| 1 | Effectiveness of Jinying capsule on pelvic inflammatory disease in patients with symptom pattern of damp and heat accumulation: a double-blinded, multicenter, randomized, placebo-controlled clinical trial显示文摘OBJECTIVE: To evaluate the therapeutic effectiveness and safety of Jinying capsule on pelvic inflammatory disease(PID) in patients with symptoms identified as the pattern of damp and heat accumulation in terms of Traditional Chinese Medicine(TCM).METHODS: We conducted a double-blinded, multicenter, randomized, placebo-controlled clinical trial which included 155 patients diagnosed with PID and identified as symptom pattern of damp and heat accumulation. They were randomly divided into experimental group(n = 78) and control group(n = 77) according to a random number table. The treatment lasted for a period of 28 d. The experimental group was given Jinying capsules and oral levofloxacin plus oral metronidazole for first 7 d.They continued with Jinying capsules and levofloxacin placebo and metronidazole placebo for another 7 d. For the remaining 14 d, they continued with Jinying capsules only. Whereas, the control group was treated with oral levofloxacin and metronidazole and Jinying capsule placebo for the first 14 d in the same way as the experimental group and then continued with Jinying capsule placebo only for the remaining 14 d. The clinical efficacy was assessed using McCormack scale, TCM symptom pattern scores, physicochemical indexes including white blood cell and erythrocyte sedimentation rate, C-reaction protein, smear of vaginal discharge,and pelvic ultrasound.RESULTS: Comparing McCormack scale between both groups after treatment, the difference in curative effect between both groups was significant (P = 0.0269). The cure rate of the experimental group and control group is 76.32% and 59.46% respectively at week 4. Comparing TCM symptom pattern scores between both groups before and after treatment, the differences in total effective rate were both significant(P < 0.05). The curative effect rate of experimental group is 2.63% and 13.70% of the control group at week 1(P = 0.0131), and73.33% of the experimental group and 56.94% of the control group at week 4(P = 0.0369). No significant differences were found between the two groups on the Physicochemical indexes(all P >0.05). | 3 | Development and Evaluation of a Loop-mediated Isothermal Amplification Assay for the Rapid Detection and Identification of Pectobacterium carotovorum on Celery in the Field显示文摘Pectobacterium carotovorum is the causal agent of bacterial soft rot in a wide range of vegetable host species.Once P.carotovorum infects the plant,the spread of the disease is difficult to control.In this study,a rapid and sensitive method based on loop-mediated isothermal amplification(LAMP)was developed for detecting P.carotovorum in celery with soft rot using a primer set designed from the pmrA conserved sequence of P.carotovorum.The specificity of the LAMP primer set for P.carotovorum was extensively validated on both P.carotovorum strains and nontarget strains.The sensitivity was 1 pg of P.carotovorum genomic DNA,which demonstrated 10 times more sensitive than the conventional PCR assay.LAMP was also used to detect P.carotovorum in bacterial suspension.The lowest detection concentration was 104 CFU·mL^−1.In addition,a LAMP assay,in conjunction with a crude DNA extraction method,was successfully performed on P.carotovorum-infected samples derived from both artificially and naturally infected plants.In summary,the LAMP assay established in this study constitutes a simple,sensitive,and rapid method for the detection of P.carotovorum,and has potential application in the control of celery soft rot disease through early detection. | Yanxia Shi Zhiwen Jin Xianglong Meng Lixue Wang Xuewen Xie Ali Chai Baoju Li | 2020 | Horticultural Plant Journal2020,6,5: | 3 |
| 4 | Safety,tolerability,and pharmacokinetics of BAT8001 in patients with HER2-positive breast cancer:An open-label,dose-escalation,phase I study显示文摘Background:The introductions of anti-human epidermal growth factor receptor-2(HER2)agents have significantly improved the treatment outcome of patients with HER2-positive breast cancer.BAT8001 is a novel antibodydrug conjugate targeting human epidermal growth factor receptor-2(HER2)-expressing cells composed of a trastuzumab biosimilar linked to the drug-linker Batansine.This dose-escalation,phase I study was designed to assess the safety,tolerability,pharmacokinetics,and preliminary anti-tumor activity of BAT8001 in patients with HER2-positive