12018 Chinese Pediatric Cardiology Society(CPCS) guideline for diagnosis and treatment of syncope in children and adolescents显示文摘Syncope belongs to the transient loss of consciousness(TLOC), characterized by a rapid onset, short duration, and spontaneous complete recovery. It is common in children and adolescents, accounting for 1% to 2% of emergency department visits.Recurrent syncope can seriously affect children's physical and mental health, learning ability and quality of life and sometimes cardiac syncope even poses a risk of sudden death. The present guideline for the diagnosis and treatment of syncope in children and adolescents was developed for guiding a better clinical management of pediatric syncope. Based on the globally recent development and the evidence-based data in China, 2018 Chinese Pediatric Cardiology Society(CPCS) guideline for diagnosis and treatment of syncope in children and adolescents was jointly prepared by the Pediatric Cardiology Society, Chinese Pediatric Society, Chinese Medical Association(CMA)/Committee on Pediatric Syncope, Pediatricians Branch, Chinese Medical Doctor Association(CMDA)/Committee on Pediatric Cardiology, Chinese College of Cardiovascular Physicians, Chinese Medical Doctor Association(CMDA)/Pediatric Cardiology Society, Beijing Pediatric Society, Beijing Medical Association(BMA). 2Effects of AT1 receptor antagonist,Iosartan,on rat hepatic fibrosis induced by CCl_4显示文摘AIM To investigate effect of losartan,an AT1receptor antagonist,on hepatic fibrosis induced byCCl;and to determine whether or not AT1receptors are expressed on hepatic stellate cells,METHODS AND RESULTS Fifty male Sprague-Dawley rats,weighing(180±20)g,wererandomized into five groups(control group,modelgroup,and three losartan treated groups),inwhich all rats were given the subcutaneousinjection of 40% CCl4(every 3 days for 6 weeks)except for rats of control group.Rats of losartan-treated groups were treated with losartan(20 mg/kg,10 mg/kg,5 mg/kg,daily gavage),After 6weeks liver tissue and serum samples of all ratswere examined.Serum hyaluronic acid(HA),procollagen typeⅢ(PCⅢ)were detected byradioimmunoassays,van Giesion collagen stainingwas used to evaluate the extracellular matrix of ratswith liver fibrosis.The expression of AT1receptors,transforming growth factor-beta(TGF-β),and alpha-smooth muscle actin(a-SMA)inliver tissue were determined byimmunohistochemical techniques.Compared withmodel group,serum ALT and AST of losartan-treated groups were significantly reduced(t=4.20,P<0.01 and t=4.57,P<0.01).Serum HAand PCⅢalso had significant differences(t=3.53,P<0.01 and t=2.20,P<0.05).Thedegree of fibrosis was improved by losartan and correlated with the expressions of AT1 receptors,TGF-β,and α-SMA in liver tissue.CONCLUSION AT1 receptor antagonist,losartan,could limit the progression of the hepatic fibrosisinduced by CCl4.The mechanism may be related tothe decrease in the expression of AT1 receptorsand TGF-β,ameliorating the injury of hepatocytes;activation of local renin-angiotensin system mightrelate to hepatic fibrosis;and during progressionof fibrosis,activated hepatic stellate cells mightexpress AT1 receptors.Hong Shan Wei Ding Guo Li Han Ming Lu Yu Tao Zhan Zhi Rong Wang Xin Huang Jing Zhang Ji Lin Cheng Qin Fang Xu Department of Gastroenterology,Xinhua Hospital,Shanghai Second Medical University,Shanghai 200092,China 2000World Journal of Gastroenterology2000,6,4:42

4The regulatory role of AT 1 receptor on activated HSCs in hepat,c fibrogenesis,effects of RAS inhibitors on hepatic fibrosis induced by CCl_4显示文摘AIM To assess the effect of ACE inhibitor andAng Ⅱ type Ⅰ(AT1)receptor antagonist inpreventing hepatic fibrosis caused by CCl4administration in rats;to investigate whether ornot there are expression of AT 1 receptors onhepatic stellate cells;and to observe the effectof Ang Ⅱ on proliferation and ECM synthesis ofcultured HSCs.METHODS Studies were conducted in maleSprague-Dawley rats.Except for thehepatofibrotic model group and the controlgroup,in three treated groups,either enalapril(5 mg/kg),or Iosartan(10 mg/kg),or enalapril+Iosartan were given to the fibrotic rats bydaily gavage,and