2Ultrasound features of extranodal extension in the metastatic cervical lymph nodes of papillary thyroid cancer:a case-control study显示文摘Objective:Extranodal extension in cervical lymph nodes is an important risk factor for the progression and prognosis of papillary thyroid cancer.The purpose of this study was to identify the common and characteristic preoperative ultrasonography features that are associated with the pathologic extranodal extension of metastatic papillary thyroid carcinoma.Methods:We retrospectively assessed and compared clinicopathologic and ultrasound features between 60 papillary thyroid cancer patients with extranodal extension and 120 control patients with papillary thyroid cancer without extranodal extension.Results:With respect to the pathological N stage and clinicopathologic features,N1b stage papillary thyroid carcinomas were more frequently found in patients who were extranodal extension-positive,in comparison with those who were extranodal extension-negative(78.3%vs.63.3%,P=0.043).Extranodal extension was detected most frequently in level VI cervical lymph nodes(48.7%).In our univariate analysis of patients with papillary thyroid carcinoma,cervical lymph nodes with extranodal extension showed higher incidences of node matting,microcalcification,cystic area,aspect ratio<2,and larger diameter than those without extranodal extension(all P<0.05).Our multivariate analysis demonstrated that node matting and cystic area were independent risk factors for the presence of extranodal extension[odds ratio(OR):4.751,95%confidence interval(CI):1.212~18.626,P=0.025;OR:2.707,95%CI:1.127~6.502,P=0.026].Conclusions:Common ultrasound features may indicate the presence of extranodal extension in patients with metastatic cervical lymph nodes of papillary thyroid carcinoma.Jiali Mu Xiaofeng Liang Fangxuan Li Juntian Liu Sheng Zhang Jing Tian 2018Cancer Biology & Medicine2018,15,2:7
3Vertical motions of tide gauge stations near the Bohai Sea and Yellow Sea显示文摘A modified Gauss-Markov model with weighted constraints was constructed by combining satellite altimeter and tide gauge records. Vertical motion rates of nine tide gauge stations around the Bohai Sea and Yellow Sea are estimated. This is the first time systematic estimates have been derived in this region. Downward trends were seen at the six tide gauge stations located at Tanggu, Longkou, Laohutan, Bayuquan, Xiaochangshan, and Yantai; with vertical motion rates of-1.82±0.50,-1.65±0.46,-0.88±0.42,-0.58±0.62,-0.13±0.43, and-0.01±0.43 mm/yr, respectively. Upward trends were seen at the three tide gauge stations located at Qinhuangdao, Huludao and Chengshantou; with vertical motion rates of 1.12±0.46, 0.55±0.49 and 0.26±0.44 mm/yr, respectively. There was significant subsidence in Tanggu and Longkou, and a rising trend in Qinhuangdao. According to our results, the rate of sea level rise calculated from these tide gauge records can be improved using a more accurate measurement of the land elevation accounting for lifting or subsidence. 4Glucose metabolism controls humanγδ-cell-mediated tumor immunosurveillance in diabetes显示文摘Patients with type 2 diabetes mellitus(T2DM)have an increased risk of cancer.The effect of glucose metabolism onγδT cells and their impact on tumor surveillance remain unknown.Here,we showed that high glucose induced Warburg effect type of bioenergetic profle in Vy9vδ2 T cells,leading to excessive lactate accumulation,which further inhibited lytic granule secretion by impairing the traffcking of cytolytic machinery to the Vy9vδ2 T-cell-tumor synapse by suppressing AMPK activation and resulted in the loss of antitumor activity in vitro,in vivo and in patients.Strikingly,activating the AMPK pathway through glucose control or metformin treatment reversed the metabolic abnormalities and restored the antitumor activity of Vy9vδ2 T cells.These results suggest that the impaired antitumor activity of Vy9vδ2 T cells induced by dysregulated glucose metabolism may contribute to the increased cancer risk in T2DM patients and that metabolic reprogramming by targeting the AMPK pathway with metformin may improve tumor immunosurveillance.Xiaofeng Mu Zheng Xiang Yan Xu Jing He Jianwen Lu Yuyuan Chen Xiwei Wang Chloe Ran Tu Yanme Wenyue Zhang Zhinan Yin Wing-hang Leung Yu-Lung Lau Yinping Liu Wenwei Tu 2022Cellular & Molecular Immunology2022,19,8:2