locally advanced or metastatic breast cancer.Methods:This trial was conducted in subjects with histologically confirmed HER2-positive breast cancer(having evaluable lesions and an Eastern Cooperative Oncology Group performance status of 0 or 1)using a 3+3 design of escalating BAT8001 doses.Patients received BAT8001 intravenously in a 21-day cycle,with dose escalation in 5 cohorts:1.2,2.4,3.6,4.8,and 6.0 mg/kg.The primary objective was to evaluate the safety and tolerability of BAT8001.Preliminary activity of BAT8001 was also assessed as a secondary objective.Results:Between March 2017 to May 2018,29 HER2-positive breast cancer patients were enrolled.The observed dose-limiting toxicities were grade 4 thrombocytopenia and grade 3 elevated transaminase.The maximum tolerated dose was determined to be 3.6 mg/kg.Grade 3 or greater adverse events(AEs)occurred in 14(48.3%)of 29 patients,including thrombocytopenia in 12(41.4%)patients,aspartate aminotransferase increased in 4(13.8%)patients,γ-glutamyl transferase increased in 2(6.9%)patients,alanine aminotransferase increased in 2(6.9%)patients,diarrhea in 2(6.9%)patients.Objective response was observed in 12(41.4%,95%confidence interval[CI]=23.5%-61.1%)and disease control(including patients achieving objective response and stable disease)was observed in 24(82.8%,95%CI=64.2%-94.2%)patients.Conclusions:BAT8001 demonstrated favorable safety profiles,with promising anti-tumor activity in patients with HER2-positive locally advanced or metastatic breast cancer.BAT8001 has the potential to provide a new therapeutic option in patients with metastatic HER2-positive breast cancer. | Ruoxi Hong Wen Xia Liye Wang Kaping Lee Qianyi Lu Kuikui Jiang Shengfeng Li Jinquan Yu Jin Wei Weijia Tang Danyang Zhou Xin An Jiajia Huang Cong Xue Xiwen Bi Yanxia Shi Zhongyu Yuan Fei Xu Shusen Wang | 2021 | Cancer Communications2021,41,2: | 2 |

| 14 | Revisit of event-based rainfall characteristics at Shapotou area in northern China显示文摘Distribution of rainfall event sizes and interval lengths between events are important characteristics of arid and semi-arid climates. Understanding their importance will contribute to our ability to understand ecosystem dynamics in these regions. Rainfall event timing and magnitude are important drivers of ecosystem processes and are instrumental in creating landscape heterogeneity in arid and semi-arid regions. Rainfall event characteristics were analyzed using an automatic tipping-bucket rain-gauge record across the entire summer monsoon season from 2008 to 2015 at the arid desert area of Shapotou in the Tengger Desert, China. Changing the minimum inter-event time(MIT) from 30 min to 24 h alters the number of rainfall events from 64 to 25 for the event depth larger than 0.1 mm. The mean rainfall intensity declined from 0.95 mm/h to 0.53 mm/h, and the geometric mean event duration rose from 0.55 h to 4.4 h. The number of rainfall events, mean rainfall intensity, and geometric mean event duration differed under different criteria of individual rainfall depths, except that for an individual rainfall depth of 0.5, 1.0, and 5.0 mm. The aforementioned features differed only at the lowest range of the mean rainfall intensity and depth for MIT=3 and 6 h. These findings suggest that identification of event-based rainfall in this specific arid region can be better achieved by setting the MIT at six hours. The wide variation in rainfall event properties indicate the need for paying more attention to the proper selection and reporting of event criteria in studies that adopt event-based data analysis. | 15 | Synergistic effects of AAGL and anti-PD-1 on hepatocellular carcinoma through lymphocyte recruitment to the liver显示文摘Objective:Therapy for hepatocellular carcinoma(HCC)is a major challenge,and targeted therapies provide only a modest benefit in terms of overall survival.Treatment with antibodies to programmed cell death protein 1(PD-1)/PD-L1 can restore the functions of tumor-infiltrating T cells in HCC and has shown clinical efficacy in 20%of patients with advanced HCC.Novel approaches are urgently needed to treat HCC and to augment the efficacy of immunotherapy.Methods:Tumor-bearing mice were treated with Agrocybe aegerita galectin(AAGL)alone or in combination with anti-PD-1,and the tumor sizes and lifespans of mice were determined.Transcriptome analysis,cytokine analysis,flow cytometry analysis of the number and proportion of immune cell subsets in the liver and spleen,and molecular and cellular analyses of tumors were used to define the underlying mechanisms.Results:AAGL