3Population Genomic Analysis and De Novo Assembly Reveal the Origin of Weedy Rice as an Evolutionary Game显示文摘Crop weediness,especially that of weedy rice(Oryza sativa f.spontanea),remains mysterious.Weedy rice possesses robust ecological adaptability;however,how this strain originated and gradually formed proprietary genetic features remains unclear?Here,we demonstrate that weedy rice at Asian high latitudes(WRAH)is phylogenetically well defined and possesses unselected genomic characteristics in many divergence regions between weedy and cultivated rice.We also identified novel quantitative trait loci underlying weedy-specific traits,and revealed that a genome block on the end of chromosome 1 is associated with rice weediness.To identify the genomic modifications underlying weedy rice evolution,we generated the first de novo assembly of a high-quality weedy rice genome(WR04-6),and conducted a comparative genomics study between WR04-6 with other rice reference genomes.Multiple lines of evidence,including the results of demographic scenario comparisons,suggest that differentiation between weedy rice and cultivated rice was initiated by genetic improvement of cultivated rice and that the essence of weediness arose through semi-domestication.A plant height model further implied that the origin of WRAH can be modeled as an evolutionary game and indicated that strategy-based selection driven by fitness shaped its genomic diversity.Jian Sun Dianrong Ma Liang Tang Minghui Zhao Guangchen Zhangh Wenjia Wang Jiayu Song Xiang Li Zimeng Liu Wenxing Zhang Quan Xu Yuncheng Zhou Jianzhong Wu Toshio Yamamoto Fei Dai Yan Lei Song Li Gang Zhou Hongkun Zheng Zhengjin Xu Wenfu Chen 2019Molecular Plant2019,12,5:9
4Tailoring the mechanical anisotropy of sintered NdFeB magnets by Pr-Cu grain boundary reconstruction显示文摘Pr_(83)Cu_(17)(wt%) grain boundary reconstruction was applied to prepare sintered NdFeB magnets. The effects of addition amount and annealing on the bending strength were investigated. The results show that Pr_(83)Cu_(17) can not only effectively enhance the bending strength, but also change the mechanical anisotropy in two directions parallel and perpendicular to c-axis. The bending strength perpendicular to c-axis reaches 404 MPa in 10 wt% addition magnet, higher than 348 MPa along parallel direction. This change is attributed to the preferred distribution of boundary phases, i.e., ductile(Nd,Pr)-Fe-Cu phase along perpendicular direction to c-axis and(Nd,Pr)-Fe phase along parallel direction. 5Sequence framework of two different kinds of margins and their response to tectonic activity during the Middle-Late Triassic, Ordos Basin显示文摘Two kinds of margin respectively occur in the Ordos Basin during the Middle-Late Triassic (Yanchang Age), one is foreland margin developed under the background of flexural subsidence by thrusting intensively in the southwest margin, and the other is intracratonic basin margin by stable subsidence in northern and central parts of the basin. The Middle-Late Triassic Yanchang Formation can be divided into four regional third-order sequences, which are separated by gentle angular unconformity or regional erosion surface, made up of lowstand system tract (LST), expanding system tract (EST) and highstand system tract (HST) from lower to upper within a sequence. But there are distinct differences of the sequence framework between the southwest margin and northern and central parts of the basin. The southwest margin develops heavy conglomerate layer and unconformity as a result of orogeny by thrusting, and the intracratonic basin margin by stable subsidence in the northern and central parts grows aggradational sandstone, conglomerate in fluvio-delta system and parallel unconformity. The depositional framework of southwest margin reflects the tectonic evolution from flexural subsidence by thrusting to rebounded uplift. The formation of sequence boundary is related to the resilient uplift and erosion. The sequence stratigraphic framework and depositional system tract configuration in the foreland basin are controlled by structural activity of the fold and thrust belt, and the sequence succession reflects episodic thrusting of the Middle-Late Triassic toward the foreland basin. The sequence evolution in northern and central parts reflects the depositional succession of fluvio-delta system under intracratonic background, composed of coarse-grained sediment in braided channel deposit at the lower, meandering channel deposit in the middle and fine-grained sediment in the flood plain at the upper, dominated by lake level fluctuation. 