| 1 | Interleukin 17-Producing γδ T Cells Increased in Patients with Active Pulmonary Tuberculosis显示文摘Although it has been known that γδ T cells may play an important role in the immune response to infection of Mycobacterium tuberculosis (M. tb), the mechanisms by which the γδ T cells participate in the innate and/or acquired immunity to tuberculosis (TB) have not been full elucidated. In the present study, 27 patients with active pulmonary TB and 16 healthy donors (HD) were performed. We found that proportion of IL-17-producing cells among lymphocyte was similar between TB patients and HD, whereas the proportions of γδ T cells in IL-17-producing cells (59.2%) and IL-17-producing cells in γδ T cells (19.4%) in peripheral blood were markedly increased in TB patients when compared to those in HD (43.9% and 7.7%, respectively). In addition, the proportions of IFN-γ-producing γδ T cells in TB patients were obviously lower than that in HD. Upon re-stimulated with M. tb heat-treated antigen (M. tb-HAg) in vitro, fewer IL-17-producing γδ T cells were generated from HD and TB patients, whereas IFN-γ-producing γδ T cells were increased in TB patients compared to that in HD. | 3 | Sedimentary environment of Ediacaran glacigenic diamictite in Guozigou of Xinjiang, China显示文摘The Talisayi Formation found in the Guozigou region on the southern side of Sayram Lake of Xinjiang, China is a Neoproterozoic glacigenic diamictite succession that has not been adequately researched. According to its isotopic geochronology and regional stratigraphic correlation, the Talisayi Formation is equivalent to the Ediacaran glacial sediments. The present paper provides the geochemical data of the Talisayi Formation that reveals the tectonic setting and sedimentary environment in this area during the Ediacaran period. The oxides of Si, K, Na and Al and their ratios of matrix materials from glacigenic diamictites and bulk rock of siltstone and argillaceous siltstones indicate that the sedimentary environment of this formation evolved from a tectonically active region to a passive continental margin. Values of CIA (carbonate-corrected Chemical Index of Alteration) ranging from 54 to 74 show a low-middle weathering action under a glacial environment. Total organic carbon (TOC), V/Cr, Ni/Co and U/Th ratios suggest that the sediments deposited in shallow water with an oxic and weak hydrodynamic condition. | 4 | Trace elements and stable isotopic geochemistry of an Early Cambrian chert-phosphorite unit from the lower Yurtus Formation of the Sugetbrak section in the Tarim Basin显示文摘A chert-phosphorite unit from the Sugetbrak section in the Tarim Basin was analyzed for rare earth elements(Ce,Eu),redox sensitive proxies,and carbon isotopic compositions(??13C carb and??13C org)in the lower Yurtus Formation of the Early Cambrian period.Redox sensitive element ratios(Th/U,V/Cr,Ni/Co,and V/Sc)were employed to determine the palaeoenvironmental conditions during this period.The ratios indicated that the depositional environment of the chert-phosphorite-black shale unit ranged from suboxic to anoxic.Negative Ce and positive Eu anomalies in the chert-phosphorite assemblages of the studied Yurtus Formation indicated the existence of a redox-stratified ocean,similar to that of South China.Overturn or upward expansion of the deep water-mass probably reached the shallow marine zone after the formation of the Yurtus phosphorites.The characteristics of the negative Ce anomaly may be due to phosphoritic inheritance from the Ce-depleted signature of the overlying water column.Subsequent hydrothermal inputs and reduced detrital supplies of the deep water caused by the upwelling affected certain redox sensitive elements in the sedimentary basin.??13C carb and??13C org negative excursions in the Yurtus chert-phosphorite unit may be related to a transgression phase when episodic basinal upwelling moved12C-and P-rich waters from the pelagic basin floor to the continental shelf.Although carbon isotopic compositions in the Yurtus chert-phosphorite assemblages may have suffered from diagenetic alteration,they can be used to probe diagenetic conditions.Multi-proxy geochemical studies indicated that the??13C carb values of the Yurtus chert-phosphorite assemblages might be considered reflections of a predominantly suboxic environment that was subsequently affected by hydrothermal inputs due to the upwelling. | YAO ChunYan MA DongSheng DING HaiFeng ZHANG XiaoYong HUANG Hui | 2014 | Science China Earth Sciences2014,57,3: | 7 |
