| 1 | Herbal decoctosome is a novel form of medicine显示文摘Traditionally, herbal medicine is consumed by drinking decoctions produced by boiling herbs with water. The functional components of the decoction are heat stable. Small RNAs(sRNAs) were reported as a new class of functional components in decoctions. However, the mechanisms by which sRNAs survive heat treatment of the decoction and enter cells are unclear.Previous studies showed that plant-derived exosome-like nanoparticles(ELNs), which we call botanosomes, could deliver therapeutic reagents in vivo. Here, we report that heat-stable decoctosomes(ELNs) from decoctions have more therapeutic effects than the decoctions in vitro and demonstrate therapeutic efficacy in vivo. Furthermore, sRNAs, such as HJT-sRNA-m7 and PGY-sRNA-6, in the decoctosome exhibit potent anti-fibrosis and anti-inflammatory effects, respectively. Decoctosome is comprised of lipids, chemical compounds, proteins, and s RNAs. A medical decoctosome mimic is called bencaosome. A single lipid sphinganine(d22:0) identified in the decoctosome was mixed and heated with the synthesized sRNAs to form the simplest bencaosome. This simple bencaosome structure was identified by critical micelle concentration(cmc) assay that sRNAs coassembled with sphinganine(d22:0) to form the lipid layers of vesicles. The heating process facilitates co-assembly of sRNAs and sphinganine(d22:0) until a steady state is reached. The artificially produced sphinganine-HJT-sRNA-m7 and sphinganinePGY-sRNA-6 bencaosomes could ameliorate bleomycin-induced lung fibrosis and poly(I:C)-induced lung inflammation, respectively, following oral administration in mice. | 4 | Review of the progress in preparing nano TiO_2: An important environmental engineering material显示文摘TiO2 nanomaterial is promising with its high potential and outstanding performance in photocatalytic environmental applications, such as CO2 conversion, water treatment, and air quality control. For many of these applications, the particle size, crystal structure and phase,porosity, and surface area influence the activity of TiO2 dramatically. TiO2 nanomaterials with special structures and morphologies, such as nanospheres, nanowires, nanotubes, nanorods,and nanoflowers are thus synthesized due to their desired characteristics. With an emphasis on the different morphologies of TiO2 and the influence factors in the synthesis, this review summarizes fourteen TiO2 preparation methods, such as the sol–gel method, solvothermal method, and reverse micelle method. | 5 | Efficacy and safety of metformin and sitagliptin based triple antihyperglycemic therapy(STRATEGY):a multicenter,randomized,controlled,non-inferiority clinical trial显示文摘Despite the current guideline's recommendation of a timely stepwise intensification therapy,the 'clinical inertia',termed as the delayed treatment intensification,commonly exists in the real world,which may be partly due to the relatively little substantial evidence and no clear consensus regarding the efficacy and safety of triple oral agents in patients inadequately controlled with dual therapy.In this clinical trial performed in 237 centers in China,5,535 type 2 diabetic patients inadequately controlled by previous therapies were treated with a stable metformin/sitagliptin dual therapy for 20 weeks.The patients who did not reach the glycated hemoglobin A1c(HbA1c) goal were then further randomized into glimepiride,gliclazide,repaglinide,or acarbose group for an additional 24-week triple therapy.A mean HbAlc reduction of 0.85%was observed when sitagliptin was added to the patients inadequately controlled with metformin in 16 weeks.Further HbAlc reductions in the 24-week triple therapy stage were 0.65%in glimepiride group,0.70%in gliclazide group,0.61%in repaglinide group,and 0.45%in acarbose group.The non-inferiority criterion for primary hypotheses was met for gliclazide and repaglinide,but not for acarbose,compared with glimepiride,when added to metformin/sitagliptin dual therapy.The incidences of adverse events(AEs) were 29.2%in the dual therapy stage and30.3%in the triple therapy stage.Metformin/sitagliptin as baseline therapy,with the addition of a third oral antihyperglycemic agent,including glimepiride,gliclazide,repaglinide,or acarbose,was effective,safe and well-tolerated for achieving an HbAlc<7.0%goal in type 2 diabetic patients inadequately controlled with previous therapies.The timely augmentation of up to three oral antihyperglycemic agents is valid and of important clinical benefit to prevent patients from exposure to unnecessarily prolonged hyperglycemia. | Wen Xu Yiming Mu Jiajun Zhao Dalong Zhu Qiuhe Ji Zhiguang Zhou Bin Yao Anhua Mao Samuel S.Engel Bin Zhao Yan Bi Longyi Zeng Xingwu Ran Juming Lu Linong Ji Wenying Yang Weiping Jia Jianping Weng | 2017 | Science China(Life Sciences)2017,60,3: | 19 |

