A NAC transcription factor, NOR-like1, is a new positive regulator of tomato fruit ripening显示文摘Ripening of the model fruit tomato(Solanum lycopersicum)is controlled by a transcription factor network including NAC(NAM,ATAF1/2,and CUC2)domain proteins such as No-ripening(NOR),SlNAC1,and SlNAC4,but very little is known about the NAC targets or how they regulate ripening. Here,we conducted a systematic search of fruit-expressed NAC genes and showed that silencing NOR-like1(Solyc07g063420)using virus-induced gene silencing(VIGS)inhibited specific aspects of ripening.Ripening initiation was delayed by 14 days when NOR-like1 function was inactivated by CRISPR/Cas9 and fruits showed obviously reduced ethylene production, retarded softening and chlorophyll loss, and reduced lycopene accumulation.RNA-sequencing profiling and gene promoter analysis suggested that genes involved in ethylene biosynthesis(SlACS2,SlACS4),color formation(SlGgpps2,SlSGR1),and cell wall metabolism(SlPG2a,SlPL,SlCEL2,and SlEXP1)are direct targets of NOR-like1.Electrophoretic mobility shift assays(EMSA),chromatin immunoprecipitation-quantitative PCR(ChIP-qPCR),and dual-luciferase reporter assay(DLR)confirmed that NOR-like1 bound to the promoters of these genes both in vitro and in vivo, and activated their expression. Our findings demonstrate that NOR-like1 is a new positive regulator of tomato fruit ripening, with an important role in the transcriptional regulatory network. | Ying Gao Wei Wei Xiaodan Zhao Xiaoli Tan Zhongqi Fan Yiping Zhang Yuan Jing Lanhuan Meng Benzhong Zhu Hongliang Zhu Jianye Chen Cai-Zhong Jiang Donald Grierson Yunbo Luo Da-Qi Fu | 2018 | Horticulture Research2018,5,1: | 9
Secalonic acid D induces cell apoptosis in both sensitive and ABCG2-overexpressing multidrug resistant cancer cells through upregulating c-Jun expression显示文摘Secalonic acid D(SAD) could inhibit cell growth in not only sensitive cells but also multidrug resistant(MDR) cells. However, the molecular mechanisms need to be elucidated. Here, we identified that SAD possessed potent cytotoxicity in 3 pairs of MDR and their parental sensitive cells including S1-MI-80 and S1,H460/MX20 and H460, MCF-7/ADR and MCF-7 cells. Furthermore, SAD induced cell G2/M phase arrest via the downregulation of cyclin B1 and the increase of CDC2 phosphorylation. Importantly, JNK pathway upregulated the expression of c-Jun in protein level and increased c-Jun phosphorylation induced by SAD, which was linked to cell apoptosis via c-Jun/Src/STAT3 pathway. To investigate the mechanisms of upregulation of c-Jun protein by SAD, the mR NA expression level and degradation of c-Jun were examined. We found that SAD did not alter the mR NA level of c-Jun but inhibited its proteasome-dependent degradation. Taken together, these results implicate that SAD induces cancer cell death through c-Jun/Src/STAT3 signaling axis by inhibiting the proteasome-dependent degradation of c-Jun in both sensitive cells and ATP-binding cassette transporter sub-family G member 2(ABCG2)-mediated MDR cells. | Hong Zhang Liyan Huang Liyang Tao Jianye Zhang Fang Wang Xu Zhang Liwu Fu | 2019 | Acta Pharmaceutica Sinica B2019,9,3: | 5

Liquid metal hydraulics paradigm:Transmission medium and actuation of bimodal signals显示文摘In this article, room temperature gallium-based fluids(RTGFs)with unique thermal and conductive properties are proposed as a transmission fluid for the force carrying medium of hydraulics, which rectify the lack of the multi-functional hydraulic roles of liquid metal(LM).The typical physical properties of RTGFs and comparative conventional hydraulic fluids(commercial hydraulic oil and deionized water),such as thermal stability and rheological characteristics, are evaluated.Experimental and numerical methods, then, were adopted to clarify the force transmission performance of RTGFs and commercial hydraulic oils,as well as the influence of temperature fields on the viscosity of fluids. The results disclosed that the advantages of inherent flame resistance, wide