saline vehicle was given tomodel and normal control rats.After 6 weeks,liver fibrosis was assessed directly by hepaticmorphometric analysis,which has beenconsidered the gold standard for thequantification of fibrosis.The expressions of AT1 receptors and(α-mooth muscle actin,α-SMA)in liver tissue or isolated hepatic stellate cells(HSCs)were detected by immunohistochemicaltechniques.The effect of Ang Ⅱ on HSCproliferation was determined by MTT method.Effect of Ang Ⅱ on collagen synthesis of HSCswas determined by 3H-proline incorporation.RESULTS Contrasted to the fibrosis in rats ofthe model group,groups of rats treated with either enalapril or Iosartan,or a combination oftwo drugs showed a limited expansion of theinterstitium(4.23±3.70 vs 11.22±4.79,P<0.05),but no difference was observedamong three treated groups(5.38±3.43,4.96±2.96,4.23±2.70,P>0.05).Expression of AT 1receptors was found in fibrotic interstitium offibrotic rats,whereas in normal control rats theywere limited to vasculature only to a very slightdegree.AT 1 receptors were also expressed onactivated HSCs in the culture.At concentrationsfrom 10-9to 10-5mol/L,Ang Ⅱ stimulated HSCproliferation in culture in a dose-dependentmanner.Increasing Ang Ⅱ concentrationsproduced corresponding increases in 3H-prolineincorporation.Differences among groups were significant.CONCLUSION Angiotensin-converting enzyme inhibitors and AT I blocker may slow the progression of hepatic fibrosis; activated HSCs express AT 1 receptors, and Ang Ⅱ can stimulate the proliferation and collagen synthesis of HSCs in a dose-dependent manner; and activation of RAS may be related to hepatic fibrogenesis induced by CCI4.Hong Shan Wei Han Ming Lu Ding Guo Li Yu Tao Zhan Zhi Rong Wang Xin Huang Ji Lin Cheng Qin Fang Xu Department of Gastroenterology,Xinhua Hospital,Shanghai Second Medical University,Shanghai 200092,China 2000World Journal of Gastroenterology2000,6,6:27
5Single-incision vs three-port laparoscopic cholecystectomy:Prospective randomized study显示文摘AIM:To compare the clinical outcome of single-inci-sion laparoscopic cholecystectomy(SILC)with threeport laparoscopic cholecystectomy(TPLC). METHODS:Between 2009 and 2011,one hundred and two patients with symptomatic benign gallbladder diseases were randomized to SILC(n=49)or TPLC (n=53).The primary end point was post operative pain score(at 6 h and 7 d).Secondary end points were blood loss,operation duration,overall complications,postoperative analgesic requirements,length of hospital stay,cosmetic result and total cost.Surgical techniques were standardized and all operations were performed by one experienced surgeon,who had performed more than 500 laparoscopic cholecystectomies. RESULTS:One patient in the SILC group required conversion to two-port LC.There were no open conversions or major complications in either treatment groups.There were no differences in terms of esti-mated blood loss(mean±SD,14±6.0 mL vs 15±4.0 mL),operation duration(mean±SD,41.8±17.0 min vs 38.5±22.0 min),port-site complications(contusion at incision:5 cases vs 4 cases and hematoma at inci- sion:2 cases vs 1 case),total cost(mean±SD,12 075 ±1047 RMB vs 11 982±1153 RMB)and hospital stay (mean±SD,1.0±0.5 d vs 1.0±0.2 d),respectively. TPLC had a significantly worse visual analogue pain score at 8 h after surgery(mean±SD,3.5±1.6 vs 2.0 ±1.5),however,the scores were similar on day 7(mean ±SD,2.5±1.4 vs 2.0±1.3).Cosmetic satisfaction, as determined by a survey at 2 mo follow-up favored SILC(mean±SD,8±0.4 vs 6±0.2). 7Clinical and experimental study on regional administration of phosphorus32 glassmicrospheres in treating hepatic carcinoma显示文摘AIM To study the therapeutical effectiveness, dosage range and toxic adverse effects of domestic phosphorus 32 glass microsphere and evaluate its clinical significance. METHODS Ⅰ. Fifty two BALB/*!c tumor bearing male nude mice were allocated into treatment group( n =38) and control group( n =14). In the former group different doses of 32 P GMS were injected into the tumor mass, while in the latter 31 P GMS or no treatment was given. The experimental animals were sacrificed in batches, and then the tumors and their nearby tissues