The pathogenicity on legumes of Cucumber mosaic virus was determined by 243 nucleotides on 2a polymerase gene of viral RNA2显示文摘We had isolated and identified two Cucumber mosaic virus (CMV) isolates, the CMV red bean (CMV-RB) isolate and the CMV pea (CMV-P1) isolate. CMV-RB induces necrotic local lesions on inoculated leaves of broad bean, pea, cowpea and bean, and could not infect these hosts systemically. However, CMV-P1 was able to infect these legumes systemically. To study the difference of pathogenic-ity on the legumes induced by these two CMV isolates, the full-length infectious cDNA clones of CMV-Fny, which induced similar symptoms as CMV-RB in the four legumes, were used. The 243 nucleotides fragment, which encodes highly conserved GDD amino acid motif on 2a replicase gene of CMV-Fny RNA2, was replaced with that of CMV-P1. The constructed chimeric virus FP could infect these legumes systemically. The exchange of this region changes the virus symptoms on the legumes, indicating that this 243 nucleotides fragment has major effect on pathogenicity of CMV on the legumes. | TAO Xiaorong ZHOU Xueping LI Guixin YU Jialin | 2002 | Chinese Science Bulletin2002,47,9: | 5

Distribution of soil aggregates and organic carbon in deep soil under long-term conservation tillage with residual retention in dryland显示文摘To ascertain the effects of long-term conservation tillage and residue retention on soil organic carbon(SOC) content and aggregate distribution in a deep soil(>20-cm depth) in a dryland environment, this paper analyzed the SOC and aggregate distribution in soil, and the aggregate-associated organic carbon(OC) and SOC physical fractions. Conservation tillage(reduced tillage with residue incorporated(RT) and no-tillage with residue mulch(NT)) significantly increased SOC sequestration and soil aggregation in deep soil compared with conventional tillage with residue removal(CT). Compared with CT, RT significantly increased the proportion of small macroaggregates by 23%–81% in the 10–80 cm layer, and the OC content in small macroaggregates by 1%–58% in the 0–80 cm layer. RT significantly increased(by 24%–90%) the OC content in mineral-SOC within small macroaggregates in the 0–60 cm layer, while there was a 23%–80% increase in the 0–40 cm layer with NT. These results indicated that:(1) conservation tillage treatments are beneficial for soil aggregation and SOC sequestration in a deep soil in a dryland environment; and(2)the SOC in mineral-associated OC plays important roles in soil aggregation and SOC sequestration. In conclusion, RT with NT is recommended as an agricultural management tool in dryland soils because of its role in improving soil aggregation and SOC sequestration. | WANG Bisheng GAO Lili YU Weishui WEI Xueqin LI Jing LI Shengping SONG Xiaojun LIANG Guopeng CAI Dianxiong WU Xueping | 2019 | Journal of Arid Land2019,11,2: | 3
The symptom difference induced by Tobacco mosaic virus and Tomato mosaic virus in tobacco plants containing the N gene is determined by movement protein gene显示文摘Tobacco mosaic virus (TMV) and Tomato mosaic virus (ToMV) are two closely related viruses in the genus Tobamovirus, but they induce obviously different sizes of necrotic lesions in tobacco plants containing the N gene. Comparison of the symptoms produced by TMV, ToMV and a chimaeric virus (T/OMP), in which the TMV movement protein (MP) gene was re- placed by the ToMV MP gene, showed T/OMP caused necrotic lesions that were similar in size to those of ToMV in tobacco plants containing the N gene. The coat protein and MP of the three viruses accumulated in planta with similar levels, and the replication level of TMV and T/OMP in protoplasts also had no difference. Comparison of the activities of defense-related enzymes (PAL, POD and PPO) induced by the three viruses also showed that the variability of enzyme activity induced by T/OMP was similar to that induced by TMV, but different from that induced by ToMV. The results indicate that the size difference of necrotic lesions induced by TMV and ToMV in to- bacco plants containing the N gene results from the functional difference of their MP genes. | YU Cui1, HU Dongwei1, DONG Jiahong2, CUI Xiaofeng1, WU Junjie1, YU Jialin2 & ZHOU Xueping1 1. | 5 | Functional analysis of a novel βV1 gene identified in a geminivirus