| 1 | 2018 Chinese Pediatric Cardiology Society(CPCS) guideline for diagnosis and treatment of syncope in children and adolescents显示文摘Syncope belongs to the transient loss of consciousness(TLOC), characterized by a rapid onset, short duration, and spontaneous complete recovery. It is common in children and adolescents, accounting for 1% to 2% of emergency department visits.Recurrent syncope can seriously affect children's physical and mental health, learning ability and quality of life and sometimes cardiac syncope even poses a risk of sudden death. The present guideline for the diagnosis and treatment of syncope in children and adolescents was developed for guiding a better clinical management of pediatric syncope. Based on the globally recent development and the evidence-based data in China, 2018 Chinese Pediatric Cardiology Society(CPCS) guideline for diagnosis and treatment of syncope in children and adolescents was jointly prepared by the Pediatric Cardiology Society, Chinese Pediatric Society, Chinese Medical Association(CMA)/Committee on Pediatric Syncope, Pediatricians Branch, Chinese Medical Doctor Association(CMDA)/Committee on Pediatric Cardiology, Chinese College of Cardiovascular Physicians, Chinese Medical Doctor Association(CMDA)/Pediatric Cardiology Society, Beijing Pediatric Society, Beijing Medical Association(BMA). | 3 | Human pescadillo induces large-scale chromatin unfolding显示文摘The human pescadillo gene encodes a protein with a BRCT domain. Pescadillo plays an important role in DNA synthesis, cell proliferation and transformation. Since BRCT do- mains have been shown to induce chromatin large-scale unfolding, we tested the role of Pesca- dillo in regulation of large-scale chromatin unfolding. To this end, we isolated the coding region of Pescadillo from human mammary MCF10A cells. Compared with the reported sequence, the isolated Pescadillo contains in-frame deletion from amino acid 580 to 582. Targeting the Pesca- dillo to an amplified, lac operator-containing chromosome region in the mammalian genome re- sults in large-scale chromatin decondensation. This unfolding activity maps to the BRCT domain of Pescadillo. | 7 | Prognostic Nomogram for Patients with Hepatitis E Virus-related Acute Liver Failure:A Multicenter Study in China显示文摘Background and Aims:Timely and effective assessment scoring systems for predicting the mortality of patients with hepatitis E virus-related acute liver failure(HEV-ALF)are urgently needed.The present study aimed to establish an effective nomogram for predicting the mortality of HEV-ALF patients.Methods:The nomogram was based on a cross-sectional set of 404 HEV-ALF patients who were identified and enrolled from a cohort of 650 patients with liver failure.To compare the performance with that of the model for end-stage liver disease(MELD)scoring and CLIF-Consortiumacute-on-chronic liver failure score(CLIF-C-ACLFs)models,we assessed the predictive accuracy of the nomogram using the concordance index(C-index),and its discriminative ability using time-dependent receiver operating characteristics(td-ROC)analysis,respectively.Results:Multivariate logistic regression analysis of the development set carried out to predict mortality revealed that γ-glutamyl transpeptidase,albumin,total bilirubin,urea nitrogen,creatinine,international normalized ratio,and neutrophil-to-lymphocyte ratio were independent factors,all of which were incorporated into the new nomogram to predict the mortality of HEV-ALF patients.The area under the curve of this nomogram for mortality prediction was 0.671(95%confidence interval:0.602-0.740),which was higher than that of the MELD and CLIF-C-ACLFs models.Moreover,the td-ROC and decision curves analysis showed that both discriminative ability and threshold probabilities of the nomogram were superior to those of the MELD and CLIF-C-ACLFs models.A similar trend was observed in the validation set.Conclusions:The novel nomogram is an accurate and efficient mortality prediction method for HEV-ALF patients. | Jian Wu Cuifen Shi Xinyu Sheng Yanping Xu Jinrong Zhang Xinguo Zhao Jiong Yu Xinhui Shi Gongqi Li Hongcui Cao Lanjuan Li | 2021 | Journal of Clinical and Translational Hepatology2021,9,6: | 2 |