were examined by light and electron microscopy. Ⅱ. Through selective catheterization of hepatic artery, 32 P GMS was infused to 5 healthy domestic pigs in a dosage equivalent to the therapeutic dose for human being, and 31 P GMS was infused to another 5 healthy domestic pigs. Two pigs infused with contrast medium served as whole course blank controls. One pig from each group was surrendered to euthanasia at week 1, 4, 8 and 16 respectively. The ultrastructural histopath ological changes in liver tissues taken from different sites were evaluated semiquan titatively. Ⅲ. One hundred and twenty seven times of 32 P GMS intrahepatic artery interventional therapies were performed on 93 patients with hepatic carcinoma, including 79 cases of primary hepatic carcinoma and 14 cases of secondary hepatic carcinoma. 32 P GMS ( n =30), and group B, 32 P GMS and half dose of trans hepatic artery embolization (TAE) ( n =49) , and 18 patients with HCC by TAE only as control group C. Fourteen patients with secondary hepatic carcinoma were treated in the same way as group B or C. RESULTS Ⅰ. Comparing with the control group, the treatment group of tumor bearing nude mice attained the tumor inhibition rates of 59 7%-93 7% ( F =579 62, P <0 01) at 14*!d . At an absorbed dose of 7320Gy, the tumor cells were completely destroyed. When the absorbed doses ranged from 1830Gy to 3660Gy, most of the tumor cells showed the evidences of injury or necrosis, but there appeared some well differentiated tumor cells and enhanced effect of the autoimmunocytes. At an absorbed dose of 366Gy or less, some tumor cells still remained active proliferative ability. The definite anticancer effect appeared as early as 3d after intratumoral injection of 32 P GMS. Ⅱ. The cumulative amount of 32 P GMS in the target tissue after trans hepatic artery instillation attained more than 90% of the total dose administrated. Semiquantitative analysis of ultrastructral morphology in the experimental group showed no statistical difference between the nuclear abnormality (N abn ) and mitochondrial variability (M var ) at week 1 or 2, but revealed prominent difference (χ 2=6 70-9 68, P <0 01 , χ 2=65 09-115 09, P <0 001 ) as compared with those in the other groups. In the experimental group the N abn in tissues showed no significant difference between week 8 and week 16. No apparent changes were found in the stomach, spleen, kidney and lung tissues of the experimental pigs. Ⅲ. The therapeutical results of HCC patients in group A were closely approximated to those of group C, no hematological toxic side effects were noted, and the systemic reaction was mild. In some patients 2*!mos - 3*!mos after treatment some secondary foci appeared around the periphery of the primary lesion. In general better effectiveness was obtained in patients with small lesion. After analyzing by RIDIT method, the therapeutic result in group B was significantly better than that in group C, and secondary foci around the original lesion were rarely seen at 3*!mos after treatment. In group C the collateral circulation was reestablished along the periphery of primary foci and the secondary foci appeared more frequently, and required to undergo several courses of treatment. 8Cancer stem cells in glioblastoma—molecular signaling and therapeutic targeting显示文摘Glioblastomas(GBMs)are highly lethal primary brain tumors.Despite current therapeutic advances in other solid cancers,the treatment of these malignant gliomas remains essentially palliative.GBMs are extremely resistant to conventional radiation and chemotherapies.We and others have demonstrated that a highly tumorigenic subpopulation of cancer cells called GBM stem cells(GSCs)promotes therapeutic resistance.We also found that GSCs stimulate tumor angiogenesis by expressing elevated levels of VEGF and contribute to tumor growth,which has been translated into a useful therapeutic strategy in the treatment of recurrent or progressive GBMs.Furthermore,stem cell-like cancer cells(cancer stem cells)have been shown to promote metastasis.Although GBMs rarely metastasize beyond the central nervous system,these highly infiltrative cancers often invade into normal