liquid temperature range, and the viscosity changing slightly with temperature made RTGFs potential in finding efficient application in the hydraulics as new working fluids. Finally, for illustration, the rigid and flexible actuators driven by RTGFs were designed as hydraulic fluid and demonstrated their capabilities in grasping objects with various shapes and weights, respectively. And the tunable stiffness of such a flexible actuator was enabled via the solid-liquid phase change of LM.Additionally,a frequency-adjustable antenna was manufactured and showcased owing to the introduced transformable electromagnetic behaviors of LM.Overall, the gallium-based LM fluids with thermoelectrical and mechanical multimodal signal medium would serve as a potential candidate for future complex multifunctional signal transmission systems. | FU JunHeng GAO JianYe QN Peng LI DongDong YU DeHai SUN Peng HE ZhiZhu DENG ZhongShan LIU Jing | 2022 | Science China(Technological Sciences)2022,65,1: | 1
Dynamic phosphorylation of CENP-N by CDK1 guides accurate chromosome segregation in mitosis显示文摘In mitosis, accurate chromosome segregation depends on the kinetochore,a supermolecular machinery that couples dynamic spin-dle microtubules to centromeric chromatin. However, the structure–activity relationship of the constitutive centromere-associated network(CCAN)during mitosis remains uncharacterized.Building on our recent cryo-electron microscopic analyses of human CCAN structure,we investigated how dynamic phosphorylation of human CENP-N regulates accurate chromosome segregation. Our mass spectrometric analyses revealed mitotic phosphorylation of CENP-N by CDK1,which modulates the CENP-L–CENP-N interaction for accurate chromosome segregation and CCAN organization.Perturbation of CENP-N phosphorylation is shown to prevent proper chromosome alignment and activate the spindle assembly checkpoint. These analyses provide mechanistic insight into a previously undefined link between the centromere–kinetochore network and accurate chromosome segregation. | Ran Liu Zhen Dou Tian Tian Xinjiao Gao Lili Chen Xiao Yuan Chunyue Wang Jiahe Hao Ping Gui McKay Mullen Felix Aikhionbare Liwen Niu Guoqiang Bi Peng Zou Xuan Zhang Chuanhai Fu Xuebiao Yao Jianye Zang Xing Liu | 2023 | Journal of Molecular Cell Biology2023,15,6: | 0

Silica Nanoparticles with Virus-Mimetic Spikes Enable Efficient siRNA Delivery In Vitro and In Vivo显示文摘Oligonucleotide-based therapy has experienced remarkable development in the past 2 decades, but its broad applications are severely hampered by delivery vectors.Widely used viral vectors and lipid nanovectors are suffering from immune clearance after repeating usage or requiring refrigerated transportation and storage, respectively. In this work,amino-modified virus-mimetic spike silica nanoparticles(NH_(2)-SSNs)were fabricated using a 1-pot surfactant-free approach with controlled spike lengths, which were demonstrated with excellent delivery performance and biosafety in nearly all cell types and mice.It indicated that NH2-SSNs entered cells by spike-dependent cell membrane docking and dynamin-dependent endocytosis. The positively charged spikes with proper length on the surface can facilitate the efficient encapsulation of RNAs,protect the loaded RNAs from degradation, and trigger an early endosome escape during intracellular trafficking, similarly to the cellular internalization mechanism of virions.Regarding the fantastic properties of NH_(2)-SSNs in nucleic acid delivery,it revealed that nanoparticles with solid spikes on the surface would be excellent vehicles for gene therapy, presenting self-evident advantages in storage, transportation, modification, and quality control in large-scale production compared to lipid nanovectors. | Jianye Fu Wenwei Han Xue Zhang Yutong Sun Rajendra Bhadane Bo Wei Li Li Liangmin Yu Jinbo Yang Jessica MRosenholm Outi MHSalo-Ahen Taojian Fan Bin Zhang Wageh Swelm Ahmed AAl-Ghamdi Lin Xia Han Zhang Meng Qiu Hongbo Zhang Xin Wang | 2023 | Research2023,,3: | 0