betasatellite显示文摘Betasatellites(DNAβ)are circular ssDNA molecules that are associated with monopartite geminiviruses and exert a positive effect on the viral infection.Betasatellites encode one protein,namedβC1,on the complementary strand;βC1 functions as a pathogenicity factor and RNA silencing suppressor. In this report,we describe the identification of another betasatellite-encoded protein,βV1,which also contributes to symptom development.TheβV1 open reading frame can be found on the viral strand of approximately 40%of reported betasatellite sequences, and is conserved in position and sequence. The presence of theβV1 transcript was observed in plants infected with Tomato yellow leaf curl China virus(TYLCCNV)along with its associated betasatellite Tomato yellow leaf curl China betasatellite(TYLCCNB).Mutant viruses unable to produceβV1 showed reduced virulence and decreased viral load.Ectopic expression of the TYLCCNB-βV1 gene in Nicotiana benthamiana plants from a PVX-based vector resulted in leaf mosaic and chlorosis. We further demonstrated that theβV1 protein could elicit hypersensitive response(HR)-type cell death in N.benthamiana leaves. Our results uncover a novel betasatellite-encoded protein that contributes to the virus infection, and this discover gives us a more complete view of the plant-geminivirus interaction landscape. | Tao Hu Yu Song Yaqin Wang Xueping Zhou | 2020 | Science China(Life Sciences)2020,63,5: | 2
Fair Efficiency Comparisons of Decoy-State Quantum Key Distribution Protocols显示文摘Taking the schemes based on different formulas of secure key rate as examples, we give a fair comparison between typical protocols under Universal composable(UC) security frame. Through analyzing the relationship of security parameters in post-processing stage and final secure key rate, we achieve the unified quantification between protocols based on Gottesman-Lo-L¨utkenhausPreskill(GLLP) formula and the ones under UC security.Based on the above research, the impact of different sending lengths and secure parameters on secure key rate is investigated. Besides, we analyze the conditions of fair comparison, and for the first time we give a fair comparison between the protocols based on GLLP formula and smooth entropy. By typical efficiency comparisons, we summarize the way for improving secure key rate in the light intensity choice and the single bit error rate estimation method. | LI Hongxin GAO Ming YAN Xueping YAN Bao HAN Yu SHAN Ling MA Zhi | 2018 | Chinese Journal of Electronics2018,27,2: | 1
Better prognostic determination of cT3 rectal cancer through measurement of distance to mesorectal fascia:A multicenter study显示文摘Objective:To forward the magnetic resonance imaging(MRI)based distance between the deepest tumor invasion and mesorectal fascia(DMRF),and to explore its prognosis differentiation value in cT3 stage rectal cancer with comparison of cT3 substage. Methods:This was a retrospective, multicenter cohort study including cT3 rectal cancer patients undergoing neoadjuvant chemoradiotherapy followed by radical surgery from January 2013 to September 2014.DMRF and cT3 substage were evaluated from baseline MRI.The cutoff of DMRF was determined by disease progression.Multivariate cox regression was used to test the prognostic values of baseline variables. Results:A total of 804 patients were included,of which 226(28.1%)developed progression.A DMRF cutoff of7 mm was chosen.DMRF category, the clock position of the deepest position of tumor invasion(CDTI)and extramural venous invasion(EMVI)were independent predictors for disease progression, and hazard ratios(HRs)were 0.26[95%confidence interval(95%CI),0.13-0.56],1.88(95%CI,1.33-2.65)and 1.57(95%CI,1.13-2.18),respectively.cT3 substage was not a predictor for disease progression. Conclusions:The measurement of DMRF value on baseline MRI can better distinguish cT3 rectal cancer prognosis rather than cT3 substage, and was recommended in clinical evaluation. | Xiaoyan Zhang Qiaoyuan Lu Xiangjie Guo Wuteng Cao Hongmei Zhang Tao Yu Xiaoting Li Zhen Guan Xueping Li Ruijia Sun Yingshi Sun | 2021 | Chinese Journal of Cancer Research2021,33,5: | 1