brain tissues preventing surgical resection,and GSCs display an aggressive invasive phenotype.These studies suggest that targeting GSCs may effectively reduce tumor recurrence and significantly improve GBM treatment.Recent studies indicate that cancer stem cells share core signaling pathways with normal somatic or embryonic stem cells,but also display critical distinctions that provide important clues into useful therapeutic targets.In this review,we summarize the current understanding and advances in glioma stem cell research,and discuss potential targeting strategies for future development of anti-GSC therapies.Zhi Huang Lin Cheng Olga A.Guryanova Qiulian Wu Shideng Bao 2010Protein & Cell2010,1,7:18
9Vascular anatomy of inferior mesenteric artery in laparoscopic radical resection with the preservation of left colic artery for rectal cancer显示文摘AIM To investigate the vascular anatomy of inferior mesenteric artery(IMA) in laparoscopic radical resection with the preservation of left colic artery(LCA) for rectal cancer. METHODS A total of 110 patients with rectal cancer who underwent laparoscopic surgical resection with preservation of the LCA were retrospectively reviewed. A 3 D vascular reconstruction was performed before each surgical procedure to assess the branches of the IMA. During surgery, the relationship among the IMA, LCA, sigmoid artery(SA) andsuperior rectal artery(SRA) was evaluated, and the length from the origin of the IMA to the point of branching into the LCA or common trunk of LCA and SA was measured. The relationship between inferior mesenteric vein(IMV) and LCA was also evaluated.RESULTS Three vascular types were identified in this study. In type A, LCA arose independently from IMA(46.4%, n = 51); in type B, LCA and SA branched from a common trunk of the IMA(23.6%, n = 26); and in type C, LCA, SA, and SRA branched at the same location(30.0%, n = 33). The difference in the length from the origin of IMA to LCA was not statistically significant among the three types. LCA was located under the IMV in 61 cases and above the IMV in 49 cases. CONCLUSION The vascular anatomy of the IMA and IMV is essential for laparoscopic radical resection with preservation of the LCA for rectal cancer. 11Effectiveness of Different Waist Circumference Cut-off Values in Predicting Metabolic Syndrome Prevalence and Risk Factors in Adults in China显示文摘Objective To study the effectiveness of waist circumference cut-off values in predicting the prevalence of metabolic syndrome(MetS) and risk factors in adults in China. Methods A cross-sectional survey was condcuted in 14 provinces(autonomous region, municipality) in China. A total of 47 325 adults aged ≥20 years were selected by multistage stratified sampling, and questionnaire survey and physical and clinical examination were conducted among them. MetS was defined according to the International Diabetes Federation(IDF) criteria and modified IDF criteria.Results The age-standardized prevalence of MetS was 24.2%(22.1% in men and 25.8% in women) and 19.5%(22.1% in men and 18.0% in women) according to the IDF criteria and modified IDF criteria respectively. The age-standardized prevalence of pre-MetS was 8.1%(8.6% in men and 7.8% in women) according to the modified IDF criteria. The prevalence of MetS was higher in urban residents than rural residents and in northern China residents than in southern China residents. The prevalence of central obesity was about 30% in both men and women according to the ethnicity-specific cut-off values of waist circumference for central obesity(90 cm for men and 85 cm for women). Multivariate regression analysis revealed no significant difference in risk factors between the two MetS definitions. 