6Structural insights into a high fidelity variant of SpCas9显示文摘The RNA-guided endonucleases of the CRISPR-Cas9 system, including the most w泊ely used Cas9 from Streptococcus pyogenes (SpCas9)z are becoming a robust genome editing tool in model organisms and hold immense promise for therapeutic applications. Many strategies have bee n employed to overcome the limitati ons caused by SpCas9zs off-target effects and its strin gent requirement for the protospacer adjacent motif (PAM) sequenee. However, the structural mechanisms underlying these strategies remain undefined. Here, we present crystal structure of a SpCas9 variant, xCas9 3.7 that has broad PAM compatibility and high DNA targeting specificity, in complex with a single-guide RNA and its double-stranded DNA targets. Structural comparison revealed that salt bridge-stabilized R1335 is critical for the stringent selection of PAM sequence by SpCas9. Unrestricted rotamerization of this residue by the E1219V mutation in xCas9 3.7 lessens the stringency for PAM recognition and allows SpCas9 to recognize multiple PAM sequences as further supported by biochemical data. Compared to those in wild-type (WT) SpCas9, REC2 and REC3 domains in xCas9 3.7 un dergo striki ng conformati onal changes, leadi ng to reduced con tact with DNA substrate. SpCas9 mutants en gineered to display less interaction with DNA and have conformationally more flexible REC2 and REC3 domains display enhanced specificity for DNA substrates in both biochemical and cellular assays. 7into the Role of Additives in Catalytic Synthesis of Insight Cyclohexylamine from Nitrobenzene显示文摘Cyclohexylamine is a versatile intermediate in various chemical industries,which can be expediently synthesized via hydrogenation of aniline or nitrobenzene.However,such processes always suffer from side reactions.Many reports have found that specific additives can suppress the formation of side products,but the mechanism is still not clear.In this work,results suggest that it is the hydroxide ion of alkali metal hydroxides,rather than the cations,which plays a key role in suppressing the side reactions on supported Ru-based catalysts.With the assistance of LiOH,the selectivity toward cy- clohexylamine increased from 85.4% to 100%.Side products,such as dicyciohexylamine,cyclohexanol and N-isopropyl cyclohexylamine,could no longer be detected.Theoretical calculations further disclosed that addition of alkali metal hydroxides inhibited the dissociation of enamine and decreased the adsorption energy of cyclohexylamine,which might be the reasons for a better selectivity.However,the addition of alkali metal hydroxides reduced the activity of nitrobenzene hydrogenation by unfolding the condensation reaction route.To recover or even further enhance the catalytic performance,a second metal component was introduced and the resultant RuNi/AC exhibited a significant improvement in activity compared with Ru/AC.Xuefeng Li Zhe Wang Shanjun Mao Yiqing chen Minghui Tang Haoran Li Yong Wang 2018Chinese Journal of Chemistry2018,36,12:4