| 5 | PADI3 induces cell cycle arrest via the Sirt2/AKT/p21 pathway and acts as a tumor suppressor gene in colon cancer显示文摘Objective:As a member of the peptidyl arginine deiminase(PAD)family,PADI3 is weakly expressed in colon cancer tissues and highly expressed in adjacent colon cancer tissues.However,the role of PADI3 in colon cancer is unclear.In this study,we investigated the function and molecular mechanism of PADI3 in colon cancer tumorigenesis.Methods:Western blot and real-time PCR were used to detect the expression levels of several genes.CCK-8,flow cytometry(FCM)and colony formation assays were used to examine cell proliferation,the cell cycle and colony formation ability.RNAsequencing analysis was used to study the molecular mechanism of PADI3 in tumorigenesis.A truncation mutation experiment was performed to determine the key functional domain of PADI3.Results:PADI3 overexpression inhibited cell proliferation and colony formation and led to G1 phase arrest in both HCT116(originating from primary colon cancer)and LoVo(originating from metastatic tumor nodules of colon cancer)cells.PADI3-expressing HCT116 cells had a lower tumor formation rate and produced smaller tumors than control cells.PADI3 significantly decreased Sirtuin2(Sirt2)and Snail expression and AKT phosphorylation and increased p21 expression,and Sirt2 overexpression partly reversed the effects induced by PADI3 overexpression.Immunocytochemistry showed that PADI3 is mainly localized in the cytoplasm.Truncation mutation experiments showed that the C-domain is the key domain involved in the antitumor activity of PADI3.Conclusions:PADI3 suppresses Snail expression and AKT phosphorylation and promotes p21 expression by downregulating Sirt2 expression in the cytoplasm,and the C-domain is the key domain for its antitumor activity. | Xiaotian Chang Zhengbin Chai Jiaorui Zou Hongxing Wang Yao Wang Yabing Zheng Hui Wu Chunyan Liu | 2019 | Cancer Biology & Medicine2019,16,4: | 4 |
| 6 | The direct electrochemistry of glucose oxidase based on the synergic effect of amino acid ionic liquid and carbon nanotubes显示文摘Amino acid ionic liquids(AAILs) have attracted much attention due to their special chemical and physical properties,especially their outstanding biocompatibility and truly green aspect.In this work,a novel electrochemical biosensing platform based on AAILs/carbon nanotubes(CNTs) composite was fabricated.AAILs were used as a novel solvent for glucose oxidase(GOD) and the GOD-AAILs/CNTs/GC electrode was conveniently prepared by immersing the carbon nanotubes(CNTs) modified glassy carbon(GC) electrode into AAILs containing GOD.The direct electrochemistry of GOD on the GOD-AAILs/CNTs/GC electrode has been investigated and a pair of reversible peaks was obtained by cyclic voltammetry.The immobilized glucose oxidase could retain bioactivity and catalyze the reduction of dissolved oxygen.Due to the synergic effect of AAILs and CNTs,the GOD-AAILs/CNTs/GC electrode shows excellent electrocatalytic activity towards glucose with a linear range from 0.05 to 0.8 mM and a detection limit of 5.5 μM(S/N = 3).Furthermore,the biosensor exhibits good stability and ability to exclude the interference of commonly coexisting uric and ascorbic acid.Therefore,AAILs/CNTs composite can be a good candidate biocompatible material for the direct electrochemistry of the redox-active enzyme and the construction of third-generation enzyme sensors. | WANG MengDong DENG ChunYan NIE Zhou XU XiaHong YAO ShouZhuo | 2009 | Science China Chemistry2009,52,11: | 4 |
| 7 | Early Cambrian carbon isotope stratigraphy in the Tarim Basin and a correlation with the Yangtze Platform显示文摘Carbon isotope chemostratigraphy has been used worldwide for stratigraphic correlation.In this study,δ13Ccarb values are estimated for the Early Cambrian Sugaitebulake section in the Tarim Basin,Xinjiang Autonomous Region,China.As a result,one positive and two negative carbon isotope excursions in the studied section were iden-tified.The δ13Ccarb values reached the maximum negative excursion(N1:-12.39‰) at the basal of the Yuertusi For-mation,and then increased to P1.After P1,δ13Ccarb values sharply decreased to about-7.06‰(N2) in the studied section.The pattern of δ13Ccarb in the Early Cambrian is comparable to the synchronous records of other sections,such as the Laolin section,the Xiaotan section and the Anjiahe section of the Yangtze Platform.It is concluded that the Early Cambrian Yuertusi Formation from the Tarim Basin is within the Nemakit-Daldynian stage,and the lower strata of the Yuertusi Formation may belong to the Zhujiaqing Formation(Meishucun Formation) of the Yangtze Platform.The Ediacaran/Cambrian boundary of the Tarim Block should be located in cherts and phosphorites suc-cessions at the basal of the Yuertusi Formation.The δ13Ccarb negative excursion N1 is just across the PC/C boundary,and may be related to certain biomass extinction due to anoxic sedimentary environment,transgression and/or the oceanic overturn.The second δ13Ccarb negative excursion N2 may account for the sea-level falling in the Early Cam-brian. | YAO Chunyan MA Dongsheng DING Haifeng ZHANG Xiaoyong | 2011 | Chinese Journal Of Geochemistry2011,30,3: | 3 |
