13Increased CD4/CD8 Lymphocyte ratio predicts favourable neoadjuvant treatment response in gastric cancer:A prospective pilot study显示文摘BACKGROUND Despite optimal neoadjuvant chemotherapy only 40%of gastric cancer tumours achieve complete or partial treatment response.In the absence of treatment response,neoadjuvant chemotherapy in gastric cancer contributes to adverse events without additional survival benefit compared to adjuvant treatment or surgery alone.Additional strategies and methods are required to optimize the allocation of existing treatment regimens such as FLOT chemotherapy(5-Fluorouracil,Leucovorin,Oxaliplatin and Docetaxel).Predictive biomarkers detected using immunohistochemistry(IHC)methods may provide useful data regarding treatment response.AIM To investigate the utility of CD4,CD8,Galectin-3 and E-cadherin in predicting neoadjuvant FLOT chemotherapy tumour response in gastric adenocarcinoma.METHODS Forty-three adult patients with gastric adenocarcinoma,of which 18 underwent neoadjuvant chemotherapy,were included in a prospective clinical cohort.Endoscopic biopsies were obtained from gastric cancer and normal adjacent gastric mucosa.Differences in expression of Galectin-3,Ecadherin,CD4^(+)and CD8^(+)molecules between tumours with and without treatment response to neoadjuvant chemotherapy were assessed with IHC.Treatment response was graded by clinical pathologists using the Tumour Regression Score according to the College of American Pathologists criteria.Treatment response was defined as complete or near complete tumour response,whereas partial or poor/no response was defined as incomplete.Digital IHC images were annotated and quantitatively assessed using QuPath 0.3.1.Biomarker expression between responsive and incomplete response tumours was assessed using a two-sided Wilcoxon test.Biomarker expression was also compared between normal and cancer tissue and between 15 paired tumour samples before and after chemotherapy.We performed a preliminary multivariate analysis and power analysis to guide future study.Statistical analyses were completed using R 4.1.2.RESULTS The ratio between CD4^(+)and CD8^(+)lymphocytes was significantly greater in treatment responsive tumours(Wilcoxon,P=0.03).In univariate models,CD4^(+)/CD8^(+)ratio was the only biomarker that significantly predicted favourable treatment response(Accuracy 86%,P<0.001).Using a glmnet multivariate model,high CD4^(+)/CD8^(+)ratio and low Galectin-3 expression were the most influential variables in predicting a favourable treatment response.Analyses of paired samples found that FLOT chemotherapy also results in increased expression of CD4^(+)and CD8^(+)tumour infiltrating lymphocytes(Paired Wilcoxon,P=0.002 and P=0.008,respectively).Our power analysis suggests future study requires at least 35 patients in each treatment response group for CD8 and Galectin-3 molecules,whereas 80 patients in each treatment response group are required to assess CD4 and E-cadherin biomarkers.CONCLUSION We demonstrate that an elevated CD4^(+)/CD8^(+)Ratio is a promising IHC-based biomarker to predict favourable treatment response to FLOT neoadjuvant chemotherapy in locally advanced gastric cancer.Daniel Skubleny Andrea Lin Saurabh Garg Ross McLean Michael McCall Sunita Ghosh Jennifer L Spratlin Daniel Schiller Gina Rayat 2023World Journal of Gastrointestinal Oncology2023,15,2:1

16Comparative performances of new and existing indices of crown asymmetry:an evaluation using tall trees of Eucalyptus pilularis(Smith)显示文摘Over the past 50 years,crown asymmetry of forest trees has been evaluated through several indices constructed from the perspective of projected crown shape or displacement but often on an ad hoc basis to address specifi c objectives related to tree growth and competition,stand dynamics,stem form,crown structure and treefall risks.Although sharing some similarities,these indices are largely incoherent and non-comparable as they diff er not only in the scale but also in the direction of their values in indicating the degree of crown asymmetry.As the fi rst attempt at devising normative measures of crown asymmetry,we adopted a relative scale between 0 for perfect symmetry and 1 for extreme asymmetry.Five existing crown asymmetry indices(CAIs)were brought onto this relative scale after necessary modifi cations.Eight new CAIs were adapted from measures of circularity for digital images in computer graphics,indices of income inequality in economics,and a bilateral symmetry indicator in plant leaf morphology.The performances of the 13 CAIs were compared over diff erent numbers of measured crown radii for 30 projected crowns of mature Eucalyptus pilularis trees through benchmarking statistics and rank order correlation analysis.For each CAI,the index value based on the full measurement of 36 evenly spaced radii of a projected crown was taken as the true value in the benchmarking process.The index(CAI 13)adapted from the simple bilateral symmetry measure proved to be the least biased and most precise.Its performance was closely followed by that of three other CAIs.The minimum number of crown radii that is needed to provide at least an indicative measure of crown asymmetry is four.For more accurate and consistent measures,at least 6 or 8 crown radii are needed.The range of variability in crown morphology of the trees under investigation also needs to be taken into consideration.Although the CAIs are from projected crown radii,they can be readily extended to individual tree crown metrics that are now commonly extracted from LiDAR and other remotely sensed data.Adding a normative measure of crown asymmetry to individual tree crown metrics will facilitate the process of big data analytics and artifi cial intelligence in forestry wherever crown morphology is among the factors to be considered for decision making in forest management.Fanlin Kong Huiquan Bi Michael McLean Fengri Li 2021Journal of Forestry Research2021,32,1:1

18Adaptable physics-based super-resolution for electron backscatter diffraction maps显示文摘In computer vision,single-image super-resolution(SISR)has been extensively explored using convolutional neural networks(CNNs)on optical images,but images outside this domain,such as those from scientific experiments,are not well investigated.Experimental data is often gathered using non-optical methods,which alters the metrics for image quality.One such example is electron backscatter diffraction(EBSD),a materials characterization technique that maps crystal arrangement in solid materials,which provides insight into processing,structure,and property relationships.We present a broadly adaptable approach for applying state-of-art SISR networks to generate super-resolved EBSD orientation maps.This approach includes quaternion-based orientation recognition,loss functions that consider rotational effects and crystallographic symmetry,and an inference pipeline to convert network output into established visualization formats for EBSD maps.The ability to generate physically accurate,high-resolution EBSD maps with super-resolution enables high-throughput characterization and broadens the capture capabilities for three-dimensional experimental EBSD datasets.Devendra K.Jangid Neal R.Brodnik Michael G.Goebel Amil Khan SaiSidharth Majeti McLean P.Echlin Samantha H.Daly Tresa M.Pollock B.S.Manjunath 2022npj Computational Materials2022,,1:0