Mechanistic action of the acute toxicity of Bajitian(Morinda officinalis)in zebrafish embryos显示文摘Objectives:To investigate acute toxicity of Bajitian(Morinda officinalis)in zebrafish embryos. Methods:Zebrafish embryos at 48-h post fertilization(hpf)were exposed to Bajitian ethanol extract for72 h.The causative action of a delay in yolk sac absorption by Bajitian was investigated by RT-PCR analysis of lipid metabolism-related microsomal triglyceride transfer protein(MTP),apolipoprotein CII(ApoC2)and lipogenesis-related liver x receptor(LXR)genes. The effect of Bajitian eliciting an inflammatory response was studied by exposing 72 hpf myeloperoxidase(MPO):GFP transgenic zebrafish embryos to Bajitian extract for 4 h.Assessment was done by TUNEL,caspase-3/7,and RT-PCR analysis of the apoptosis related pathway B-cell lymphoma 2 associated X protein(Bax),Nuclear factor kappa-lightchain-enhancer of activated B cells(NF-k B)genes, neutrophil development-related stem cell leukaemia(SCL)and transcription factor PU.1 genes,to reveal the causative action of Bajitian reducing neutrophils.Results:RT-PCR analysis found that Bajitian extract had no effect on the expression of MTP or ApoC2 genes, but upregulated LXR gene, which might explain the delay in yolk sac absorption.Analysis of the inflammatory response showed that compared with negative controls,Bajitian extract significantly(P<.05)reduced the number of neutrophils in MPO:GFP embryos.TUNEL,caspase-3/7,and RT-PCR analysis of Bax and NF-k B genes found that Bajitian extract did not trigger the cell apoptosis.Further RT-PCR analysis found that Bajitian extract did not affect SCL expression, but did lead to down-regulation of PU.1.The inhibition of neutrophil development/differentiation may explain the decline in the total number of neutrophils following Bajitian treatment, which could be attributed to the anti-inflammatory effects found clinically for this drug. Conclusions:This study demonstrated that Bajitian caused a delay in yolk sac absorption and a decrease neutrophil in zebrafish embryos, which may be related to the inhibition of neutrophil development. | Lirong Tan Min Cheng Gigi Chi Ting Auyeung Oscar Yu Suen Chan Xueping Chen | 2020 | Journal of Traditional Chinese Medical Sciences2020,7,3: | 1

Intercellular transfer of activated STING triggered by RAB22A-mediated non-canonical autophagy promotes antitumor immunity显示文摘STING,an endoplasmic reticulum(ER)transmembrane protein, mediates innate immune activation upon cGAMP stimulation and is degraded through autophagy. Here,we report that activated STING could be transferred between cells to promote antitumor immunity,a process triggered by RAB22A-mediated non-canonical autophagy.Mechanistically,RAB22A engages PI4K2A to generate PI4P that recruits the Atg12–Atg5–Atg16L1 complex, inducing the formation of ER-derived RAB22A-mediated non-canonical autophagosome,in which STING activated by agonists or chemoradiotherapy is packaged. This RAB22A-induced autophagosome fuses with RAB22A-positive early endosome, generating a new organelle that we name Rafeesome(RAB22A-mediated non-canonical autophagosome fused with early endosome).Meanwhile,RAB22A inactivates RAB7 to suppress the fusion of Rafeesome with lysosome, thereby enabling the secretion of the inner vesicle of the autophagosome bearing activated STING as a new type of extracellular vesicle that we define as R-EV(RAB22A-induced extracellular vesicle).Activated STING-containing R-EVs induce IFNβrelease from recipient cells to the tumor microenvironment, promoting antitumor immunity.Consistently,RAB22A enhances the antitumor effect of the STING agonist diABZI in mice, and a high RAB22A level predicts good survival in nasopharyngeal cancer patients treated with chemoradiotherapy. Our findings reveal that Rafeesome regulates the intercellular transfer of activated STING to trigger and spread antitumor immunity, and that the inner vesicle of non-canonical autophagosome originated from ER is secreted as R-EV,providing a new perspective for understanding the intercellular communication of organelle membrane proteins. | Ying Gao Xueping Zheng Boyang Chang Yujie Lin Xiaodan Huang Wen Wang Shirong Ding Weixiang Zhan Shang Wang Beibei Xiao Lanqing Huo Youhui Yu Yilin Chen Run Gong Yuanzhong Wu Ruhua Zhang Li Zhong Xin Wang Qiuyan Chen Song Gao Zhengfan Jiang Denghui Wei Tiebang Kang | 2022 | Cell Research2022,32,12: | 1
