.This study finds that the academic performance of universities in China is advancing rapidly,with many universities coming out near the top of the rankings,especially in terms of quantity of papers.The quantitative performance of universities in China is remarkable.The research results show that there is a considerable gap between quality and quantity of research output at the top universities of China,with research influence concentrated on some specific fields for selected universities.Engineering,Chemistry and Material Science are the fields with relatively outstanding performance in the top universities.曹燕 Tong Hefeng Zhang Xu Chen Darzen Huang Muhsuan 2013High Technology Letters2013,19,2:0