| 8 | Carbon Isotope Features of the Sugetbrak Section in the Aksu-Wushi Area, Northwest China: Implications for the Precambrian/Cambrian Stratigraphic Correlations显示文摘The upper Qigeblaq Formation(Fm) dolostones and the Yurtus Fm phosphatic cherts, black shales, limestones, and dolostones are widely distributed in the Precambrian/Cambrian transitional succession of the Aksu-Wushi area. Negative δ13C excursion above the Yurtus Fm/Qigeblaq Fm boundary was determined in this study. The pronounced negative carbon isotope excursion occurs in the phosphatic chert layers at the bottom of the Cambrian Yurtus Fm, below which the first appearance of the Asteridium- Heliosphaeridium-Comasphaeridium(AHC) acritarch assemblage zone. The δ13C curve of the lower part of the Yurtus Fm in the Aksu-Wushi area was found to be correlated with the early Cambrian δ13C curves of the Zhujiaqing Fm(Daibu Member), the lower part of the Yanjiahe Fm on the Yangtze Platform in China, the lower Tal Fm in India, the Sukharikha Fm in Siberia, and the upper part of the Tsagaan Oloom Fm in Mongolia through biostigraphy. The lower part of the Yurtus Fm in the Tarim Basin is at the Nemakit-Daldynian stage, and the Precambrian/Cambrian boundary of the Aksu-Wushi area may be located in the phosphatic chert unit which just below the first appearance AHC acritarch assemblage zone. | 10 | Poly(PEGMA) magnetic nanogels:Preparation via photochemical method,characterization and application as drug carrier显示文摘One-pot synthesis of magnetic nanogels with excellent biocompatibility via the photochemical method is reported in this paper.Poly(PEGMA) modified superparamagnetic nanogels(poly(PEGMA) magnetic nanogels) were synthesized by in-situ polymerization using poly(ethylene glycol) methacrylate(PEGMA) as the monomer and N,N'-methylene-bis-(acrylamide)(MBA) as the cross-linking agent in magnetite aqueous suspension under UV irradiation.The surface functional groups and components of magnetic nanogels were analyzed by Fourier transform infrared spectroscopy(FTIR) and a thermogra-vimetric analyzer(TGA).The results indicated that the poly(PEGMA) magnetic nanogels were synthe-sized successfully by coating poly(PEGMA) on the Fe3O4 nanoparticles under UV irradiation,and the Fe3O4 nanoparticles content in this nanogels was above 50 wt%.The morphology,size,zeta-potential and magnetic property were also characterized.The magnetic nanogels had a nearly spherical shape and core-shell structure,the average size in aqueous system measured by photon correlation spec-troscopy(PCS) was 68.4 nm,which was much bigger than that in the dry state,the nanogels behaved superparamagnetically with saturated magnetization of 58.6 emu/g,and the zeta-potential was-16.3-_17.3 mV at physiological pH(6.8-7.4) which could help to maintain stability in blood.The preliminary application as drug carrier was made and the doxorubicin-loaded magnetic nanogels had an excellent property in slow-release.The experiment indicated that the magnetic nanogel was an ideal candidate carrier in target drug delivery systems and other biomedical application. | SUN HanWen ZHANG LianYing ZHU XinJun KONG ChunYan ZHANG CunLan YAO SiDe | 2009 | Science China Chemistry2009,52,1: | 2 |

| 12 | Assembly of layer-by-layer films of superoxide dismutase and gold nanorods:A third generation biosensor for superoxide anion显示文摘Based on the layer-by-layer self-assembly of positively charged cetyltrimethylammonium bromide (CTAB) wrapped gold na- norods (AuNRs) and negatively charged superoxide dismutase (SOD) from their aqueous solutions on cysteine modified gold electrode (Cys/Au), a third generation electrochemical biosensor ((SOD/AuNRs)2/Cys/Au) for superoxide anion (O2·-) was developed. The two layers assembly of SOD/AuNRs can significantly enhance the direct electron transfer between SOD and the electrode. The functional enzymatic activities of the SOD offer an electrochemical approach to the determination of O2·-.