12Movement and strain conditions of active blocks in the Chinese mainland显示文摘The definition of active block is given from the angles of crustal deformation and strain. The movement and strain parameters of active blocks are estimated according to the unified velocity field composed of the velocities at 1598 GPS stations obtained from GPS measurements carried out in the past years in the Chinese mainland and the surrounding areas. The movement and strain conditions of the blocks are analyzed. The active blocks in the Chinese mainland have a consistent E-trending movement component, but its N and S components are not consistent. The blocks in the western part have a consistent N-trending movement and the blocks in the eastern part have a consistent S-trending movement. In the area to the east of 90°E, that is the area from Himalayas block towards NE, the movement direction of the blocks rotates clockwisely and the movement rates of the blocks are different. Generally, the movement rate is large in the west and south and small in the east and north with a difference of 3 to 4 times between the rates in the west and east. The distributions of principal compressive strain directions of the blocks are also different. The principal strain of the blocks located to the west of 90oE is basically in the SN direction, the principal compressive strain of the blocks in the northeastern part of Qingzang plateau is roughly in the NE direction and the direction of principal compressive strain of the blocks in the southeastern part of Qingzang plateau rounds clockwisely the east end of Himalayas structure. In addition, the principal strain and shear strain rates of the blocks are also different. The Himalayas and Tianshan blocks have the largest principal compressive strain and the maximum shear strain rate. Then, Lhasa, Qiangtang, Southwest Yunnan (SW Yunnan), Qilian and Sichuan-Yunan (Chuan-Dian) blocks followed. The strain rate of the blocks in the eastern part is smaller. The estimation based on the stain condition indicates that Himalayas block is still the area with the most intensive tectonic activity and it shortens in the NS direction at the rate of 15.2±1.5 mm/a. Tianshan block ranks the second and it shortens in the NS direction at the rate of 10.1±0.9 mm/a. At present, the two blocks are still uprising. It can be seen from superficial strain that the Chinese mainland is predominated by superficial expansion. Almost the total area in the eastern part of the Chinese mainland is expanded, while in the western part, the superficial compression and expansion are alternatively distributed from the south to the north. In the Chinese mainland, most EW-trending or proximate EW-trending faults have the left-lateral or left-lateral strike-slip relative movements along both sides, and most NS-trending faults have the right-lateral or right-lateral strike-slip relative movements along both sides. According to the data from GPS measurements the left-lateral strike-slip rate is 4.8±1.3 mm/a in the central part of Altun fault and 9.8±2.2 mm/a on Xianshuihe fault. The movement of the fault along the block boundary has provided the condition for block movement, so the movements of the block and its boundary are consistent, but the movement levels of the blocks are different. The statistic results indicate that the relative movement between most blocks is quite significant, which proves that active blocks exist. Himalayas, Tianshan, Qiangtang and SW Yunnan blocks have the most intensive movement; China-Mongolia, China-Korea (China-Korea), Alxa and South China blocks are rather stable. The mutual action of India, Pacific and Philippine Sea plates versus Eurasia plate is the principal driving force to the block movement in the Chinese mainland. Under the NNE-trending intensive press from India plate, the crustal matter of Qingzang plateau moves to the NNE and NE directions, then is hindered by the blocks located in the northern, northeastern and eastern parts. 13Methylation of Dickkopf-3 as a prognostic factor in cirrhosis-related hepatocellular carcinoma显示文摘AIM:To investigate the prevalence and time of Dickkopf(DKK) family methylation and its clinical significance in hepatocarcinogenesis. METHODS:Methylation of DKK family genes was quantitatively analyzed in 115 liver tissue samples,including 50 pairs of primary hepatocellular carcinoma(HCC) and matched noncancerous cirrhotic tissue samples,as well as 15 liver cirrhosis biopsy samples. RESULTS:The methylation level of DKK3 was significantly higher in HCC tissue samples than in matched noncancerous cirrhotic tissue samples(P<0.0001) orin