| 8 | Clue to a thorough understanding of terahertz pulse generation by femtosecond laser filamentation显示文摘In this work, it has been demonstrated that in order to fully understand the terahertz(THz) pulse generation process during femtosecond laser filamentation, the interaction between THz wave and air plasma has to be taken into account. This interaction is mainly associated with the spatial confinement of the THz pulse by the plasma column, which could be described by the one-dimensional negative dielectric(1DND) waveguide model. By combining the 1 DND model with the conventional four-wave mixing(4WM) and photocurrent(PC) models,the variation of THz spectral amplitude and width obtained in experiments could be better understood. | 9 | Sequential fate-switches in stem-like cells drive the tumorigenic trajectory from human neural stem cells to malignant glioma显示文摘Glioblastoma(GBM)is an incurable and highly heterogeneous brain tumor,originating from human neural stem/progenitor cells(hNSCs/hNPCs)years ahead of diagnosis.Despite extensive efforts to characterize hNSCs and end-stage GBM at bulk and single-cell levels,the de novo gliomagenic path from hNSCs is largely unknown due to technical difficulties in early-stage sampling and preclinical modeling.Here,we established two highly penetrant hNSC-derived malignant glioma models,which resemble the histopathology and transcriptional heterogeneity of human GBM.Integrating time-series analyses of whole-exome sequencing,bulk and single-cell RNA-seq,we reconstructed gliomagenic trajectories,and identified a persistent NSC-like population at all stages of tumorigenesis.Through trajectory analyses and lineage tracing,we showed that tumor progression is primarily driven by multi-step transcriptional reprogramming and fate-switches in the NSC-like cells,which sequentially generate malignant heterogeneity and induce tumor phenotype transitions.We further uncovered stage-specific oncogenic cascades,and among the candidate genes we functionally validated C1QL1 as a new glioma-promoting factor.Importantly,the neurogenic-to-gliogenic switch in NSC-like cells marks an early stage characterized by a burst of oncogenic alterations,during which transient AP-1 inhibition is sufficient to inhibit gliomagenesis.Together,our results reveal previously undercharacterized molecular dynamics and fate choices driving de novo gliomagenesis from hNSCs,and provide a blueprint for potential early-stage treatment/diagnosis for GBM. | Xiaofei Wang Ran Zhou Yanzhen Xiong Lingling Zhou Xiang Yan Manli Wang Fan Li Chuanxing Xie Yiming Zhang Zongyao Huang Chaoqiong Ding Kaidou Shi Weida Li Yu Liu Zhongwei Cao Zhen-Ning Zhang Shengtao Zhou Chong Chen Yan Zhang Lu Chen Yuan Wang | 2021 | Cell Research2021,31,6: | 8 |
| 10 | Metallogenic model and prognosis of the Shuiyindong super-large strata-bound Carlin-type gold deposit, southwestern Guizhou Province, China显示文摘The Shuiyindong deposit is one of the largest (more than 100 tonnes of Au) and highest grade (more than 7×10-6-10×10-6), strata-bound Carlintype gold deposits in southwestern Guizhou Province, China. The deposit is controlled by both structure and favorable lithology. It is situated near the axis of the striking Huijiabao anticline and is hosted in bioclastic limestone of the Permian Longtan Formation. Gold mineralization occurred under low temperature with Th of 220℃± and is closely associated with decarbonation, silicification, sulfidation and dolomitization. The deposit has a characteristic elemental assemblage of Au-As-Hg-Tl. Studies of geochemistry and isotope compositions indicated that the ore-bearing materials and fluids of the gold deposit mainly originated from a plutonic source, and possess a mixing feature with the strata matter