Construction of a high-density mutant population of Chinese cabbage facilitates the genetic dissection of agronomic traits显示文摘Chinese cabbage(Brassica rapa ssp.pekinensis)is an economically important vegetable crop throughout the world, especially in Asia.High-quality genome sequences are available for Chinese cabbage, but gene functional studies remain challenging. To promote functional genomic studies of Chinese cabbage,we generated an ethyl methane sulfonate(EMS)mutant population of~8000 M_(2) plants using the double haploid inbred line A03 as the parent. The genome of A03 was sequenced and used as a reference for high-throughput functional characterization of gene mutations at the whole-genome level.A total of 300 M_(2) to M_(5) EMS mutants were phenotypically screened and then sequenced, revealing 750629 SNPs and 46272 InDel mutations that cover 98.27%of all predicted genes in the A03 genome.A forward-genetics approach was successfully used to identify two genes with chloroplast-related functions that are responsible for the yellow leaf mutant trait.A reverse-genetics approach was also used to identify associations between mutations in five genes of the glucosinolate biosynthetic pathway and variations in glucosinolate content of the mutant plants. In addition,we built the Chinese cabbage EMS mutation database(CCEMD,www.bioinformaticslab.cn/EMSmutation/home)to increase the usability of this mutant population resource. In summary,we performed large-scale screening of a heading Chinese cabbage EMS mutant collection at the phenotypic and genotypic levels, which will facilitate gene mining of Chinese cabbage and might also be useful for the study of other Brassica crops. | Xiaoxue Sun Xing Li Yin Lu Shan Wang Xiaomeng Zhang Kang Zhang Xiangjie Su Mengyang Liu Daling Feng Shuangxia Luo Aixia Gu Yu Fu Xueping Chen Shuxin Xuan Yanhua Wang Donghui Xu Shumin Chen Wei Ma Shuxing Shen Feng Cheng Jianjun Zhao | 2022 | Molecular Plant2022,15,5: | 1
Plant and animal positive-sense single-stranded RNA viruses encode small proteins important for viral infection in their negative-sense strand显示文摘Positive-sense single-stranded RNA(+ssRNA)viruses, the most abundant viruses of eukaryotes in nature, require the synthesis of negative-sense RNA(-RNA)using their genomic(positive-sense)RNA(+RNA)as a template for replication.Based on current evidence, viral proteins are translated via viral+RNAs,whereas-RNA is considered to be a viral replication intermediate without coding capacity. Here,we report that plant and animal+ssRNA viruses contain small open reading frames(ORFs)in their-RNA(reverse ORFs[rORFs]).Using turnip mosaic virus(TuMV)as a model for plant+ssRNA viruses,we demonstrate that small proteins encoded by rORFs display specific subcellularlocalizations, and confirm the presence of rORF2 in infected cells through mass spectrometry analysis. The protein encoded by TuMV rORF2 forms punctuate granules that are localized in the perinuclear region and co-localized with viral replication complexes. The rORF2 protein can directly interact with the viral RNA-dependent RNA polymerase, and mutation of rORF2 completely abolishes virus infection, whereas ectopic expression of rORF2 rescues the mutant virus.Furthermore,we show that several rORFs in the-RNA of severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)have the ability to suppress type l interferon production and facilitate the infection of ve-sicular stomatitis virus. In addition,we provide evidence that TuMV might utilize internal ribosome entry sites to translate these small rORFs.Taken together, these findings indicate that the-RNA of+ssRNA vi-ruses can also have the coding capacity and that small proteins encoded therein play critical roles in viral infection, revealing a viral proteome larger than previously thought. | Pan Gong Qingtang Shen Mingzhen Zhang Rui Qiao Jing Jiang Lili Su Siwen Zhao Shuai Fu Yu Ma Linhao Ge Yaqin Wang Rosa Lozano-Durán Aiming Wang Fangfang Li Xueping Zhou | 2023 | Molecular Plant2023,16,11: | 0