| 15 | Synthesis and Applications of Tetra- functional Branched Poly(ethylene glycol) Derivative for the Decellularized Valve Leaflets Cross-linking显示文摘To investigate the effects of polyethylene glycol cross-linking on the mechanical properties, 80 porcine aortic valves were harvested, decellularized, and introduced with sulfhydryl. Then the valves were randomly assigned into 5 experimental groups and 1 control group(n=16). For the valves in those experimental groups, branched polyethylene glycol diacrylate(PEG) of 5 different molecular weights(3.4, 8, 12, 20, 40 k Da) were synthesized and cross-linked with them respectively. The efficiency of the cross-linking was determined by measuring the amount of residual thiol group and the mechanical properties of the cross-linked valve leaflets were assessed by uni-axial planar tensile testing. The efficiency of the PEG 20 k Da group was 70.72±2.33%, obviously superior to that of the other groups(p<0.05). Tensile test proved that branched PEG cross-linking can significantly enhance the mechanical behaviors of the decellularized valve leaflet and the Young's modulus of each group was positively correlated with the molecular weight of PEG. | 18 | Fast-response fluorescent probe with favorable water solubility for highly sensitive imaging of endogenous tyrosinase in living cells and zebrafish model显示文摘Tyrosinase(TYR) is an important polyphenolic oxidase enzyme and usually regards as a biomarker of melano ma cancer.Highly effective tracking TYR activity in vivo will help to study the mechanism of TYR in living organisms and forecasts related diseases.In this study,we present a novel TYR-activatable fluo rescent probe(CHMC-DOPA) for tracking TYR activity in vitro and in vivo.CHMC-DOPA is constructed by incorporating dopamine(DOPA) moiety into a fluorescent chloro-hydroxyl-merocyanine(CHMC)scaffold.Upon exposure to TYR,the dopamine unit in CHMC-DOPA is oxidized to a dopaquinone derivative,and an intramolecular photo-induced electron transfer(PET) process between CHMC fluorophore and o-dopaquinone will take place,the fluorescence of CHMC-DOPA is quenched rapidly.Therefore,the evaluation of TYR activity is established in terms of the relationship between fluorescence quenching efficiency and TYR activity.In our experiments,CHMC-DOPA shows various advantages,such as fast response(8 min),low concentration of TYR activation(0.5 U/mL),good water-solubility,as well as the lowest detection limit(0.003 U/mL) compared with previously reported works.Furthermore,CHMCDO PA also exhibits excellent cell membrane permeability and low cytotoxicity,which is successfully used to monitor endogenous TYR activity in living cancer cells and zebrafish models.CHMC-DOPA performs well,and we anticipate that this newly designed novel platform will provide an alternative for high effective monitoring TYR activity in biosystems. | Zheng Li Xiaofeng Xia Yu You Cuifen Lu Guichun Yang Chao Ma Junqi Nie Qi Sun Shuilin Wu Jun Ren Feiyi Wang | 2021 | Chinese Chemical Letters2021,32,5: | 1 |

Thirteen intracellular induced genes were identified with a HeLa cell infection model. Of these, two were identified as alkylation-related genes; one was related to metabolism; one encoded a transcriptional regulator; three were identified as insertion elements; three ap- peared to be antisense to genes involved in the transmethylation,biosyntheseis, and phos- photransferase system;and three were predicted to encode polypeptides with unknown functions. Intracellular survival assavs showed that the mutants of alkA,citC and wcaJ genes had lower capability of intracellular replication or survival than the the wild-type strain.The results indicated that alkA, citC and wcaJ genes could take part in the intracellular survival or replication of S. flexneri 2a and the capability of intracellular survival or replication could be one of the major virulence elements. However, the yaiC mutant was able to survive in the murine infection assay but almost not in HeLa cell infection assay. Very possibly, yaiC gene was involved in the other mechanism of S. flexneri virulence.