during transportation from mantle to crust. Fluid inclusions in vein quartz from the gold deposit are rich in volatile flux, indicating that metallogenic fluid is an overpres-sured one. The activity and geothermal state of the Earth's crust in the long period of time are favorable for the formation of overpressured fluids in a large area, and extensive structures would drive the fluids into ore-forming sys-tem and make gold deposits formed. The complexity of structural movement in the upper crust of southwestern Guizhou Province resulted in complicated gold mineralization. | 11 | Focal adhesion protein Kindlin-2 regulates bone homeostasis in mice显示文摘Our recent studies demonstrate that the focal adhesion protein Kindlin-2 is critical for chondrogenesis and early skeletal development. Here, we show that deleting Kindlin-2 from osteoblasts using the 2.3-kb mouse Col1 a1-Cre transgene minimally impacts bone mass in mice, but deleting Kindlin-2 using the 10-kb mouse Dmp1-Cre transgene, which targets osteocytes and mature osteoblasts, results in striking osteopenia in mice. Kindlin-2 loss reduces the osteoblastic population but increases the osteoclastic and adipocytic populations in the bone microenvironment. Kindlin-2 loss upregulates sclerostin in osteocytes,downregulates β-catenin in osteoblasts, and inhibits osteoblast formation and differentiation in vitro and in vivo. Upregulation ofβ-catenin in the mutant cells reverses the osteopenia induced by Kindlin-2 deficiency. Kindlin-2 loss additionally increases the expression of RANKL in osteocytes and increases osteoclast formation and bone resorption. Kindlin-2 deletion in osteocytes promotes osteoclast formation in osteocyte/bone marrow monocyte cocultures, which is significantly blocked by an anti-RANKLneutralizing antibody. | 14 | Synthesis of low-cost porous ceramic microspheres from waste gangue for dye adsorption显示文摘Low-cost porous ceramic microspheres from waste gangue were prepared by simple spray drying and subsequent calcination. Effects of calcination temperature on phase and microstructure evolution, specific surface area, pore structure, and dye adsorption mechanism of the microspheres were investigated systematically. Results showed that the microspheres were spherical, with some mesopores both on the surface and inside the spheres. The phase kept kaolinite after calcined at 800 and 900 ℃ and transformed into mullite at 1000 ℃. The microspheres calcined at 800 ℃ showed larger adsorption capacity and removal efficiency than those calcined at higher temperatures. Methylene blue(MB) and basic fuchsin(BF) removal efficiency reached 100% and 99.9% with the microsphere dosage of 20 g/L, respectively, which was comparable to that of other low-cost waste adsorbents used to remove dyes in the literature. Adsorption kinetics data followed the pseudosecond-order kinetic model, and the isotherm data fit the Langmuir isotherm model. The adsorption process was attributed to multiple adsorption mechanisms including physical adsorption, hydrogen bonding, and electrostatic interactions between dyes and gangue microspheres. | 15 | The YDA-MKK4/MKK5-MPK3/MPK6 Cascade Functions Downstream of the RGF1-RGI Ligand-Receptor Pair in Regulating Mitotic Activity in Root Apical Meristem显示文摘The mitotic activity of root apical meristem(RAM)is critical to primary root growth and development.Previous studies have identified the roles of ROOT GROWTH FACTOR 1(RGF1),a peptide ligand,and its receptors,RGF1 INSENSITIVEs(RGIs),a clade of five leucine-rich-repeat receptor-like kinases,in promoting cell division in the RAM,which determines the primary root length.However,the downstream signaling components remain elusive.In this study,we identify a complete mitogen-activated protein kinase(MAPK or MPK)cascade,composed of YDA,MKK4/MKK5,and MPK3/MPK6,that functions downstream of the RGF1-RGI ligand-receptor pair.Similar to the rgi1/2/3/4/5 quintuple mutant,loss-of-function mutants of MPK3 and MPK6,MKK4 and MKK5,or YDA show a short-root phenotype,which is associated with reduced mitotic activity and lower expression of PLETHORA 1(PLT1)/PLT2 in the RAM.Furthermore,MPK3/MPK6 activation in response to exogenous RGF1 treatment is impaired in the rgi1/2/3/4/5 quintuple,yda single,and mkk4 m kk5 double mutants.Epistatic analyses demonstrated that the expression of constitutively active MKK4,MKK5,or YDA driven by the RGI2 promoter can rescue the short-root phenotype of the rgi1/2/3/4/5 mutant.Taken together,these results suggest that the YDA-MKK4/MKK5-MPK3/MPK6 cascade functions downstream of the RGF1-RGI ligand-receptor pair and upstream of PLT1/PLT2 to modulate the stem cell population and primary root growth in Arabidopsis. | Yiming Shao Xinxing Yu Xuwen Xu Yong Li Wenxin Yuan Yan Xu Chuanzao Mao Shuqun Zhang Juan Xu | 2020 | Molecular Plant2020,13,11: | 6 |