8Induced Pluripotent Stem Cell-derived Mesenchymal Stem Cell Seeding on Biofunctionalized Calcium Phosphate Cements显示文摘Induced pluripotent stem cells(iPSCs)have great potential due to their proliferation and differentiation capability.The objectives of this study were to generate iPSC-derived mesenchymal stem cells(iPSC-MSCs),and investigate iPSC-MSC proliferation and osteogenic differentiation on calcium phosphate cement(CPC)containing biofunctional agents for the first time.Human iPSCs were derived from marrow CD34+cells which were reprogrammed by a single episomal vector.iPSCs were cultured to form embryoid bodies(EBs),and MSCs migrated out of EBs.Five biofunctional agents were incorporated into CPC:RGD(Arg-Gly-Asp)peptides,fibronectin(Fn),fibronectin-like engineered polymer protein(FEPP),extracellular matrix Geltrex,and platelet concentrate.iPSC-MSCs were seeded on five biofunctionalized CPCs:CPC-RGD,CPC-Fn,CPC-FEPP,CPC-Geltrex,and CPC-Platelets.iPSC-MSCs on biofunctional CPCs had enhanced proliferation,actin fiber expression,osteogenic differentiation and mineralization,compared to control.Cell proliferation was greatly increased on biofunctional CPCs.iPSC-MSCs underwent osteogenic differentiation with increased alkaline phosphatase,Runx2 and collagen-I expressions.Mineral synthesis by iPSC-MSCs on CPC-Platelets was 3-fold that of CPC control.In conclusion,iPSCs showed high potential for bone engineering.iPSC-MSCs on biofunctionalized CPCs had cell proliferation and bone mineralization that were much better than traditional CPC.iPSC-MSC-CPC constructs are promising to promote bone regeneration in craniofacial/orthopedic repairs.WahWah TheinHan Jun Liu Minghui Tang Wenchuan Chen Linzhao Cheng Hockin H.K.Xu 2013Bone Research2013,1,4:3

11A Variational Binary Level Set Method for Structural Topology Optimization显示文摘This paper proposes a variational binary level set method for shape and topology optimization of structural.First,a topology optimization problem is presented based on the level set method and an algorithm based on binary level set method is proposed to solve such problem.Considering the difficulties of coordination between the various parameters and efficient implementation of the proposed method,we present a fast algorithm by reducing several parameters to only one parameter,which would substantially reduce the complexity of computation and make it easily and quickly to get the optimal solution.The algorithm we constructed does not need to re-initialize and can produce many new holes automatically.Furthermore,the fast algorithm allows us to avoid the update of Lagrange multiplier and easily deal with constraints,such as piecewise constant,volume and length of the interfaces.Finally,we show several optimum design examples to confirm the validity and efficiency of our method.Xiaoxia Dai Peipei Tang Xiaoliang Cheng Minghui Wu 2013Communications in Computational Physics2013,13,5:1
12Ferroelectricity in dopant-free HfO_(2) thin films prepared by pulsed laser deposition显示文摘As a high-k material,hafnium oxide(HfO_(2))has been used in gate dielectrics for decades.Since the discovery of polar phase in Si-doped HfO_(2) films,chemical doping has been widely demonstrated as an effective approach to stabilize the ferroelectric phase in HfO_(2) based thin films.However,the extra capping layer deposition,post-growth annealing and wake-up effect are usually required to arouse the ferroelectricity in HfO_(2) based thin films,resulting in the increase of complexity for sample synthesis and the impediment of device application.In this study,the ferroelectricity is observed in non-capped dopant-free HfO_(2) thin films prepared by pulsed laser deposition(PLD)without post-growth annealing.By adjusting the deposited temperature,oxygen pressure and thickness,the maximum polarization up to 14.7 m C/cm^(2) was obtained in 7.4 nm-thick film.The fraction of orthorhombic phase,concentrations of defects and size effects are considered as possible mechanisms for the influences of ferroelectric prop-erties.This study indicates that PLD is an effective technique to fabricate high-quality ferroelectric HfO_(2) thin films in the absence of chemical doping,capping layer deposition and post-growth annealing,which may boost the process of nonvolatile memory device application.Yongjian Luo Zhenxun Tang Xiaozhe Yin Chao Chen Zhen Fan Minghui Qin Min Zeng Guofu Zhou Xingsen Gao Xubing Lu Jiyan Dai Deyang Chen Jun-Ming Liu 2022Journal of Materiomics2022,8,2:1