5Inhibitory effect of a new gossypol derivative apogossypolone (ApoG2) on xenograft of human prostate cancer cell line PC-3显示文摘Objective: To investigate the inhibitory effect of apogossypolone (ApoG2) on prostate cancer cell line PC-3 in vivo, and explore its mechanism. Methods: The models of transplantation tumors in Balb/c nu/nu mice were established via subcutaneous injection of PC-3 cells and the tumor-transplanted mice were divided into 4 groups: control group and three ApoG2 treatment groups, with 10 mice in each group. Volumes of the tumor were estimated every 2 d and the morphology of tumor tissues was observed. Immunohistochemistry was employed to observe the expression of Bcl-2, PCNA, CD31, caspase-3 and caspase-8 in tumor tissues. Results: ApoG2 (2.5 mg/kg-10 mg/kg) given intraperitoneally once a day can obviously inhibit the growth of subcutaneous prostatic carcinoma implant. The tumor volume decreased obviously when the treatment dosage was bigger than 5.0 mg/kg (P<0.01). Meanwhile, ApoG2 decreased the expression of PCNA and CD31, and enhanced the expression of caspases-3, caspase-8 in tumor tissues. 6CD137 costimulation enhances the antiviral activity of Vγ9Vδ2-T cells against influenza virus显示文摘Influenza epidemics and pandemics are constant threats to global public health.Although strategies including vaccines and antiviral drugs have achieved great advances in controlling influenza virus infection,the efficacy of these strategies is limited by the highly frequent mutations in the viral genome and the emergence of drug-resistant strains.Our previous study indicated that boosting the immunity of human Vγ9Vδ2-T cells with the phosphoantigen pamidronate could be a therapeutic strategy to treat seasonal and avian influenza virus infections.However,one notable drawback ofγδ-T cell-based immunotherapy is the rapid exhaustion of proliferation and effector responses due to repeated treatments with phosphoantigens.Here,we found that the expression of CD137 was inducible in Vγ9Vδ2-T cells following antigenic stimulation.CD137^(+)Vγ9Vδ2-T cells displayed more potent antiviral activity against influenza virus than their CD137−counterparts in vitro and in Rag2^(-/-)γc^(-/-)mice.We further demonstrated that CD137 costimulation was essential for Vγ9Vδ2-T cell activation,proliferation,survival and effector functions.In humanized mice reconstituted with human peripheral blood mononuclear cells,CD137 costimulation with a recombinant human CD137L protein boosted the therapeutic effects of pamidronate against influenza virus.Our study provides a novel strategy of targeting CD137 to improve the efficacy of Vγ9Vδ2-T cell-based immunotherapy.Yujun Pei Kun Wen Zheng Xiang Chunyu Huang Xiwei Wang Xiaofeng Mu Liyan Wen Yinping Liu Wenwei Tu 2020Signal Transduction and Targeted Therapy2020,5,1:1

16Identification of a TGF-β1 homologue in the large yellow croaker(Larimichthys crocea)revealed its role in regulation of immune response显示文摘Mammalian TGF-β1 plays a crucial role in maintaining immune tolerance and inhibiting autoimmunity.Although several TGF-β1 homologues have been identified in teleosts,their immunosuppressive functions remain unclear.Herein,we identified and characterized LcTGF-β1,a TGF-β1 homologue from the large yellow croaker(Larimichthys crocea).LcTGF-β1 possesses several features characteristic of the TGF-βsuperfamily,including an RGD integrin binding site,an RSKR tetrabasic cut site,and nine conserved cysteines in the mature peptide.The LcTGFβ1 gene