| 18 | Photoinduced large polaron transport and dynamics in organic-inorganic hybrid lead halide perovskite with terahertz probes显示文摘Organic-inorganic hybrid metal halide perovskites(MHPs)have attracted tremendous attention for optoelectronic applications.The long photocarrier lifetime and moderate carrier mobility have been proposed as results of the large polaron formation in MHPs.However,it is challenging to measure the effective mass and carrier scattering parameters of the photogenerated large polarons in the ultrafast carrier recombination dynamics.Here,we show,in a one-step spectroscopic method,that the optical-pump and terahertz-electromagnetic probe(OPTP)technique allows us to access the nature of interplay of photoexcited unbound charge carriers and optical phonons in polycrystalline CH_(3)NH_(3)PbI_(3)(MAPbI_(3))of about 10μm grain size.Firstly,we demonstrate a direct spectral evidence of the large polarons in polycrystalline MAPbI_(3).Using the Drude-Smith-Lorentz model along with the Frӧhlich-type electron-phonon(e-ph)coupling,we determine the effective mass and scattering parameters of photogenerated polaronic carriers.We discover that the resulting moderate polaronic carrier mobility is mainly influenced by the enhanced carrier scattering,rather than the polaron mass enhancement.While,the formation of large polarons in MAPbI_(3)polycrystalline grains results in a long charge carrier lifetime at room temperature.Our results provide crucial information about the photo-physics of MAPbI3 and are indispensable for optoelectronic device development with better performance. | Zuanming Jin Yan Peng Yuqing Fang Zhijiang Ye Zhiyuan Fan Zhilin Liu Xichang Bao Heng Gao Wei Ren Jing Wu Guohong Ma Qianli Chen Chao Zhang Alexey VBalakin Alexander PShkurinov Yiming Zhu Songlin Zhuang | 2022 | Light(Science & Applications)2022,11,8: | 4 |

| 20 | The efficacy and safety of pramipexole ER versus IR in Chinese patients with Parkinson’s disease: a randomized, double-blind, double-dummy, parallel-group study显示文摘Objective:To evaluate the non-inferiority of pramipexole extended-release(ER)versus immediate-release(IR)in Chinese patients with Parkinson’s disease(PD)in a double-blind,randomized,parallel-group study.Methods:Subjects were Chinese patients with idiopathic PD with diagnosis≥2 years prior to trial,age≥30 years old at diagnosis,and Modified Hoehn and Yahr score 2-4 during‘on’-time.Subjects received treatment with pramipexole ER(n=234)or IR(n=239).Non-inferiority was based on the primary endpoint,the change from baseline to end of maintenance(week 18)in the UPDRS(Parts II+III)total score.Results:For the primary endpoint,the adjusted mean changes(standard error)of UPDRS Parts II+III at week 18 were−13.81(0.655)and−13.05(0.643)for ER and IR formulations,respectively,using ANCOVA adjusted for treatment and centre(fixed effect)and baseline(covariate).The adjusted mean between group difference was 0.8 for the 2-sided 95%CI(−1.047,2.566).Since the lower limit of the 2-sided 95%CI(−1.047)for treatment difference was higher than the non-inferiority margin of−4,non-inferiority between pramipexole ER and IR was demonstrated.The incidence of adverse events(AEs)was 68.8%in the ER arm and 73.6%in the IR arm with few severe AEs(ER:2.1%;IR:3.8%).Conclusion:Based on the UPDRS II+III score,pramipexole ER was non-inferior to pramipexole IR.The safety profiles of pramipexole ER and IR were similar.These results were based on comparable mean daily doses and durations of treatment for both formulations. | Ying Wang Shenggang Sun Suiqiang Zhu Chunfeng Liu Yiming Liu Qing Di Huifang Shang Yan Ren Changhong Lu Mark Forrest Gordon Nolwenn Juhel Shengdi Chen for the Pramipexole ER Study Team | 2014 | Translational Neurodegeneration2014,3,1: | 3 |