liver cirrhosis biopsy samples(P=0.0139) .Receiver operator characteristic curve analysis confirmed that the percent of methylated reference(PMR) values of DKK3 could effectively discriminate HCC tissue samples from noncancerous tissue samples(AUC=0.8146) or liver cirrhosis biopsy samples(AUC=0.7093) .Kaplan-Meier survival curves revealed that the progression-free survival time of patients with a higher DKK3 methylation level(PMR >1%) was significantly shorter than that of those with a lower DKK3 methylation level(PMR≤1%)(P=0.0255) . Multivariate Cox analysis indicated that methylated DKK3 was significantly and independently related with a shorter survival time(relative risk=2.527,95%CI:1.063-6.008,P=0.036) of HCC patients. 14The Prospective Two-Dimensional Graphene Nanosheets: Preparation, Functionalization, and Applications显示文摘Graphene, as an intermediate phase between fullerene and carbon nanotube, has aroused much interests among the scientific community due to its outstanding electronic, mechanical, and thermal properties.With excellent electrical conductivity of 6000 S/cm, which is independent on chirality, graphene is a promising material for high-performance nanoelectronics, transparent conductor, as well as polymer composites. On account of its Young's Modulus of 1 TPa and ultimate strength of 130 GPa, isolated graphene sheet is considered to be among the strongest materials ever measured. Comparable with the single-walled carbon nanotube bundle,graphene has a thermal conductivity of 5000 W/(m·K), which suggests a potential application of graphene in polymer matrix for improving thermal properties of the graphene/polymer composite. Furthermore, graphene exhibits a very high surface area, up to a value of 2630 m^2/g. All of these outstanding properties suggest a wide application for this nanometer-thick, two-dimensional carbon material. This review article presents an overview of the significant advancement in graphene research: preparation, functionalization as well as the properties of graphene will be discussed. 16Satellite SAR interferometric observations of displacements associated with urban subsidence in Suzhou, Eastern China显示文摘SAR interferometry (InSAR) has a high potential for surface displacement mapping in the range from millimeter to meter. In this paper the potential of ERS-1/2 SAR interferometry for mapping subtle land subsidence has been investigated. A time series of ERS-1/2 SAR data from February 1993 to February 2000 is collected from measurements taken in Suzhou city, Jiangsu Province, China, eight ERS-1/2 SAR images are used to create seven interferograms, and three differential interferograms are produced using the three-pass method, which clearly show the spatial extent of land subsidence. The deformation maps are validated by leveling surveys, the correlation coefficient and standard deviation between them are 0.943 and 0.1706 respectively. Based on seven benchmarks, the subsidence rates are estimated, the overall trends are in close agreement with InSAR results. 17THE EFFECTS OF RADIATION STRESS ON WAVE HEIGHTS AND SEA LEVEL IN THE INTERACTION OF COUPLED WAVE-TIDE-SURGE IN THE COASTAL AREA显示文摘Based on coastal high resolution (2′×2′) coupled wave tide surge interaction numerical model, the effects of radiation stress on wave heights and sea level in the coastal area of Huanghe Delta were studied. By comparisons of simulated and measured wave heights and sea level for two mediately strong weather cases, it is demonstrated that the results simulated by coupled wave tide surge model are closer to the measured and particularly in excellent agreement in the extreme values of the wave heights and set up. This study shows that the radiation stress can increase the wave heights maximally to 67cm and sea level to 40cm. It is also found that there are areas of over 50cm wave height increase and an area of over 20cm sea level increase in the Huanghe Delta coastal area, and this find may be very important in engineering. 