significantly inhibited the growth of liver tumors in a dose-dependent manner.Furthermore,AAGL increased the expression of multiple cytokines and chemokines in tumor-bearing mouse livers;this effect was associated with the activation and migration of T cells and macrophages,in agreement with the in vitro results.Importantly,the aggregation of T cells and macrophages induced by AAGL in tumor-bearing mouse livers clearly enhanced the response to PD-1 blockade immunotherapy.Conclusions:The results showed that AAGL induced the activation and migration of lymphocytes to the liver,and that the combination of AAGL and anti-PD-1 may be a promising strategy for HCC treatment. | Xiangdong Ye Xueqing Wang Wenhui Yu Qing Yang Yan Li Yanxia Jin Yanting Su Jiaqi Song Bo Xu Hui Sun | 2021 | Cancer Biology & Medicine2021,18,4: | 0 |
| 16 | Effects of illness perception on negative emotions and fatigue in chronic rheumatic diseases:Rumination as a possible mediator显示文摘BACKGROUND Illness perception has long been hypothesized to be linked to psychological well-b eing in patients with rheumatic diseases,although substantial evidence is lacking,and the contribution of ruminative coping style to this relationship is unclear.AIM To investigate the roles of illness perception and rumination in predicting fatigue and negative emotions in patients with chronic rheumatic diseases.METHODS Illness perception,rumination,fatigue and negative emotions(i.e.depression,anxiety and stress)were assessed by the Illness Perception Questionnaire-Revised,Stress Reactive Rumination Scale,Multidimensional Assessment of Fatigue,and the Depression,Anxiety and Stress Scale respectively.Multivariate regression analysis,the Sobel test,and the bootstrap were used to identify the mediating effect of rumination.RESULTS All five subscales of illness perception,including perceived illness identity,chronicity,cyclical nature,consequences and coherence of illness,were significantly associated with fatigue and negative emotions.In mediational analysis,rumination was found to mediate three components of illness perception(the identity,cyclical nature and consequences of illness)and negative emotions/fatigue.CONCLUSION Perceived identity,cyclical nature,and consequences of illness are significantly associated with fatigue and negative emotions in patients with chronic rheumatic diseases and these associations are mediated by rumination.Our findings suggest that psychological intervention should target rumination to improve physical and emotional well-being of patients with chronic rheumatic diseases. | Yanxia Lu Xia Jin Li-Wei Feng CSK Tang Michelle Neo Roger C Ho | 2022 | World Journal of Clinical Cases2022,10,34: | 0 |
| 17 | Closed-system pyrolysis-based hydrocarbon generation simulation and gas potential evaluation of the Shanxi Formation shales from the Ordos Basin, China显示文摘The Shanxi Formation(Shan 1 and Shan 2 Members)shales show good prospects in shale gas development in the Yan'an area of Ordos Basin.Based on the simulation experiment of hydrocarbon generation of low maturity shale samples,the hydrocarbon generation characteristics of shale samples was studied systematically.Then,combined with the geochemical analysis of shale and gas generation simulation,shale gas potential was evaluated.The results reveal that Shan 1 and Shan 2 shale samples are favorable for shale gas enrichment by and large,with C_(1)-C_(5) maximum yields of 146.96-160.83 mg/g TOC and 148.48-148.67 mg/g TOC respectively at a heat rate of 20℃/h and 2℃/h.The Shan 1 and Shan 2 shales are basically the same in terms of organic carbon production potential of each unit.The carbon isotopic composition of alkane gas reveals that heteroatomic compounds(NSOs)cracking is an important mechanism for shale gas generation of Shanxi Formation shales,and conducive to gas generation at highto over-mature stages.Given thermal history and kinetic parameters of hydrocarbon generation,the shales of Shanxi Formation reached the maximum gas production potential in the Late Cretaceous,with a maximum yield of 160.3 mg/g TOC under present geological conditions.During geological history,the Shanxi Formation shales went through high-to over-maturity evolution,mainly producing dry gas,and their gas generation capacity was controlled by the organic matter abundance and cracking capacity.The gas generation potential of Shan 2 shale is higher than that of Shan 1,due to its higher TOC. | Xiaobo Guo Baohong Shi Yu Li Yanxia Li Jianbo Sun Gang Liu Jintao Yin Hongzhu Wu Xi Jin | 2022 | Energy Geoscience2022,3,1: | 0 |