was constitutively expressed in all examined tissues,with the highest expression in the spleen,which is rich in immune cells.LcTGF-β1 was significantly upregulated in the head kidney and spleen after Vibrio alginolyticus stimulation.LcTGF-β1 was also expressed in primary head kidney lymphocytes(PKLs),monocytes/macrophages(MO/Mφs),and granulocytes(PKGs),and was significantly upregulated in the MO/Mφs after lipopolysaccharide(LPS)stimulation.Recombinant LcTGF-β1 protein(rLcTGF-β1)significantly suppressed reactive oxygen species(ROS)and nitrogen oxide(NO)production in LPS-activated head kidney MO/Mφs.Furthermore,in these MO/Mφs,rLcTGF-β1 strongly inhibited the expression of pro-inflammatory cytokine(TNFα)and inducible NO synthase(iNOS),while it induced anti-inflammatory cytokines(TGF-β1 and IL-10),arginase1,and CD206.The phagocytic activity of LPS-activated MO/Mφs was also increased by rLcTGF-β1.Finally,rLcTGF-β1 significantly inhibited the proliferation of T cells(ZAP70þcells)and downregulated T-cell activation genes(CD3ζ,TCRα,and ZAP70),T-cell marker genes(CD4 and CD8),and IL-2 in PKLs.These results thus provide strong evidence that LcTGF-β1 promotes the alternative activation of head kidney MO/Mφs and inhibits T-cell proliferation in PKLs,highlighting its role in the regulation of immune response in large yellow croaker.Xiaofeng Li Pengfei Mu Yan Teng Xiaomeng Li Yinanzi Wu Xinhua Chen 2022Water Biology and Security2022,1,1:0

18Development of a high-power blue diode laser for precision vaporization of prostate tissue for the surgical management of benign prostatic hyperplasia显示文摘This paper explores the advancement and application of high-power blue diode lasers in treating benign prostatic hyperplasia(BPH).Addressing the challenges posed by existing techniques,the study focuses on optimizing tissue removal methods.Energy platform for BPH should balance a range of factors,such as operative time,patient conditions,urinary functions,complications,durability,accessibility,and cost,all while prioritizing patient care.Blueray Medical's innovation of high-power blue diode laser systems for BPH surgery is explored,with emphasis on achieving a balance among these considerations.By illustrating the biomedical effects of lasers and their interaction with soft tissues,particularly emphasizing the role of photon absorption by biomolecules and proteins in tissue behavior,this study outlines the advantages of the highpower blue diode laser system.The initial laboratory experiments and clinical results consistently align with our theoretical predictions,especially in terms of tissue vaporization efficiency,tissue coagulation,and bleeding control.In conclusion,blue diode lasers hold potential to enhance surgical outcomes for BPH.Their unique properties offer benefits like improved tissue removal rate and reduced thermal damages.Integrating blue laser technology into BPH protocols could lead to shorter hospital stays,cost savings,and expanded patient eligibility,although rigorous clinical studies are needed to fully understand their benefits and limitations.Liyue Mu Xiang Zhu Bing Yang Lin Yang Xiaofeng Xu Yongwei Zhao Dalin He 2023UroPrecision2023,1,3:0
19Apogossypolone对人前列腺癌LNCaP移植瘤的生长抑制作用(英文)显示文摘Objective:The aim of this study was to investigate the inhibitory effect of apogossypolone (ApoG2) on subcutaneous implants of human LNCaP prostatic carcinoma cells, and explore its mechanism. Methods:To establish human LNCaP prostatic carcinoma cell line subcutaneous xenograft models and observe the inhibitory effect of ApoG2 on the tumor model. Immunohistochemistry was employed to observe the expression of Bcl-2, PCNA, CD31, caspase-3 and-8 in tumor tissues. The microvessel density was calculated. Results:ApoG2 could obviously inhibit the growth of subcutaneous prostatic carcinoma implant. ApoG2 decreased the expression of PCNA and CD31, and increased the expression of caspases-3,-8 in tumor tissues.