18Prognostic value of tertiary lymphoid structure and tumour infiltrating lymphocytes in oral squamous cell carcinoma显示文摘Tertiary lymphoid structures(TLS)are ectopic lymphoid structures in cancers that are largely associated with favourable prognosis.However,the prognostic value of TLSs in oral squamous cell carcinoma(OSCC)is largely unknown,and the association between tumour infiltrating lymphocytes(TILs)and TLSs has been rarely explored in OSCC.In this study,associated markers of TLS,including peripheral node address(PNAd)in high endothelial venules,CD20 in B cells and CD3 in T cells,were examined in 168 OSCC patients,and survival analysis was performed between TLS-positive and TLS-negative cohorts.We detected the presence of TILs by staining CD8+cytotoxic T cells and CD57+NK cells as well.TLSs appeared as highly organized structures in 45(26.8%)cases.TLSpositive patients had a better 5-year overall survival(OS)rate(88.9%vs.56.1%,P<0.001)and relapse-free survival(RFS)rate(88.9%vs.63.4%,P=0.002).Moreover,the presence of TLS was an independent prognostic factor for both the 5-year OS rate(hazard ratio[HR]=3.784;95%confidence interval[CI],1.498–9.562)and RFS rate(HR=3.296;95%CI,1.279–8.490)in multivariate analysis.Furthermore,a higher density of CD8+T cells and CD57+NK cells was found in TLS-positive sections than in TLS-negative counterparts(P<0.001),and their combination provided a higher predictive accuracy(AUC=0.730;95%CI,0.654–0.805).In conclusion,our results suggest that TLS is an independent positive prognostic factor for OSCC patients.These findings provide a theoretical basis for the future diagnostic and therapeutic value of TLSs in OSCC treatment.Qunxing Li Xiangqi Liu Dikan Wang Yanqiong Wang Huanzi Lu Shuqiong Wen Juan Fang Bin Cheng Zhi Wang 2020International Journal of Oral Science2020,12,3:12
19Hybrid bioartificial liver support in cynomolgus monkeys with D-galactosamine-induced acute liver failure显示文摘AIM:To evaluate a hybrid bioartificial liver support system(HBALSS)in cynomolgus monkeys with acute liver failure.METHODS:To establish a model of acute liver failure,0.3 g/kg of D-galactosamine was injected intravenously into cynomolgus monkeys.Chinese human liver cells were introduced into a perfusion bioreactor to carry out hybrid bioartificial liver support treatment.Forty-eight hours after the injection,one group of cynomolgus monkeys received HBALSS care,and a second experimental group received no treatment.Clinical manifestations of all animals,survival time,liver and kidney functions and serum biochemistry changes were recorded.Simultaneous detection of the number,viability and function of hepatocytes in the hybrid bioartificial liver were also performed.RESULTS:Forty-eight hours after the injection of D-galactosamine,serum biochemistry levels were significantly increased,whereas albumin levels and the Fischer index were significantly reduced compared to baseline(all P s<0.05).Of the ten monkeys in the HBALSS treatment group,five survived,with an average duration of survival of 128±3 h.All cynomolgus monkeys in the control group died,with a duration of survival of112±2 h.Survival time was significantly longer with HBALSS treatment(P<0.05).Moreover,the number,viability and function of hepatocytes were maintained at a high level with HBALSS.CONCLUSION:The novel hybrid bioartificial liver plays a significant role in liver support by significantly reducing serum biochemistry levels and extending animal survival time.Zhi Zhang Yi-Chao Zhao Yuan Cheng Guo-Deng Jian Ming-Xin Pan Yi Gao 2014World Journal of Gastroenterology2014,20,46:12
20Adoptive therapy with CAR redirected T cells for hematological malignancies显示文摘The survival of patients with hematological malignancies has been significantly improved due to the development of new therapeutic agents. However, relapse remains a major matter for concern. Recently, T cells engineered with chimeric antigen receptor(CAR) were reported to show unprecedented responses in a range of hematological malignancies. The persistence of the CAR-T cell can last for years and tends toward long-term antitumor memory by which relapses can be effectively prevented. The primary side effects that appear in most clinical trials are cytokine release syndrome and neurotoxicity. However, these symptoms can be treated and reversed.