Extruded monolith MnO_(x)-CeO_(2)-TiO_(2) catalyst for NH_(3)-SCR of low temperature flue gas from an industry boiler:Deactivation and recovery显示文摘The selective catalytic reduction(SCR) of NO_(x) with NH_(3)(NH_(3)-SCR) technology has been widely applied for reducing NO_(x) emissions from stationary and mobile sources. In this work, the extruded monolith MnO_(x)-CeO_(2)-TiO_(2) catalyst was installed in a cement kiln for NH_(3)-SCR of NO_(x),where the flue gas temperature was 110-140℃.It is found that the monolith catalyst is severely deactivated after operating for about 200 h with almost no NO_(x) conversion at 160℃ under GHSV of 50000 h^(-1),while the fresh monolith catalyst remains 60% NO_(x) conversion.Scanning electron microscopy-energy dispersive spectroscopy(SEM-EDS),X-ray photoelectron spectroscopy(XPS),temperature-programmed desorption of SO_(2)(SO_(2)-TPD) and thermogravimetric-differential thermal analysis(TG-DTG) experiments reveal that both MnO_(x) and CeO_(2) oxides in monolith are severely sulfated to manganese sulfate and cerium sulfate, and the external monolith walls are covered by massive ceria sulfate and little ammonium nitrate. In situ diffuse reflectance infrared Fourier trans form spectroscopy(DRIFTS) analysis demonstrates that the formation of nitrates at low temperatures is inhibited due to the occupation of active sites in MnO_(x)-CeO_(2)-TiO_(2) by sulfates, resulting in the decrease of low temperature activity. After washing with water, the activity of deactivated monolith catalyst can be partially recovered, together with significant loss of manganese and cerium from monolith. | Wei Shi Jingjing Liu Ying Zhu Lin Zhao Yonggang Wang Zhaohuan Cheng Xueping Peng Xiaoyan Shi Yunbo Yu Hong He | 2023 | Journal of Rare Earths2023,41,9: | 0
A Retrospective Study of the Epidemiologic and Clinical Characteristics of COVID-19 Among Hospitalized Patients in Quanzhou,China显示文摘Coronavirus disease 2019(COVID-19)has spread throughout China. However, information about COVID-19 in cities and regions outside Wuhan is limited and the indicators that predict the length of hospital stay for patients with COVID-19 are unclear. Therefore,we collected clinical data from 47 patients with COVID-19 in Quanzhou City. The median age was 38 years[interquartile range(IQR):31–50 years],and 24(51%)were male. There were 8 mild,36 moderate, and 3 severe/critical cases. The median interval from exposure to disease onset was 13 days(IQR:8–18 days). The incidence of severe/critical cases was 33%(3/10)in patients with hypertension.Common symptoms included fever(83%),cough(77%),fatigue(40%),a sore, dry throat(28%),and diarrhea(21%).One patient(2%)developed respiratory distress syndrome on day 13 of inpatient treatment.Six patients had leukopenia,17 had elevated C-reactive protein(CRP),and 8 had lymphocytopenia and elevated lactate dehydrogenase(LDH).The median length of hospitalization was 22 days(IQR:16-30 days).Dynamic monitoring of LDH,CRP,and neutrophil-lymphocyte ratio predicted whether length of hospitalization would exceed 21 days. Most patients presented with mild and moderate disease.Patients with hypertension were more likely to become severe or critical.Dynamic monitoring of LDH,CRP,and neutrophil-lymphocyte ratio levels can help predict delayed discharge from the hospital. | Wenhuang Chen Yijian Lin Hongbo Huang Maosheng Cai Dongheng Lin Milong Su Zhijun Su Xibin Zhuang Xueping Yu | 2021 | Infectious Microbes & Diseases2021,3,1: | 0
Discussion on the Selection of ACEI and ARB for the Patients with COVID-19 and Hypertension from ACE2显示文摘With the wide spread of COVID-19,some studies have confirmed that novel coronavirus enters the cell through the binding of spike protein and ACE2 protein, which has the risk of causing the virus to enter the cell to accelerate its transmission.ACEI and ARB are the key drugs for the treatment of hypertension and are widely used in clinic. They are good for ventricular and vascular remodeling in patients with hypertension, coronary heart disease and heart failure.ACEI and ARB drugs may increase the expression of ACE2 in lung tissue and increase the risk of aggravation of the disease.According to the advice of hypertension experts:for mild ordinary COVID-19 patients with hypertension,we can consider discontinuing ACEI and ARB drugs, while for severe COVID-19 patients with hypertension,ACEI and ARB drugs should be stopped immediately.During the discontinuation period, temporary replacement therapy with diuretics, dipine and vasodilatation hypotensive drugs can be considered. | Jinfeng CHEN Zhongyi LEI Chaofeng LIU Hong FAN Peng LEI Xueping WU Xiaoyong YU Yanfen ZHOU Jiejun HOU | 2020 | Medicinal Plant2020,11,4: | 0