14Crystallization Behavior of Nd_2O_3 Doped Na_2O–CaO–SiO_2 Laser Glass-ceramics显示文摘For the desirable laser optical property, transition metals or rare-earths are always doped into parent glasses as active ions, and this doping will affect the crystallization of the precursor glasses inevitably.In this work, crystallization behavior of Na O–Ca O–Si O2 system glasses doped with Nd2O3 was investigated. The crystallization kinetic parameters including the crystallization apparent activation energy(E)and the Avrami parameter(n) were also measured. The results show that the Na O–Ca O–Si O2 system glassceramics with the Na2Ca2Si3O9 crystal as primary phase can be highly crystalized as above 90%. 16Rhesus monkeys exhibiting spontaneous ritualistic behaviors resembling obsessive-compulsive disorder显示文摘Obsessive-compulsive disorder(OCD)is a chronic and debilitating psychiatric disorder that affects∼2%–3%of the population globally.Studying spontaneous OCD-like behaviors in non-human primates may improve our understanding of the disorder.In large rhesus monkey colonies,we found 10 monkeys spontaneously exhibiting persistent sequential motor behaviors(SMBs)in individual-specific sequences that were repetitive,time-consuming and stable over prolonged periods.Genetic analysis revealed severely damaging mutations in genes associated with OCD risk in humans.Brain imaging showed that monkeys with SMBs had larger gray matter(GM)volumes in the left caudate nucleus and lower fractional anisotropy of the corpus callosum.The GM volume of the left caudate nucleus correlated positively with the daily duration of SMBs.Notably,exposure to a stressor(human presence)significantly increased SMBs.In addition,fluoxetine,a serotonergic medication commonly used for OCD,decreased SMBs in these monkeys.These findings provide a novel foundation for developing better understanding and treatment of OCD.Rongwei Zhai Geya Tong Zheqin Li Weichen Song Yang Hu Sha Xu Qiqi Wei Xiaocheng Zhang Yi Li Bingbing Liao Chenyu Yuan Yinqing Fan Ge Song Yinyin Ouyang Wenxuan Zhang Yaqiu Tang Minghui Jin Yuxian Zhang He Li Zhi Yang Guan Ning Lin Dan JStein Zhi-Qi Xiong Zhen Wang 2023National Science Review2023,10,11:1

20Transcriptome analysis suggests mechanisms for a novel flowering type: Cleistogamous wheat显示文摘Wheat is one of the most important staple crops worldwide.Fusarium head blight severely reduces wheat yield and quality.Cultivation of a novel type of cleistogamous wheat mutant,ZK001,which was created by static magnetic field treatment,is a new strategy for controlling Fusarium head blight.However,little is known about the mechanism of cleistogamy in wheat.The present study demonstrated that anthers of ZK001 were retained on the glumes at all flowering stages,whereas those of YM18 were extruded from the paleae and lemmae.There was a clear difference in the morphological characteristics of lodicules between YM18 and ZK001.Lodicule calcium and potassium contents were significantly higher in YM18 than in ZK001 from white to yellow anther stages.In Fusarium head blight resistance,the diseased kernel rate and deoxynivalenol content of ZK001 were markedly lower than those of YM18 and QM725.Comparative transcriptome analysis of YM18 and ZK001 was performed to identify regulatory mechanisms of cleistogamy.The main differentially expressed genes identified in the spikelets of YM18 and ZK001 at the green anther stage were associated with cell walls,carbohydrates,phytohormones,water channel,and ion binding,transport,and homeostasis.These differentially expressed genes may play an important role in regulating cellular homeostasis,osmotic pressure,and lodicule development.The results indicate that ZK001 lost the ability to push the lemmae and paleae apart during the flowering stage because of the thin lodicules.ZK001 was speculated to provide structural barriers for Fusarium head blight during the flowering stage.The thin lodicule of ZK001 results from low levels of soluble sugar,calcium ions,and potassium ions in the lodicules.These levels are regulated by differentially expressed genes.Caiguo Tang Minghao Li Minghui Cao Ruiju Lu Huilan Zhang Chenghong Liu Shengwei Huang Pingping Zhang Hao Hu Weiwei Zhao Lifang Wu 2020The Crop Journal2020,8,2:1
